Abstract
The invention discloses Lactobacillus plantarum, the strain name of which is Lactobacillus plantarum subsp.plantarum FLPL135, the thallus is short rod-shaped gram-positive bacteria, the bacterial colony is milky white, shows smooth and neat, is stored in Guangdong province microbial strain storage center, and the storage number is GDMCC No: 61028. the invention belongs to the technical field of microorganisms, and provides lactobacillus plantarum GDMCC No: 61028 has strong antioxidant effect, can remarkably increase collagen content in aged skin, and has remarkable inhibiting effect on acne bacillus.
Description
Technical Field
The invention belongs to the technical field of microorganisms, and relates to a novel Lactobacillus plantarum subsp.
Background
The lactobacillus plantarum belongs to one of lactic acid bacteria, is gram-positive, does not produce spores, is anaerobic or facultative anaerobic, is in a straight rod shape, is in a single, paired or chain-forming shape, belongs to homofermentation and chemotropy heterogeneous lactic acid bacteria, can ferment gluconate or pentose, has lactic acid as the most fermentation end product, reaches more than 85 percent, and can produce special lactobacillus in the propagation process. The lactobacillus plantarum has good adaptability and can live in various environments, different strains have different life habits, and the lactobacillus plantarum is the only microorganism which can live in various ecological environments. Comparative genomics analysis shows that the lactobacillus plantarum is in a nomadic life style in nature. The adaptability of the bacteria determines the wide distribution of the bacteria, including intestinal tracts of mammals, saliva, breast milk, fermented products (pickles, wine, traditional fermented dairy products) and the like.
The skin is the largest organ of the human body and is also an important organ in which the body and the outside world are in direct contact. As the first line of defense of immune response, it is mainly used to protect the body from various physical, chemical and pathogenic biological damages. However, the skin is disturbed by factors such as external environment, self-aging and the like, and the skin morphology can be obviously changed after the skin is in middle age, which is characterized by skin thinning, elasticity reduction, wrinkle appearance, dryness, roughness and the like. Skin aging is associated with oxygen free radical damage, hypoimmunity, decreased telomerase activity, inflammatory infiltration, etc. Usually, the beauty-loved people areThere is a great risk in delaying skin aging, for example, in treating skin aging by surgical methods such as botulinum toxin injection, collagen subcutaneous injection, and skin tightening. However, the use of probiotics can perfectly avoid the defects and promote human health while beautifying. Lactobacillus plantarum is reported to have antioxidant capacity in its thallus and metabolites. Such as Lactobacillus plantarum C88, has high DPPH and hydroxyl radical scavenging ability, and can be used for treating H2O2Has strong tolerance capability, and can remarkably improve the activity of superoxide dismutase (SOD) in serum, improve the activity of glutathione peroxidase (GSH-PX) in liver and reduce the content of Malondialdehyde (MDA) after intervening in a D-galactose-induced aged mouse. Lactobacillus plantarum MA2 tolerates 2.0mM H2O2Has strong reducibility and lipid peroxidation inhibition capability, and the fermentation supernatant and the cell homogenate have glutathione peroxidase activity and superoxide dismutase activity. Lactobacillus plantarum metabolites, such as Exopolysaccharides (EPS), also have a corresponding free radical scavenging capacity. The lactobacillus plantarum is used as a natural antioxidant, and the strain resource of the lactobacillus plantarum needs to be further mined and expanded.
Disclosure of Invention
The invention identifies a new lactobacillus plantarum FLPL135 with the preservation number GDMCC No. 61028, which is an anaerobic gram-positive bacterium without producing spores, the bacterium body is in a long rod shape, and is in a white, round and smooth colony in an MRS agar plate culture medium. The lactobacillus plantarum GDMCC No: 61028 can be grown in MRS broth and are gram positive bacteria.

Detailed Description
The present invention is further explained below with reference to examples, but the examples are only for illustrating the present invention, and the application range of the present invention is not limited in any way by these examples.
Example 1: lactobacillus plantarum GDMCC No: 61028 isolation and identification
Separating from pickle, crushing, diluting by gradient 10, 100, 1000 and 10000, taking 100 mu L of the diluted product, coating the 100 mu L of the diluted product on an MRS solid culture medium containing 1% of vancomycin, carrying out inversion culture at 37 ℃ for 24h, picking single colony, scribing on the MRS solid culture medium containing 1% of vancomycin, carrying out inversion culture at 37 ℃ for 24h, picking single colony, carrying out colony PCR, and carrying out sequencing comparison on PCR results.
The 16S rRNA base sequence of the isolated strain was analyzed and sequenced, and the sequence was as follows:
according to the identification results, the separated strain is confirmed to belong to Lactobacillus plantarum, Lactobacillus plantarum subsp. 61028.
Example 2: effect of Lactobacillus plantarum GDMCC61028 on moisture content of aged mouse skin
Lactobacillus plantarum GDMCC No: 61028 can significantly maintain the moisture content of the skin of an aging mouse. The mice of 17 months were gavaged 10 daily9CFU/mL Lactobacillus plantarum GDMCC No: 61028 the fermented milk of 100 μ L, after 3 months, that is, 20 months, the skin moisture content was measured by a skin moisture tester, and found to be 22.95%, andthere was essentially no significant difference in skin moisture content of 23.39% in young 4-month mice, whereas the moisture content was significantly increased compared to aged 20-month mice (20.69%), and the results are shown in fig. 1.
Example 3: effect of Lactobacillus plantarum GDMCC61028 on collagen content of aged mouse skin
Lactobacillus plantarum GDMCC No: 61028 the collagen content of the skin of aged mice is significantly maintained. The mice of 17 months were gavaged 10 daily9CFU/mL Lactobacillus plantarum GDMCC No: 61028, 100 μ L of fermented milk, and after 3 months, collagen staining was performed on skin samples, and 20-month mice were used as controls, as shown in fig. 2 and 3. FIG. 3 shows that the content of collagen is very significantly increased after the administration of Lactobacillus plantarum.
Example 4: lactobacillus plantarum GDMCC No: 61028 Effect on acne bacilli
Lactobacillus plantarum GDMCC No: 61028 inhibit the growth of acne bacilli. Selection of propionibacterium acnes ATCC No: 6919 indicator bacteria, adjusting the concentration of the bacterial suspension by turbidimetry to 1 × 107cfu/mL. Add 1mL of bacterial suspension to the solid medium and spread well with a sterile triangular glass spatula. The Oxford cup is clamped by a pair of tweezers and is placed in the solid culture medium coated with the bacterial suspension, and the Oxford cup is lightly pressed to be in good contact with the culture medium. 100 μ L of Lactobacillus plantarum GDMCC No: 61028 fermented milk and MRS medium supernatant, 10% skim milk and MRS medium as blank control. Putting the culture dish into an anaerobic incubator, culturing for 48h at 37 ℃, and finally measuring the diameter of the bacteriostatic transparent ring by using a vernier caliper. The results show that whether the lactobacillus plantarum GDMCC No: 61028 fermented milk and MRS culture supernatant can effectively inhibit the growth of acne bacillus, and there is no significant difference between them.
The foregoing is a more detailed description of the invention in connection with specific preferred embodiments and it is not intended that the invention be limited to these specific details. For those skilled in the art to which the invention pertains, several simple deductions or substitutions can be made without departing from the spirit of the invention, and all shall be considered as belonging to the protection scope of the invention.
