Abstract

Background
Alopecia is pathological alopecia and physiological alopecia, and hair falling off by 100 normal people every day is normal phenomenon, which belongs to physiological alopecia. Pathologic alopecia is excessive hair loss. Alopecia refers to abnormal or excessive hair loss, the main symptoms of which are greasy hair or scorched hair, lack of gloss, difficulty in fixing pale yellow scales, or flying off grey scales. Modern medicine considers that alopecia has a very close relationship with genetic factors, hormone levels and nutritional factors. Alopecia mainly comprises the following types according to the reasons: (1) seborrheic alopecia: currently Western medicine treats male seborrheic alopecia because male alopecia is associated with in vivo dihydrotestosterone concentration. It has been shown that female seborrheic alopecia is associated with elevated levels of scalp testosterone, and not elevated levels of dihydrotestosterone, indicating that female seborrheic alopecia should be dominated by anti-androgenic medications. (2) Pathological alopecia: mainly because viruses, bacteria and high heat damage hair mother cells, the hair follicle is prevented from normal division, and hair follicle is in shock state to cause alopecia, such as acute infectious disease, long-term administration of certain medicine, etc. (3) Chemical hair loss: damage to scalp tissue, hair follicle cells by harmful chemicals results in hair loss. The effective rate of the composition for treating 480 patients suffering from seborrheic alopecia is 91.2%, the curative effect of 210 patients suffering from alopecia areata is 89.3%, and the effect is remarkable, but the medicine application principle of the composition clearly indicates that the pregnant woman is forbidden. Presumably, the adverse effects of ganoderma lucidum, ginseng, wolfberry fruit, angelica and the like on the fetus cannot be eliminated due to the too strong medicinal properties.
Chinese patent application CN105412758A discloses a traditional Chinese medicine composition for promoting hair loss recovery, comprising the following components in parts by weight: comprises 20-25 parts of gentian, 15-20 parts of radix rehmanniae, 15-20 parts of galangal, 10-15 parts of trollius chinensis, 10-15 parts of sandalwood, 10-15 parts of donkey-hide gelatin, 5-10 parts of dayflower, 4-8 parts of loquat leaf and 3-6 parts of ginkgo leaf. The cure rate of 100 patients is 72%, the effective rate is 98%, and the failure rate is 2%. Therefore, it is desirable to develop products for pregnant women to treat or prevent hair loss.

Further, the composition comprises the following preparation raw materials in parts by weight:
30 parts of red date, 50 parts of beef bones, 20 parts of kelp, 18 parts of ginger, 8 parts of cinnamon and 0.5 part of folic acid compounds.

s1: respectively crushing the red dates, the beef bones and the kelp in parts by weight, adding the crushed red dates, the beef bones and the kelp into water, then adding hydrochloric acid to adjust the pH to be acidic, heating and refluxing for extraction, and carrying out solid-liquid separation after the extraction is finished to obtain an extract A;
s2: concentrating the extract A under reduced pressure to obtain a part of solvent, adding sodium hydroxide to adjust pH to alkaline, and stirring to obtain extract B;
s3: concentrating the extract B to obtain a part of solvent, desalting with a chromatographic column with C18 as filler and water as eluent, collecting the fraction with water and ethanol mixture as eluent, concentrating, and drying to obtain extract;
s4: drying rhizoma Zingiberis recens and cortex Cinnamomi, pulverizing, mixing with the extract and folic acid compounds, and grinding into powder.

The composition according to any one of claims 1 or 2, wherein the composition further comprises at least one pharmaceutically acceptable excipient; the excipient is a diluent, binder, disintegrant, lubricant, colorant, flavoring agent, pH adjustor, buffering agent, homogenizing agent, preservative, anti-adhesive agent, glidant, acidulant, sweetener, or any combination thereof.
6. A process for preparing a composition according to any one of claim 1 to 5, comprising the steps of,
s1: respectively crushing the red dates, the beef bones and the kelp in parts by weight, adding the crushed red dates, the beef bones and the kelp into water, then adding hydrochloric acid to adjust the pH to be acidic, heating and refluxing for extraction, and carrying out solid-liquid separation after the extraction is finished to obtain an extract A;
s2: concentrating the extract A under reduced pressure to obtain a part of solvent, adding sodium hydroxide to adjust pH to alkaline, and stirring to obtain extract B;
s3: concentrating the extract B to obtain a part of solvent, desalting with a chromatographic column with C18 as filler and water as eluent, collecting the fraction with water and ethanol mixture as eluent, concentrating, and drying to obtain extract;
s4: drying and pulverizing rhizoma Zingiberis recens and cortex Cinnamomi, mixing with the extract and folic acid compounds, and grinding into powder to obtain the composition;
