Abstract

Description
技术领域
本发明涉及一种包含胶囊化的挥发性精油的农用制剂,其中,所述胶囊化精油包含溶解在其中的聚合物以改善所述胶囊化的挥发性精油的释放特性。
具体地,本发明的农用制剂还包含(但不限于)天然来源的非挥发性赋形剂,例如天然昆虫生长调节剂(IGR),诸如印楝素-印楝 (Azadirachtin-neem)。为了防止非挥发性赋形剂的分解和氧化,本发明的制剂还包含抗氧化剂,例如醋酸生育酚(维生素E乙酸酯)。特别地,本发明的制剂除了包含挥发性精油和溶解在所述挥发性精油中的聚合物之外,还包含天然的非挥发性赋形剂和抗氧化剂。
背景技术
在现代化学和制药学工业出现之前,在日常生活的许多领域中,精油通常被用作药物和化妆品中的防腐剂和消毒剂材料,例如抗菌剂和杀幼虫剂。研究还发现,具有广谱抗菌活性的精油类试剂对哺乳动物是相对无毒的。尽管精油存在上述优点,但由于合成化学品(诸如抗生素)更加便宜、高效且能够以较低浓度使用,因此精油逐渐被类似的合成化学品所替代。然而,随着时间的推移,鉴于合成化学品的毒性及其对环境带来的负面影响,现在人们正在致力于用原先被替换掉的精油类试剂来取代合成化学品。
已经报道了精油类试剂在农业生产中的应用。本申请发明人的 PCT申请WO 04/098767(申请号PCT/IL2004/000384)披露了下列精油微胶囊:例如作为用于消费者市场的消毒剂产品、硬表面清洁剂、衣物洗涤剂和软化剂、杀虫剂、驱虫剂以及抗病毒剂或抗真菌剂。当精油微胶囊施用于给定基底时,其中所包含的精油在一段时间内以恒定速率释放。所述微胶囊的功效仅仅取决于与该微胶囊本身有关的参数,例如大小、包封膜的厚度、其中所含精油的持续释放能力等,而与将它们递送到目标环境并随后立即干燥的水性介质无关。
中国专利申请No.1430981(申请号2002-144675)公开了一种包括莪术(Curcumazedoaria)精油、半合成脂肪酸甘油酯、聚乙二醇和表面活性剂的抗病毒胶囊。将加热的精油过滤并与熔融的聚乙二醇和表面活性剂混合以获得用于填充胶囊的胶囊化精油。该申请还公开了含有精油的包衣且缓释的片剂和药膏。
本申请发明人的另一件专利申请EP 1845786B1报道了如果作为杀虫剂、驱虫剂、杀卵剂和抗病毒剂的胶囊化的挥发性精油悬浮或分散在非挥发性赋形剂(可包含诸如高沸点或高熔点的精油和脂质等试剂)中,则精油的功效可得到增强。更具体地,已经确定如果这些胶囊化的挥发性精油在施用于基底或目标环境时,例如在水性介质蒸发时可保持与非挥发性试剂(能够强化施用效果)接触,则其功效可得到增强。而且还令人惊异的是,发现非挥发性赋形剂可以产生加和效应或协同效应。然而,发明人最近发现,如在本申请中所述,在挥发性精油与溶解在所述精油中的聚合物共胶囊化时,通过聚合物与精油之间的化学和物理相互作用,所述聚合物通过控制挥发性精油的释放速率而显著延长了所述挥发性精油活性的持续时间且提高了挥发性精油的活性。
发明内容
在本发明的一个方面,提供了一种用于驱虫、杀虫、特别是杀幼虫、杀卵或抗病毒的农用制剂,所述制剂包含:
(i)至少一种胶囊化的挥发性精油,其包含溶解在所述挥发性精油中的聚合物;和
(ii)非挥发性赋形剂,其用于携载所述至少一种胶囊化的挥发性精油,其中所述非挥发性赋形剂为固体或液体的形式。

优选地,所述聚合物选自由以下组成的组中的一种或多种:聚乙烯吡咯烷酮、聚环氧乙烷及其与聚环氧丙烷的共聚物、单独的聚环氧丙烷及其共聚物、聚乙酸乙烯酯、其它烷基化物以及多糖。其中,所述多糖为纤维素衍生物。所述纤维素衍生物选自羟基烷基醚和羟基烷基酯中的一种或两种。所述羟基烷基醚为羟乙基纤维素。
优选地,所述非挥发性赋形剂选自非挥发性植物油、非挥发性液体萜烯或固体萜烯、脂质或其任意组合。
优选地,所述非挥发性赋形剂选自:芝麻油、除虫菊油、甘油衍生的脂质、甘油脂肪酸衍生物或其任意组合。
优选地,所述非挥发性赋形剂为天然的昆虫生长调节剂。其中,所述天然的昆虫生长调节剂选自:(1)保幼激素模拟物;(2)蜕皮激素抑制剂;和/或(3)几丁质合成抑制剂。其中,所述保幼激素模拟物选自吡丙醚、苯氧威、烯虫乙酯或其任意组合;所述蜕皮激素抑制剂为印楝素;所述几丁质合成抑制剂为灭蝇胺。
优选地,所述聚合物的含量为所述挥发性精油重量的0.2-20重量%。优选地,所述聚合物的含量为所述挥发性精油重量的1-15重量%。更优选地,所述聚合物的含量为所述挥发性精油重量的5-10 重量%。
优选地,所述至少一种胶囊化的挥发性精油与所述非挥发性赋形剂的重量比为1.5:1至4:1。
优选地,本发明的农用制剂还包含作为所述赋形剂的一部分或在胶囊内的至少一种添加剂,所述至少一种添加剂选自由以下组成的组中的一种或多种:抗氧化剂、佐剂、粘合剂、防水剂、表面活性剂、防止微胶囊聚集的空间位阻聚合物和胶凝破坏剂。

优选地,所述农用制剂以下列剂型存在:乳化性浓缩剂、可湿性粉剂、流动型制剂、悬浮剂、颗粒剂、粉尘剂、熏蒸剂或溶液剂。其中,所述可湿性粉剂为粒状可湿性粉剂。所述溶液剂为水性溶液剂。
优选地,所述农用制剂以单制剂存在,其中所述胶囊化的挥发性精油与非挥发性赋形剂预混合;或者以双组分制剂存在,其包含作为第一组分的胶囊化的挥发性精油以及作为第二组分的非挥发性赋形剂。
在本发明的另一方面,提供了一种制备本发明的农用制剂的方法,所述方法包括:将至少一种胶囊化的挥发性精油分散在非挥发性赋形剂或包含所述非挥发性赋形剂的介质中;或者将非挥发性赋形剂添加至含有胶囊化的挥发性精油的水性溶液中,其中,所述胶囊化的挥发性精油包含溶解在其中的聚合物。其中,所述胶囊化的挥发性精油通过界面聚合制备。所述界面聚合通过在所述挥发性精油液滴的周围形成聚脲和/或聚氨酯膜进行所述挥发性精油的胶囊化。
在本发明的另一方面,提供了一种用于控制害虫种群的方法,包括将本发明的农用制剂施用于目标环境或害虫种群或害虫栖所,其中,所述目标环境为原生环境或次生环境。

在本发明的又一方面,提供了一种试剂盒,其包括第一容器,其容纳有作为第一组分的至少一种胶囊化的挥发性精油,其中,所述挥发性精油包含溶解在其中的聚合物;和第二容器,其容纳有作为第二组分的非挥发性赋形剂。
在本发明的又一方面,提供了本发明的农用制剂在制备用于防治粉虱、番茄蠹蛾和胡椒象甲的药剂中的用途。
具体实施方式
在本文中,表述“胶囊化(encapsulated)精油”或“胶囊化 (encapsulation)”或其任何语言上的变型指能够在其中容纳一种以上具有溶解的聚合物的精油的任何形状和大小的颗粒形式。该胶囊化的一个实例是微胶囊化。尽管本申请的农用制剂可以采用本领域已知的任何胶囊化精油颗粒形式而不会影响其有益之处,但优选的形式为微胶囊。在本发明中,基于微胶囊的总重量计,微胶囊具有30-98重量%、优选地60-95重量%的含有溶解的聚合物的挥发性精油,其中所述微胶囊可以通过异氰酸酯的界面聚合而制备。例如,在申请WO 04/098767中所披露的,所述界面聚合可提供聚氨酯、聚脲或其组合的微胶囊壳。微胶囊通常具有0.1微米至100微米的平均尺寸。其他适合的微胶囊可通过例如在WO 94/13139、EP0252897、US5576009 和US5925464中披露的方法制备。
在本发明中,溶解在挥发性精油中的聚合物起到了控制精油的释放速率以及物理上稳定在非挥发性赋形剂表面和内部的微胶囊的作用。根据聚合物的化学性质以及浓度,可以控制精油的释放速率。聚合物溶解的过程实际上是精油与聚合物之间相互作用的过程,这种相互作用延长了精油活性的持续时间。在本发明中,可使用的聚合物的具体实例包括合成或天然来源的:聚乙烯吡咯烷酮(PVP)、聚环氧乙烷(PEO)及其与聚环氧丙烷(PPO)的共聚物、单独的聚环氧丙烷及其共聚物、聚乙酸乙烯酯(PVAc)、其它烷基化物以及多糖,例如纤维素衍生物,如羟基烷基醚(如羟乙基纤维素(HEC))或羟基烷基酯,所有这些聚合物都可溶于精油。具体地,基于挥发性精油的重量计,聚合物的含量为挥发性精油重量的0.2-20重量%、优选 1-15重量%、更优选5-10重量%。当聚合物的含量高于挥发性精油重量的20重量%时,会引起过高的粘度,从而使精油与聚合物的混合变得困难,同时对精油产生稀释作用,难以实现精油有效的胶囊化。
在一个实施方式中,本发明的农用制剂还包含非挥发性赋形剂,其可以是合成来源的赋形剂、天然来源的赋形剂、或者其两者。
在一个优选的实施方式中,使用增强本发明的农用制剂的药效的来自天然来源(实质上非合成)的非挥发性赋形剂。具体而言,非挥发性赋形剂为天然的昆虫生长调节剂。使用天然的昆虫生长调节剂的一个优势是:根据美国环保局的规定,如果要使用合成的昆虫生长调节剂,则需要通过残留物测试,其中不仅要测试合成的昆虫生长调节剂本身,还要测试其分解产物。然而,对于天然组分来说,这些测试不是必需的。在本发明中,天然的昆虫生长调节剂可以选自以下类别: (1)保幼激素(JH)模拟物,例如吡丙醚(pyriproxyfen)、苯氧威 (fenoxycarb)和烯虫乙酯(hydroprene);(2)蜕皮激素抑制剂,例如印楝素(Azadirachtin);和/或(3)几丁质合成抑制剂,例如环丙氨嗪(cyromazine)。优选地,天然的昆虫生长调节剂为印楝素和烯虫乙酯,更优选地印楝素。印楝素是通过紫外线分解的抗饲料昆虫生长调节剂。另外,也可以使用在世界范围内登记的、非天然的其他昆虫生长调节剂,例如烯虫酯(methoprene)、蚊蝇醚(pyriproxyphen) 和杀铃脲(triflumuron)。
在本发明中,术语“非挥发性赋形剂”通常是指施用后与微胶囊一起停留在目标环境并可发挥加和效应或协同效应的有机试剂。该赋形剂可以为具有高沸点或高熔点的液体或固体(纯物质或混合物),并且在施用于目标环境后其离开目标环境表面的蒸发速度小于挥发性精油的蒸发速度。具体地,该赋形剂可为非挥发性植物油、非挥发性液体萜烯或固体萜烯、以及脂质。
如果需要,水可以作为本发明制剂的主要组分。在该示例性情况中,当在水性溶液中制备微胶囊时、或者当加入水以使得具有更好的流动性和喷雾性时、或者当在水中包装或保存制剂时,可以将非挥发性赋形剂添加至用作介质并且不会对制剂的驱虫、杀虫、特别是杀幼虫或杀卵特征产生不利效果的水性溶液中。在一些情况下,本发明的制剂在施用后水蒸发,留下微胶囊悬浮在非挥发性赋形剂中。在其他实施方式中,水可能不是必需的。
当所述赋形剂是非挥发性植物油或脂质时,该术语是指沸点高于 300℃的那些物质。例如,高沸点的非挥发性植物油可以为除虫菊酯类。脂质的实例是芝麻油或棉籽油。
术语“固体赋形剂”是指能够与微胶囊混合的固体试剂(纯物质或混合物),并且在对目标环境施用前其可以溶解、悬浮或均匀分散在诸如水的液体介质中。例如,固体赋形剂可以为粉末的形式。
术语“液体赋形剂”是指纯液体、或者多种试剂(每一种试剂在混合前均可为固体、液体或气体)的均匀液体混合物或者非均匀液体混合物,如液体赋形剂可以为悬浮液的形式。微胶囊在液体赋形剂或通过将固体赋形剂溶解或分散在诸如水的适宜介质中而制备的溶液中的悬浮液应当不影响微胶囊中的挥发性精油的稠度、分布、物理状态或浓度等特性。另外,液体赋形剂还具有这样的特性:其携载的微胶囊不会发生溶解、劣化、分解、精油浸出或经历任何其他物理或化学的变化。
在一个实施方式中,所述非挥发性赋形剂包括至少一种非挥发性植物油。其中,在不影响挥发性精油的性能的情况下,还可以对赋形剂进行一定的变化,例如可以对诸如脂质,如不同的甘油三酸酯进行改变。
在一个实施方式中,至少一种胶囊化的挥发性精油与非挥发性赋形剂的重量比在1.5:1至4:1的范围内。
在一个实施方式中,本发明的农用制剂测试针对番茄病虫害中常见的粉虱或粘虫进行。本发明的制剂长时间内不仅对成年虫有活性,而且对幼虫和若虫也有活性。
在本发明中,术语“挥发的”、“中度挥发的”和“非挥发的”是指在环境温度和压力下化学试剂蒸发能力的等级。一般来说,试剂的沸点越低,则认为该试剂的挥发性越高。关于精油,挥发性的低沸点精油指定义为沸点低于约250℃的那些精油。中度挥发的精油指定义为沸点为250℃-300℃的那些精油。非挥发的或较少挥发的精油指定义为沸点高于300℃的那些精油。
在本发明中,术语“植物油”指由植物制得的不溶于水的天然复杂的混合油状物。已经从植物体的多个部分(根、茎、叶、花、果实和种子)获取植物油,并且所述植物油还在某些特殊细胞或细胞组织 (如腺体)中富集。术语“精油”指通常赋予植物特有的气味、香味或其他类似性质的那些物质。通常,根据产生精油的部分的性质,精油可以是通过各种处理方法由植物获得的复杂混合物。所述处理方法例如为压缩、蒸汽蒸馏、油溶出(提取)或油吸收、或者加压浸渍。
尽管术语“精油”和“植物油”在不同的文献来源中可交替使用,但在本发明的范围内,后者是指还包括脂质的更大的化合物族群。
在本文中,术语“脂质”包括脂肪酸、甘油衍生的脂质(包括脂肪和油以及磷脂)、鞘氨醇衍生的脂质(包括神经酰胺、脑苷脂、神经节苷脂和鞘磷脂)、类固醇及其衍生物、萜烯类及其衍生物、某些芳香族化合物和长链醇及蜡。另外,术语“脂质”还指脂蛋白(与蛋白质或碳水化合物结合的脂质)、脂多糖以及维生素(诸如脂溶性维生素)。
在本发明中,挥发性精油选自肉桂油、雪松油、蓖麻油、丁香油、天竺葵油、柠檬草油、薄荷油、百里香油、姜黄油、冬青油、迷迭香油、小茴香油、小豆蔻油、春黄菊油、芫荽油、枯茗油、莳萝油、欧芹油、罗勒油、樟脑油、香茅油、桉树油、茴香油、姜油、葡萄柚油、柠檬油、橘皮油、橙油、松针油、胡椒油、玫瑰油、甜橙油、红桔油、茶树油、薰衣草油、茶籽油、葛缕子油、蒜油、胡椒薄荷油、洋葱油和留兰香油。优选地,挥发性精油选自香茅油、天竺葵油、茶树油、薰衣草油、丁香油、松树油和桉树油。
在优选的实施方式中,植物油选自芝麻油或除虫菊(Pyrethrum cinerariifolium)油、天然来源的甘油衍生的脂质或甘油脂肪酸衍生物。

为了防止来自天然来源的非挥发性赋形剂的分解和氧化,可以添加抗氧化剂。抗氧化剂可以选自非酶类物质,例如谷胱甘肽、脯氨酸、α-生育酚、类胡萝卜素和类黄酮以及维生素C(抗坏血酸)等;或酶蛋白,例如超氧化物歧化酶、过氧化氢酶谷胱甘肽过氧化物酶和谷胱甘肽还原酶。优选地,抗氧化剂是非酶促脂溶性的抗氧化剂,例如生育酚(例如α-生育酚)、类胡萝卜素或类黄酮。更优选地,抗氧化剂是生育酚,如醋酸生育酚(维生素E乙酸酯)。
佐剂可用于例如延长保质期、喷雾性和对基底的吸附作用。所述佐剂可以选自天然的聚合物或合成的聚合物,例如聚乙烯醇、聚乙烯吡咯烷酮、聚环氧乙烷、乙烯或马来酸酐的共聚物、甲基乙烯基醚- 马来酸酐共聚物、水溶性纤维素、水溶性聚酰胺或聚酯、丙烯酸的共聚物或均聚物、水溶性淀粉和改性淀粉、天然胶(诸如藻酸盐)、糊精以及蛋白质(诸如明胶和酪蛋白)等。

本发明的农用制剂的物理状态,即作为固体或液体的状态,取决于非挥发性赋形剂是液体还是固体,或者非挥发性赋形剂和微胶囊是否悬浮或分散在诸如水的不能混溶的液体中。不管其物理状态如何,本发明的制剂还可以通过将其进一步形成所需的剂型而使用,所述剂型如乳化性浓缩剂、可湿性粉剂(如粒状可湿性粉剂)、流动型制剂、悬浮剂、颗粒剂、粉尘剂和熏蒸剂等。根据例如下列参数可以确定剂型的性质:目标环境、施用的方法、施用的条件、微胶囊在非挥发性赋形剂中的相对浓度等。
尽管可以控制挥发性精油在非挥发性赋形剂中的浓度,但挥发性精油的浓度可以根据存储条件、气候条件、剂型、施用的方法、施用的位置、待被控制的目标害虫和目标作物等变化。按重量计,挥发性精油在本发明农用制剂中的浓度可以在0.01%-90%范围内变化,优选为0.1%-25%。对于不同的目标环境,诸如作物地块(field)的施用量,挥发性精油以及其中溶解的聚合物的含量可以根据作物地块的种植类型、作物生长发育时期、当地的气候条件、制剂剂型以及目标害虫等条件而相应调整。
不希望受到理论的束缚,当将例如水性制剂施用于目标环境时,其中的水在表面上干燥,留下其中封装有胶囊化的挥发性精油的非挥发性赋形剂层。所述非挥发性赋形剂对处理过的环境施加初期效果;之后缓慢地通过自发释放或借助任何其他因子的引发释放,微胶囊开始释放其内容物,由此二者对环境产生加和效应或协同效应。
本发明的制剂可适合于微胶囊的四种释放曲线,即(1)快速释放曲线;(2)持续或延迟释放曲线;(3)所谓的“突释(knock down)”式生物效应曲线,在该曲线下微胶囊将其内容物作为一个部分释放或在相对短的时间内释放;和(4)残留曲线,其中在所谓的“突释”之后跟随着持续释放曲线。
根据所用的具体精油,本发明的制剂可以用于各种农学、园艺和农艺学目的。术语“农用制剂”还可以指园艺和农学制剂。不希望受到理论和操作方式的束缚,本发明的制剂可以通过将害虫驱逐而非使其随后死亡用作驱虫剂;通过杀死昆虫或其他害虫的一部分或整个种群用作杀虫剂(或除虫剂);以及通过杀死昆虫卵的一部分或整个种群用作杀卵剂。术语“害虫(pest)”可以指昆虫类、线虫类、以及任何其他对人类、动物及植物品种有害的动物。害虫的实例例如为属于半翅目(Hemiptera)的异翅亚目(Heteroptera)、同翅目(Homoptera)、鞘翅目(Coleoptera)、鳞翅目(Leopidoptera)、缨翅目(Thysanoptera)、膜翅目(Hymenoptera)、等翅目(Isoptera)、直翅目(Orthoptera)、蜱螨目(Acarina)、矛线目(Dorylaimida)、腹足纲(Gastropoda) 和垫刃目(Tylenchida)的那些害虫。

通过影响能够对目标环境造成损害的昆虫种群,本发明的制剂通过限制昆虫媒介所导致的病毒传播还有助于减少由植物病毒造成的损害。对目标环境造成最严重的损害是通过植物病毒(主要是双生病毒(begomovirus))传播发生的,对番茄最具破坏性的一种病毒是番茄黄化卷叶病毒(TYLCV)。
此外,在进行本发明制剂的研究过程中已经发现,非挥发性赋形剂可以增强由挥发性精油所发挥的驱虫、杀虫、杀卵或抗病毒的效果,而且非挥发性赋形剂施加的效果也可以通过挥发性精油所增强。
在一个实施方式中,本发明的农用制剂还具有一定的抗病毒效果,因此也可用作预防、减轻或消除病毒危害的抗病毒剂。
在一个实施方式中,本发明的制剂可用作抗昆虫、诸如银叶粉虱等的驱虫剂,以及用作抗害虫,诸如粉虱、番茄蠹蛾和胡椒象甲等的虫卵的杀卵剂。
在一个具体的实施方式中,提供一种抗粉虱的驱虫制剂,所述驱虫制剂包含胶囊化的香茅油和/或天竺葵油和/或茶树油和/或熏衣草油和/或丁香油,以及含有除虫菊油和芝麻油的液体赋形剂。
在另一个具体的实施方式中,抗粉虱的驱虫制剂包含胶囊化的姜油和含有棉籽油的液体赋形剂。
在又一个具体的实施方式中,提供一种抗番茄蠹蛾的驱虫制剂,所述驱虫制剂包含香茅油和/或天竺葵油和/或茶树油和/或熏衣草油和/或丁香油,以及含有除虫菊油和芝麻油的液体赋形剂。
在另一个实施方式中,提供一种抗胡椒象甲的杀卵制剂或驱虫制剂,所述制剂包含香茅油和/或天竺葵油和/或茶树油和/或熏衣草油和 /或丁香油,以及含有除虫菊油和芝麻油的液体赋形剂。
在另一个实施方式中,可以使用本发明的制剂来防治能够作为传染用病毒媒介的病毒传播型害虫。术语“病毒媒介”指如本文定义并例示的任何能够携带和传播植物病毒(致病生物体)的害虫。
正如在上文中所叙述的,本发明的制剂可以包含通过任何已知方法制备的微胶囊中的任何精油。如此制备的微胶囊可以从反应混合物中回收并再次悬浮在非挥发性赋形剂中,或者处于包含该赋形剂的溶液中。作为另一种选择,可以对任何包含微胶囊的介质(水性介质或其他介质)进行处理,而无需首先从所述介质(其中微胶囊通过使用至少一种非挥发性赋形剂而制造)中分离。在大多数情况下,介质是水。
在优选进行分离的情况下,根据微胶囊的尺寸,所述微胶囊的回收可以如下实现:进行离心分离或过滤,并用多份适宜溶剂(例如蒸馏水)进行洗涤以从表面除去游离的反应物。必要时,还可以在减压下加热微胶囊以进一步除去来自微胶囊内部的任何残留的反应物。优选地,通过在高于构成微胶囊壳的聚合物的平均玻璃化转变温度的温度加热微胶囊以进一步除去来自微胶囊内部的任何残留的反应物。
这些微胶囊可以分散或悬浮在所述非挥发性的液体赋形剂或固体赋形剂中。在一些情况下,所述非挥发性赋形剂是粒状固体,如粉末,为此优选地通过使有效量的干燥微胶囊与赋形剂混合来制备分散体。在一些情况下,所述非挥发性赋形剂是液体,为此优选的通过在赋形剂中机械搅拌有效量的微胶囊来制备悬浮液。本文中使用的术语“有效量”是指根据经验确定的量,施用该有效量的本发明制剂可以对目标环境产生如下文所述的效果。
本发明进一步提供用于制造本发明的制剂的方法,所述方法包括将至少一种胶囊化的挥发性精油分散在非挥发性赋形剂或包含该非挥发性赋形剂的介质中,胶囊化的挥发性精油包含溶解在其中的聚合物,其中非挥发性赋形剂和聚合物增强了挥发性精油的驱虫、杀虫(特别是杀幼虫)、杀卵或抗病毒的效果。
在一个实施方式中,本发明还提供用于制备本发明的制剂的方法,如上文所定义的,所述方法包括将非挥发性赋形剂添加至本发明的胶囊化的挥发性精油的水性溶液中,所述胶囊化的挥发性精油包含溶解在其中的聚合物。
所述胶囊化的挥发性精油的制备方法包括如下步骤:将固体聚合物加入至液体挥发性精油中,并在环境温度下搅拌直至其溶解,最终呈透明或轻微浑浊的溶液。制备该胶囊化的挥发性精油的典型方法包括但不限于:使用常规的顶置式搅拌器或高剪切混合器进行半小时到一小时的混合。可替选地,所述聚合物可被溶解于溶剂中,所述溶剂是精油的共溶剂,包括但不限于丙酮、乙酸乙酯、四氢呋喃、乙醇和丁醇。然后,所述溶剂可以在不蒸发精油的情况下被蒸发掉。
表述“胶囊化的挥发性精油的水性溶液”是利用本领域的技术人员所熟知的任何方法制造的溶液。优选地,包含溶解于其中的聚合物的所述胶囊化的挥发性精油的水性溶液是根据WO04/98767中要求保护的方法制造的制剂,所述方法主要包括将二异氰酸酯或多异氰酸酯溶解在精油中,在包含二胺或多胺和/或二羟基化合物或多羟基化合物的水性溶液中乳化所得的混合物,从而通过界面聚合来实施所述精油的胶囊化,由此在所述精油液滴的周围形成聚脲和/或聚氨酯膜,该膜能够增加精油的稳定性、降低其蒸发速率并控制其在施用于基底后的释放速率。
一旦制得,本发明的农用制剂(例如可溶性固体)可被一直保存直到通过某些方式将其释放至环境中。通常,最方便的是将包含胶囊化精油的悬浮液装入瓶中或罐中,在该情况下将添加剂添加至该悬浮液中是理想的。可以加入诸如密度平衡剂、表面活性剂、增稠剂、杀生物剂、分散剂、防冻剂和盐等“添加剂”,以提高悬浮液的稳定性和施用的简易性。所述添加剂可以以相对于悬浮液的重量计约0.01 重量%-约30重量%的浓度添加至微胶囊的悬浮液中。
所述制剂的制造方法还可包括将离子型或非离子型表面活性剂添加至液体赋形剂或包含液体赋形剂的介质(例如水)中。所述表面活性剂可以在制造微胶囊的过程中加入以便于控制微胶囊的尺寸、或者也可以在制造微胶囊后加入以破坏微胶囊化所生成的凝胶并提供具有流动性的非凝胶制剂。一种特别优选的表面活性剂是十二烷基硫酸钠(SDS)。其中,相对于微胶囊的总重量,所述表面活性剂优选地以0.1%-10%的浓度、更优选地以0.5%-5%的浓度添加。
除了表面活性剂之外的其他添加剂的非限制性实例例如是有助于维持颗粒分离的空间位阻聚合物,例如聚乙烯吡咯烷酮(PVP)、聚乙烯醇(PVA)和聚(乙氧基)壬基苯酚。在一些情况下,理想的添加剂例如当成品微胶囊的溶液与其他农药结合时能够调节该成品微胶囊制剂的pH。可使用用于调节溶液酸性或碱性的常用试剂作为添加剂,其包括例如盐酸、氢氧化钠、碳酸钠和碳酸氢钠。
在本发明的另一个方面中,提供用于控制害虫种群的方法,所述方法包括将本发明的制剂施用于目标环境、或害虫种群或其栖所。
“控制害虫种群”指下列含义中的一种或多种:将部分或全部害虫种群从选定的目标环境中驱散;控制害虫后代;消灭部分或全部害虫种群;消除害虫繁殖地;在害虫侵染之前使目标环境曝露于驱虫制剂或杀虫制剂;在害虫侵染之前或在害虫侵染造成损害之后限制害虫的成虫、幼虫或虫卵;以及减少害虫损害的程度。术语“害虫种群”指成虫种群、幼虫或若虫种群、虫卵种群、混合种群或其任意组合,其中,种群中的害虫的数目可以为一百万至数百万。
本文使用的术语“目标环境”指易于受到害虫种群侵染的原生环境,以及环绕该原生环境的且不包括任何农学或园艺上理想目标的次生环境。所述原生环境例如是下列植物的叶、茎、果实、花、种子或根:稻谷、大麦、小麦、黑麦、燕麦、玉米等谷类;大豆、红豆、蚕豆、豌豆、菜豆、花生等菜豆类和豌豆类;苹果、柑橘、梨、葡萄、桃、李、樱桃、胡桃、栗、扁桃、香蕉、草莓等果树;甘蓝、番茄、菠菜、花椰菜、莴苣、洋葱、青葱、西班牙辣椒、茄子、胡椒等叶菜类和果菜类;胡萝卜、马铃薯、甘薯、芋头、萝卜、莲藕、芜箐、牛蒡、大蒜等根类作物;棉花、亚麻、甜菜、啤酒花、甘蔗、糖用甜菜、橄榄、树胶、咖啡、烟草、茶等加工类作物;南瓜、黄瓜、香瓜、西瓜、甜瓜等葫芦科植物;鸭茅、高粱、猫尾草、三叶草、紫花苜蓿等牧场植物;草坪草;香料作物;花卉及观赏植物;银杏树、樱桃树、金叶树(gold-leaf plant)等园林树木;以及白杉、银杉、松树、斧叶柏树(hatchet-leaved arbor-vitae)、柳杉、日本扁柏等材木。
目标环境还可以是某一害虫种群(为抗该种群可施用本发明的制剂)。
为控制害虫发生,本发明的农用制剂可施用于预期会出现害虫侵染的植物,所述制剂可以直接施用或者以有效控制害虫种群的浓度在适当量的水等中的稀释液或悬浮液的形式施用。例如,为控制害虫在诸如果树、谷类和蔬菜等可食用农产品上的出现,所述制剂可施用于其周围而不直接施用于所述目标环境。所述制剂还适用于叶子处理、种子处理(诸如将种子浸没在种衣用制剂中)、拌入土壤中、灌穴处理、育苗箱施用等。
本发明的农用制剂可以作为单剂存在、存储、包装或施用,其中胶囊化的挥发性精油与非挥发性赋形剂预混合;或者作为双组分制剂,其包含作为第一组分的胶囊化的挥发性精油,例如在单独的容器中或单独施用,以及作为第二组分的非挥发性赋形剂。
因此,提供一种用于控制害虫种群的方法,所述方法包括:将包含至少一种胶囊化的挥发性精油的第一组分施用于目标环境,该挥发性精油包含溶解于其中的聚合物;和将包含非挥发性赋形剂的第二组分施用于所述目标环境。
具体地,所述胶囊化的包含聚合物的挥发性精油通过如下方法制备:将固体聚合物加入至液体挥发性精油中,并在环境温度下搅拌直至其溶解,最终呈透明或轻微浑浊的溶液。典型的制备方法包括但不限于:使用常规的顶置式搅拌器或高剪切混合器进行半小时到一小时的混合,但是根据所使用的聚合物的分子量和所使用的挥发性精油的性质,混合时间可进行适当变化。
所述第二组分的施用可以在施用第一组分后立即进行,或者在第一试剂施用后的任何时间进行。对于具体情况,本领域的技术人员能够根据特定的情况来从本文所公开的用于控制害虫种群的两种方法中选择一个更为合适的方法。
本发明的制剂可以通过本领域技术人员熟知的任何方法递送至目标环境。递送方法包括但不限于:a)通过将用例如水稀释或者未用水稀释的液体制剂施用于种植基底(plant base)而将所述制剂人工或机械地施用于土壤;b)将诸如粉尘剂或可湿性粉剂等粒状制剂施用于种植基底、种植穴或种植行;c)将液体制剂由地面或空中喷施于整个地块或具体选定的区域;d)将制剂埋置在表土层中等。本发明的制剂可施用于收获前或收获后的水果和蔬菜,另外还可施用于储粮害虫、室内害虫、卫生害虫和森林害虫。此外,还可以利用熏蒸、饵料等施用于房屋建筑材料。
精油从微胶囊中释放的模式和随后对处理过的环境产生的效果取决于微胶囊的物理特性。根据本发明,活性挥发性精油是由微胶囊膜封装并由非挥发性赋形剂携载的液体库(liquid depot),所述非挥发性赋形剂可加和地或者协同地增强精油所发挥的效应。据认为,在将制剂递送至目标环境时,由于在微胶囊的内部与外部之间存在挥发性精油的浓度梯度,因此当施用于目标环境时微胶囊的内容物开始释放。该释放过程及其动力学受到例如下列因素的影响:a)微胶囊的干燥;b)与水性介质(例如水或雨水)的接触,从而导致微胶囊壳的缓慢分解;c)高温;和d)直射日光。然而,由于微胶囊的内容物可能自发而独立地释放,因此释放过程也可能不受上述因素的影响。
本发明的农用制剂还可用于控制小庭院、室内苗圃、菜圃和花床中以及少数特定样本(如室内植物)中的害虫种群。
在一个实施方式中,在农用制剂为双组分制剂的情况下,本发明的试剂盒包括第一容器,其容纳有作为第一组分的至少一种胶囊化的挥发性精油;和第二容器,其容纳有作为第二组分的非挥发性赋形剂。可替选地,本发明的试剂盒还可包括如何将所述两种组分施用于目标环境从而实现所需效果的说明书,例如通过已经预先吸收制剂的海绵或布块,或者通过手提式喷雾器而将制剂施用于目标环境。
在试剂盒中,本发明的双组分制剂或单组分制剂可以以固体或液体的形式以及浓缩态或稀释态存在。
以下通过具体的实施例对本发明进行描述。尽管下文描述了本发明的特定实施方式,但本领域的技术人员应当理解本发明并不限于此,在不脱离由所附的权利要求所限定的范围的条件下,可进行各种细节的修改、变更或者组合。
实施例
实施例1:本发明的农用制剂的制备
如下表1-3示出了本发明的农用制剂在施用后3周对银叶粉虱的防治效果。其中,以来自本申请发明人的另一专利EP1845786B1中的制剂1和3-9作为对照制剂(即比较例),而本发明的农用制剂与对照制剂1和3-9的区别在于分别加入含量为5%或10%的聚合物(该百分含量为相对于挥发性精油的重量计),聚合物例如为聚乙酸乙烯酯(PVAc)、聚乙烯吡咯烷酮(PVP)、羟乙基纤维素(HEC)、聚环氧乙烷(PEO)、聚环氧丙烷(PPO)或甲基纤维素。具体地,按照如下方法制备本发明的农用制剂:
本发明的制剂1:将17.5g TDI(甲苯二异氰酸酯)与125g香茅油混合,所述香茅油中溶解有聚乙酸乙烯酯(PVAc)、聚乙烯吡咯烷酮(PVP)或羟乙基纤维素(HEC)6.25g或12.5g,并使用高剪切力搅拌器将其添加至250g含有2.5g PVA的水中。向其中添加70ml 含有27.8g PEG 4000的水。在室温下持续混合2小时。将0.4g黄原胶(rodopol)和2g杀真菌剂(nefocide)加入该分散剂中。为破坏该乳化液的水凝胶特性,加入5g SDS(1%的十二烷基硫酸钠)。其中,可采用上述文中描述的任何方法将聚合物溶解在香茅油中。
然后向在水中的所述微胶囊悬浮液以相对于每89g悬浮液添加 10g的除虫菊油(50%浓缩剂)和lg芝麻油。
除了如下所述的本发明的制剂3-9中的挥发性精油和/或非挥发性赋形剂与制剂1不同以外,使用与制剂1相同的步骤制备本发明的制剂3-9,并使得最终的聚合物含量相对于挥发性精油的重量计为5%或10%:本发明的制剂3,使用天竺葵油代替香茅油;本发明的制剂4,使用茶树油代替香茅油;本发明的制剂5,使用熏衣草油或丁香油代替香茅油;本发明的制剂6,使用24.3ml棉籽油和0.62ml胶囊化姜油;本发明的制剂7,使用23.7ml棉籽油和1.25ml胶囊化姜油;本发明的制剂8,使用22.5ml棉籽油和2.5ml胶囊化姜油;本发明的制剂9,使用21.2ml棉籽油和3.7ml胶囊化姜油。
实施例2:将农用制剂施用至目标环境
以成年银叶粉虱及其卵作为目标害虫,分别测试本发明的制剂以及对照制剂的防治效果。另外还使用以下溶液作为对照:1)水;2) Ultra-Fine油(含有石蜡油的市售品);3)除虫菊油。
将实施例1中制备的各个制剂以及对照制剂分别处理番茄种苗的叶子的顶部或底部,基于处理过的叶片上的被驱除的粉虱成虫的数目以及产卵数目,测定本发明的农用制剂的功效。
如下表1-3所示,在挥发性精油中加入聚合物明显增加了农用制剂的药效和活性。具体而言,在施用后3周本发明的含有溶解的聚合物的制剂比不含有该聚合物的制剂的药效和活性增加了两倍以上。对于添加不同的聚合物:聚乙酸乙烯酯(PVAc)、聚乙烯吡咯烷酮(PVP) 或羟乙基纤维素(HEC)的制剂,其均表现出改善的药效。结果表明,聚合物的添加能够通过影响挥发性精油的释放性能而提高挥发性精油的活性,进而显著地增强了本发明的制剂对于粉虱的防治功效。
发明人还用聚环氧乙烷及其与聚环氧丙烷的共聚物、聚环氧丙烷和甲基醚纤维素替代聚乙酸乙烯酯(PVAc)、聚乙烯吡咯烷酮(PVP) 或羟乙基纤维素(HEC)进行测试也全部实现了较好的防治效果,其结果均显示相对于未包含聚合物的制剂相比,药效和活性增加了两倍以上。
另外,发明人还测试了添加印楝素作为非挥发性赋形剂和2重量%的醋酸生育酚作为抗氧化剂的制剂的药效。结果显示,与不含印楝素和醋酸生育酚的其他制剂相比,使用印楝素和醋酸生育酚的制剂在抵抗粉虱幼虫和若虫方面表现出同样优异的效力。
本发明并限于本申请所述的具体实施方式,所述实施方式意图作为本发明个体方面的单个说明。在不偏离本发明的实质和范围的情况下,可以对本发明进行许多修饰和改变,这对本领域技术人员而言是显而易见的。根据前述描述,除了本文列举的以外,本发明范围内的功能等同的方法和设备,对本领域技术人员而言应是显而易见的。这类修饰和改变落入所附权利要求的范围内。本发明仅受到所附权利要求以及这类权利要求所拥有的等同物的全部范围的限制。应当理解的是,本发明并不限于具体的方法、时间、化合物、组合物或生物体系,它们当然可以变化。还应当理解的是,本文所用的术语仅拥有描述具体实施方式的目的,并非意图限制。
Claims (10)
1.一种农用制剂,所述农用制剂包含:
(i)至少一种胶囊化的挥发性精油,其包含溶解在所述挥发性精油中的聚合物;和
(ii)非挥发性赋形剂,其携载所述至少一种胶囊化的挥发性精油,其中,所述非挥发性赋形剂为固体或液体形式。
2.如权利要求1所述的农用制剂,其中,所述挥发性精油选自由以下组成的组中的一种或多种:肉桂油、雪松油、蓖麻油、丁香油、天竺葵油、柠檬草油、百里香油、姜黄油、冬青油、迷迭香油、小茴香油、小豆蔻油、春黄菊油、芫荽油、枯茗油、莳萝油、薄荷油、欧芹油、罗勒油、樟脑油、香茅油、桉树油、茴香油、姜油、葡萄柚油、柠檬油、橘皮油、橙油、松针油、胡椒油、玫瑰油、甜橙油、红桔油、茶树油、薰衣草油、茶籽油、葛缕子油、蒜油、胡椒薄荷油、洋葱油和留兰香油。
3.如权利要求1或2所述的农用制剂,其中,所述聚合物选自由以下组成的组中的一种或多种:聚乙烯吡咯烷酮、聚环氧乙烷及其与聚环氧丙烷的共聚物、单独的聚环氧丙烷及其共聚物、聚乙酸乙烯酯、其它烷基化物以及多糖。
4.如权利要求3所述的农用制剂,其中,所述多糖为纤维素衍生物。
5.如权利要求4所述的农用制剂,其中,所述纤维素衍生物选自羟基烷基醚和羟基烷基酯中的一种或两种。
6.如权利要求5所述的农用制剂,其中,所述羟基烷基醚为羟乙基纤维素。
7.如权利要求1所述的农用制剂,其中,所述非挥发性赋形剂选自非挥发性植物油、非挥发性液体萜烯或固体萜烯、脂质或其任意组合。
8.如权利要求7所述的农用制剂,其中,所述非挥发性赋形剂选自:芝麻油、除虫菊油、甘油衍生的脂质、甘油脂肪酸衍生物或其任意组合。
9.如权利要求7所述的农用制剂,其中,所述非挥发性赋形剂为天然的昆虫生长调节剂。
10.如权利要求9所述的农用制剂,其中,所述天然的昆虫生长调节剂选自:(1)保幼激素模拟物;(2)蜕皮激素抑制剂;和/或(3)几丁质合成抑制剂。
