本发明提供一种基于聚六亚甲基胍的消毒用涂膜剂及其制备方法,所述基于聚六亚甲基胍的消毒用涂膜剂的配方如下:聚六亚甲基胍:0.25‑0.5;中药材提取物:0.1‑5.0;成膜材料:5.0‑15.0;增塑剂:2.0‑4.0;保湿剂/致孔剂:10‑15;乙醇:20‑75;余量为精制水;该基于聚六亚甲基胍的消毒用涂膜剂以多组分形成的复方消毒用涂膜剂,能够扩大杀菌及抗病毒作用,利用成膜材料的成膜特性,涂布在手臂上能够形成一层不溶于水的保护膜,具有隐形手套的作用,同时加入了保湿剂/致孔剂,不影响皮肤的生理功能及手指的正常工作,具有杀菌迅速、维持时间长的特点。

本发明属于涂膜剂技术领域,特别涉及一种基于聚六亚甲基胍的消毒用涂膜剂及其制备方法。
背景技术
药剂学中的一种剂型-涂膜剂,系指药物溶解或分散于成膜材料的溶剂中,涂布于患处后形成薄膜的外用制剂。用时涂布于患处,溶剂挥发后形成薄膜,对患处有保护作用,同时逐渐释放所含药物起到治疗作用。环保型高分子聚合物杀菌消毒剂-聚六亚甲基胍,具有杀菌广谱、速效、有效浓度低、作用速度快、性质稳定、易溶于水的优良性能;可在常温下使用,长期抑菌、无副作用、无腐蚀性、无色、无嗅、无毒;不燃、不爆、使用安全;价格适中;运输方便,可以说是最佳的杀菌剂,聚六亚甲基胍杀菌的原理是通过作用于细菌及真菌的细胞壁和细胞质的膜系统,与细胞膜磷脂中的脂肪酸结合,迅速击穿它们的细胞质膜,攻破病菌细胞的核心,使病菌体内的生命物质外流,从而达到杀菌效果。与其它杀菌剂联合使用,可以发挥极佳的消毒效果。由于聚六亚甲基胍具有一种非特定的杀菌模式,因此细菌对其产生抗药性的可能性非常小。
中药材苦参、蛇床子、大黄、地肤子和百部等已经被证实在人体内外皆具有杀菌和抑制病毒的作用。
乙醇是一种常用的医用消毒杀菌剂,其杀菌作用是使细菌等病原体的蛋白质的变性,其特点是作用迅速。
然而,现有技术中尚未提供能够快速杀菌并有持续抑菌作用的成膜制剂。在当前抗疫形势依然严峻的大环境下,亟需具有持续消毒杀菌效果的涂膜剂,作为其他个人卫生用品的辅助及替代用品,为此,本发明提出一种基于聚六亚甲基胍的消毒用涂膜剂及其制备方法。
发明内容
为了解决现有技术存在的问题,本发明提供了一种基于聚六亚甲基胍的消毒用涂膜剂及其制备方法,本发明的基于聚六亚甲基胍的消毒用涂膜剂以多组分形成的复方消毒用涂膜剂,能够扩大杀菌及抗病毒作用,利用成膜材料的成膜特性,涂布在手臂上能够形成一层不溶于水的保护膜,具有隐形手套的作用,同时加入了保湿剂/致孔剂,不影响皮肤的生理功能及手指的正常工作,具有杀菌迅速、维持时间长的特点。
为了实现上述目的,本发明是通过如下的技术方案来实现:一种基于聚六亚甲基胍的消毒用涂膜剂,配方如下:聚六亚甲基胍:0.25-0.5;中药材提取物:0.1-5.0;成膜材料:5.0-15.0;增塑剂:2.0-4.0;保湿剂/致孔剂:10-15;乙醇:20-75;余量为精制水。
作为本发明的一种优选实施方式,所述成膜材料为聚乙烯醇缩丁醛(PVB)、聚维酮(PVP)、聚乙烯醇(PVA)、乙烯-醋酸乙烯共聚物(EVA)和羟丙甲纤维素中的一种或几种的组合,优选为聚乙烯醇缩丁醛或乙烯-醋酸乙烯共聚物,最优选为聚乙烯醇缩丁醛。
作为本发明的一种优选实施方式,所述增塑剂为邻苯二甲酸脂、柠檬酸酯和癸二酸酯中的一种或几种的组合,优选为柠檬酸三乙酯、邻苯二甲酸二丁脂或乙酰柠檬酸三丁酯,最选为柠檬酸三乙酯,所述邻苯二甲酸脂包括但不限于邻苯二甲酸二丁脂和邻苯二甲酸苄丁酯,所述柠檬酸酯包括但不限于柠檬酸三乙酯和乙酰柠檬酸三丁酯,所述癸二酸酯包括但不限于癸二酸二丁酯。
作为本发明的一种优选实施方式,所述保湿剂/致孔剂为甘油、丙二醇或聚乙二醇-400中的一种或几种的组合,优选甘油。
作为本发明的一种优选实施方式,所述中药材提取物的原料为苦参、蛇床子、大黄、地肤子和百部中的一种或几种的组合,所述中药材提取物是将一种以上中药材混合经水提-醇沉法提取,回收乙醇后经浓缩得到的流浸膏。
一种基于聚六亚甲基胍的消毒用涂膜剂的制备方法,具体步骤如下:
步骤一:根据配方取用适量的各种原料备用;
步骤二:将成膜材料加入乙醇中,搅拌使其溶解后加入增塑剂,搅拌均匀;
步骤三:将聚六亚甲基胍和中药材提取物溶于精制水中并加入保湿剂/致孔剂,搅拌均匀;
步骤四:将步骤三所得液体加到步骤二所得液体中搅拌均匀,得到基于聚六亚甲基胍的消毒用涂膜剂。
本发明的有益效果为:
1、本发明的一种基于聚六亚甲基胍的消毒用涂膜剂以多组分形成的复方消毒用涂膜剂,能够扩大杀菌及抗病毒作用,利用成膜材料的成膜特性,涂布在手臂上能够形成一层不溶于水的保护膜,具有隐形手套的作用,同时加入了保湿剂/致孔剂,不影响皮肤的生理功能及手指的正常工作,具有杀菌迅速、维持时间长的特点。
2、本发明的一种基于聚六亚甲基胍的消毒用涂膜剂均匀涂抹在手和身体皮肤上能够在1-2分钟内形成保护膜,达到快速杀菌的效果并有持续抑菌作用,不影响皮肤正常生理功能,对皮肤没有刺激性,使用方便。
3、本发明的一种基于聚六亚甲基胍的消毒用涂膜剂将聚六亚甲基胍、中药提取物、聚乙烯醇缩丁醛、乙醇联合应用,组成复方消毒用涂膜剂,具有在手臂上可快速形成保护膜,快速杀菌并维持长效的特点,甘油、丙二醇和聚乙二醇具有优良的保湿、润滑和致孔性质,不影响皮肤正常生理功能,对皮肤没有刺激性。

具体实施方式
为使本发明实现的技术手段、创作特征、达成目的与功效易于明白了解,下面结合具体实施方式,进一步阐述本发明。
请参阅图1,本发明提供一种技术方案:
实施例1
将10克聚乙烯醇缩丁醛(PVB)加入到80克乙醇中,搅拌溶解后加入3.0克柠檬酸三乙酯搅拌溶解,制成溶液1;取0.5克聚六亚甲基胍置于5克精制水中,加入苦参、蛇床子、大黄、地肤子和百部的混合提取物5克,甘油10克,搅拌使溶解,制成溶液2;将溶液2加到溶液1中搅拌均匀,补足余量精制水至100克,即得聚六亚甲基胍与中药提取物组合的消毒用涂膜剂。
实施例2
将3克乙烯-醋酸乙烯共聚物(EVA)加入到80克乙醇中,搅拌溶解,再加入4克乙酰柠檬酸三丁酯,制成溶液1;取0.5克聚六亚甲基胍置于5克精制水中,加入苦参、蛇床子、大黄、地肤子和百部的混合提取物5克,丙二醇10克、搅拌使溶解,制成溶液2;将溶液2加到溶液1中搅拌均匀,补足余量精制水至100克,即得聚六亚甲基胍与中药提取物组合的消毒用涂膜剂。
消毒用涂膜剂的杀菌试验:上述实施例1-2的消毒用涂膜剂,在19-21℃条件下,对金黄色葡萄球菌、大肠埃希菌、白色念珠菌、铜绿假单胞菌等作用2-3分钟时的平均杀灭对数值均>5.00。
消毒用涂膜剂的抑菌试验:将实施例1和2的涂膜剂均匀涂布在相应培养基上,在1、2、3和4小时,观察对金黄色葡萄球菌、大肠埃希菌、白色念珠菌、铜绿假单胞菌等的杀灭效果时,显示均具有抑制细菌生长作用;
以胰酪大豆胨琼脂培养基制备平板,凝固后在培养基表面涂布实施例1和2的涂膜剂,待抑菌剂形成薄膜后,分别在0小时、1小时、2小时和4小时后在薄膜表面滴加金黄色葡萄球菌,铜绿假单胞菌菌液1滴,30-35℃培养,观察菌落生长情况,具体如下表:

结论:本发明的消毒用涂膜剂在胰酪大豆胨琼脂培养基表面形成薄膜,阻止了细菌与培养基的接触,抑制了细菌的生长,没有生成菌落,2小时的抑菌实验效果与1和4小时的结果相同。
消毒用涂膜剂的稳定性试验:实施例1-2的涂膜剂的按照《中国药典》2020版中药物稳定性实验项下的“加速试验”结果显示,40±2℃的实验条件下,放置6个月,聚六亚甲基胍含量及乙醇含量下降率均小于5%,预测产品的有效期大于2年。

Claims (6)
1.一种基于聚六亚甲基胍的消毒用涂膜剂,其特征在于,配方如下:聚六亚甲基胍:0.25-0.5;中药材提取物:0.1-5.0;成膜材料:5.0-15.0;增塑剂:2.0-4.0;保湿剂/致孔剂:10-15;乙醇:20-75;余量为精制水。
2.根据权利要求1所述的一种基于聚六亚甲基胍的消毒用涂膜剂,其特征在于:所述成膜材料为聚乙烯醇缩丁醛(PVB)、聚维酮(PVP)、聚乙烯醇(PVA)、乙烯-醋酸乙烯共聚物(EVA)和羟丙甲纤维素中的一种或几种的组合,优选为聚乙烯醇缩丁醛或乙烯-醋酸乙烯共聚物,最优选为聚乙烯醇缩丁醛。
3.根据权利要求1所述的一种基于聚六亚甲基胍的消毒用涂膜剂,其特征在于:所述增塑剂为邻苯二甲酸脂、柠檬酸酯和癸二酸酯中的一种或几种的组合,优选为柠檬酸三乙酯、邻苯二甲酸二丁脂或乙酰柠檬酸三丁酯,最选为柠檬酸三乙酯,所述邻苯二甲酸脂包括但不限于邻苯二甲酸二丁脂和邻苯二甲酸苄丁酯,所述柠檬酸酯包括但不限于柠檬酸三乙酯和乙酰柠檬酸三丁酯,所述癸二酸酯包括但不限于癸二酸二丁酯。
4.根据权利要求1所述的一种基于聚六亚甲基胍的消毒用涂膜剂,其特征在于:所述保湿剂/致孔剂为甘油、丙二醇或聚乙二醇-400中的一种或几种的组合,优选甘油。
5.根据权利要求1所述的一种基于聚六亚甲基胍的消毒用涂膜剂,其特征在于:所述中药材提取物的原料为苦参、蛇床子、大黄、地肤子和百部中的一种或几种的组合,所述中药材提取物是将一种以上中药材混合经水提-醇沉法提取,回收乙醇后经浓缩得到的流浸膏。
6.根据权利要求1-5任一所述的一种基于聚六亚甲基胍的消毒用涂膜剂的制备方法,其特征在于,具体步骤如下:
步骤一:根据配方取用适量的各种原料备用;
步骤二:将成膜材料加入乙醇中,搅拌使其溶解后加入增塑剂,搅拌均匀;
步骤三:将聚六亚甲基胍和中药材提取物溶于精制水中并加入保湿剂/致孔剂,搅拌均匀;
步骤四:将步骤三所得液体加到步骤二所得液体中搅拌均匀,得到基于聚六亚甲基胍的消毒用涂膜剂。
