Abstract
茶树己糖激酶CsHXK3基因在调控植物生长发育和增强抗寒能力中的应用,属于生物基因工程技术领域。本发明包括茶树己糖激酶CsHXK3基因在调控植物生长发育和增强抗寒能力中的应用;一种利用茶树己糖激酶CsHXK3基因调控拟南芥生长发育和增强抗寒能力的方法。本发明通过过表达实验首次验证了CsHXK3基因的生物学功能,可以为茶树新品种选育提供基因资源。本发明提供了一个重要的并且可能具有普遍性的调控植物生长发育和抗寒能力的基因资源,为培育抗寒性的植物品种提供了优秀的候选基因。
Description
技术领域
本发明属于生物基因工程技术领域,具体涉及茶树己糖激酶CsHXK3基因在调控植物生长发育和增强抗寒能力中的应用。
背景技术
茶树[Camellia sinensis (L.) O. Kuntze]是我国重要的经济叶用型作物。其原产于我国云、贵、川等低纬度地区,形成了喜温畏寒的生理特性,冬季低温冻害和春季霜冻“倒春寒”严重威胁着茶树的生长萌发和地理分布。茶树品种的叶片大小(特大叶型、大叶型、中叶型、小叶型)暗示着其耐寒能力的强弱,通常叶片越小,抵御低温冻害的能力越强。中小叶种茶树可生存的最低温度为-14℃至-16℃,而大叶种茶树仅为-6℃。茶树的抗寒能力与茶叶产量和茶产业经济收益紧密相关。因此,研究茶树中参与低温抗性的重要基因,阐明茶树低温响应的调控机制,选育强抗寒性茶树品种,对茶产业经济发展具有重要的促进作用。
植物体内的可溶性糖,如葡萄糖和果糖(己糖),不仅作为碳和能量物质参与机体的代谢,而且作为重要的信号分子,调控植物的生长发育与逆境胁迫响应。植物己糖激酶(Hexokinase,HXK)是双功能蛋白,具有磷酸化己糖和介导糖信号的关键性作用。源库组织内的己糖,经HXK磷酸化形成己糖-6-磷酸,进而参与糖酵解、呼吸作用、合成代谢等过程,为植物的生命活动提供能量和代谢底物;此外,HXK可作为糖传感蛋白,参与糖信号转导,感知光照、养分、胁迫等信号,调控植株的基因表达和生长发育。本发明涉及的CsHXK3基因序列已在NCBI中公开(accession number: KX078245),且前期的研究表明,CsHXK3属于缺乏己糖磷酸化活性的HXKL类蛋白,其基因表达响应低温胁迫而迅速诱导,但其功能未见任何报道。
茶树为多年生木本植物,采用常规杂交、辐射诱变等育种方法,具有育种程序复杂、育种周期长、育种效率低的特征。此外,茶树转基因技术体系尚不成熟,而拟南芥是植物生物学研究领域的模式植物。因此,通过农杆菌介导的转基因技术可以在拟南芥中过量表达CsHXK3基因,进而研究CsHXK3基因在植物生长发育及低温胁迫下的生物学功能。己糖激酶CsHXK3基因的克隆及功能研究为茶树优良新品种培育提供宝贵的基因资源。
发明内容
针对上述现有技术中存在的问题,本发明的目的在于设计提供茶树己糖激酶CsHXK3基因在调控植物生长发育和增强抗寒能力中的应用。本发明通过在拟南芥中过量表达该基因进行生物学功能研究,证明所述CsHXK3基因通过介导植物体内的糖浓度信号,调控拟南芥的生长发育和增强拟南芥的低温抗性,为培育耐寒性植物新品种提供了基因资源,具有极好的潜在应用价值。

1)使植物包含CsHXK3基因;

所述的茶树己糖激酶CsHXK3基因在调控植物生长发育和增强抗寒能力中的应用,其特征在于所述方式2)采用农杆菌介导法将CsHXK3基因转入到植物中,得到CsHXK3基因过量表达的转基因植物。
一种利用茶树己糖激酶CsHXK3基因调控拟南芥生长发育和增强抗寒能力的方法,其特征在于包括以下步骤:
(1)提取茶树叶片总RNA,反转录得到cDNA,设计上游引物F和下游引物R,以茶树cDNA为模板进行PCR扩增,得到CsHXK3基因扩增产物,将扩增产物连接到具有35S启动子的植物表达载体PH7FWG2上,得到重组表达载体;
(2)将所述重组表达载体转化至农杆菌,利用转化后的农杆菌侵染拟南芥花序,获得转基因拟南芥株系。
所述的一种利用茶树己糖激酶CsHXK3基因调控拟南芥生长发育和增强抗寒能力的方法,其特征在于所述上游引物F的核苷酸序列如SEQ ID NO. 3所示,下游引物R的核苷酸序列如SEQ ID NO.4所示。
本发明通过过表达实验首次验证了CsHXK3基因的生物学功能。通过转基因技术使CsHXK3基因在拟南芥中过量表达,获得的转CsHXK3基因植株具有株型、叶片变小,地上部分鲜重降低,以及抗寒能力增强的表型。所以,茶树CsHXK3基因的克隆和功能验证可以为茶树新品种选育提供基因资源。本发明提供了一个重要的并且可能具有普遍性的调控植物生长发育和抗寒能力的基因资源,为培育抗寒性的植物品种提供了优秀的候选基因。

具体实施方式
下面结合说明书附图和具体实施例对本发明进行进一步说明,以便于更好地理解,但不用来限制本发明的范围。若无特殊说明,实施例均按照常规实验方法(可参照Sambrook J. & Russell D. W., Molecular cloning: a laboratory manual, 2001)或依据生产厂商说明书进行。
以下实施例中pENTRTM/D-TOPO为常用克隆载体,可市购;pH7FWG2为常用表达载体,可市购;拟南芥野生型品种为哥伦比亚生态型;农杆菌GV3101是常用菌株,多数分子生物学实验室均有保存。
以下实施例中的主要试剂是:pENTRTM/D-TOPO克隆载体试剂盒、LR反应试剂盒、植物总RNA提取TRIzol试剂等,购自于Invitrogen公司;高保真PrimeSTAR® HS DNAPolymerase酶、dNTP、反转录试剂盒等,购自于TaKaRa公司;质粒提试剂盒和琼脂糖凝胶回收试剂盒,购自于Axygen公司;潮霉素、SYBR green等,购自于Roche公司;琼脂糖、卡那霉素、壮观霉素、利福平等,购自Sigma公司。实施例中使用的各种其它化学试剂均为进口或国产分析纯试剂。
实施例中需要合成的引物和需要测序的载体均由上海华津生物科技有限公司提供服务完成。
实施例1:茶树CsHXK3基因的克隆和过表达载体构建
根据NCBI数据库中公布的CsHXK3序列,其核苷酸序列如SEQ ID NO. 1所示,其编码蛋白质的氨基酸序列如SEQ ID NO. 2所示,设计上下游引物F和R,以茶树叶片cDNA为模板,PCR扩增得到CsHXK3基因。将扩增出的序列连接到pENTRTM/D-TOPO载体上,通过LR反应最终连接到pH7FWG2植物表达载体上。
所用上游引物 F:5’-CACCATGGGGAAGGTAGGGTTG-3’,为SEQ ID NO. 3所示;下游引物R:5’-TAGCAACTGTACGGTATCCACATT-3’,为SEQ ID NO. 4所示。
实施例2:CsHXK3基因过表达植株的构建与筛选
将实施例1中成功构建的载体转化至农杆菌GV3101菌株中,再用浸花法转化至野生型拟南芥中,收获得到转基因拟南芥T0代的种子。pH7FWG2载体携带的筛选抗性基因为潮霉素,因此,使用潮霉素对拟南芥转基因幼苗进行阳性筛选,将具有潮霉素抗性的T1代阳性苗进行单株收种,再对T1代的种子进行潮霉素抗性筛选,鉴定分离比,选择抗性苗:无抗性苗约为3:1的株系,说明在该株系中,携带CsHXK3基因的载体序列是以单拷贝形式插入植物基因组。将这些株系中具有潮霉素抗性的T2代幼苗移出,再进行单株收种,再进行潮霉素抗性筛选,如果没有分离,说明该转基因株系为纯合体,该T3代纯合体可以用于后续繁种和生理实验。
筛选获得过表达株系OE-1、OE-2和OE-3。提取野生型、OE-1、OE-2和OE-3的总RNA,反转录合成cDNA,利用qRT-PCR方法检测过表达株系OE-1、OE-2和OE-3中CsHXK3的表达水平,结果见图1,可以看出CsHXK3基因均高表达于OE-1、OE-2和OE-3中,且表达量依次增多。
实施例3:过表达CsHXK3植物的生长发育表型
将野生型、OE-1、OE-2和OE-3拟南芥种子播种于1.5%葡萄糖和6.0%葡萄糖以及1.5%果糖和6.0%果糖的1/2 MS固体培养基上,种子萌芽生长状态见图2A、B、C和D。野生型种子在含1.5%和6.0%葡萄糖的培养基上,均能健康萌展,存活率无显著性差异(图2E),而三个过表达株系在含6.0%葡萄糖的培养基上的种子存活率显著性低于1.5%葡萄糖(图2E)。同样地,相较于野生型,三个过表达株系在含6.0%果糖的培养基上的种子存活率显著低于1.5%果糖(图2F)。结果表明,CsHXK3能感知糖浓度信号而参与调控植物种子的生长萌发。
再将野生型、OE-1、OE-2和OE-3拟南芥种子播种至育苗块中生长,培养至23天苗龄时拍照记录,见图3A;并进行了地上部分鲜重称量(每2株作为一个生物学重复,共8个生物学重复),见图3B。结果发现,三个过表达植株相较野生型均表现出叶片面积和地上部分鲜重降低的表型,其中过表达量高的株系OE-2和OE-3达到显著性减小。
实施例4:过表达CsHXK3植物的耐低温能力
将野生型、OE-1、OE-2和OE-3拟南芥种子播种至育苗块中生长,每个育苗块种植2株苗子。常温正常条件下培养21天,生长状态见图4A;随后进行4℃低温处理6小时,发现过表达株系与野生型有着显著的低温表型差异,即野生型表现出叶片完全萎蔫和坍塌,过表达株系则叶片坚挺,如图4B。
检测植株相对电导率,发现在常温条件下及低温4℃处理3天、0℃处理8小时后,3个过表达株系的相对电导率均显著性低于野生型,如图4C,表明植物内过表达CsHXK3基因有利于细胞膜的稳定。
常温条件下,OE-2和OE-3株系内的葡萄糖和果糖含量均显著性高于野生型,如图4E和F。经低温4℃处理7天后,3个过表达株系内的可溶性总糖、葡萄糖、果糖含量相较野生型出现不同程度的增加,其中OE-2株系内的糖含量均达到显著性上升,见图4D、E和F。因此,低温胁迫下,提高CsHXK3基因的表达量能增加植物细胞内可溶性糖组分的含量。
综合植株表型以及生理指标变化,证明了CsHXK3基因通过介导植物组织内的糖浓度信号正向调控植物的抗寒能力。需要注意的是,上文中描述的仅是本发明的若干个具体实施方案。显然,本发明不限于以上实施例,还可在本发明基础上做出一些改动。因此,本领域技术人员在不偏离本发明精神的基础上所做出的所有改动,均应认为是本发明的保护范围。

Claims (5)
1.茶树己糖激酶CsHXK3基因在调控植物生长发育和增强抗寒能力中的应用,所述茶树己糖激酶CsHXK3基因的核苷酸序列如SEQ ID NO. 1所示,茶树己糖激酶CsHXK3基因编码蛋白质的氨基酸序列如SEQ ID NO. 2所示,所述调控植物生长发育为调控植物株型、叶片变小,地上部分鲜重降低。
2.如权利要求1所述的茶树己糖激酶CsHXK3基因在调控植物生长发育和增强抗寒能力中的应用,其特征在于所述植物包括木本植物和草本植物。
3.如权利要求1所述的茶树己糖激酶CsHXK3基因在调控植物生长发育和增强抗寒能力中的应用,其特征在于包括以下方式:
1)使植物包含CsHXK3基因;
或2)使植物过量表达CsHXK3基因。
4.如权利要求3所述的茶树己糖激酶CsHXK3基因在调控植物生长发育和增强抗寒能力中的应用,其特征在于所述方式2)采用农杆菌介导法将CsHXK3基因转入到植物中,得到CsHXK3基因过量表达的转基因植物。
5.一种利用茶树己糖激酶CsHXK3基因调控拟南芥生长发育和增强抗寒能力的方法,其特征在于包括以下步骤:
(1)提取茶树叶片总RNA,反转录得到cDNA,设计上游引物F和下游引物R,所述上游引物F的核苷酸序列如SEQ ID NO. 3所示,下游引物R的核苷酸序列如SEQ ID NO.4所示,以茶树cDNA为模板进行PCR扩增,得到CsHXK3基因扩增产物,将扩增产物连接到具有35S启动子的植物表达载体PH7FWG2上,得到重组表达载体;
(2)将所述重组表达载体转化至农杆菌,利用转化后的农杆菌侵染拟南芥花序,获得转基因拟南芥株系。
