Abstract
本发明提供一种含海洋硫酸软骨素的营养强化麦片食品及其制备方法。本发明的方法包括鲨鱼软骨破碎、蒸煮、固液分离、酶解、除腥、过滤、干燥、配料、混合、包装步骤。采用生物酶解偶联发酵技术处理,直接水解软骨提取硫酸软骨素、骨胶原低聚肽,在酶解的同时达到祛除腥味的效果,节约了时间成本;分子量保持完整、生物活性不受破坏,避免了有机溶剂的使用。制备所得麦片食品富含硫酸软骨素、骨胶胶原蛋白、磷脂,冲调性好,口感滑润,能提高人体免疫力和保护或修护关节、满足增强体质的需求。

技术领域
本发明属于水产品加工技术领域,具体涉及一种含海洋硫酸软骨素的营养强化麦片食品及其制备方法。
背景技术
软骨鱼类即软骨鱼纲,是仅存最古老的有颌脊椎动物,主要包括鲨鱼、魟鱼、鳐和银鲛等,是海洋生态系统主要的组成部分之一,软骨鱼软骨中富含多肽、低分子量蛋白、糖蛋白和多糖等活性物质,可以作为新生血管生成抑制因子、肿瘤细胞抑制因子、免疫调节因子和抗入侵因子等用于肿瘤的治疗和预防。
鲨鱼生活在海洋,性凶猛,行动敏捷,捕食其他鱼类,具有很高的营养价值,其蛋白质不仅含量高,而且结构特殊,含粘多糖(氨基多糖)等富含生物活物质,日本学者在《鲨鱼软骨素神奇药效》一书中更详细介绍有关治病、防病的生动实例。V.B.Krylov等研究发现的鲨鱼硫酸软骨素具有抗炎活性,还表现出一些抗凝血的性质。鲨鱼硫酸软骨素相对于A类软骨素(猪、牛、鸡等),还具有独特的功能:对人和动物具有抗血管新生和抗癌活性,而被广泛地应用于膳食补充剂中;而且鲨鱼软骨还具有治疗骨关节炎、风湿性关节炎、进行性系统性硬化症和神经血管性青光眼的疗效果。
骨胶原肽不仅是一种潜在的钙源,还可以促进人体吸收膳食中的钙,提高人体对钙的生物利用度,增强骨密度和预防骨质疏松。鲨鱼中有机态钙含量高,吸收率远高于无机态钙,对骨质疏松发病率较高的中老年人群,是较好的补钙来源。
来源于海洋水产动物的硫酸软骨素、胶原蛋白在许多方面明显优于陆生动物来源的硫酸软骨素、胶原蛋白,具有安全性强、低过敏性和低抗原性等优点。由于疯牛病、口足疫等许多高危病毒及病原微生物的发现,其安全性受到质疑,使得动物来源的生物制品的应用受到严格限制,因而深海软骨鱼来源的硫酸软骨素和胶原蛋白产品具有更广阔的应用前景。
软骨鱼主要加工成冷冻制品,但占成鱼体重近15%的头骨和翅骨,在软骨鱼加工过程中经常被当作废弃物丢弃,未能得到充 分的利用。该技术为软骨鱼加工的综合利用开拓了下游产品,变废为宝,避免了环境的污染 和资源的浪费,大大提高产品的附加值。不仅可以实现软骨类资源的综合利用,还可以为开发补钙产品提供新的资源。实现软骨鱼鱼体利用价值的最大化,使软骨鱼原料经济价值得到全方位的提升。
发明内容
本发明的目的在于提供一种含海洋硫酸软骨素的营养强化麦片食品及其制备方法。
为实现上述目的,本发明采用如下技术方案:
一种含海洋硫酸软骨素的营养强化麦片食品,其原料按重量百分比计,包括以下组分:超细骨粉20%、熟麦片50%、魔芋葡甘聚糖3%、低聚果糖3%,鱼卵磷脂0.9%,奶粉23 %,复合抗氧化剂0.1%。
一种含海洋硫酸软骨素的营养强化麦片食品的制备方法,包括如下步骤:
(1)原料预处理:鲨鱼软骨除肉、清洗,鲨鱼软骨在4-8℃冷水,0.5%~1.0%的H2O2浓液中减菌处理10~20min,并破碎成0.1-0.5mm的细微颗粒;
(2)蒸煮:将处理好的软骨和纯水以1:3-5 的重量比例混合;在95~100℃水中煮50min,并冷却至60~70℃,超声波功率为400~800W,频率为40~50kHz,处理时间为50~60min;
(3)固液分离:将步骤(2)蒸煮处理后的鲨鱼软骨溶液,使用卧螺式离心机和碟片式离心机连续除渣,收集软骨汤液;
(4)提取:将步骤(3)收集的软骨汤液,按照占软骨汤液重量0.20%~0.50% 的比例加入酶,于50~55℃,pH6.2~7.0 条件下酶解反应4~6h ,得酶解液;
(5)发酵:加入酶解液重量0.2%的菌种和0 .1%的白砂糖溶液,混合均匀,42℃保温2小时;
(6)除臭,脱色:在步骤(5)的发酵液加热至70-80℃,再加入发酵液重量0.3%-0.5%的葡甘聚糖和2%-3%的活性炭,在pH 6.2—7.0处理0.5-1h;
(7)过滤:加硅藻土板框过滤机过滤;
(8)浓缩、干燥脱水:步骤(7)过滤所得溶液,脉冲干燥,产品水分≤10%,硫酸软骨素的≥40%,胶原蛋白≥55%;
(9)超微粉碎:步骤(8)干燥所得的产物300目超微粉碎,获得超细骨粉;
(10)配料:按重量百分比为超细骨粉20%、熟麦片50%、魔芋葡甘聚糖3%、低聚果糖3%,鱼卵磷脂0.9%、奶粉23 %、复合抗氧化剂0.1%配料;
(11)定量、包装:根据要求定量包装,制备获得含海洋硫酸软骨素的营养强化麦片。
上述步骤(4)中,所述酶为胰蛋白酶、枯草杆菌碱性蛋白酶、氨肽酶和中性蛋白酶按质量比为1:1:1:1的比例混合。
上述步骤(5)中,所述菌种为质量比为酵母菌:双歧杆菌=1:1的混合菌种;所述白砂糖添加前用35℃温水溶解。
上述步骤(9)中,所述超细鲨鱼硫酸软骨素粉的粒径范围在15 ~ 30 μm之间。
上述步骤(10)中,复合抗氧化剂由抗坏血酸钠、茶多酚和维生素E 柠檬酸组成;抗坏血酸钠、茶多酚和维生素E的重量比为2 :1 :1。
制得的营养强化麦片中硫酸软骨素≥8.0wt .%,分子量≤10000的鱼骨胶原蛋白≥10wt .%。
本发明具有如下有益效果:
1.采用生物酶解偶联发酵技术处理,直接水解软骨提取硫酸软骨素、骨胶原低聚肽,在酶解的同时达到祛除腥味的效果,节约了时间成本;分子量保持完整、生物活性不受破坏,避免了有机溶剂的使用,
2.在常温下干燥,较好地保留了软骨鱼(鲨鱼)的硫酸软骨素、骨胶原低聚肽活性,加入了帮助肠道调节功能的双歧杆菌,复配鱼卵磷脂、魔芋葡甘聚糖、奶粉、低聚果糖、维生素、抗坏血酸钠而成的富含硫酸软骨素、骨胶原低聚肽、磷脂营养强化蛋白粉。该产品具有富含硫酸软骨素、骨胶胶原蛋白、磷脂,冲调性好,口感滑润等特点,特别适合不同阶段运动人群和老年人食用,
3.能提高人体免疫力和保护或修护关节、满足增强体质、健脑等多种需求。
4.该产品同时含有帮助肠道调节功能的双歧杆菌。魔芋葡甘聚糖是可溶性膳食纤维,不含热量,有饱腹感,有减脂瘦身的功效。
具体实施方式
为了使本发明所述的内容更加便于理解,下面结合具体实施方式对本发明所述的技术方案做进一步的说明,但是下述的实例仅仅是本发明其中的例子而已,并不代表本发明所限定的权利保护范围,本发明的权利保护范围以权利要求书为准。
实施例1
一种含海洋硫酸软骨素的营养强化麦片食品的制备方法,包括如下步骤:
(1)原料预处理:鲨鱼软骨除肉、清洗,鲨鱼软骨在4℃冷水,0.5%的H2O2浓液中减菌处理10min,并破碎成0.1-0.5mm的细微颗粒;
(2)蒸煮:将预处理好的鲨鱼软骨和纯水以1:3的重量比例混合;在95℃水中煮50min,并冷却至60℃,超声波功率为400W,频率为40kHz,处理时间为50min;
(3)固液分离:将步骤(2)蒸煮处理后的鲨鱼软骨溶液,使用卧螺式离心机和碟片式离心机连续除渣,收集软骨汤液;
(4)提取:将步骤(3)收集的软骨汤液,按照占软骨汤液重量0.20% 的比例加入酶混匀后,于50℃,pH6.2 条件下酶解反应4h ;所述酶为胰蛋白酶、枯草杆菌碱性蛋白酶、氨肽酶和中性蛋白酶按质量比为1:1:1:1 的比例混合:
(5)发酵:加入酶解液重量0.2%(酵母菌:双歧杆菌=1:1)的菌种和0 .1%的白砂糖(用35℃温水溶解),混合均匀,42℃保温2 .0小时;
(6)除臭,脱色: 在步骤(5)的发酵液加热至70℃,再加入发酵液重量0.3%的葡甘聚糖和2%的活性炭,在pH 6.2处理0.5h;
(7)过滤:加硅藻土板框过滤机过滤;
(8)浓缩、干燥脱水:步骤(7)过滤所得溶液,脉冲干燥,产品水分≤10%,鲨鱼硫酸软骨素的纯度≥40%,胶原蛋白≥60%;
(9)超微粉碎:步骤(8)干燥所得的产物300目超微粉碎,获得超细骨粉;所述超细骨粉的粒径范围在15 ~ 30 μm之间。
(10)配料:按重量百分比为超细骨粉20%、熟麦片50%、魔芋葡甘聚糖3%、低聚果糖3%,鱼卵磷脂0.9%、奶粉20 %、复合抗氧化剂0.1%配料;所述复合抗氧化剂由抗坏血酸钠、茶多酚和维生素E 柠檬酸组成;抗坏血酸钠、茶多酚和维生素E的重量比为2 :1 :1。
(11)定量、包装:根据要求定量包装,制备获得含海洋硫酸软骨素的营养强化麦片。
实施例2
一种含海洋硫酸软骨素的营养强化麦片食品的制备方法,包括如下步骤:
(1)原料预处理:鲨鱼软骨除肉、清洗,鲨鱼软骨在6℃冷水,0.8%的H2O2浓液中减菌处理15min,并破碎成0.1-0.5mm的细微颗粒;
(2)蒸煮:将预处理好的鲨鱼软骨和纯水以1:4的重量比例混合;在98℃水中煮50min,并冷却至65℃,超声波功率为600W,频率为45kHz,处理干燥时间为55min;
(3)固液分离:将步骤(2)蒸煮处理后的鲨鱼软骨溶液,使用卧螺式离心机和碟片式离心机连续除渣,收集软骨汤液;
(4)提取:将步骤(3)收集的软骨汤液,按照占软骨汤液重量0.3% 的比例加入酶混匀后,于50℃,pH6.8条件下酶解反应5h ;所述酶为胰蛋白酶、枯草杆菌碱性蛋白酶、氨肽酶和中性蛋白酶按质量比为1:1:1:1 的比例混合;
(5)发酵:加入酶解液重量0.2%(酵母菌:的双歧杆菌=1:1)的菌种和0 .1%的白砂糖(用35℃温水溶解),混合均匀,42℃保温2.0小时;
(6)除臭,脱色: 在步骤(5)的发酵液加热至75℃,再加入发酵液重量0.4%的葡甘聚糖和2.5%的活性炭,在pH6.8处理0.5h;
(7)过滤:加硅藻土板框过滤机过滤;
(8)浓缩、干燥脱水:步骤(7)过滤所得溶液,脉冲干燥,产品水分≤10%,鲨鱼硫酸软骨素的纯度≥40%,胶原蛋白≥60%;
(9)超微粉碎:步骤(8)干燥所得的产物300目超微粉碎,获得超细骨粉;所述超细骨粉的粒径范围在15 ~ 30 μm之间。
(10)配料:按重量百分比为超细骨粉20%、熟麦片50%、魔芋葡甘聚糖3%、低聚果糖3%,鱼卵磷脂0.9%、奶粉20 %、复合抗氧化剂0.1%配料;所述复合抗氧化剂由抗坏血酸钠、茶多酚和维生素E 柠檬酸组成;抗坏血酸钠、茶多酚和维生素E的重量比为2 :1 :1。
(11)定量、包装:根据要求定量包装,制备获得含海洋硫酸软骨素的营养强化麦片。
实施例3
一种含海洋硫酸软骨素的营养强化麦片食品的制备方法,包括如下步骤:
(1)原料预处理:鲨鱼软骨除肉、清洗,鲨鱼软骨在8℃冷水, 1.0%的H2O2浓液中减菌处理20min,并破碎成0.5mm的细微颗粒;
(2)蒸煮:将预处理好的鲨鱼软骨和纯水以1:5 的重量比例混合;在100℃水中煮50min,并冷却至70℃,超声波功率为800W,频率为50kHz,处理干燥时间为60min;
(3)固液分离:将步骤(2)蒸煮处理后的鲨鱼软骨溶液,使用卧螺式离心机和碟片式离心机连续除渣,收集软骨汤液;
(4)提取:将步骤(3)收集的软骨汤液,按照占软骨汤液重量0.50% 的比例加入酶,酶于55℃,pH7.0 条件下酶解反应6h ;所述酶为胰蛋白酶、枯草杆菌碱性蛋白酶、氨肽酶和中性蛋白酶按质量比为1:1:1:1 的比例混合;
(5)发酵:加入酶解液重量0.2%(酵母菌:的双歧杆菌=1:1)的菌种和0 .1%的白砂糖(用35℃温水溶解),混合均匀,42℃保温2 .0小时;
(6)除臭,脱色: 在步骤(5)的发酵液加热至80℃,再加入发酵液重量0.5%的葡甘聚糖和3%的活性炭,在pH7.0处理1h;
(7)过滤:加硅藻土板框过滤机过滤;
(8)浓缩、干燥脱水:步骤(7)过滤所得溶液,脉冲干燥,产品水分≤10%,鲨鱼硫酸软骨素的纯度≥40%,胶原蛋白≥60%;
(9)超微粉碎:步骤(8)干燥所得的产物300目超微粉碎,获得超细骨粉;所述超细骨粉的粒径范围在15 ~ 30 μm之间。
(10)配料:按重量百分比为超细骨粉20%、熟麦片50%、魔芋葡甘聚糖3%、低聚果糖3%,鱼卵磷脂0.9%、奶粉20 %、复合抗氧化剂0.1%配料;所述复合抗氧化剂由抗坏血酸钠、茶多酚和维生素E 柠檬酸组成;抗坏血酸钠、茶多酚和维生素E的重量比为2 :1 :1。
(11)定量、包装:根据要求定量包装,制备获得含海洋硫酸软骨素的营养强化麦片。
实施例4
SPF级雌性SD大鼠80只,体重约280~300g。实验条件为屏障环境,实验期间实验环境温度22°C-24°C,湿度52%-56%。实验动物随机分为4组:实施例1-3组以及空白对照组,分别取实施例1-3海洋硫酸软骨素的营养强化麦片3.33g加1%羧甲基纤维素至100mL,空白对照组给予等体积的1%羧甲基纤维素,分别给予受试动物灌胃,每天灌胃一次,灌胃体积为1mL,共90天。实验结束时,颈椎脱臼处死 大鼠,剥离右股骨,烘烤至恒重,测量股骨中点处和远心端的骨密度(BMD)。结果以平均值表示。

实施例5
将实施例2制得的海洋硫酸软骨素的营养强化麦片进行食用效果验证。采取自愿原则,选取年龄50-70阶段,男性女性分别30人,每种性别中具有腕关节疼、膝关节疼各15人。
临床实验时,停服其他药物和营养保健品,食用本发明制作的海洋硫酸软骨素的营养强化麦片食品。每份:50克海洋硫酸软骨素的营养强化麦片,加入150毫升热水;1份/每天·人。食用三个月后,症状有所缓解,六个月后,症状明显改善,其中女性腕关节疼痛改善的有8人,膝关节疼痛改善的14人;男性腕关节疼痛改善的9人,膝关节改善的13人。
以上所述仅为本发明的较佳实施例,凡依本发明申请专利范围所做的均等变化与修饰,皆应属本发明的涵盖范围。
Claims (6)
1.一种含海洋硫酸软骨素的营养强化麦片食品,其特征在于,其原料按重量百分比计,包括以下组分:超细骨粉20%、熟麦片50%、魔芋葡甘聚糖3%、低聚果糖3%,鱼卵磷脂0.9%,奶粉23 %,复合抗氧化剂0.1%。
2.如权利要求1所述一种含海洋硫酸软骨素的营养强化麦片食品的制备方法,其特征在于,包括如下步骤:
(1)原料预处理:鲨鱼软骨除肉、清洗,鲨鱼软骨在4-8℃冷水,0.5%~1.0%的H2O2浓液中减菌处理10~20min,并破碎成0.1-0.5mm的细微颗粒;
(2)蒸煮:将预处理好的鲨鱼软骨和纯水以1:3-5 的重量比例混合;在95~100℃水中煮50min,并冷却至60~70℃,超声波功率为400~800W,频率为40~50kHz,处理时间为50~60min;
(3)固液分离:将步骤(2)蒸煮处理后的鲨鱼软骨溶液,使用卧螺式离心机和碟片式离心机连续除渣,收集软骨汤液;
(4)提取:将步骤(3)收集的软骨汤液,按照占软骨汤液重量0.20%~0.50% 的比例加入酶,于50~55℃,pH6.2~7.0 条件下酶解反应4~6h ;
(5)发酵:加入酶解液重量0.2%的菌种和0 .1%的白砂糖溶液,混合均匀,42℃保温2 .0小时;
(6)除臭,脱色: 在步骤(5)的发酵液加热至70-80℃,再加入发酵液重量0.3-0.5%的葡甘聚糖和2-3%的活性炭,在pH 6.2—7.0处理0.5-1h;
(7)过滤:加硅藻土板框过滤机过滤;
(8)浓缩、干燥脱水:步骤(7)过滤所得溶液,脉冲干燥,产品水分≤10%,鲨鱼硫酸软骨素的纯度≥40%,胶原蛋白≥60%;
(9)超微粉碎:步骤(8)干燥所得的产物300目超微粉碎,获得超细骨粉;
(10)配料:按重量百分比为超细骨粉20%、熟麦片50%、魔芋葡甘聚糖3%、低聚果糖3%,鱼卵磷脂0.9%、奶粉23 %、复合抗氧化剂0.1%配料;
(11)定量、包装:根据要求定量包装,制备获得含海洋硫酸软骨素的营养强化麦片。
3.根据权利要求3所述的一种含海洋硫酸软骨素的营养强化麦片食品的制备方法,其特征在于:步骤(4)中,所述酶为胰蛋白酶、枯草杆菌碱性蛋白酶、氨肽酶和中性蛋白酶按质量比为1:1:1:1 的比例混合。
4.根据权利要求3所述的一种含海洋硫酸软骨素的营养强化麦片食品的制备方法,其特征在于:步骤(5)中,所述菌种为质量比为酵母菌:双歧杆菌=1:1的混合菌种;所述白砂糖添加前用35℃温水溶解。
5.根据权利要求3所述的一种含海洋硫酸软骨素的营养强化麦片食品的制备方法,其特征在于:步骤(9)中,所述超细鲨鱼硫酸软骨素粉的粒径范围在15 ~ 30 μm之间。
6.根据权利要求3所述的一种含海洋硫酸软骨素的营养强化麦片食品的制备方法,其特征在于:步骤(10)中,复合抗氧化剂由抗坏血酸钠、茶多酚和维生素E 柠檬酸组成;抗坏血酸钠、茶多酚和维生素E的重量比为2 :1 :1。
