本发明公开了一种二裂酵母发酵产物溶胞物的制备方法,涉及微生物化妆品原料相关生产技术领域,包括如下步骤:将质量分数1%‑4%的大豆和质量分数1%‑2%的焦香麦芽于纯化水中煮沸2小时,冷却至室温后,使用800目筛子过滤,然后加入质量分数5%‑10%的葡萄糖、0.1%‑1%的酵母浸粉、0.1%‑0.5%的磷酸氢二钾,并于115℃灭菌20min;冷却至室温,接种质量分数1.5%‑3.5%的二裂酵母菌液,于37℃发酵48小时;达到发酵终点后,发酵液进行离心收集菌体,加入0.2倍‑2倍无菌超纯水复溶;加入质量分数0.1%‑1.0%的溶菌酶;加入质量分数5%‑20%的海藻糖作为保护剂,并在80℃‑100℃进行热裂解;冷却后,常温保藏。从而能够解决二裂酵母发酵产物溶胞物生产过程中清除自由基物质活性降低的问题。

技术领域
本发明涉及微生物化妆品原料相关生产技术领域,尤其是一种二裂酵母发酵产物溶胞物的制备方法。
背景技术
二裂酵母是人体肠道菌群之一,肠道黏附性强,定植于肠道的能力强,并具有激活巨噬细胞、刺激免疫因子释放、维持肠道菌群平衡、抗肿瘤、促进微量元素吸收、控制内毒素的产生等重要的生理保健功能,因此二裂酵母的应用价值极高。二裂酵母发酵产物溶胞物是一种应用广泛的天然活性化妆品原料,该原料经菌种培养、灭活及分解得到代谢产物、细胞质片段、细胞壁组分及多糖复合体;该原料具有很强的抗免疫抑制活性并能促进DNA修复,可有效保护皮肤,不受紫外线引起的损伤,用于乳化、水基及水醇体系的护肤、防晒及晒后护理产品,帮助预防表皮及真皮的光老化;另外可以加强角质层的代谢,还能捕获自由基,抑制脂质的过氧化,具有美白、抗衰老的功能;其中富含的营养物质,有滋养皮肤的功能。
现有技术中由于生产工艺的限制,生产得到的微生物化妆品原料,其自由基清除率衰减很大,过度剧烈的生产工艺会导致大量清除自由基的活性物质失去活性,从而导致其抗氧化活性的降低,同样活性的情况下,增加了生产成本,该现状亟待解决。
发明内容
为了克服上述技术问题,本发明提供一种二裂酵母发酵产物溶胞物的制备方法,能够解决二裂酵母发酵产物溶胞物生产过程中清除自由基物质活性降低的问题。
本发明解决其技术问题所采用的技术方案是:一种二裂酵母发酵产物溶胞物的制备方法,包括如下步骤:
S1: 将质量分数1%-4%的大豆和质量分数1%-2%的焦香麦芽于纯化水中煮沸2小时,冷却至室温后,使用800目筛子过滤,然后加入质量分数5%-10%的葡萄糖、0.1%-1%的酵母浸粉、0.1%-0.5%的磷酸氢二钾,并于115℃灭菌20min;
S2: 灭菌后,冷却至室温,接种质量分数1.5%-3.5%的二裂酵母菌液,于37℃发酵48小时;
S3: 发酵终点为发酵液pH值小于4.0,若48小时pH值未达到4.0以下,继续发酵;
S4: 达到发酵终点后,发酵液进行离心收集菌体,离心转速为3000 r/min – 8000 r/min ,离心时间1min-10min,加入0.2倍-2倍无菌超纯水复溶;
S5:复溶之后,加入质量分数0.1%-1.0%的溶菌酶,30℃-40℃酶解10min-60min;
S6:酶解完成后,加入质量分数5%-20%的海藻糖作为保护剂,并在80℃-100℃进行热裂解;
S7:冷却后,得到最终产品,于常温保藏。
本发明的有益效果是,与现有技术相比:
(1)该二裂酵母发酵产物溶胞物的制备方法,通过增加酶解工艺,使菌种的破壁更加充分,增加了细胞内容物的溶出比例,提高了二裂酵母溶胞物的制备效率,同时,也提高了单位量产品的自由基清除率。
(2)该二裂酵母发酵产物溶胞物的制备方法,通过增加保护剂之后的热裂解,保证了细胞在温和的环境中被破壁,使溶液中清除自由基的活性物质不被高温环境所破坏,有效的保证了产品的清除自由基物质活性。
具体实施方式
为了使本发明的目的、技术方案及优点更加清楚明白,以下结合实施例,对本发明进行进一步详细说明。
本实施例提供一种二裂酵母发酵产物溶胞物的制备方法,包括如下步骤:
S1: 将质量分数2%的大豆和质量分数1%的焦香麦芽于纯化水中煮沸2小时,冷却至室温后,使用800目筛子过滤,然后加入质量分数6%的葡萄糖、1%的酵母浸粉、0.2%的磷酸氢二钾,并于115℃灭菌20min;
S2: 灭菌后,冷却至室温,接种质量分数3%的二裂酵母菌液,于37℃发酵48小时;
S3: 发酵终点为发酵液pH值小于4.0,若48小时pH值未达到4.0以下,继续发酵;
S4: 达到发酵终点后,发酵液进行离心收集菌体,离心转速为3000 r/min,离心时间10min,加0.2倍无菌超纯水复溶;
S5:复溶之后,加入质量分数0.1%的溶菌酶,37℃酶解30min;
S6:酶解完成后,添加质量分数20%的海藻糖作为保护剂,并在90℃进行热裂解;
S7:冷却后,常温保藏。
对二裂酵母发酵产物溶胞物DPPH自由基清除率测试,测试使用DPPH自由基检测试剂盒,制备后的原液和十倍稀释液,分别进行自由基清除率测试。测试结果见下表。
对二裂酵母发酵产物溶胞物羟自由基清除率测试,测试使用羟自由基检测试剂盒,制备后的原液和十倍稀释液,分别进行自由基清除率测试。测试结果见下表。
对二裂酵母发酵产物溶胞物超氧阴离子自由基清除率测试,测试使用超氧阴离子自由基检测试剂盒,制备后的原液和十倍稀释液,分别进行自由基清除率测试。测试结果见下表。
对二裂酵母发酵产物溶胞物美白功效测试,测试使用酪氨酸还原法,同时使用5mg/mL的熊果苷作为阳性对照。测试结果见下表。

1.一种二裂酵母发酵产物溶胞物的制备方法,其特征在于,包括如下步骤:
S1: 将质量分数1%-4%的大豆和质量分数1%-2%的焦香麦芽于纯化水中煮沸2小时,冷却至室温后,使用800目筛子过滤,然后加入质量分数5%-10%的葡萄糖、0.1%-1%的酵母浸粉、0.1%-0.5%的磷酸氢二钾,并于115℃灭菌20min;
S2: 灭菌后,冷却至室温,接种质量分数1.5%-3.5%的二裂酵母菌液,于37℃发酵48小时;
S3: 发酵终点为发酵液pH值小于4.0,若48小时pH值未达到4.0以下,继续发酵;
S4: 达到发酵终点后,发酵液进行离心收集菌体,离心转速为3000 r/min – 8000 r/min ,离心时间1min-10min,加入0.2倍-2倍无菌超纯水复溶;
S5:复溶之后,加入质量分数0.1%-1.0%的溶菌酶,30℃-40℃酶解10min-60min;
S6:酶解完成后,加入质量分数5%-20%的海藻糖作为保护剂,并在80℃-100℃进行热裂解;
S7:冷却后,常温保藏。
