Adding 150g of flaky bletilla striata into pure water which is 10 times of the weight of the raw materials, extracting for 3 hours at 70 ℃, filtering and removing slag while the solution is hot to obtain coarse bletilla striata filtrate, adjusting the temperature of the coarse filtrate to 60 ℃, adding 5 wt% of acid protease, preserving heat and performing enzymolysis for 1 hour, heating to 80 ℃ after the enzymolysis is finished, adding 5 wt% of high-temperature amylase, and preserving heat and performing enzymolysis for 1 hour. And after enzymolysis, adding 15 wt% of white diatomite into the enzymolysis liquid for filtration assistance to obtain a fine filtrate, adding ion exchange resin (preferably 717 resin) accounting for 15 wt% of the weight of the raw materials into the fine filtrate, stirring and adsorbing for 1 hour, filtering after adsorption, adding HCl into the filtrate, and adjusting the pH value to 6. Adjusting the pH of the filtrate to 70 deg.C, adding active carbon 8 wt% of the raw materials into the filtrate, stirring and decolorizing for 3 hr under heat preservation to obtain 1180g of rhizoma bletilla root extract product with solid content of 3.39%, and the filtrate is white to light yellow.
Example 2
Adding 150g of flaky bletilla striata into pure water which is 15 times of the weight of the raw materials, extracting for 1 hour at 90 ℃, filtering and removing slag while the solution is hot to obtain coarse bletilla striata filtrate, adjusting the temperature of the coarse filtrate to 40 ℃, adding 0.05 wt% of acid protease, preserving heat and performing enzymolysis for 6 hours, heating to 80 ℃ after the enzymolysis is finished, adding 0.05 wt% of high-temperature amylase, and preserving heat and performing enzymolysis for 6 hours. And after the enzymolysis is finished, adding 4 wt% of white diatomite into the enzymolysis liquid for filtration assistance to obtain a fine filtrate, adding ion exchange resin (preferably 717 resin) with the weight of 4 wt% of the raw material into the fine filtrate, stirring and adsorbing for 6 hours, filtering after the adsorption is finished, adding HCl into the filtrate, and adjusting the pH of the filtrate to be 6. Adjusting the pH of the filtrate to 50 deg.C, adding activated carbon 2 wt% of the raw materials into the filtrate, stirring and decolorizing for 1 hr under heat preservation to obtain 1324g of rhizoma bletilla root extract product with solid content of 3.3%, and the filtrate is white to light yellow.
Example 3
Adding 150g of flaky bletilla striata into pure water which is 12 times of the weight of the raw materials, extracting for 2 hours at 80 ℃, filtering and removing slag while the solution is hot to obtain coarse bletilla striata filtrate, adjusting the temperature of the coarse filtrate to 50 ℃, adding 2.5 wt% of acid protease, preserving heat and performing enzymolysis for 3 hours, heating to 90 ℃, adding 2.5 wt% of high-temperature amylase, preserving heat and performing enzymolysis for 3 hours. And after the enzymolysis is finished, adding 10 wt% of white diatomite into the enzymolysis liquid for assisting filtration to obtain a fine filtrate, adding ion exchange resin (preferably 717 resin) accounting for 10 wt% of the weight of the raw materials into the fine filtrate, stirring and adsorbing for 3 hours, filtering after adsorption is finished, adding HCl into the filtrate, and adjusting the pH value of the filtrate to be 6. Adjusting the pH of the filtrate to 60 deg.C, adding 6 wt% active carbon into the filtrate, stirring and decolorizing for 2 hr to obtain 1218g rhizoma bletilla extract product with solid content of 3.1%, and white to light yellow filtrate.
Example 4
Adding 150g of flaky bletilla striata into pure water 13 times of the weight of the raw materials, extracting for 2 hours at 90 ℃, finishing extraction, filtering and removing slag while the mixture is hot to obtain coarse bletilla striata filtrate, adjusting the temperature of the coarse filtrate to 55 ℃, adding 0.5 wt% of acid protease, preserving heat and performing enzymolysis for 2 hours, finishing enzymolysis, heating to 95 ℃, adding 0.5 wt% of high-temperature amylase, and preserving heat and performing enzymolysis for 2 hours. And after the enzymolysis is finished, adding 8 wt% of white diatomite into the enzymolysis liquid for filtration assistance to obtain a fine filtrate, adding ion exchange resin (preferably 717 resin) accounting for 5 wt% of the weight of the raw materials into the fine filtrate, stirring and adsorbing for 2 hours, filtering after the adsorption is finished, adding HCl into the filtrate, and adjusting the pH value of the filtrate to be 6. Adjusting the pH of the filtrate to 60 deg.C, adding 6 wt% active carbon into the filtrate, stirring and decolorizing for 1 hr to obtain 1250g rhizoma bletilla root extract product with solid content of 3.2%, and white to light yellow filtrate.
Example 5
Adding 150g of flaky bletilla striata into pure water which is 15 times of the weight of the raw materials, extracting for 3 hours at 80 ℃, filtering and removing slag while the solution is hot to obtain coarse bletilla striata filtrate, adjusting the temperature of the coarse filtrate to 50 ℃, adding 3 wt% of acid protease, preserving heat and performing enzymolysis for 3 hours, heating to 95 ℃ after the enzymolysis is finished, adding 3 wt% of high-temperature amylase, and preserving heat and performing enzymolysis for 3 hours. And after the enzymolysis is finished, adding 10 wt% of white diatomite into the enzymolysis liquid for assisting filtration to obtain a fine filtrate, adding ion exchange resin (preferably 717 resin) accounting for 10 wt% of the weight of the raw materials into the fine filtrate, stirring and adsorbing for 3 hours, filtering after adsorption is finished, adding HCl into the filtrate, and adjusting the pH value of the filtrate to be 6. Adjusting the pH of the filtrate to 70 deg.C, adding 6 wt% active carbon into the filtrate, stirring and decolorizing for 2 hr to obtain 1350g rhizoma bletilla root extract product with solid content of 3.6%, and white to light yellow filtrate.
Comparative example 1
The preparation method in comparative example 1 was the same as that of example 2 except that the ion exchange resin adsorption was not performed.
Comparative example 2
The preparation method in comparative example 2 was the same as that of example 2 except that the ion exchange resin adsorption and the decoloring treatment were not performed.
Comparative example 3
The preparation method in comparative example 3 was the same as that of example 2 except that the treatment order of acid protease and high temperature amylase was changed.
Comparative example 4
The preparation method of comparative example 4 was the same as that of example 2 except that the extraction temperature was 100 ℃.
Comparative example 5
The preparation method of comparative example 5 was the same as that of example 2 except that the extraction temperature was 60 ℃.
Stability test method: and (3) placing the sample in a constant-temperature drying oven at 60 ℃ and 80 ℃, observing whether the sample is precipitated or not, and comparing the sample with the initial sample. The test was carried out for 10 days, and the product was observed on day 5 and day 10, respectively. The results are shown in Table 1.

Immunoregulation effect of bletilla striata root extract on keratinocyte HACAT
Cell collection: when the keratinocyte plating rate reaches 60-75%, the culture solution in the culture bottle is discarded, PBS is used for cleaning twice, 1mL of pancreatin digestive cells with the concentration of 0.25% is added, the cells are digested for 3-5min at 37 ℃, 5mL of DMEM culture solution containing serum is added to stop the digestion when about 80% of the cells are in a suspension state under an inverted microscope, the cells in the culture bottle are collected in a centrifuge tube, and the cells are centrifuged for 3min at 1000 rpm/min.
Cell counting: after centrifugation, the supernatant was discarded, 5mL of PBS was added, the cells were flushed through an elbow pipette and counted in a cell counter.
Cell inoculation: after centrifugation at 1000rpm for 3min to discard PBS, the cell suspension was seeded into 12-well plates at a volume of 1mL per well. After completion of the inoculation, the cells were placed in an incubator (37 ℃ C., 5% CO)295% RH) for 24h +/-2 h.
Grouping experiments: the experiment required setting blank control group (no LPS induction), negative control group (LPS induction), sample group (LPS induction) and positive control group (LPS induction + dexamethasone 100 μ g/mL), each group set up 3 multiple wells.
Sample preparation: then diluted with cell culture medium to the corresponding concentration (0.5%, 0.1%).
Administration: according to experimental grouping, drug administration operation is carried out, a sample group is added with culture solution containing a sample, a blank control group and a negative control group are added with cell culture solution, and a positive control group is added with cell culture solution containing 100 mu g/mL dexamethasone, wherein each well is 2 mL. After the administration, the 6-well plate was placed in an incubator (37 ℃, 5% CO2, 95% RH) and incubated for 2 h. + -. 15 min.
And (3) LPS induction: after the incubation culture is finished, replacing fresh cell culture solution for a blank control group, replacing LPS culture solution containing 1 mu g/mL for the rest groups, adding cell culture solution containing LPS into a negative control group, adding culture solution containing a sample and LPS into a sample group, and adding cell culture solution containing 100 mu g/mL dexamethasone and LPS into a positive control group, wherein each well is 2 mL. After the administration, the 6-well plate was placed in an incubator (37 ℃, 5% CO2, 95% RH) and incubated for 22 h. + -. 1 h.
Collecting cell supernatant: after the incubation culture is finished, collecting cell culture supernatant into a 1.5mL centrifuge tube, and placing the centrifuge tube in an ultra-low temperature refrigerator at-80 ℃ for freezing and storing.
Detecting the content of IL-1 beta, IL-6, IL-8 and TNF-alpha: detection was performed according to the instructions of the test kit.
The bletilla striata root extract can reduce the inflammatory reaction of cells, thereby improving the immunologic function of skin and leading the skin to have better effect of resisting the environment to the skin. Specific results are shown in table 2.

Similarly, the bletilla striata root extract obtained according to method 2, method 3, method 4 and method 5 in example 1 also had a repairing effect.
The details not described in the specification of the present application belong to the common general knowledge of those skilled in the art.

"substantially" means within an acceptable error range, and a person skilled in the art can solve the technical problem within a certain error range to substantially achieve the technical effect.

Claims (10)
1. A preparation method of bletilla striata extract with repairing and whitening effects is characterized by comprising the following steps:
a) extraction: adding water 10-15 times the weight of rhizoma Bletillae into rhizoma Bletillae, extracting at 70-90 deg.C for 1-3 hr, and filtering to obtain crude filtrate;
b) enzymolysis: adding 0.05-5 wt% of acid protease into the coarse filtrate of bletilla striata, performing heat preservation enzymolysis for 1-6 hours, adding 0.05-5 wt% of high-temperature amylase after enzymolysis is finished, and performing heat preservation enzymolysis for 1-6 hours; after the enzymolysis is finished, adding a filter aid, and filtering to obtain a fine filtrate;
c) resin adsorption: adding ion exchange resin 4-15 wt% of the fine filtrate into the fine filtrate, treating for 1-6 hr, filtering, and adjusting pH of the filtrate to 5-7;
d) and (3) decoloring: adding a decoloring agent into the filtrate treated in the step c) for decoloring to obtain the bletilla extract.
2. The method of claim 1, wherein:
the bletilla striata in the step a) is flaky;
in the step b), before adding the acid protease, the temperature of the coarse filtrate is adjusted to 40-60 ℃, and the temperature is 40-60 ℃ when the acid protease is used for enzymolysis; before adding the high-temperature amylase, the temperature of the acidic protease enzymolysis liquid is adjusted to 80-95 ℃, and the temperature of the high-temperature amylase enzymolysis liquid is 80-95 ℃.
3. The method of claim 2, wherein:
in the step b), before adding the acid protease, the temperature of the coarse filtrate is adjusted to 50-60 ℃, and the temperature is 50-60 ℃ when the acid protease is used for enzymolysis; before adding the high-temperature amylase, the temperature of the acid protease zymolyte is adjusted to 85-95 ℃, and the temperature is 85-95 ℃ when the high-temperature amylase is used for enzymolysis.
4. The method of claim 2, wherein:
the filter aid in the step b) is diatomite.
5. The method of claim 2, wherein:
in the step b), the addition amount of the filter aid is 4-15 wt% of the weight of the product after the enzymolysis of the high-temperature amylase.
6. The method of claim 5, wherein:
in the step b), the addition amount of the filter aid is 8-12 wt% of the weight of the product after the enzymolysis of the high-temperature amylase.
7. The method of claim 2, wherein:
in the step c), the addition amount of the ion exchange resin is 8-12 wt% of the weight of the fine filtrate, and the pH value is adjusted by adopting a hydrochloric acid solution.
8. The method of claim 2, wherein:
in the step d), the decoloring temperature is 50-70 ℃.
9. The method of claim 2, wherein:
in the step d), the decolorizing agent is activated carbon, and the adding amount of the decolorizing agent is 2-8 wt% of the weight of the filtrate.
10. A bletilla striata extract with repairing and whitening effects, which is characterized by being prepared by the preparation method of any one of claims 1-9.
