Abstract
本发明涉及皮肤病中药治疗领域,具体而言,涉及一种用于治疗湿疹的中药组合物。该中药组合物以重量计,包括以下组份:地黄5~15份、石膏10~20份、黄芩5~15份、龙胆草1~5份、地榆5~15份、乌梅3~8份、牡丹皮3~10份、栀子1~5份、炒白术5~15份、豨莶草5~15份、丹参5~15份和甘草3~8份。本发明原料均为天然药物,治疗效果好,复发率低,用药后副作用少。相对于目前市场上同类产品来说,更加安全、有效、无依赖性(无抗生素和化学药物成分);且本发明中药组合物药味数目少,原材料丰富,制备工艺简单,成本低效果佳,具有很好的市场前景。

技术领域
本发明涉及皮肤病中药治疗领域,具体而言,涉及一种用于治疗湿疹的中药组合物。
背景技术
湿疹在临床上是一种非常常见的皮肤性疾病。它使皮肤发生炎症,使患者瘙痒难忍,并且久治不愈。可发生于可发生于体表的任何部位,且多为对称分布,常见于手足、四肢、阴囊、肛门等部位。饮酒、抓挠、肥皂洗或是热水浸泡都可导致患者的瘙痒症状加重,严重者甚至无法入睡,对患者的正常生活造成了较大的影响。
湿疹的一般性分类是按皮损表现分为急性、亚急性、慢性三类。其中急性湿疹皮损初为多数密集的粟粒大小的丘疹、丘疱疹或小水疱,基底潮红,逐渐融合成片,由于搔抓,丘疹、丘疱疹或水疱顶端抓破后呈明显的点状渗出及小糜烂面,边缘不清;亚急性湿疹即为急性湿疹炎症减轻后,皮损以小丘疹、结痂和鳞屑为主,仅见少量丘疱疹及糜烂,但仍伴有剧烈瘙痒;而慢性湿疹大多是由湿疹反复发作不愈转化所致。
在临床上,西医治疗效果多不理想,西医在治疗湿疹上,能够在短期内控制病情,缓解症状,目前西医药治疗湿疹主要以皮质类固醇药物、抗组胺药物、受体阻断剂、免疫调节剂等为主,虽然治疗见效快,但长期使用会引发多种不良反应,并对药物产生依赖性,若患者停止使用药物,病情可能会加重。而中医治疗湿疹效果明显,多以标本兼顾,内外并治为治疗原则,因此,有必要开发一种治疗湿疹上在消退皮损和瘙痒、改善症状、缓解病情、防止复发等方面具有显著疗效的中药组合物。
有鉴于此,特提出本发明。
发明内容
本发明涉及一种用于治疗湿疹的中药组合物,以重量计,其包括以下组份:
地黄5~15份、石膏10~20份、黄芩5~15份、龙胆草1~5份、地榆5~15份、乌梅3~8份、牡丹皮3~10份、栀子1~5份、炒白术5~15份、豨莶草5~15份、丹参5~15份和甘草3~8份。
本发明的第二方面涉及制备如上所述的用于治疗湿疹的中药组合物的方法,包括:
提取各原料中的活性成分;
用所述活性成分制备所需剂型。
本发明的第三方面涉及一种护肤组合物,含有如上所述的治疗湿疹的中药组合物。

本发明原料均为天然药物,标本兼治,复发率低,用药后副作用少。相对于目前市场上同类产品来说,更加安全、有效、无依赖性(无抗生素和化学药物成分);且本发明中药组合物原材料丰富,制备工艺简单,成本低效果佳,具有很好的市场前景。

本发明的发明人将上述药物进行配伍并摸索出最佳用量。发现将各位药物组合内服治疗湿疹效果显著。其中地榆可以起到凉血止血,解毒敛疮、滋阴生津的功效,使得泻火而不损肝之体,含有的主要成分可以促进皮损消退;龙胆草能清肝胆实火而除下焦湿热,泻火除湿,地黄、石膏、乌梅、丹皮均可以凉血收敛;黄芩、栀子清肝泻火除湿;炒白术补气健脾,燥湿利水,既可以顾护脾胃,防止前药苦寒伤脾,又可以助其除湿;豨莶草清解疮毒,祛风湿,既治疮毒,又解瘙痒;丹参主入血分,功擅活血祛瘀,畅行血脉,能祛瘀生新而不伤正,与解毒除湿药配伍,可增强消散痈肿的作用;甘草甘平,药性和缓,能升能降,能浮能沉,为使药,有缓和药性,调和诸药之功。各组分之间协同作用,共同实现治疗湿疹的目的。
一、原料(组分)
地黄为玄参科植物生地Rehmannia glutinosa Libosch.的干燥块根。味甘,性寒。归心、肝肾经。具有清热凉血,养阴生津的功效。对于因血热、血虚、风燥所致湿疹,具有长久的治疗作用。现代药理作用表明生地黄水提液有抗炎、降压、镇静、抗过敏及抑制真菌等作用。地黄的添加量可以为5~15重量份,例如7、8、10、12、13重量份。
石膏为硫酸盐类矿物硬石膏族石膏,主含含水硫酸。味甘、辛,性大寒,钙具有清热泻火、除烦止渴的功效。可用于湿疹,疮疡溃后不敛。地黄的添加量可以为10~20重量份,例如12、14、15、16、18重量份。
黄芩为唇形科植物黄芩Scutellaria baicalensis Georgi.的干燥根。味苦,性寒。归肺、胆、脾、大肠、小肠经。功效为清热燥湿、泻火解毒、止血、安胎。黄芩善泻肺火及上焦实热、还可泻火解毒,善治湿热痞满、肺热血热以及痈肿疮毒等,可用于治疗皮肤病中的湿热之证。现代药理作用表明黄芩具有抗炎、抗敏、抗菌及增强机体免疫等作用。黄芩的添加量可以为5~15重量份,例如7、8、10、12、13重量份。
龙胆草为龙胆科植物条叶龙胆Gentiana manshurica Kitag.、龙胆Gentianascabra Bge.、三花龙胆Gentiana rigescens Pall.的干燥根和根茎。味苦,性寒。归肝、胆经。具有清热燥湿、泻肝胆火的功效,善清下焦湿热,可用于治疗湿热下注,阴肿阴痒,带下,湿疹瘙痒,湿疹瘙痒。现代药理作用表明龙胆草具有显著抗炎、抗过敏作用,对常见的细菌、真菌都有不同程度的抑制作用。龙胆草的添加量可以为1~5重量份,例如2、3、4重量份。
地榆为蔷薇科楦物地榆Sanguisorba officinalis L.或长叶地偷Sanguisorbaofficinalis L.var.longifolia(Bert.)Yüet Li的干燥根。味苦、酸、涩,性微寒。归肝、大肠经。具有凉血止血、解毒敛疮、消炎止痛、杀菌止痒的作用,并且可收敛减少渗出。地榆的添加量可以为5~15重量份,例如7、8、10、12、13重量份。
乌梅为蔷薇科植物梅Prunus mume(Sieb.)Sieb.et Zucc的干燥近成熟果实。味酸、涩,性平。归肝、脾、肺、大肠经。具有敛肺止咳、涩肠止泻、安蛔止痛、生津止渴的功效。具有广泛的抗菌,抗真菌,抗过敏的作用,且可收敛以减少渗出。乌梅的添加量可以为3~8重量份,例如4、5、6、7重量份。
牡丹皮为毛茛科植物牡丹Paeonia suffruticosa Andr.的干燥根皮。味苦、辛,性微寒。归心、肝、肾经。具有清热凉血、活血化瘀的功效。可用于热入营血,瘟毒发斑,吐血衄血,夜热早凉,无汗骨蒸,经闭痛经,跌扑伤痛,痈肿疮毒,能够有效缓解由湿疹引起的皮肤红肿、瘙痒等。牡丹皮的添加量可以为3~10重量份,例如4、5、6、7、8、9重量份。
栀子为茜草科植物栀子Gardenia jasminoides Ellis的干燥成熟果实。味苦,性寒。归心、肺、三焦经。能够清泻三焦火邪,为治热病躁扰不宁之要药。具有泻火除烦、清热利湿、凉血解毒的功效。可用于治疗湿热郁蒸之黄疸,临床多用于治疗血热淋痛、吐血、目赤肿痛、火毒疮疡等。现代药理作用表明,栀子煎剂及醇提取物有镇静、降压、提高免疫功能、抑制多种皮肤真菌及细菌的作用。栀子的添加量可以为1~5重量份,例如2、3、4重量份。
炒白术为菊科植物白术Atractylodes macrocephala Koidz.的干燥根茎用蜜炙麸皮炒制而成。味苦、甘、温。归脾、胃经。具有健脾益气、燥湿利水、止汗,安胎的功效。临床常用于治疗脾虚湿盛型的亚急性、慢性湿疹。炒白术的添加量可以为5~15重量份,例如7、8、10、12、13重量份。
豨莶草为菊科植物豨莶Siegesbeckia orientalis L.、腺梗豨莶Siegesbeckiapubescens Makino或毛梗豨莶Siegesbeckia glabrescens Makino的干燥地上部分。味辛、苦,性寒。归肝、肾经。具有祛风湿,利关节,解毒的功效。用于风湿痹痛,筋骨无力,腰膝酸软,四肢麻痹,半身不遂,风疹湿疮。豨莶草的添加量可以为5~15重量份,例如7、8、10、12、13重量份。
丹参为唇形科植物丹参Salvia miltiorrhiza Bge.的干燥根和根茎。味苦,性微寒。归心、肝经。具有活血祛瘀、通经止痛、清心除烦、凉血消痈的功效,临床长于用慢性湿疹,湿热久郁而致血瘀者,常与清热解毒、活血止痛药配伍治疗疮痈肿痛。丹参的添加量可以为5~15重量份,例如7、8、10、12、13重量份。
甘草为豆科植物甘草Glycyrrhiza uralensis Fisch.、胀果甘草Glycyrrhizainflata Bat.、或光果甘草Glycyrrhiza glabra L.的干燥根和根茎。味甘,性平。归心、肺、脾、胃经。具有补脾益气、清热解毒、祛痰止咳、缓急止痛、调节诸药的功效,临床上亦常用于治疗痈肿疮毒。现代药理作用表明,生甘草具有抗炎、抗病毒、抗肿瘤和保肝解毒及增强免疫功能等作用,且甘草提取物有抗过敏作用。甘草的添加量可以为3~8重量份,例如4、5、6、7重量份。
原料中还可以包含药学上可接受的辅料。
药学上可接受的辅料成分的实例包括结合剂(糖浆、阿拉伯树胶、明胶、山梨醇、黄芪胶(tragacanth)、聚乙烯吡咯烷酮等)、填充剂(乳糖、蔗糖、淀粉、磷酸钙、山梨糖醇、甘氨酸等)、润滑剂(硬脂酸镁、滑石、聚乙二醇等)、崩解剂(淀粉、微晶纤维素(microcrystalline cellulose)等)、湿润剂(十二烷基硫酸钠(sodium lauryl sulphate)等)、悬浮剂(山梨糖醇、糖浆、甲基纤维素、葡萄糖浆(glucose syrup)、明胶、氢化食用脂肪等)、乳化剂(卵磷脂、山梨醇单油酸酯、阿拉伯树胶等)、非水性载体(杏仁油、分馏椰子油或甘油、丙二醇、乙醇等疏水酯等)、防腐剂(对羟基苯甲酸甲酯或对羟基苯甲酸丙酯、山梨酸等)、芳香剂(合成香料、天然香料等)、甜味剂(蔗糖、甜叶菊、木糖醇等)、pH调节剂(碳酸氢钠、碳酸钾等)、着色剂(色素、染料等)、增稠剂(阿拉伯树胶、甲基纤维素等)、抗氧化剂(维生素C、维生素E等)等等。
二、形式和剂型
在一种实施方式中,本发明的中药组合物为简单混合物。制备过程为将各药材(原料)简单混合即可,这种简单混合物适于制备简单的水剂。
在另一种实施方式中,本发明的药物还可以做成中成药的形式。中成药的剂型包括但不限于散剂、膏剂、丹剂、酒剂、锭剂、水剂、气雾剂或巴布剂;
(1)散剂
散剂是一种或多种药材混合制成的粉末状制剂,分内服散剂和外用散剂,是我国古代剂型之一。散剂治疗范围广,服用后分散快,奏效迅速,且具有制作方便、携带方便、节省药材等优点。
本发明采用外用散剂。
(2)膏剂
膏剂系药物剂型之一。膏剂是将药物用水或植物油煎熬浓缩而成的剂型。有内服和外用两种。外用膏有软膏、硬膏两种。外用的软膏称油膏,一般药膏,是把蜂蜡加入棉子油或花生油中,加热熔化,乘热加入应用的药物细粉,不断搅拌,待冷凝即成。膏剂优选为软膏、乳膏剂或贴膏剂。
本发明优选为外用软膏。
(3)丹剂
丹剂有内服和外用两种,没有固定剂型。有的将药物研成细末即成,有的再加糊或黏性药汁制成各种形状,有的丹剂也是丸剂的一种。因多用精炼药品或贵重药品制成,所以不称丸而称丹。外用丹剂,仅供外科用。
(4)酒剂
酒剂古称“酒醴”,后世称为“药酒”。是以酒为溶媒,一般以白酒或黄酒浸制药物,或加温同煮,去渣取液供内服或外用。酒剂不宜于阴虚火旺的病人。
(5)锭剂
将药物研成细末,单独或加适当的糊粉、蜂蜜与赋型剂混合后制成不同形状的一种固体制剂。本发明采用外用,磨汁涂敷患处。若制成饼状则称为饼剂。
(6)水剂
将药物冲泡后外用。其可以为洗剂或搽剂。
(7)气雾剂
系指药物与适宜抛射剂封装于具有特制阀门系统的耐压容器中制成的的制剂,使用时,借助抛射剂的压力将内容物以定量或非定量喷出,药物喷出多为雾状气溶胶,其雾滴一般小于50μm。
(8)巴布剂
巴布剂(cataplasma)是一种新型的外用制剂,系指将药物溶解或混合于水溶性高分子材料基质中,摊涂于裱背材料上,供皮肤贴敷的外用剂型。
(9)凝胶剂
系指药物与能形成凝胶的辅料制成溶液、混悬或乳状液型的稠厚液体或半固体制剂。通常凝胶剂限局部用于皮肤及体腔(如鼻腔、阴道和直肠)。
三、制备方法
所述组合物中的活性物质可以通过分别提取中药原料得到,也可以通过共同提取中药原料药材得到,也可以通过其他方式得到,如:通过粉碎、压榨、研磨、过筛、渗漉、萃取、水提、醇提、层析等方法得到,这些活性物质可以是浸膏形式的物质,可以是干浸膏也可以是流浸膏,根据剂型的不同需要决定制成不同的浓度。
1、简单混合物的制备
可以将上述原料简单混合在一起,从而制备本发明的简单混合物。
在一些实施方式中,制备该药物的方法包括以下步骤:将各组分粉碎,混合在一起。
2、中成药的制备
在一些实施方式中,制备该药物的方法包括:
提取各原料中的活性成分;
用所述活性成分制备所需剂型。
在一些实施方式中,提取各原料中的活性成分的方法包括水煎煮法、浸渍法、渗漉法、改良明胶法、回流法、溶剂提取法、水蒸气蒸馏法和升华法中的一种,或其组合。
本发明治疗湿疹的中药组合物,其药物活性物质是经过提取加工制得的,如可以用以下方法加工:
方法1:按处方量称取所有药材经过6~12倍水(或50%~95%乙醇以及其他可接受溶剂)回流(或浸渍)提取1~3次,每次0.5~2h,合并滤液,过100~600目滤布,55℃~85℃浓缩至相对密度为1.05~1.30的稠膏。
进一步优选:按处方量称取所有药材经过8倍水回流提取2次,每次1h,合并滤液,过300目滤布,60℃浓缩至相对密度为1.20的稠膏。
方法2:按处方量称取所有药材经过6~12倍水回流提取1~3次,每次0.5~2h,合并滤液,过100~600目滤布,55℃~85℃浓缩至相对密度为1.05~1.30的稠膏。加70~90%乙醇醇沉,收集醇沉液,55℃~85℃浓缩至相对密度为1.05~1.30的稠膏。
进一步优选:按处方量称取所有药材经过8倍水回流提取2次,每次1h,合并滤液,过300目滤布,60℃浓缩至相对密度为1.20的稠膏。加80%乙醇醇沉,收集醇沉液,65℃浓缩至相对密度为1.20的稠膏。
方法3:按处方量称取所有药材经过6~12倍水回流提取1~3次,每次0.5~2h,合并滤液,过100~600目滤布,55℃~85℃浓缩至相对密度为1.05~1.30的稠膏。上大孔吸附树脂柱,3~8倍水洗,弃去水液,50~90%乙醇洗脱,收集50~90%乙醇洗脱部分,55℃~85℃浓缩至相对密度为1.05~1.30的稠膏。
进一步优选:按处方量称取所有药材经过8倍水回流提取2次,每次1h,合并滤液,过300目滤布,70℃浓缩至相对密度为1.15的稠膏。上大孔吸附树脂柱,5倍水洗,弃去水液,80%乙醇洗脱,收集80%乙醇洗脱部分,70℃浓缩至相对密度为1.25的稠膏。
方法4:按处方量称取所有药材经过6~12倍水回流提取1~3次,每次0.5~2h,合并滤液,过100~600目滤布,55℃~85℃浓缩至相对密度为1.05~1.30的稠膏。上大孔吸附树脂柱,3~8倍水洗,弃去水液,50~90%乙醇洗脱,收集50~90%乙醇洗脱部分,回收乙醇,加0.1~3%活性炭脱色0.5~2h,过0.1~0.45μm滤膜,55℃~85℃浓缩至相对密度为1.05~1.30的稠膏。
进一步优选:按处方量称取所有药材经过8倍水回流提取2次,每次1h,合并滤液,过300目滤布,70℃浓缩至相对密度为1.15的稠膏。上大孔吸附树脂柱,5倍水洗,弃去水液,80%乙醇洗脱,收集80%乙醇洗脱部分,回收乙醇,加1%活性炭脱色1h,过0.22μm滤膜,70℃浓缩至相对密度为1.25的稠膏。
方法5:按处方量称取牡丹皮,经过3~18小时水蒸气蒸馏提取得到蒸馏液;另按处方量称取其他药材经过6~12倍水(或50%~95%乙醇以及其他可接受溶剂)回流(或浸渍)提取或上述药材与牡丹皮提蒸馏液后药渣合并后经水(或醇以及其他可接受溶剂)提取1~3次,每次0.5~2h,合并滤液,过100~600目滤布,55℃~85℃浓缩至相对密度为1.05~1.30的稠膏,稠膏与蒸馏液混合即得药物活性物质。上述稠膏也可经过醇沉、滤膜、色谱柱等其他方式进行精制处理。
进一步优选:按处方量称取牡丹皮经过12小时水蒸气蒸馏提取得到蒸馏液;另按处方量称取其他药材经过8倍水(或80%乙醇以及其他可接受溶剂)回流提取或上述药材与牡丹皮提蒸馏液后药渣合并后经水(或醇以及其他可接受溶剂)提取2次,每次1h,合并滤液,过100~600目滤布60℃浓缩至相对密度为1.20的稠膏,稠膏与蒸馏液混合即得药物活性物质。上述稠膏也可经过醇沉、滤膜、色谱柱等其他方式进行精制处理。
进一步优选:所述方法包括:
a)将所述牡丹皮用水蒸气蒸馏法处理,得到蒸馏液和药渣;
所述蒸馏液经低温沉淀、固液分离得到提取液和沉淀物,滤取所述沉淀物得丹皮酚提取物;
b)所述地黄、石膏、黄芩、龙胆草、地榆、乌梅、栀子、炒白术、豨莶草、丹参、甘草及所述药渣煎煮得到煎煮液,将所述煎煮液及所述提取液过滤浓缩得到稠膏;
c)将所述稠膏与所述挥发油及所述丹皮酚提取物混合。
在一些实施方式中,所述水蒸气蒸馏法的时间为3~18小时,例如12小时。
在一些实施方式中,所述煎煮所用溶剂为水或者乙醇(50%~95%乙醇,优选70%~90%,更优选75%~85%,更优选80%)。
在一些实施方式中,所述煎煮所用溶剂为被煎煮药材种类的6~12倍水,优选8倍。
在一些实施方式中,所述煎煮的次数为1~3次,进一步优选每次煎煮的时间为0.5h~4h,例如1h、2h、3h。
在一些实施方式中,所述稠膏也可经过醇沉、滤膜、色谱柱等其他方式进行精制处理。
四、护肤组合物
本发明还提供护肤组合物,其含有如上所述的用于治疗湿疹的中药组合物。
在一种实施方式中,该护肤组合物另外包含生理学上可接受的介质和选自以下的至少一种化合物:增稠剂、防腐剂、芳香剂、杀/抑菌剂、有机溶剂(特别包括C1-C6醇和C2-C10羧酸酯)、和基于碳的油和/或硅油(无论它们是矿物、动物和/或植物来源)。
该护肤组合物另外包含生理学上可接受的介质,即没有令人不快的气味、颜色或外观,并且不会引起使用者任何不适(如不可接受的刺痛或紧绷)的介质。特别地,该组合物适合局部施用于皮肤。
因此,“生理学上可接受的介质”应理解为意指与人角蛋白材料、并且特别是与身体或面部皮肤相容的介质。
根据本发明的组合物可以包含通常用于所设想的施用领域的任何化妆品辅助剂。
根据本发明的组合物可以大体上是流体并且可以具有呈白色或着色的乳膏、软膏、面霜、身体霜、乳液、洗液(特别是洗面奶、沐浴剂)、精华液、糊剂、凝胶或泡沫的外观。它可以任选地以气溶胶形式施用。它还可以处于固体形式,并且特别是处于棒状物形式。它还可以在支持物上,例如在擦拭物(如卸妆擦拭物)上;又例如为面膜。
五、医药用途
本发明还涉及如上所述的中药组合物在制备治疗湿疹的药物中的应用。
本发明还涉及治疗湿疹的方法,其包括用有效量的如上所述的中药组合物局部接触皮肤。
如本文中所用,术语“治疗”包括抑制病理状态、病症或疾病,例如阻止或减少病理状态、病症或疾病或其临床症状的发展;或缓解病理状态、病症或疾病,例如致使病理状态、病症或疾病或其临床症状消退。这些术语还涵盖治疗和治愈。治疗意指改善或以其它方式有益地改变病理状况、病症或疾病的症状的任何方式。优选地,需要此类治疗的受试者为哺乳动物,更优选为人。
下面将结合实施例对本发明的实施方案进行详细描述。
实施例1~7
一种治疗湿疹的中药组合物,配方如下:
制备方法1:
按处方量称取处方中所有药材加8倍水煎煮3次,每次1小时,合并煎煮液,过滤,滤液浓缩至相对密度为1.20的稠膏。
制备方法2:
按处方量称取处方中所有药材加8倍水煎煮3次,每次1小时,合并煎煮液,过滤,滤液浓缩至相对密度为1.20的稠膏,加80%乙醇醇沉,收集醇沉液,65℃浓缩至相对密度为1.20的稠膏。
制备方法3:
按处方量称取处方中所有药材加8倍80%乙醇煎煮2次,每次1.5小时,合并煎煮液,过滤,滤液浓缩至相对密度为1.15的稠膏,上大孔吸附树脂柱,3倍水洗,弃去水液,50%乙醇洗脱,收集50%乙醇洗脱部分,85℃浓缩至相对密度为1.20的稠膏。
制备方法4:
按处方量称取处方中所有药材加8倍80%乙醇煎煮2次,每次1.5小时,合并煎煮液,过滤,滤液浓缩至相对密度为1.15的稠膏,上大孔吸附树脂柱,3倍水洗,弃去水液,50%乙醇洗脱,收集50%乙醇洗脱部分,加1%活性炭脱色1h,过0.22μm滤膜,70℃浓缩至相对密度为1.20的稠膏。
制备方法5:
(1)按处方量称取牡丹皮,用水蒸气蒸馏提取10小时得蒸馏液,蒸馏液及药渣备用。
(2)按处方量称取处方中另11味药材及(1)中药渣加6倍水煎煮3次,每次1小时,合并煎煮液及(1)中的提取液,过滤,55℃浓缩至相对密度为1.20的稠膏,与(1)中蒸馏液混合,即得。
制备方法6:
(1)按处方量分别称取牡丹皮用水蒸气蒸馏提取12小时得蒸馏液,蒸馏液备用。
(2)按处方量称取处方中另11味药材加10倍80%乙醇煎煮2次,每次2小时,合并煎煮液,过滤,滤液浓缩至相对密度为1.15的稠膏,加80%乙醇醇沉,收集醇沉液,65℃浓缩至相对密度为1.20的稠膏,与(1)中蒸馏液混合,即得。
制备方法7:
(1)按处方量分别称取牡丹皮用水蒸气蒸馏提取12小时得蒸馏液,蒸馏液备用。
(2)按处方量称取处方中另11味药材加10倍80%乙醇煎煮2次,每次2小时,合并煎煮液,过滤,滤液浓缩至相对密度为1.15的稠膏,上大孔吸附树脂柱,8倍水洗,弃去水液,90%乙醇洗脱,收集90%乙醇洗脱部分,加1%活性炭脱色1h,过0.22μm滤膜,55℃浓缩至相对密度为1.20的稠膏,与(1)中蒸馏液混合,即得。
其中,实施例1-7分别采用制备方法1-7制备。
对比例1~8
对比例1~8制备方法:
按处方量称取处方中所有药材加8倍水煎煮3次,每次1小时,合并煎煮液,过滤,滤液浓缩至相对密度为1.20的稠膏,加80%乙醇醇沉,收集醇沉液,65℃浓缩至相对密度为1.20的稠膏。
本发明提供了上述实施例及对比例中制品的治疗湿疹的试验研究:
实验动物分组及造模:SPF级KM小鼠共170只,雌雄各半,体重20±2g。随机分成17组:空白对照组、模型对照组、实施例1-7组、对比例1-8组,每组10只。小鼠实验前1d于小鼠背部选2cm×2cm大小区域用剃毛刀去毛。第1天,以7%DNCB 100μl外涂小鼠去毛背部致敏;第2天,同法强化涂抹1次,再次致敏;第7天,在小鼠右耳双侧用移液枪外涂1%DNCB 20μl激发。激发后肉眼观察小鼠右耳明显肿胀,局部毛细血管扩张,或有少许渗出物,提示急性湿疹模型成功建立。
给药方法:造模成功后第2天开始对各组给药,实施例组及对比例组按编号每日灌胃给予对应的样品0.87g/kg BW,2次/天;空白对照组及模型对照组每日灌胃纯净水0.87g/kg BW,2次/天;各组均连续给药14d。给药期间用正常饲料喂养。
测定方法:末次给药后24h,用游标卡尺测量双耳厚度(取3个点测量取去均值),计算厚度差;小鼠脱颈椎处死,剪下双耳,用圆形打孔器(直径为6mm)打下左右耳相同部位,称双耳片重,计算两耳片重之差。计算厚度差=右耳厚度-左耳厚度,重量差=右耳重量-左耳重量。
由实施例1-7以及对比例1-8制得的药物组合物对小鼠左右耳厚度差和左右耳重量差比较结果如下表1所示:
注:与空白对照组比较,**P<0.01;与模型对照组比较,aP<0.01,bP<0.05
模型对照组小鼠造模侧耳部发红、肿胀,正常对照组小鼠两耳色泽及厚度相近,其余各给药组右耳红肿程度情况不一。
模型对照组与空白对照组比较,左右耳厚度差及重量差有明显增加,有统计学意义(P<0.01),说明KM小鼠急性湿疹模型成立。与模型对照组比较,实施例样品能显著改善耳部肿胀(P<0.05)。对比例样品虽然能抑制耳廓肿胀,但不具有统计学意义。上述试验结果表明本发明提供的组合物具有良好的治疗湿疹的作用。
以上所述实施例的各技术特征可以进行任意的组合,为使描述简洁,未对上述实施例中的各个技术特征所有可能的组合都进行描述,然而,只要这些技术特征的组合不存在矛盾,都应当认为是本说明书记载的范围。
以上所述实施例仅表达了本发明的几种实施方式,其描述较为具体和详细,但并不能因此而理解为对发明专利范围的限制。应当指出的是,对于本领域的普通技术人员来说,在不脱离本发明构思的前提下,还可以做出若干变形和改进,这些都属于本发明的保护范围。因此,本发明专利的保护范围应以所附权利要求为准。
Claims (5)
1.一种内服的用于治疗湿疹的中药组合物,其特征在于,以重量计,由以下组份组成:
地黄7~13份、石膏12~18份、黄芩7~13份、龙胆草2~4份、地榆7~13份、乌梅4~7份、牡丹皮4~9份、栀子2~4份、炒白术7~13份、豨莶草7~13份、丹参7~13份和甘草4~7份。
2.根据权利要求1所述的内服的用于治疗湿疹的中药组合物,其特征在于,以重量计,由以下组份组成:
地黄8~12份、石膏14~16份、黄芩8~12份、龙胆草2~4份、地榆8~12份、乌梅5~6份、牡丹皮5~8份、栀子2~4份、炒白术8~12份、豨莶草8~12份、丹参8~12份和甘草5~6份。
3.根据权利要求2所述的内服的用于治疗湿疹的中药组合物,其特征在于,以重量计,由以下组份组成:
地黄9~11份、石膏14~16份、黄芩9~11份、龙胆草2~4份、地榆9~11份、乌梅5~6份、牡丹皮6~7份、栀子2~4份、炒白术9~11份、豨莶草9~11份、丹参9~11份和甘草5~6份。
4.制备根据权利要求1~3任一项所述的内服的用于治疗湿疹的中药组合物的方法,其特征在于,包括:
提取各原料中的活性成分;
用所述活性成分制备所需剂型。
5.根据权利要求4所述的方法,其特征在于,提取各原料中的活性成分的方法包括水煎煮法、浸渍法、渗漉法、改良明胶法、回流法、溶剂提取法、水蒸气蒸馏法和升华法中的一种,或其组合。
