Description
Technical Field
The invention belongs to the technical field of disinfection, and relates to a bacteriostatic composition and application thereof.
Background
The transmission pathway of bacterial and viral infections is common as contact transmission, with hand being the most important transmission medium, and hand disinfection is therefore the most important measure for effective prevention and control of nosocomial infections. In the numerous medical and nursing work of medical staff, many work is completed by hands, the number of various bacteria on the hands of the medical staff is often more than that of other people, and the microbial contamination of the hands is serious. Cross-infection caused by hand contamination of medical personnel is one of the major risk factors for nosocomial infections. Daily adherence of medical personnel to high quality hand washing and disinfection can reduce the incidence of nosocomial infections. The problem of nosocomial infection is increasingly prominent, and the nosocomial infection becomes a public health problem which is urgently needed to be solved by hospitals.
At present, the hand disinfectants in the market are various in types, and the disinfection effect and the practicability are good and uneven; conventional hand washing methods use soap and water to remove dirt and harmful microorganisms from the skin surface, however if the hand water is not completely dried through, the residual water can become a harborage of bacteria. This situation poses no serious threat to the general population, but the possibility of infection to the population in the hospital is high, which makes it highly desirable to minimize the risk of contamination.
The quick-drying hand disinfectant is recommended to be a preferred hand disinfection method because of the advantages of quick disinfection, no residue, no irritation, convenient use, working time saving, work efficiency improvement and the like, and the gel type hand quick disinfectant is a hand disinfection product sold in the market in recent years, has the characteristics of good sterilization effect, quick action speed, good skin adaptability, no leakage and no splashing in use, is well received by medical staff, and is considered as an important measure for ensuring that the medical staff keeps hand hygiene constantly. However, with the development of medical technology, higher requirements are put forward on skin disinfection gel used in cooperation, and virus propagation and infection caused by pathogenic bacteria residue are reduced to the maximum extent, so that the traditional skin disinfection gel cannot meet the development of medical requirements, and the improvement of the sterilization effect and the disinfection effect becomes a problem to be solved urgently in development and application.
CN105362212A relates to a hand-washing-free disinfection gel and a preparation method thereof, which comprises 0.05-2% of cationic bactericide, 0.0005-0.02% of anionic surfactant or zwitterionic surfactant, 50-80% of ethanol, 0.05-1.5% of thickening agent, 0.1-1% of skin-care humectant, 0.2-1% of neutralizer and the balance of water, but the invention has no lasting antibacterial effect and needs to be further improved. CN106215142A provides a traditional Chinese medicine medical disinfection gel, which is prepared from the following main raw materials in parts by weight: 5-10 parts of mint, 5-10 parts of cucurbita pepo, 10-20 parts of fructus cnidii, 10-15 parts of coptis chinensis, 5-12 parts of dried ginger, 13-16 parts of golden cypress, 5-9 parts of rhizoma cimicifugae, 4-8 parts of gypsum, 5-8 parts of mung bean, 8-15 parts of exocarpium benincasae, 30-45 parts of hydroxypropyl methyl cellulose, 38-43 parts of polyvinyl alcohol, 10-20 parts of abelmoschus manihot gum and 3-5 parts of glycerol. The invention adds too many traditional Chinese medicine components, has bad smell and bad use experience. CN104547275A discloses a hand essential oil skin-care disinfecting gel, which is prepared from liquid raw materials and solid raw materials, wherein the liquid raw materials comprise the following raw materials in percentage by volume: 55-70% of ethanol, 0.5-1% of an essential oil mixture, 1-5% of glycerol, 0.05-0.1% of triethanolamine and 29-40% of purified water; the solid material comprises triclosan and carbomer, wherein 0.3-0.6g triclosan and 0.15-0.4g carbomer are added into 100ml liquid material. But the invention takes chemical components as main sterilization factors, has single effect and has a non-wide bacteriostatic range.
Therefore, the long-acting antibacterial skin-moistening hand-washing-free disinfection gel is developed, is applied to the fields of daily life and medical care, and has wide application prospect and great market value.

Skin irritation test
Selecting 9 adult healthy SD rats, and shearing the back hairs on two sides of the spine of the experimental animal, wherein the hair removing range is 1.5cm multiplied by 1.5 cm; 0.3mL of the gel prepared in examples 1-3 was applied directly to the skin, covered and fixed with two layers of gauze (2 cm. times.2 cm) and one layer of non-irritating oilpaper, the other side serving as a control. After 2h of stimulation, removing residual test substances by using deionized water; the application is carried out for 7 days 1 time per day, and the result is observed 12h after each application.
The result shows that after the gel is continuously applied for 7 days, no erythema or red swelling is seen on the skin on the administration side, and no abnormality is seen compared with the skin on the control side, which indicates that the gel is mild and non-irritant to the skin.
Disinfection effect monitoring
The disinfection effect detection is carried out on the examples and the comparative examples according to the carrier soaking quantitative sterilization test in the technical specification 2002 edition of the disinfectant, the average value of the killing time and the killing rate of three bacteria including staphylococcus aureus, escherichia coli and pseudomonas aeruginosa is calculated, meanwhile, the bacteriostasis time with the bacteria proliferation rate lower than 10% is detected, and the result is shown in the table 1;
TABLE 1
As can be seen from Table 1, the gels prepared in examples 1-3 have short killing time and high killing rate, wherein the gel prepared in example 1 has the best effect, the killing rate is as high as 97.25%, the bacteriostasis time is as long as 23h, while the gel prepared in comparative example 1 lacks ginger extract, and the killing rate is still higher than 88.43%; the gel of comparative examples 2 to 5 lacks any one of the four components, the bacteriostasis time is obviously shortened, and the killing rate is obviously reduced, while the addition amount of the components in the gel of comparative examples 6 to 9 exceeds the range, the bacteriostasis time and the sterilization rate of the gel are also influenced, which shows that in the technical scheme of the application, the four components of the composition lack one and can not cooperate with each other in a specific range, the synergy is realized, and the bacteriostasis time and the sterilization rate are improved.
Sensory evaluation test
Selecting 120 men and women with skin problems at 20-50 years old, randomly dividing into 12 groups, continuously using the products of examples 1-3 and comparative examples 1-9 for 1 month, and evaluating the using effect, wherein the results are shown in Table 2;
TABLE 2
As can be seen from table 2, the total sensory evaluation scores of the gels prepared according to the technical scheme provided by the invention in examples 1-3 are excellent, and the skin repairing capability is excellent, while the different indexes of the sensory evaluation are reduced in different degrees due to the lack of any component or the exceeding of the addition of the component in comparative examples 1-9, and the core formula component is adopted in comparative example 7, only the amount of trehalose is increased, and the low irritation and good moistening and moisturizing effects can be still realized; the comparative examples 1 to 5 lack any component, so that the active effects of the raw material components cannot be exerted to the maximum extent, and the repair performance of the product is obviously reduced, which shows that the composition provided by the invention is lack of any component and has synergistic effect.
In conclusion, the invention provides the antibacterial composition and the application thereof, the active ingredients are selected, the formula is optimized, the components are matched with each other to promote, synergize and inhibit bacteria widely and durably, and the antibacterial composition is used for preparing the disinfection gel, is convenient and quick, has good disinfection effect, wide application prospect and great market value.
