white peony root extract, bletilla tuber extract, red clover extract and poncirus trifoliata extract can quickly permeate to the muscle bottom to generate the functions of antioxidation, anti-inflammatory and blood circulation promotion, so that the darkness and dullness of skin are improved, uneven skin color is repaired, the skin color is lightened, the skin elasticity is increased, and finally the skin is fair and glossy, and the texture is warm and fine;

Further, the method comprises the following steps of: 1-1000 parts of solvent, 1-100 parts of thickener, 1-100 parts of humectant, 1-100 parts of skin conditioning agent, 1-100 parts of surfactant, 1-100 parts of chelating agent, 1-100 parts of softener, 1-100 parts of film forming agent, 1-100 parts of preservative and 1-100 parts of pH regulator.
The preparation method of the moisturizing and skin-tendering essence lotion comprises the following steps of:
s1: optimizing a fibroin gene sequence by using a pichia pastoris conventional codon on the gene level, synthesizing the gene artificially, integrating the gene into a yeast chromosome, constructing a recombinant pichia pastoris engineering bacterium with constitutive secretory expression, transferring the recombinant pichia pastoris engineering bacterium into a fermentation medium, actively secreting and expressing the recombinant fibroin into a fermentation broth along with the vital activity of the engineering bacterium, centrifuging to collect a supernatant, adsorbing the recombinant fibroin by using resin under a non-deformation condition, washing impurities by using a washing solution, eluting the recombinant fibroin, collecting an eluent, desalting and concentrating, and finally performing vacuum freeze drying to obtain the recombinant fibroin for later use;
s2: pulverizing radix Paeoniae alba, rhizoma Bletillae, herba Portulacae, flos Lonicerae, fructus Ponciri Trifoliatae, herba Trifoliatae, fructus Muscovitum, lac Regis Apis, and wheat embryo respectively, sieving, adding 10-20 times of solvent (75-85% ethanol) by weight, soaking at 35-45deg.C for 7-9 hr, performing low temperature ultrasonic extraction for 5-7 hr, centrifuging, filtering to obtain supernatant, filtering again, and lyophilizing to obtain corresponding plant extract, mixing;
s3: placing water, a softening agent, a film forming agent and a solvent in a clean container, heating to 45-55 ℃, stirring and dissolving for later use;
s4: adding the recombinant fibroin prepared in the step S1 and the plant extract prepared in the step S2 into a container of the step S3, simultaneously adding methyl glucitol polyether-20 and PEG-40 hydrogenated castor oil, heating to 60-65 ℃ at the rotating speed of 60-90rpm, preserving heat and stirring for 15-25 min, homogenizing for 2-5 min until the materials are uniformly dispersed;
s5: adding saccharide isomerides, hydrolyzed sodium hyaluronate, trehalose and sodium polyglutamate raw materials into S4, homogenizing for 2-5 minutes under the stirring condition, starting a vacuum device to enable the vacuum degree of a container to be 0.04 Pa-0.1 MPa, and keeping the vacuum state to keep the temperature and stirring for 10 minutes;
s6: cooling to 35-40 ℃, adding acrylic acid (ester)/C10-30 alkanol acrylate cross-linked polymer and the recombinant silk fibroin raw material prepared in the step S1 into the mixture prepared in the step S5, and uniformly stirring;
s7: adding lysine and magnesium ascorbyl phosphate into the mixture prepared in the step S6, and uniformly stirring;
s8: cooling to 18-25deg.C, adding antiseptic octaethylene glycol and phenoxyethanol, and stirring; adding pH regulator arginine and sodium citrate, and stirring uniformly; and adding chelating agent EDTA disodium, and stirring uniformly to obtain the moisturizing and skin-tendering essence.
Preferably, the natural plant components are soaked for 8 hours at 40 ℃ and subjected to low-temperature ultrasonic extraction for 6 hours.
Further, the stirring speed in S3, S4 and S5 is 60-90rpm; the stirring speed in S6, S7 and S8 is 20-40rpm, and the homogenizing speed in S4 and S5 is 2000-3000rpm.
Further, the solvent comprises at least one of purified water, butanediol, dipropylene glycol, propylene glycol, glycerol, and ethanol. Preferably, the solvent is ethanol at a concentration of 80% by volume.
Further, the thickener includes, but is not limited to carbomers, acrylic acid (esters) based/C10-30 alkanol acrylate cross-linked polymers, hydroxyethyl acrylate/sodium acryloyldimethyl taurate copolymer, xanthan gum, hydroxyethyl cellulose, and the like.
Further, the skin conditioning agent comprises at least one of lysine, ascorbic acid, vitamin C, ceramide, magnesium ascorbyl phosphate, ascorbyl glucoside, ascorbyl palmitate, glutathione, dipotassium glycyrrhizinate, glabridin, and lecithin.
Further, the humectant comprises at least one of saccharide isomer, sodium hyaluronate, hydrolyzed sodium hyaluronate, 1, 2-hexanediol, sodium polyglutamate, squalane, chitosan, trehalose, urea, allantoin, polyether glycerol-26, sodium lactate, sodium pyrrolidone carboxylate, xylitol, and biological gum-1.
Further, the surfactant includes, but is not limited to, methyl glucitol polyether-20, PEG-40 hydrogenated castor oil, PEG-60 hydrogenated castor oil, di-C12-15 alkanol polyether-8 phosphate, hydrogenated lecithin, polyglycerol-6 polyricinoleate, polyglycerol-10 laurate, and the like.

Example 1
The moisturizing and skin-tendering essence lotion comprises the following components in parts by weight: 40 parts of recombinant silk fibroin, 12 parts of white peony root extract, 15 parts of bletilla striata extract, 10 parts of purslane extract, 11 parts of honeysuckle flower extract, 15 parts of poncirus trifoliata extract, 8 parts of red clover extract, 10 parts of papaya fruit extract, 15 parts of royal jelly extract, 20 parts of wheat germ extract, 400 parts of solvent, 3 parts of thickener, 30 parts of humectant, 20 parts of skin conditioner, 6 parts of surfactant, 10 parts of chelating agent, 3 parts of softener, 3 parts of film forming agent, 3 parts of preservative and 3 parts of pH regulator.

The preparation method of the moisturizing and skin-tendering essence lotion comprises the following steps:
s1: the recombinant pichia pastoris engineering bacteria with constitutive secretory expression are constructed by optimizing the gene sequence of the fibroin by using the conventional codon of pichia pastoris on the gene level and synthesizing the gene artificially and integrating the gene into yeast chromosome, and after the recombinant pichia pastoris engineering bacteria are transferred into a fermentation medium, the recombinant fibroin is actively secreted and expressed in a fermentation broth along with the vital activity of the engineering bacteria, the supernatant is collected by centrifugation, the recombinant fibroin is adsorbed by resin under the non-deformation condition, the impurities are washed by using a washing liquid, the recombinant fibroin is eluted, the eluent is collected, desalted and concentrated, and finally the recombinant fibroin is obtained by vacuum freeze drying for standby.
S2: pulverizing radix Paeoniae alba, rhizoma Bletillae, herba Portulacae, flos Lonicerae, fructus Ponciri Trifoliatae, herba Trifoliatae, fructus Muscovitum, lac Regis Apis, and wheat embryo respectively, sieving, adding 15 times of 80% ethanol by weight, soaking at 40deg.C for 8 hr, performing low-temperature ultrasonic extraction for 6 hr, centrifuging supernatant, filtering, refluxing ethanol, filtering again, and lyophilizing to obtain corresponding plant extract mixture;
s3: placing water, polyethylene glycol-8, polyquaternium-51 and glycerin in a clean container, heating to 50 ℃ by using external heating equipment, and stirring and dissolving for later use;
s4: adding the natural component mixture prepared in the step S2, methyl glucitol polyether-20 and PEG-40 hydrogenated castor oil into a container of S3, stirring and heating to 62 ℃ at a rotating speed of 70rpm, preserving heat and stirring for 20min, and homogenizing for 3 min until the materials are uniformly dispersed;
s5: uniformly stirring saccharide isomer, hydrolyzed sodium hyaluronate, trehalose and sodium polyglutamate raw materials, adding into the container in the step S4, homogenizing for 3 minutes under the stirring condition, starting a vacuum device to ensure that the vacuum degree of the container is 0.06MPa, keeping the vacuum state, and continuously preserving heat and stirring for 10 minutes;
s6: when the temperature is reduced to 38 ℃, adding acrylic acid (ester)/C10-30 alkanol acrylate crosslinked polymer and recombinant silk fibroin raw material into the mixture prepared in the step S5, and uniformly stirring;
s7: adding lysine and magnesium ascorbyl phosphate raw materials into the mixture prepared in the step S6, and uniformly stirring;
s8: cooling to 22deg.C, adding antiseptic octaethylene glycol and phenoxyethanol, and stirring; adding pH regulator arginine and sodium citrate, and stirring uniformly; and adding chelating agent EDTA disodium, and stirring uniformly to obtain the moisturizing and skin-tendering essence.
Example 2
This embodiment differs from embodiment 1 in that: the plant components in the moisturizing and skin-tendering essence lotion comprise the following components in parts by weight: 30 parts of recombinant silk fibroin, 20 parts of white peony root extract, 16 parts of bletilla striata extract, 15 parts of purslane extract, 10 parts of honeysuckle extract, 10 parts of poncir trifoliata extract, 8 parts of red clover extract, 15 parts of papaya extract, 20 parts of royal jelly extract and 15 parts of wheat germ extract.
Example 3
This embodiment differs from embodiment 1 in that: the plant components in the moisturizing and skin-tendering essence lotion comprise the following components in parts by weight: 35 parts of recombinant silk fibroin, 15 parts of white peony root extract, 15 parts of bletilla striata extract, 16 parts of purslane extract, 16 parts of honeysuckle extract, 20 parts of poncir trifoliata extract, 5 parts of red clover extract, 15 parts of papaya extract, 10 parts of royal jelly extract and 10 parts of wheat germ extract.

This embodiment differs from embodiment 1 in that: the plant components in the moisturizing and skin-tendering essence lotion comprise the following components in parts by weight: 100 parts of recombinant silk fibroin, 1 part of white peony root extract, 1 part of bletilla striata extract, 100 parts of purslane extract, 100 parts of honeysuckle extract, 1 part of poncir trifoliata extract, 100 parts of red clover extract, 1 part of papaya extract, 100 parts of royal jelly extract and 1 part of wheat germ extract.
Experimental example 1
According to cosmetic safety technical Specification (2015 edition), the moisturizing and skin-tendering essence lotions prepared in examples 1-3 were subjected to microorganism and heavy metal detection. The test results are shown in Table 1:

As can be seen from the data in Table 1, the moisturizing and skin-tendering essence prepared in the examples 1-3 of the invention do not detect microorganisms, and the heavy metal content accords with the standard, which indicates that the moisturizing and skin-tendering essence prepared in the invention is safe and reliable.
Experimental example 2
The physical and chemical indexes of the moisturizing and skin-tendering essence prepared in the embodiments 1-3 of the invention are tested, and the results are as follows:
2.1 traits: the product is milky semitransparent essence with proper viscosity, fineness, uniformity and good spreadability.
2.2pH check: a small amount of essence is dipped by wet PH test paper, and the PH is 6-7.
2.3 cold and hot test: the moisturizing and skin-tendering essence prepared in examples 1-3 is packaged in a transparent cosmetic bottle, and no layering phenomenon is observed when the cosmetic bottle is refrigerated for about one week in a refrigerator at 4 ℃. The mixture is placed in a constant temperature box at 55 ℃ for 24 hours, and the phenomena of layering, smell change and the like are avoided.
2.3 centrifugation test: the moisturizing and skin-tendering essence prepared in the examples 1-3 is packaged in a test tube, and centrifuged for 20min at 3000r/min without layering phenomenon.
2.4 irritation test, allergy test: 1) Animal experiment: the back of the mice is cut off, the moisturizing and skin-tendering essence prepared in the examples 1-3 is respectively smeared on the parts with the hair cut off, and compared with the parts without the hair cut off, the moisturizing and skin-tendering essence has no irritation and anaphylactic reaction; 2) Human body experiment: the moisturizing and skin-tendering essence prepared in examples 1-3 is smeared on the hand of volunteers (18-66 years old, 200 people) and observed after 30 minutes, the phenomena of redness, eruption and blistering do not occur.
2.5 room temperature standing experiments: the moisturizing and skin-tendering essence prepared in examples 1-3 was placed in a cosmetic bottle, and left standing at room temperature for 6 months without layering, without change in feeling after use, and without change in smell.
Experimental example 3
3.1 Experimental purposes: the moisturizing and skin-tendering essence lotions prepared in examples 1-3 were subjected to long-term moisturizing effect tests, and the moisture content of human skin under the same environment was tested.
3.2 subjects: 120 women aged 18-40 were selected and divided into four groups: blank (purified water), examples 1-3 (moisturizing skin-rejuvenating serum prepared in examples 1-3). No cosmetic or topical medicine can be used 2-3 days before the test.
3.3 experimental method:
before the test: the face is cleaned, the subject sits still for 30min under different dehumidifying environments before formal test, the skin moisture content value of the blank area before use is tested by using a CK skin tester, and the tested part is the forehead rich in sebaceous glands.
Test treatment: treatment with purified water was performed on the face of subjects in the placebo group; the faces of subjects in the group of examples 1 to 3 were treated with the moisturizing and skin-rejuvenating lotions prepared in examples 1 to 3 of the present invention, respectively; and the moisture content of the skin surface is measured at the measured part by using a CK skin tester at different time intervals (6 time, 12 time and 24 time), the working environment temperature is 22-24 ℃, the relative humidity is 33-46%, and the unit of the skin moisture content is au (arbitrary unit). The test results are shown in Table 2:

As can be seen from the data in Table 2, the moisturizing and skin-tendering essence prepared in examples 1-3 of the present invention has a greatly improved moisturizing effect compared with the blank control group, which indicates that the moisturizing and skin-tendering essence prepared in the present invention has a long-acting moisturizing effect.
Experimental example 4
4.1 experimental purposes: comprehensive effects of long-term use are studied aiming at the moisturizing and skin-tendering essence prepared in the examples 1-3.
4.2 subjects: selecting 150 women in 18-66 years old as trial objects by using a poll scoring method, and dividing the women into three groups: example 1, example 2 and example 3 (moisturizing skin-rejuvenating lotions prepared in examples 1-3, respectively).
4.3 experimental method:
the skin care essence is applied to the face of a subject, and is used twice a day in the morning and evening for two months. Finally, investigation and research on moisture retention, skin tendering, fineness, wrinkle resistance, whitening and antiallergic property are carried out. Wherein, each use effect is totally divided into 5 points: a score of 5 being the highest score indicates that it is very satisfactory; 4 is satisfied; a score of 3 being acceptable; a score of 3 or less is not good and unacceptable. The test results are shown in Table 3:

As can be seen from the data in Table 3, the moisturizing and skin-tendering essence lotions prepared in examples 1-3 of the present invention have good effects on moisturizing, skin tendering, fineness, wrinkle resistance, whitening and antiallergic properties. The skin care essence lotion used by the tested volunteers is smooth and comfortable and has good touch feeling. No special reaction is caused on the skin no matter what the skin is used, the moisturizing and skin tendering effects are good, and the skin is obviously glossy and ruddy after the skin is used.

the method for obtaining the recombinant fibroin comprises the following steps: the recombinant pichia pastoris engineering bacteria with constitutive secretory expression are constructed by optimizing the gene sequence of the fibroin by using the conventional codon of pichia pastoris on the gene level and synthesizing the gene artificially, the recombinant pichia pastoris engineering bacteria are transferred into a fermentation medium, the recombinant fibroin is actively secreted and expressed in a fermentation broth along with the vital activity of the engineering bacteria, the supernatant is collected by centrifugation, the recombinant fibroin is adsorbed by resin under the non-deformation condition, the impurities are washed by using a washing solution, the recombinant fibroin is eluted, the eluent is collected, desalted and concentrated, and finally the recombinant fibroin is obtained by vacuum freeze drying.

The moisturizing skin-rejuvenating lotion according to any one of claims 1 to 4, further comprising, in parts by weight: 1-1000 parts of solvent, 1-100 parts of thickener, 1-100 parts of humectant, 1-100 parts of skin conditioning agent, 1-100 parts of surfactant, 1-100 parts of chelating agent, 1-100 parts of softener, 1-100 parts of film forming agent, 1-100 parts of preservative and 1-100 parts of pH regulator.
6. A method for preparing the moisturizing and skin-tendering essence according to any one of claims 1 to 5, comprising the following steps:
s1: the recombinant fibroin prepared in the claim 1 is reserved;
s2: pulverizing radix Paeoniae alba, rhizoma Bletillae, herba Portulacae, flos Lonicerae, fructus Ponciri Trifoliatae, herba Trifoliatae, fructus Muscovitum, lac Regis Apis, and wheat embryo respectively, sieving, adding 10-20 times of solvent, soaking at 35-45deg.C for 7-9 hr, performing low temperature ultrasonic extraction for 5-7 hr, centrifuging, filtering to obtain supernatant, filtering again, lyophilizing to obtain corresponding plant extract, and mixing;
s3: placing water, a softening agent, a film forming agent and a solvent in a clean container, heating to 45-55 ℃, stirring and dissolving for later use;
s4: adding the plant extract prepared in the step S2 into a container of the step S3, adding a surfactant, and heating and homogenizing at a rotating speed of 60-90rpm until the material is uniformly dispersed;
s5: adding the humectant into the S4, stirring and homogenizing, and keeping the vacuum state and continuing to keep the temperature and stirring for 10 minutes;
s6: cooling to 35-40deg.C, adding thickener and recombinant fibroin prepared in step S1 into the mixture prepared in step S5, and stirring;
s7: adding a skin conditioner into the mixture prepared in the step S6, and uniformly stirring;
s8: cooling to 18-25deg.C, adding antiseptic, pH regulator, and chelating agent, and stirring to obtain the final product.

The method for preparing moisturizing and skin-tendering essence according to claim 6, wherein the humectant comprises at least one of saccharide isomer, sodium hyaluronate, hydrolyzed sodium hyaluronate, 1, 2-hexanediol, sodium polyglutamate, squalane, acetylglucosamine, trehalose, urea, allantoin, polyether glycerol-26, sodium lactate, sodium pyrrolidone carboxylate, xylitol, and biogel-1.
