背景技术
皮肤主要由三层组成,即从最表层开始,表皮、真皮和皮下。
特别地,表皮由角质形成细胞(大多数)、黑素细胞(涉及皮肤色素沉着)和朗格汉斯细胞组成。它的功能是保护人体免受外部环境影响并确保其完整性,特别是减慢微生物或化学物质的渗透,并防止皮肤中所包含的水的蒸发。

发明简述
申请人成功开发了一种制备富含多酚并且基本上不含单糖的植物提取物的方法。事实上,申请人已经证明,通过用酿酒酵母贝氏变种(Saccharomyces cerevisiaevar.bayanus)类型的酵母对所述提取物进行发酵步骤,可以去除植物提取物中的单糖并增加多酚含量。
申请人进一步证明,通过快速和完全消耗植物提取物中所含的单糖,这种品种酵母的特定用途可以获得最佳的发酵动力学。
因此,本发明涉及一种制备富含多酚和/或基本不含单糖的植物提取物的方法,该方法包括用属于酿酒酵母贝氏变种品种的酵母发酵植物提取物的步骤。
本发明还涉及可通过上述方法获得的植物提取物。该植物提取物的特征在于基本上不含单糖,但也包含高含量的多酚,例如黄酮类。

发明详述
根据第一方面,本发明涉及一种制备富含多酚和/或基本不含单糖的植物提取物的方法,该方法包括用属于酿酒酵母贝氏变种品种的酵母发酵植物提取物的步骤。
多酚是包含至少两个酚基的分子,即包含至少两个带有羟基官能团的苯环。多酚的具体实例包括羟基苯甲酸、羟基肉桂酸和香豆素、萘醌、芪类或黄酮类。黄酮类是植物界中最常见的多酚,特别包括黄酮、黄酮醇和二氢黄酮醇、黄烷酮、橙酮、查耳酮和二氢查耳酮、黄烷醇、黄烷二醇、花青素及其糖杂体(heterosides)。多酚因其抗氧化作用而闻名,这使其成为特别有利的抗老化活性成分。另外,多酚中大量羟基的存在使皮肤中的水被捕获,从而减慢了其蒸发。因此,多酚还具有非常有利的保湿作用。

在本发明的上下文中,“基本上不含单糖”的提取物是包含小于20.00重量%,特别是小于10.00重量%,优选地小于5.00重量%,甚至更优选小于1.00重量%的提取物。
在一个具体实施方案中,根据本发明的提取物完全不含单糖,即它包含0重量%的单糖。
本发明的方法包括用属于酿酒酵母贝氏变种品种的酵母发酵植物提取物的步骤。该发酵步骤减少或去除了植物提取物中所含的单糖,还富集了植物提取物中的多酚含量。
申请人证实(参见下面的实施例)使用酿酒酵母贝氏变种品种的酵母为植物提取物提供了优化的发酵动力学,而且最重要的是比使用其他酵母品种获得的发酵动力学要好得多。酿酒酵母贝氏变种酵母特别地将植物提取物的发酵时间,即消耗提取物中的糖所需的时间减少到小于24小时。因此,这些酵母快速且有效地消耗了植物提取物中所含的单糖。
酿酒酵母贝氏变种品种的酵母是已知的,并且特别用于酿酒学领域。本领域技术人员知道如何获得这种酵母以实施本发明。在一个具体实施方案中,酵母酿酒酵母贝氏变种选自以IOC Fizz+、IOC Divine、IOC 18-2007或其混合物销售的酵母菌株。所有这些菌株均由Institut de la Vigne et du Vin列出,并且对于技术人员而言是容易获得的。
IOC Fizz+酵母菌株是分别以保藏号LYCC 6022:LA CLAIRE CGC62和LYCC 6039:LA CLAIRE SP665保藏于巴斯德研究所的两种酵母的混合物。
IOC Divine菌株对应于以保藏号LYCC 7000保藏于巴斯德研究所的菌株,且菌株18-2007对应于以保藏号CNCM I-5320保藏于巴斯德研究所的菌株。
根据本发明的酵母菌株可以是干燥形式、液体形式或酵母乳形式。
在一个具体实施方案中,酿酒酵母贝氏变种菌株为干燥形式。
典型地,发酵步骤使用相对于待发酵的溶液为1-5质量%,特别是相对于待发酵的溶液为2-3质量%,更特别是相对于待发酵的溶液为2质量%的酿酒酵母贝氏变种进行。
发酵步骤的持续时间必须足以使植物提取物中的单糖含量小于20重量%,特别是10重量%,优选小于5重量%,甚至更优选小于1重量%。典型地,发酵步骤进行1-72小时,特别是12-60小时,优选18-48小时。
在一个具体实施方案中,发酵步骤可以借助于发酵剂(leaven)进行。该发酵剂由用酿酒酵母贝氏变种菌株发酵浓缩的植物提取物(所述植物提取物与本发明的方法中使用的植物提取物相同)来制备。典型地,将用于发酵剂的植物提取物浓缩,直到获得约5-30%的干物质含量。
根据该实施方案,根据本发明方法的发酵步骤通过将该发酵剂添加到植物提取物来进行。发酵剂的使用使得可以启动发酵,并因此改进根据本发明方法的发酵步骤的动力学。
典型地,通过在室温下(或在37℃下)用酿酒酵母贝氏变种将在水中浓缩至5-50%,特别是15-30%干物质的植物提取物发酵约2小时来制备发酵剂。将1-5重量%,优选2重量%的发酵剂添加到用于根据本发明方法的发酵步骤的植物提取物中。
根据本发明的方法使用的植物提取物可以通过本领域技术人员已知的任何提取方法获得。
典型地,制备植物提取物的方法包括以下步骤:
a)用至少一种醇溶剂和/或水提取植物;
b)例如通过筛分过滤a)中获得的混合物,以去除植物残余物,
c)任选地,使步骤b)中获得的混合物脱色;和
d)去除溶剂并浓缩提取物。
因此,根据实施方案,本发明涉及一种制备富含多酚和/或基本上不含单糖的植物提取物的方法,包括以下步骤:
a)用至少一种醇溶剂和/或水提取植物;
b)例如通过筛分过滤a)中获得的混合物,以去除植物残余物;
c)任选地,使步骤b)中获得的混合物脱色;
d)例如通过蒸发去除溶剂,并浓缩提取物;和
e)用属于酿酒酵母贝氏变种品种的酵母发酵由步骤d)获得的提取物。
步骤a)中使用的“植物”可以是属于植物界的任何生物。该植物是木本陆生植物或草本植物。优选地,该植物是显花植物或“开花植物”。例如,它可以选自百合(Liliumcandidum)、勿忘我(Myosotis sylvatica)、接骨木(Sambucus sp.)、茶花(Camellia sp.)、报春花(Primula sp.)、晚香玉(Polianthes tuberosa)、素馨(Jasminum grandiflorum)、车前草(Plantago lanceolata)、榅桲(Cydonia oblonga)、杏(Prunus armeniaca)或鸡蛋花(Plumeria sp.)。
在步骤a)中提取的植物部分可以是完整植物,也可以是其仅一部分,例如花、叶、茎或根。在优选实施方案中,提取物是花提取物。
步骤a)中使用的植物可以为“新鲜”形式,即在收获后48小时内,特别是在24小时内,甚至更特别地在12小时内使用。
步骤a)中使用的植物也可以是干燥形式。在这种情况下,新鲜植物要在温和的条件下(在室温下避光或在低于35℃的温度下在通风干燥机中)脱水。优选将植物干燥至干物质含量大于80%,优选大于85%。
步骤a)可以进一步包括研磨植物,典型地是其花和/或叶,以获得小于5cm,优选小于2cm的粒径。
提取步骤用至少一种包含醇溶剂和/或水的提取溶剂进行。提取溶剂取决于所选的植物。典型的植物/溶剂比为1-10(重量/重量)。
提取溶剂是水和/或醇溶剂。在具体实施方案中,醇溶剂是醇,例如乙醇,特别是96°乙醇。
提取溶剂可包含0-100体积%的水和0-100体积%的醇溶剂。在具体实施方案中,提取溶剂包含100%的水,在另一个实施方案中,其包含100%的乙醇。提取溶剂还可包含50体积%的水和50体积%的乙醇。
在具体实施方案中,水可以是去离子水。
提取步骤持续至少1小时,优选至少2小时,特别是至少3小时,并且可以重复一次或两次。
如上所述,提取过程可包括其中将来自步骤b)的混合物脱色的步骤。该步骤的目的是去除提取物中存在的色素,例如叶绿素和叶黄素。本领域技术人员熟悉几种去除这些色素的方法。例如,脱色可以通过使混合物与活性炭接触来进行。脱色后,将混合物过滤以去除碳残余物。
典型地,步骤d)涉及将提取物浓缩至干物质与水之比为5-30%,优选15-30%。
来自步骤d)的植物提取物,即浓缩的植物提取物,可以用于制备上述发酵剂。
上述方法还可以在步骤d)和e)之间包括步骤d'),其中将包含浓缩提取物的混合物精细过滤(典型地以2μm)以去除细颗粒和残留细菌。
方法可以进一步包括步骤f),其中将发酵提取物过滤,然后稀释以获得包含1-10重量%干物质的提取物。该提取物可以是澄清且稳定的水溶液形式。
在具体实施方案中,根据本发明的方法包括以下步骤:
a)在60-75℃的温度下,用乙醇或水或两者的混合物将研磨至细度小于2cm的花粉提取两次,至少2小时;
b)将混合物过滤至4μm;
c)通过与活性炭接触1小时使所得混合物进行脱色,然后进行低至1μm的微滤以去除碳残余物;
d)通过蒸发和浓缩提取物去除提取溶剂,直至水中的干物质含量达到约5-15%(重量/重量);
d’)以0.2μm过滤混合物;
e)在37℃下用2重量%的发酵剂发酵混合物24-48小时,所述发酵剂通过将步骤d)获得的部分提取物浓缩至在水中30%干物质并添加10重量%的干燥酿酒酵母贝氏变种酵母,在37℃第一次发酵2小时而获得;
f)然后,将发酵提取物以1μm过滤,然后稀释以获得包含1-10重量%干物质的提取物,向其中添加基于二醇和/或其他物质的防腐剂系统。
根据另一方面,本发明涉及可通过上述方法获得的植物提取物,即富含多酚和/或基本不含单糖的植物提取物。
因此,在这种情况下,本发明的主题是植物提取物,其特征在于它包含:
-5.00-100.00重量%的多酚,特别是5.00-85.00重量%的多酚,更特别是55.00-85.00重量%的多酚;和/或
-小于20.00重量%,特别是小于10.00重量%,优选小于5.00重量%,甚至更优选小于1.00重量%的单糖。
根据本发明的植物提取物用于美容目的,以滋润人皮肤或防止其干燥。得益于多酚的抗氧化作用,该提取物还可用于对抗老化迹象,例如皱纹和细纹,由于表皮和/或真皮中的组织损失而导致的紧致和弹性下降;由于微循环减少和表皮细胞更新减慢而导致的光泽损失,与黑色素合成异常相关的色素沉着斑点的出现,或由于角质层屏障功能降低和表皮更新减慢而导致的皮肤干燥。
事实上,抗氧化剂以其捕获自由基的能力而闻名,自由基是促进皮肤老化的主要因素之一。
本发明还涉及包含如上所述的植物提取物的化妆品组合物。
优选地,所述提取物以组合物总重量的0.001-90%的比例,特别是以组合物总重量的0.01-10%的比例,优选以0.1-10%存在于化妆品或皮肤病学组合物中。所述化妆品或皮肤病学组合物可以特别适合于局部应用。
有利地,所述化妆品或皮肤病学组合物可以是粉末、乳液、微乳液、纳米乳液、悬浮液、溶液、洗剂、霜剂、水或水醇凝胶、泡沫、精华液(serum)、用于气雾剂的溶液或分散体,或脂质囊泡的分散体的形式。
在乳液的情况下,它可以是油包水或水包油乳液。
根据本发明的化妆品或皮肤病学组合物还可包含根据各种成分和施用形式选择的溶剂。
实例包括水(优选为软化水)、醇(如乙醇)或二甘醇醚(乙氧基二甘醇单甲基醚或二甘醇单甲基醚)如。
除了根据本发明的提取物外,所述化妆品组合物还可包含至少一种本领域常用的添加剂,例如至少一种选自润肤剂或湿润剂、胶凝剂和/或增稠剂、表面活性剂、油、活性剂、染料、防腐剂、抗氧化剂、活性剂、有机或无机粉末、防晒剂和香料的化合物。
特别地,所述组合物可以包含:
-一种或多种润肤剂或湿润剂,其可以选自例如甘油、乙二醇、水溶性硅氧烷(例如以名称KF6011(Shin Etsu)销售的那些)和水溶性荷荷巴油(诸如以名称Resplanta Jojoba(Res Pharma)销售的那些)。
相对于组合物的总重量,所述润肤剂或湿润剂可以以0-30重量%,优选2-10重量%的含量存在于组合物中。
-用于水相的一种或多种胶凝剂和/或增稠剂,例如选自纤维素衍生物、植物来源的胶(瓜尔豆、刺槐豆、藻酸盐、角叉菜胶、果胶),微生物来源的胶(黄原胶)、粘土(锂蒙脱石)、由INCI名称“丙烯酰二甲基牛磺酸铵/vp共聚物”和“丙烯酰基二甲基牛磺酸铵/山嵛醇聚醚-25甲基丙烯酸酯共聚物”标识的材料(例如,以名称Aristoflex AVC和HMB由Clariant销售的那些)。
相对于组合物的总重量,所述胶凝剂和/或增稠剂可以以0-10重量%的含量存在于组合物中。
-一种或多种表面活性剂,优选非离子表面活性剂,其相对于组合物的总重量,以0-8重量%,优选0.5-3重量%的含量存在。
-一种或多种挥发性或非挥发性的室温液体脂肪(通常称为油);直链、环状或支链的烃或聚硅氧烷,例如异十二烷、环戊二甲基硅氧烷、二甲基聚硅氧烷、异壬酸异壬酯或季戊四醇四异硬脂酸酯,相对于组合物的总重量,优选其量为0-约10重量%,优选0.5-5重量%。
-一种或多种具有生物活性的天然或合成来源的活性剂,例如选自维生素、痕量元素、尿囊素、植物蛋白、植物提取物、保湿剂、抗老化剂、抗氧化剂、光泽增强剂及其混合物。特别地,所述活性剂选自香荚兰(Vanilla planifolia)果水、烟酰胺、透明质酸及其衍生物、酵母提取物及其混合物。
-一种或多种水溶性染料,例如丽春红二钠盐、茜素绿二钠盐、喹啉黄、苋属植物三钠盐、酒石黄二钠盐、若丹明一钠盐、品红或叶黄素二钠盐,相对于组合物的总重量,优选其量为0-约2重量%。
通常用于化妆品中的其他添加剂也可以存在于根据本发明的组合物中,例如在技术领域中众所周知的防腐剂、抗氧化剂或香料。
技术人员能够从所有这些可能的添加剂中选择待添加到组合物中的那些添加剂的性质和量,以使其保留其所有性能。
最后,根据另一方面,本发明还涉及酿酒酵母贝氏变种品种的酵母用于去除植物提取物中的单糖的用途。
现在将通过以下非限制性实施例说明本发明。

具体实施方式
实施例1:根据本发明的百合的提取方法:
1)将新鲜的百合花在60℃下用96°乙醇提取两次,最少2小时,植物/溶剂之比为1-10(重量/重量);
2)将混合物以100μm筛分以去除植物残余物,然后静置过夜,并且再次过滤至4μm;
3)通过与活性炭接触1小时使所得混合物进行脱色以去除色素(例如叶绿素和叶黄素);
4)通过微滤(至1μm)将脱色的混合物与碳残余物分离;
5)通过蒸发去除提取乙醇。浓缩提取物直至水中的干物质含量达到约5%(重量/重量);
6)以0.2μm过滤混合物以去除细颗粒和残留细菌;
7)为了制备用于发酵提取物的发酵剂,将步骤6)之前获得的一部分提取物浓缩至在水中30%干物质,然后添加10重量%的干酵母酿酒酵母贝氏变种。将混合物在37℃下搅拌2小时。该混合物构成发酵剂;
8)将2重量%的发酵剂添加至来自步骤6)的混合物;
9)在37℃下搅拌并在黑暗中发酵24-48小时后,将发酵提取物静置过夜以去除形成的气泡。
10)然后将发酵提取物以1μm过滤,然后稀释以获得含有1%干物质的提取物。添加0.7%的苯氧基乙醇或20%的1,3-丙二醇用于更好的保存,并将整体以0.2μm过滤。
百合花提取物的含量在下表1中给出。

如上所述,根据本发明的方法完全从百合花提取物中去除了单糖。根据本发明的方法极大提高了提取物的多酚含量。
实施例2:酿酒酵母贝氏变种Fizz+、Divine和18-2007的干酵母的发酵动力学允许花提取物的优化发酵。
进行了发酵动力学研究,以确定哪种酵母更有效。
通常,花提取物的发酵可分为三个连续步骤(M.H.Akin,“Evolution du pHpendant la fermentation alcoolique dede raisins:modélisation etinterprétation métabolique[“Change in pH during alcoholic fermentation ofgrape musts:modelling and metabolic interpretation”](2008)136):
-步骤1:将蔗糖水解为果糖和葡萄糖(持续15-30分钟)
-步骤2:开始消耗葡萄糖(持续高达5小时)
-步骤3:开始消耗果糖和消耗其他单糖(因菌株而异)
为了选择允许优化发酵的酵母菌株,我们用各种商业酵母菌株评估了来自百合提取物的第3步的总糖消耗率。
通过总糖的相对含量的线性回归获得斜率。斜率越低,动力学越大(图1)。
表2:不同酵母菌株的总糖消耗率的比较。
因此,图1和表2的结果表明,Fizz+、Divine、18-2007和参考(ref.)菌株每小时分别消耗百合花提取物中总糖的6.8%、5.3%、4.7%和2.1%。
然后,该比较使得可以按照以下方式根据其糖消耗速度对4个菌株进行分类:Fizz+>Divine>18-2007>参考非贝氏。
不属于酿酒酵母贝氏变种品种的参考菌株消耗总糖比属于酿酒酵母贝氏变种品种的Fizz+、Divine和18-2007菌株慢两到三倍。
酿酒酵母贝氏变种酵母使植物提取物具有优化的发酵动力学。
实施例3:化妆品组合物
对于本领域技术人员,可以以传统方式制备以下组合物。以下所示的量以重量百分比表示。大写字母中的成分根据INCI标识进行标识。

Claims (15)
1.一种制备富含多酚和/或基本不含单糖的植物提取物的方法,该方法包括用属于酿酒酵母贝氏变种品种的酵母发酵植物提取物的步骤。
2.根据权利要求1的方法,在发酵步骤之前,其包括通过以下步骤制备植物提取物:
a)用至少一种醇溶剂和/或水提取至少一种植物;
b)过滤a)中获得的混合物以去除植物残余物,
c)任选地,使步骤b)中获得的混合物脱色;和
d)去除溶剂并浓缩所获得的提取物。
3.根据权利要求1或2的方法,其中所述酿酒酵母贝氏变种品种的酵母选自下组:菌株IOC Fizz+、IOC Divine、IOC 18-2007及其混合物的酵母。
4.根据权利要求3的方法,其中所述酿酒酵母贝氏变种品种的酵母是IOC Fizz+菌株的酵母。
5.根据权利要求3的方法,其中所述酿酒酵母贝氏变种品种的酵母是IOC Divine菌株的酵母。
6.根据权利要求3的方法,其中所述酿酒酵母贝氏变种品种的酵母是IOC 18-2007菌株的酵母。
7.根据权利要求1-6中任一项的方法,其中所述植物提取物是植物的提取物,优选地是开花植物的提取物,更优选地是花的提取物。
8.根据权利要求7的方法,其中所述花是百合(Lilium candidum)、勿忘我(Myosotissylvatica)、接骨木(Sambucus sp.)、茶花(Camellia sp.)、报春花(Primula sp.)、晚香玉(Polianthes tuberosa)、素馨(Jasminum grandiflorum)、车前草(Plantagolanceolata)、榅桲(Cydonia oblonga)、杏(Prunus armeniaca)或鸡蛋花(Plumeria sp.)的花。
9.根据权利要求1-8中任一项的方法,其中所述发酵步骤通过用酿酒酵母贝氏变种菌株发酵浓缩的植物提取物制备的发酵剂来进行。
10.可通过权利要求1-9中任一项的方法获得的植物提取物。
11.植物提取物,其特征在于其包含:
-5.00-100.00重量%的多酚,特别是5.00-85.00重量%的多酚,更特别是55.00-85.00重量%的多酚;和/或
-小于20.00重量%,特别是小于10.00重量%,优选小于5.00重量%,甚至更优选小于1.00重量%的单糖。
12.根据权利要求11的植物提取物,其中所述提取物包含:
-5.00-100.00重量%的多酚,特别是5-85重量%的多酚,更特别是55-85重量%的多酚;和/或
-小于20.00重量%,特别是小于10.00重量%,优选小于5.00重量%,甚至更优选小于1.00重量%的单糖。
13.根据权利要求11或12的植物提取物,其中所述提取物不含单糖。
14.化妆品组合物,其包含根据权利要求10-13中任一项的植物提取物。
15.酿酒酵母贝氏变种品种的酵母用于去除植物提取物中的单糖的用途。
