Abstract
The invention discloses a polypeptide that can reduce the sensitization of β-lactoglobulin in cow's milk, which is characterized in that its sequence: amino-terminal-VAGTWYSL-carboxy-terminal. The anti-allergenic effect of this peptide was determined by β-lactoglobulin-linked immunosorbent assay kit. The results show that the peptide has a good inhibitory effect on the sensitization of β-lactoglobulin, and can be further developed into an anti-allergic drug or functional food additive of β-lactoglobulin.
Description
The technical field is as follows:
the invention relates to a polypeptide capable of reducing protein sensitization, in particular to a polypeptide capable of reducing cow milk beta lactoglobulin sensitization.
Background art:
food allergy is a disease most common in infants and young children in the world today, affecting about 8% of infants and 4% of adults, while among children food allergy is most common Cow's Milk Allergy (CMA), which accounts for almost 2.5% of food allergy, and cow's milk allergy is closely linked to atopic eczema and allergic asthma. Research shows that 20% of cow milk allergy is caused by beta lactoglobulin and can cause other concomitant diseases of children, and research shows that some natural small molecules such as flavonoids can change covalent or non-covalent actions of lactoglobulin, so that the secondary structure or the tertiary structure of the beta lactoglobulin is changed, the allergic sites of the lactoglobulin are changed, and the anti-allergic effect is generated. Many small molecules even have certain toxicity or side effects, so the development of polypeptide antiallergic polypeptides which are nontoxic and harmless and can increase the nutrient absorption of human bodies has important practical significance.
The invention content is as follows:
in view of the above, in order to solve the above problems, we provide a polypeptide capable of reducing bovine milk beta lactoglobulin sensitization, the amino acid sequence of which is: the amino terminal-VAGTWYSL-carboxyl terminal can be tried to be developed into an antiallergic drug and a functional food additive of the bovine beta lactoglobulin.
Description of the drawings:
in order to make the objects, technical solutions and advantages of the present invention more apparent, the present invention will be further described in detail with reference to the accompanying drawings, in which:
FIG. 1 shows the results of the measurement of the antiallergic effect of the newly designed peptides on bovine beta lactoglobulin at a concentration of 1X 10-5mol/L, the mol ratio of the newly designed peptide to the beta lactoglobulin is 1:1,2:1,3:1,4:1,5:1 respectively, the reaction time is 1.5h, and the pH is 7.0; the pH set experiment is set to be respectively 5.0, 6.0, 7.0, 8.0 and 9.0, the reaction time is 1.5h, and the molar ratio is 3: 1; the reaction time groups are respectively set to be 0.5h, 1.0h, 1.5h, 2.0h and 2.5h, the molar ratio is 3:1, and the reaction pH is 7.0.
The specific implementation mode is as follows:
the antiallergic effect is measured by adopting a bovine beta lactoglobulin (beta-LG) enzyme-linked immunosorbent assay kit and adopting a single-factor experiment, taking out the required lath from an aluminum foil bag after the room temperature is balanced for 60min, and sealing the rest laths by using a sealing bag and putting back the laths at 4 ℃. Standard substance holes, blank holes and poplar holes are arranged, and 50 microliters of standard substances with different concentrations are added in the standard substance holes respectively. 50 microliters of sample to be tested is added into the poplar well, and 50 microliters of dilution is added into the blank well. Blank wells, standard wells and sample wells were loaded with 50 microliters of this diluent. 100 microliters of detection antibody labeled with horseradish peroxidase (HRP) was added to each of the blank wells, the standard wells and the sample wells, the reaction wells were sealed with a sealing plate film, and incubated in a 37 ℃ water bath or incubator for 60 min. Discarding the liquid, patting dry on absorbent paper, filling 350 microliters of washing liquid into each hole, standing for 1min, throwing off the washing liquid, patting dry on the absorbent paper, and repeating the plate washing for 5 times. 50. mu.l each of the A, B substrate was added to the cells and incubated at 37 ℃ for 15min in the absence of light. Stop solution was added to each well in an amount of 50. mu.l, and the OD value of each well was measured at a wavelength of 450nm within 15 min. According to the ELISA result, the best antiallergic effect is that when the molar ratio is 4:1, the antiallergic effect of the newly designed peptide is the best, and reaches 38.71%; when the pH value is 9.0, the antiallergic effect of the newly designed peptide is optimal and reaches 27.94%; when the reaction time is 1.0h, the antiallergic effect is best, and 37.72% is achieved. Table 1 shows the anti-allergic activity of each experimental group. The result shows that the newly designed peptide has good effect of reducing beta lactoglobulin sensitization, and the sequence is as follows: amino-terminal-VAGTWYSL-carboxyl-terminal.
TABLE 1 antiallergic Activity
The above description is only a preferred embodiment of the present invention and is not intended to limit the present invention, and it is apparent that those skilled in the art can make various changes and modifications to the present invention without departing from the spirit and scope of the present invention. Thus, if such modifications and variations of the present invention fall within the scope of the claims of the present invention and their equivalents, the present invention is also intended to include such modifications and variations.
