Description

Traditional Chinese medicine composition for treating gastric ulcer and preparation method thereof
Technical Field
The invention relates to the technical field of medicines, in particular to a traditional Chinese medicine composition for treating gastric ulcer and a preparation method thereof.
Background
Gastric ulcer occurs in the digestive system and is a common stomach disease in clinic. It is a disease in which open ulcers appear on the inner wall of the stomach, and the incidence of the disease is high because of many pathogenic factors. Among them, helicobacter pylori infection is the most common pathogenic factor, and non-steroidal anti-inflammatory drugs, poor dietary habits, poor mood and the like are taken for a long time. The clinical symptoms of gastric ulcers are mainly seen in the digestive system, increasing patient discomfort, such as: stomach pain (aggravated after eating), abdominal distention, belching, nausea, anorexia, dyspepsia, emesis, hematochezia, and weight loss. If the clinical symptoms are serious, the breathing condition of the patient can be affected, the breathing difficulty of the patient is induced, and the life safety is threatened.
Gastric ulcer is mainly stomach mucosa tissue defect and necrotic lesion caused by stomach mucosa inflammation and helicobacter pylori infection, and the lesion can reach the mucosal muscularis deeply. The main clinical treatment mode of gastric ulcer is drug therapy, and drugs are adopted to protect gastric mucosa, resist bacteria and inhibit acid. Most of the existing medicines for treating gastric ulcer are western medicines, although the medicines have certain curative effect, the side effects are large, the recurrence rate is high after the medicines are stopped, once the state of an illness is repeated, the incidence rate of adverse reactions can be improved, severe conditions such as gastric perforation and gastrorrhagia can be caused, and adverse effects are caused on the life quality and life safety of patients. Some patients have persistent gastric ulcer or recurrent gastric ulcer, which is easy to become cancerous.
At present, a plurality of medicines can only eliminate the symptoms of surface pain without removing the root cause of the symptoms, and a plurality of patients are treated by medicines for a long time, the symptoms are only relieved, the symptoms cannot be radically eradicated, the symptoms are frequently relapsed, the side effects on the human body after long-term use are large, the symptoms are easy to relapse after treatment, and the symptoms are difficult to radically cure.

The rats are first placed in a quiet animal room to be kept in cage, the room temperature is controlled to be 22 +/-2 ℃, the natural rhythm illumination is carried out, free food and water are taken, standard feed is fed, after the rat is adapted for 1 week, formal experiments are carried out, and fasting is carried out for 24 hours before the experiment, and water is fed freely.
(2) Grouping experiments: experimental rats were randomly divided into a normal group, a model group, a paulownia leaf group, a rhubarb group, a patrinia group, a hemp oil group, a composition 1 group, a composition 2 group, a composition 3 group, and an omeprazole group.
(3) Preparing a medicament: medicaments with different doses are prepared, and the medicament dose of each group is shown in table 1, wherein the composition 1 group, the composition 2 group and the composition 3 group are prepared by the preparation method of the traditional Chinese medicine composition for treating gastric ulcer provided by the invention.
(4) Preparation of acetic acid gastric ulcer model in rat
Each group of rats was anesthetized with 10% chloral hydrate solution according to 2mL/kg standard, attached to the operating table, the thoracic and abdominal areas were exposed, the left flank and abdominal area were disinfected with iodophors, 3cm incisions were cut, the muscular layers were exposed, the muscular layers were cut open, and the stomach was pulled out of the abdominal cavity with forceps and flattened. Injecting 100% glacial acetic acid 0.02mL at the junction of antrum and body of stomach until the serosa layer of stomach has white ulcer ring of about 3mm, placing the stomach in abdominal cavity, suturing muscle layer and skin layer by layer, and sterilizing with iodophor. The rats in the normal group were subjected to a sham operation, which was performed as before, but only the abdominal cavity was opened, and after exposing the stomach, glacial acetic acid was not injected and then sutured.
(5) Administration: and starting intragastric administration 24h after the model building, wherein the normal group and the model group are infused with distilled water with the same volume, and the total administration time is 15 days.
(6) And (3) determining the ulcer index: after 2h of the last administration, 10% chloral hydrate is used for intraperitoneal injection and anesthesia, the abdomen is cut open, then the stomach is taken out, the stomach is cut along the greater curvature of the stomach, the content of the stomach is slightly washed away by normal saline, the inner side surface of the stomach wall is flattened, mucus and blood clots on the gastric mucosa are slightly wiped away by a small cotton ball, and the injury degree of the gastric mucosa is observed. The longest diameter and widest diameter of the gastric ulcer site were measured with a ruler, and the Ulcer Index (UI) was determined as longest diameter × shortest diameter × 1/4 × pi.
TABLE 1 measurement of ulcer index in rats
Figure BDA0003033504200000061
Figure BDA0003033504200000071
Note: p < 0.05, P < 0.01 compared with the model group; compared with omeprazole group, the delta P is less than 0.05.
As a result: compared with the model control group, the ulcer index of the composition 3 group is reduced (P < 0.05), the ulcer index of the composition 1 group, the ulcer index of the composition 2 group and the ulcer index of the omeprazole group are obviously reduced (P < 0.01), and the ulcer index of the composition 1 group is improved to a degree superior to that of the omeprazole group (P < 0.05) and each component is singly used.
