Abstract
The invention provides a lactobacillus facial mask essence capable of inhibiting propionibacterium acnes and a preparation method thereof; the method comprises the following steps: 5 parts of propylene glycol, 2 parts of PEG-40 hydrogenated castor oil, 1.4 parts of sodium hyaluronate, 2 parts of dimethyl glycyrrhizinate, 0.2 part of carbomer, 0.1 part of hydroxyethyl cellulose, 0.2 part of chlorphenesin, 0.05 part of geranium oil, 0.2 part of phenoxyethanol, 2.1 parts of triethanolamine, 0.5 part of inulin, 0.84 part of inactivated freeze-dried powder and the balance of water. The invention provides a lactobacillus facial mask essence for removing acnes, which is proved by tests to have an obvious inhibition effect on propionibacterium acnes, the diameter of a bacteriostatic ring can reach 17.94mM, the lactobacillus facial mask essence is insensitive to temperature and illumination, the lactobacillus facial mask essence has good stability under room temperature and natural illumination, and the diameter change of the bacteriostatic ring is less than 2mM when the lactobacillus facial mask essence is stored for two months under the room temperature and natural illumination conditions.

Technical Field

Background
Acne is a chronic inflammatory dermatosis of hair follicle sebaceous glands, has the morbidity of 70-87 percent, is good at adolescence, and has clinical manifestations mainly including blackheads and whiteheads of face, chest and back, inflammatory papules and pustules.
The propionibacterium acnes can induce proinflammatory factors, particularly IL-1 α, to be released due to natural immunity mediated by Toll-like receptor (TLR), and acquired immunity to amplify the inflammatory process with the development of diseases, further cause inflammatory factor release and neutrophil aggregation, and can also generate polypeptide substances to directly induce or aggravate inflammation.

(1) the invention provides a lactobacillus facial mask essence for removing acnes, which is proved by tests to have an obvious inhibition effect on propionibacterium acnes, the diameter of a bacteriostatic ring can reach 17.94mM, the lactobacillus facial mask essence is insensitive to temperature and illumination, the lactobacillus facial mask essence has good stability under room temperature and natural illumination, and the diameter change of the bacteriostatic ring is less than 2mM when the lactobacillus facial mask essence is stored for two months under the room temperature and natural illumination conditions.
(2) Compared with antibiotics, the lactobacillus facial mask essence added with the inactivated freeze-dried powder of the lactobacillus rhamnosus LR863 fermentation product is safer and milder, and can play a role in repairing skin barriers while inhibiting propionibacterium acnes, so that the effect of removing acne is achieved.

Example 1

Test materials and methods
1 test Material
(1) The strain is as follows: lactobacillus rhamnosus LR 863; wherein the registration number of the strain is CGMCC No.14410, China general microbiological culture Collection center Beijing city, Shangyang district, Xilu No.1 Hospital No. 3. Propionibacterium acnes was purchased from North Nara, accession No. BNCC 336649.
(2) Culture medium
Improving MRS liquid culture medium: 20g of glucose, 10g of peptone, 6.5g of beef powder, 5g of yeast extract powder, 5g of sodium acetate, 2g of diammonium hydrogen citrate, 2g of dipotassium hydrogen phosphate, 0.58g of MgSO4 & 7H2O 0.58, 0.25g of MnSO4 & H2O 0.25, 0.5g of cysteine hydrochloride, 801mL of Tween and 1000mL of distilled water.
Thioglycollate medium (containing agar): 15.0g of casein, 5.0g of yeast extract powder, 5.0g of glucose, 0.5g of sodium thioglycolate, 0.5g of L-cystine, 0.001g of resazurin, 2.5g of sodium chloride, 1000mL of distilled water and pH7.1 +/-0.2.
2 test method
2.1 Strain culture methods and sample preparation
Preparation of inactivated freeze-dried powder of lactobacillus rhamnosus LR863 fermentation product: cultured in modified MRS medium and incubated at 37 deg.C overnight. Inoculating lactobacillus rhamnosus LR863 seed solution into 10ml of improved MRS liquid culture medium according to the inoculation amount of 3%, shaking and uniformly mixing, culturing at 37 ℃ for 24h, sequentially and continuously activating for three generations, then inoculating into 250ml of improved MRS liquid culture medium according to the inoculation amount of 3%, carrying out amplification culture, sterilizing the cultured fermentation liquor at 70 ℃ for 3h, centrifuging to collect bacterial sludge, emulsifying, and freeze-drying to obtain LR863 fermentation product inactivated freeze-dried powder (hereinafter referred to as inactivated freeze-dried powder).
Culturing propionibacterium acnes: the culture was carried out anaerobically in a liquid thioglycollate medium (containing agar) at 37 ℃ in a incubator for 48 hours. The method comprises the following specific steps: inoculating the Propionibacterium acnes seed liquid to 10ml of liquid thioglycollate medium (containing agar) according to the inoculation amount of 3%, shaking and mixing uniformly, carrying out anaerobic culture at 37 ℃ for 48h, and sequentially and continuously activating for three generations for later use.
2.2 preparation of the lactobacillus facial mask:
the formula of the facial mask comprises: 5 parts of propylene glycol, 2 parts of PEG-40 hydrogenated castor oil, 1.4 parts of sodium hyaluronate, 2 parts of dimethyl glycyrrhizinate, 0.2 part of carbomer, 0.1 part of hydroxyethyl cellulose, 0.2 part of chlorphenesin, 0.05 part of geranium oil, 0.2 part of phenoxyethanol, 2.1 parts of triethanolamine, 0.5 part of inulin, 0.84 part of inactivated freeze-dried powder and the balance of water.
The preparation method comprises the following steps: the material A comprises the components except carbomer, hydroxyethyl cellulose and inactivated freeze-dried powder, which can be mixed uniformly in advance for later use. And the material B, carbomer and hydroxyethyl cellulose are respectively dissolved in water, stirred and heated to 80 ℃ for full dissolution, and then cooled to room temperature for later use. C, material C: the inactivated lyophilized powder should be dissolved in purified water and stirred well. And (3) respectively preparing the A, B and the material C, and uniformly mixing to obtain the lactobacillus mask essence.
3. Bacteriostasis test
(1) Cooling the agar culture medium containing 2.0% of the extract to about 50 ℃, pouring 10mL of the agar culture medium into a sterile plate, and airing the agar culture medium in a clean workbench;
(2) placing sterilized Oxford cup in a dish with agar, and standing for 10 min;
(3) add 1mL Propionibacterium acnes to 100mL of liquid thioglycollate medium (containing agar) cooled to 50 ℃ and mix well;
(4) pouring 10mL of the liquid thioglycollate culture medium (containing agar) on the flat plate with the Oxford cup, and clamping the Oxford cup after solidification to form an Oxford cup hole;
(5) 0.1mL of test sample is injected into each Oxford cup hole, tetracycline hydrochloride is used as a positive control, physiological saline is used as a negative control, standing is carried out for anaerobic culture for 48-72h at 37 ℃, the diameter of a bacteriostatic ring is measured by a vernier caliper, and two groups are arranged in parallel.
Preferably, the inactivated freeze-dried powder is lactobacillus rhamnosus LR863 fermentation product inactivated freeze-dried powder or propionibacterium acnes inactivated freeze-dried powder.
