Abstract
本发明公开了属于生物工程技术领域的一种Cry3Bb兔多克隆抗体的制备方法,所述多克隆抗体的制备方法,包括以下步骤:a)通过原核表达得到Cry3Bb重组蛋白片段;b)将Cry3Bb重组蛋白片段作为抗原免疫日本大耳白兔;c)效价检测;d)多克隆抗体纯化及western验证。所述多克隆抗体为实现抗虫蛋白Cry3Bb在转基因作物中的定性、定量检测奠定基础。
Description
技术领域
本发明涉及生物工程领域,具体涉及到一种Cry3Bb兔多克隆抗体的制备方法。
背景技术
苏云金杆菌(Bacillus thuringiensis,Bt)是一种广泛存在于土壤中的革兰氏阳性菌分泌的具有高抗虫活性的晶体蛋白。抗虫蛋白Cry3Bb对鳞翅类(Lepidoptera)昆虫有广泛毒性,当昆虫取食Cry3Bb蛋白后,该蛋白经中肠蛋白酶活化,活化的蛋白穿透围食膜,与中肠细胞的上皮细胞顶膜特异蛋白结合并形成肠穿孔,从而破坏细胞的渗透平衡,引起细胞肿胀裂解,最终导致死亡。因此Cry3Bb基因已成为植物基因工程及转基因育种应用中最具有潜力和应用前景的抗虫基因。其中Cry3Bb基因已成功的被用于转基因作物。
转基因作物的飞速发展在带来巨大经济利益的同时,人们也越来越关注转基因作物可能带来的不可预期的食用安全及环境安全性问题,越来越多的国家要求对转基因产品进行检测以实行标识制度。
公开于该背景技术部分的信息仅仅旨在增加对本发明的总体背景的理解,而不应当被视为承认或以任何形式暗示该信息构成已为本领域一般技术人员所公知的现有技术。
发明内容
本发明的目的在于提供一种Cry3Bb兔多克隆抗体的制备方法,得到其的多抗为实现抗虫蛋白Cry3Bb在转基因作物中的定性、定量检测奠定基础。
为实现上述目的,本发明提供了一种Cry3Bb多克隆抗体的制备方法,包括以下步骤:
a)抗原制备:通过原核表达得到Cry3Bb重组蛋白片段;
b)免疫动物:将Cry3Bb重组蛋白片段作为抗原免疫日本大耳白兔;
c)效价检测:采用间接ELISA法,对抗体进行梯度稀释并检测抗体的效价和灵敏度;
d)多克隆抗体纯化:采用Protein G亲和层析,得到多克隆抗体。
上述多克隆抗体通过间接ELISA检测所得到的效价为0.6ng/mL。
本发明还提供了上述多克隆抗体在检测Cry3Bb抗虫蛋白中的应用。
本发明还提供了上述多克隆抗体在制备ELISA法检测Cry3Bb抗虫蛋白的试剂中的应用。
本发明的有益效果:本发明利用工程菌株表达、纯化得到的高纯度抗虫蛋白Cry3Bb作为抗原,通过免疫日本大耳白兔获得抗血清纯化后得到的抗体间接ELISA效价为0.6ng/mL,所述多克隆抗体可以特异识别原核表达的重组Cry3Bb蛋白及转基因玉米中的内源Cry3Bb蛋白,Cry3Bb抗虫蛋白的兔多克隆抗体的构建为转基因作物中该蛋白的检测提供了物质和技术支撑。

下述实施例中所使用的实验方法如无特殊说明,均为常规方法。
下述实施例中所用的材料、试剂等,如无特殊说明,均可从商业途径可购得。
实施例1
1.免疫抗原的制备
(1)重组蛋白Cry3Bb片段表达菌株构建、菌体培养及裂解
Cry3Bb重组蛋白片段氨基酸序列:

用高保真Fastpfu进行PCR扩增,扩增到的PCR产物经(1.0%)琼脂糖凝胶电泳鉴定后,将目的条带切胶回收,用Universal DNA纯化回收试剂盒(天根)回收,回收产物经限制性内切酶Bam HI和Xho I酶切、经胶回收与进行同样酶切、胶回收处理的pET28a质粒片段于4℃过夜连接,转化Trans10感受态细胞(北京全式金),挑取克隆,进行菌落PCR鉴定,阳性克隆由北京六合华大基因公司进行测序。
将鉴定正确的重组表达载体pET28a-Cry3Bb转化大肠杆菌BL21(DE3)感受态细胞,菌液涂布于含卡那霉素(50μg/mL)的LB平板上,37℃过夜培养。次日在平板上挑取单克隆接种含相同浓度卡那霉素的液体LB培养基中,于37℃过夜培养。所得的表达菌株菌液与50%的甘油溶液等体积混匀,于-80℃冻藏。
将保存的重组蛋白Cry3Bb表达菌株复苏培养,菌液涂布于含卡那霉素(50μg/mL)的LB平板上,37℃过夜培养。挑取单克隆接种液体LB(含卡那霉素50μg/mL)培养基中37℃过夜培养。第二天以1%接种量接种,于37℃培养至OD600为0.8左右,加入1mM IPTG于16℃过夜诱导表达蛋白。诱导结束将菌液冰浴,于4℃,4000rpm,10min离心收集菌体,弃上清,菌体经PBS洗涤后用裂解液悬浮;超声破碎(2s on/4s off,Amp:45%,Time:3min);4℃15000rpm离心1hr,收集上清。
(2)重组蛋白纯化
菌体裂解液上清与经裂解液平衡的Ni Sepharose 6FF Beads(GE Healthcare)混合,于4℃结合2-3hrs,3000rpm离心3min弃上清;已结合蛋白的Ni Sepharose 6FF beads经清洗液洗2-3次;再用洗脱液洗脱。收集洗脱液即为蛋白粗纯溶液。
将亲和纯化得到的蛋白样品透析至蛋白储存溶液,并用经相同溶液平衡的Superdex 200Increase 10/30GL(GE Healthcare)进一步纯化,收集蛋白峰组分并经SDS-PAGE检测蛋白样品的纯度,通过图1可得,经过凝胶过滤最后获得电泳纯的Cry3Bb蛋白,分子量约为43KDa。
2.免疫动物
用上述质控合格的Cry3Bb重组蛋白片段作为抗原免疫2只3-5月龄的SPF级日本大耳白兔(购置于湖北省实验动物研究中心,许可证号:SCXK(鄂)2015-0018),抗原与等体积完全弗氏佐剂(首免)和不完全弗氏佐剂(加强免疫)混合并进行乳化,充分混合至油包水状态进行皮下多点免疫,2-3次加强免疫,每次免疫间隔周期2周,之后进行效价检测,高于>1:10000后1周内进行腹腔冲击,直接将免疫剂量的抗原溶于250μL的PBS中,具体免疫次数及免疫剂量如表1所示:

3多克隆抗体纯化
收集后的抗血清经过预处理后选用Protein G-琼脂糖亲和层析柱纯化,具体步骤如下:
1)用10倍柱体积的PBS(或者TBS,下同)洗涤平衡层析柱。
2)含抗体的血清或其它体液高速离心后,将上清与等体积2×PBS缓冲液混合,调整pH以及离子浓度后,缓慢加入层析柱。
3)用10倍柱体积以上的PBS洗涤,至流出液无蛋白检出。
4)加入2倍柱体积0.1M柠檬酸(Citrate Acid,pH 2.7),夹住流出管,静置5分钟后收集穿出液,重复三次。测定OD280估算抗体浓度,如果所得抗体较多可以使用SDS-PAGE检测纯度。也可以选择0.1M甘氨酸(Glycine,pH 3.0)洗脱。
5)洗脱后的抗体加入2/5体积的1M Tris,pH 8.0中和,使用Millipore蛋白浓缩管切换到所需缓冲液,通常为2×PBS含有0.02%NaN3以及1mM EDTA。
6)浓缩到所需体积,经过SDS-PAGE检测纯度(见图2),-20℃保存,避免冻结。
4.多克隆抗体的效价测定
将纯化好的多克隆抗体,以重组蛋白Cry3Bb为抗原,用间接ELISA方法检测其效价,通过图3可知,经ELISA测定,纯化得到的多克隆抗体抗滴度为0.6ng/mL。
5.多克隆抗体特异性检测
分别提取转基因玉米及其为转化亲本叶片的蛋白,跑SDS-PAGE胶,转膜用纯化多克隆抗体作为一抗,Alexa FlurorTM 680goat anti-rabbit IgG(H+L)(Invitrogen)为二抗,用Odyssey infrared740 imager(9120,Li-COR Biosciences,Lincolin,NE)红外扫描仪western检测结果,通过图4可知,纯化得到的多克隆抗体能特异识别内源样本中的Cry3Bb和重组蛋白Cry3Bb;
其中,转基因玉米叶片蛋白提取方法:
组织液氮速冻,磨碎,加1mL(按样品量,一般0.5g加1-2mL)蛋白提取液,4℃混匀30分钟,(12,000rpm 4℃离心15min,取上清),蛋白提取液配方见表2:

Claims (5)
1.一种Cry3Bb兔多克隆抗体的制备方法,其特征在于,包括以下步骤:
a)抗原制备:通过原核表达得到Cry3Bb重组蛋白片断;
b)免疫动物:将Cry3Bb重组蛋白片段作为抗原免疫白兔;
c)效价检测:采用间接ELISA法,对抗体进行梯度稀释并检测抗体的效价和灵敏度;
d)多克隆抗体纯化:采用Protein G亲和层析,得到多克隆抗体。
2.根据权利要求1所述的制备方法,其特征在于,所述的Cry3Bb重组蛋白片段免疫的白兔为日本大耳白兔。
3.根据权利要求2所述的多克隆抗体,其特征在于所产生的多克隆抗体通过间接ELISA检测所得到的效价为0.6ng/mL。
4.权利要求1所述的多克隆抗体在检测Cry3Bb抗虫蛋白中的应用。
5.权利要求1所述的多克隆抗体在制备ELISA法检测Cry3Bb抗虫蛋白的试剂中的应用。
