本发明涉及一种利用微生物发酵制备的多糖。
某些微生物多糖具有较强的免疫调节作用,如真菌中的香菇多糖、茯苓多糖、灵芝多糖、裂褶菌多糖,细菌中脂多糖、黄原胶多糖等。CN1003454号专利公开一种裂褶菌的培养工艺及其利用,它将从腐木上经组织分离的裂褶菌南大843菌株经纯化后,用黄豆粉和葡萄糖等培养基质进行发酵培养,得到含菌丝体及其多糖较高的裂褶菌发酵液,并从发酵液中提取抗肿瘤、促升白、抗菌素增效作用、抗炎的裂褶菌多糖等。CN1003305号专利公开一种生产黄单胞多糖的培养基,它在由Xanthomonas campestris NK-01菌株直接利用淀粉原料发酵时,不同的氮源和无机盐类对多糖的产量影响很大,用豆饼粉和鱼粉作为复合氮源,用CaCO3为无机盐配制的淀粉培养基进行发酵,多糖产量达22.4~26g/L。CN1034086号专利公开一种杂多糖S-657,它是由野油菜黄单胞菌的一个新菌株ATCC53159经发酵而制备的,主要由碳水化合物、约12%蛋白质和7%酰基(以0-乙酰基计算)组成。碳水化合物含有19%葡萄糖醛酸以及中性糖鼠李糖和葡萄糖,两者的摩尔比约为2∶1。
本发明旨在提供一种海洋细菌胞外多糖的生产工艺。
该菌株是从海洋植物浒苔分离获得的土壤杆菌1202菌株(Agrobacterium sp.1202)CCTCC No.M200019。土壤杆菌1202菌株(Agrobacterium sp.1202)已于2000年5月18日保藏在中国典型培养物保藏中心,其简称为CCTCC,保藏编号为CCTCC No.M 200019。土壤杆菌的形态、生理生化等鉴定特征如下(参见J.G霍尔特主编,刘复今等编译,简明第八版伯杰细菌鉴定手册,山东大学出版社,1988年,94~95)。
细胞杆状,0.8×1.5~3.0μm,通过1~4根周生鞭毛运动。如果只有一根鞭毛,则侧生较极生更为常见,没有芽孢,革兰氏阴性,普遍具有纤毛。在含有碳水化合物的培养基上生长通常伴随大量的胞外多糖粘液。菌落无色素,通常平滑,随着培养时间的延长,倾向于产生条纹,但许多菌株产生粗糙菌落。
化能有机营养:呼吸代谢,以分子氧作为最终电子受体。根癌土壤杆菌与放射形土壤杆菌都产生3-酮基乳酸。在几周内慢慢液化明胶或完全不液化。不水解酪蛋白。利用多种简单碳水化合物、有机酸和氨基酸作为碳源,但不利用纤维素、淀粉、琼脂或几丁质。释放有机酸和(或)CO2到培养基中,从而表明了碳水化合物的利用,所释放的酸和CO2,通常在含有少量有机氮化合物的合成培养基表面产生酸反应,但在含有蛋白胨或富含氨基酸的其它物质的培养基中,由于释放碱性物质反应可能被遮盖。在亚硫酸铋琼脂培养基上,一般产生硫化氢。在肉膏或酵母膏蛋白胨琼脂上迅速生长,在24~36小时内,除悬钩于土壤杆菌(A.rubi)外,一般形成浑浊并具有膜。某些种以铵盐、硝酸盐和某些氨基酸作为氨源,其它种需要添加生长因子的氨基酸。正常情况下,氧化酶阳性。
好氧,但在植物组织内,氧压降低的情况下也能生长。最适温度25~30℃,pH范围4.3~12.0,最适pH范围6.0~9.0。
除放射形土壤杆菌外,这个属的菌株当寄生于植物细胞间时,均能在不同植物上刺激茎部增生,在那里,细菌以胞外寄生状态存在。细菌通过原有的病斑或伤口进入寄主组织,菌株缺乏直接侵入寄生植物组织的能力。寄主范围的亲缘关系并没有严格的限制。根据常规生化试验鉴定菌株,组织穿刺接种是很必要的。
DNA的G+C含量范围在59.6~62.8克分子之间,(熔解温度)。
模式种:根癌土壤杆菌[Agrobacterium tumefaciens(Smith and Townsend)Conn]。
土壤杆菌1202菌株最适生长的NaCl浓度为2%~4%,产胞外多糖的NaCl浓度为0.1%~0.3%,具有较高的海洋性。
海洋细菌胞外多糖(以下简称1202多糖)生产工艺步骤为:
1)分离散得生产菌株:土壤杆菌1202菌株可从海洋植物浒苔分离获得。
2)制作种子培养液:将菌株斜面接种子牛肉膏蛋白胨液体培养基中,pH:6.8~7.4,在温度24~28℃、搅拌速度180rpm、通风比1∶1培养48~72小时,获种子培养液。
3)发酵生产:种子培养液以5%~10%的接种量接种于多糖发酵培养基中,培养基组成中各成分的重量比为:蔗糖60~120g,酵母膏2.0~4.5g,玉米浆5~25g,CaCO3 1.0~2.5g,KH2PO4 0.5~1.0g,MgSO4 0.5~1.0g,FeSO4 0.08~0.15g,NaCl 1.0~3.0g,pH7.0~7.4,自来水定容至1000mL。在温度24~28℃、搅拌速度240rpm、通风比1∶0.3~0.5发酵5~7天。
4)提取:将发酵液经离心去除菌体,上清液用2~3倍的乙醇沉淀,沉淀物调节成0.2%~0.4%的多糖水溶液,以多糖液∶氯仿∶正丁醇=30∶10∶1(v/v)沉淀去除蛋白,离心获得上清液,用2~3倍乙醇沉淀,沉淀物经等量的70%乙醇洗涤二遍,在65~70℃烘干,再粉碎后得1202多糖产品。
用上述条件发酵条件生产的1202多糖一般含多糖92%以上,该多糖为水溶性的酸性胞外多糖,由葡萄糖和葡萄糖醛酸组成,两者的含量比例为6∶1。
1202多糖的用途是,它作为单一有效活性组分用于制备增强免疫剂药物等。
由于充分分析了该菌株的海洋性,又筛选了先进的配方与工艺,因此本发明所得的1202多糖,具有良好的增强机体的免疫功能,采用口服方式,在10~100mg/Kg的服用剂量时,能显著地增强机体的淋巴细胞增殖能力、提高抗体生成积数、促进鸡红细胞吞噬能力、提高迟发型超敏反应能力以及NK细胞产量。具有细胞免疫、体液免疫和非特异性的免疫增强活性。1202多糖的LD50(小鼠口服)≥15g/Kg,属无毒级产品,可作为单一有效活性组分用于制备增强免疫作用的药物、保健品或直接作为食品添加剂的原料,可制成冲剂、片剂、胶囊或液体饮品。其次本工艺的产量高,成本低,含多糖在92%以上。
以下给出1202多糖的免疫增强及抗肿瘤活性实验
1)1202多糖的免疫增强活性测定
Bal B/C小鼠雌雄各半分组,每组10只,腹腔注射不同浓度的1202多糖。连续注射多糖8天后,以足跖肿胀法测定小鼠迟发型超敏反应;血凝法测定血清溶血素抗体;称重法测定小鼠免疫器官的重量指数;鸡红细胞吞噬实验测定吞噬能力;乳酸脱氢酶测定法测定NK细胞活性;MTT法测定脾淋巴细胞的转化能力,结果见表1,2。

2)1202多糖对S-180实体瘤的抑制活性
Bal B/C小鼠雌雄各半分组,每组10只,以105S-180瘤细胞/mL荷瘤小鼠后,次日腹腔注射不同浓度的1202多糖。连续给药10天后,分别称重测定实验组和对照组的瘤重,以对照组瘤重-实验组瘤重/对照组平均瘤重计算抑瘤率。结果见表3。

实施例1:
将土壤杆菌1202菌株斜面菌种接种于牛肉膏蛋白胨液体培养基中(pH:6.8~6.9),温度24~25℃、搅拌速度180rpm、通风比1∶1培养48~50小时,得种子培养液。种子培养液以5%~6%的接种量接种于多糖发酵培养基中(培养基组成:蔗糖60g,酵母膏4.5g,玉米浆25g,CaCO3 1.0g,KH2PO4 1.0g,MgSO4 0.5g,FeSO4 0.15g,NaCl 2.0g,pH7.0,自来水定容至1000mL。)24~25℃、搅拌速度240rpm、通风比1∶0.3~0.5,发酵7天。发酵液经3000rpm离心30min去除菌体,上清液用2.5倍的乙醇沉淀,沉淀物调节成0.3%的多糖水溶液,以多糖液∶氯仿∶正丁醇=30∶10∶1(v/v)沉淀去除蛋白,离心获得的上清液用3倍乙醇沉淀,沉淀物经70%乙醇洗涤二遍,70℃烘干,粉碎后得含多糖92%以上的多糖产品。
实施例2:
土壤杆菌1202菌株斜面接种于牛肉膏蛋白胨液体培养基中(pH7.2),温度26℃、搅拌速度180rpm、通风比1∶1培养72小时,得种子培养液。种子培养液以8%的接种量接种于多糖发酵培养基中(培养基组成:蔗糖100g,酵母膏2.0g,玉米浆5g,CaCO3 2.5g,KH2PO40.8g,MgSO4 0.8g,FeSO4 0.5g,NaCl 1.0g,pH7.2,自来水定容至1000mL)28℃、搅拌速度240rpm、通风比1∶0.3~0.4发酵6天。发酵液经2800rpm离心35min去除菌体,上清液用3倍的乙醇沉淀,沉淀物调节成0.2%的多糖水溶液,以多糖液∶氯仿∶正丁醇=30∶10∶1(v/v)沉淀去除蛋白,离心获得的上清液用2倍乙醇沉淀,沉淀物经等量的70%乙醇洗洗二遍,65℃烘干,粉碎得含多糖92%以上的多糖产品。
实施例3:
土壤杆菌1202菌株斜面接种于牛肉膏蛋白胨液体培养基中(pH7.4),28℃、搅拌速度180rpm、通风比1∶1培养56小时,得种子培养液。种子培养液以10%的接种量接种于多糖发酵培养基中(培养基组成:蔗糖120g,酵母膏3.5g,玉米浆15g,CaCO31.5g,KH2PO40.5g,MgSO4 1.0g,FeSO4 0.08g,NaCl 3.0g,pH7.4,自来水定容至1000mL)26℃、搅拌速度240rpm、通风比1∶0.4~0.5发酵5天。发酵液经3500rpm离心28min去除菌体,上清液用2倍的乙醇沉淀,沉淀物调节成0.4%的多糖水溶液,以多糖液∶氯仿∶正丁醇=30∶10∶1(v/v)沉淀去除蛋白,离心获得的上清液用2.5倍乙醇沉淀,沉淀物经70%的乙醇洗涤二遍,68℃烘干,粉碎后得含多糖92%以上的多糖产品。
实施例4:1202多糖制备免疫增强剂药物的实例
1)冲剂:取1202多糖(上述工艺制成的原料)10%、乳糖61.5%、蔗糖25%、柠檬酸1.5%、甜味素2%。各原料预先粉碎,过100目筛网后,在混合器中混合均匀。以粉料∶70%食用酒精=1∶2.5(W/V),边搅拌边缓缓加入食用酒精,使混合均匀。在造粒机上过16~20目筛网造粒。取湿粒摊于烘盘上于65℃烘干至含水量<6%。整粒(取烘干的颗粒过20目筛网,取筛下物过80目筛网,得筛上物),分装于密闭的食用包装容器中,即为1202多糖冲剂成品。
2)片剂:取1202多糖45.5%、乳糖50.0%、柠檬酸1.5%、甜味素3%。各原料预先粉碎,过100目筛网后,在混合器中混合均匀。以粉料∶70%食用酒精=1∶2.5(W/V),边搅拌边缓缓加入食用酒精,使混合均匀。在造粒机上过16~20目筛网造粒。取湿粒摊于烘盘上于65℃烘干至含水量<4%。整粒(取烘干的颗粒过20目筛网,取筛下物过80目筛网,得筛上物),加入1%的硬脂酸镁,混合均匀,于压片机上压片,分装于密闭的食用包装容器中,即为1202多糖冲剂成品。
3)胶囊:取1202多糖45.5%、乳糖50.0%、柠檬酸1.5%、甜味素3%。各原料预先粉碎,过100目筛网后,在混合器中混合均匀。以粉料∶70%食用酒精=1∶2.5(W/V),边搅拌边缓缓加入食用酒精,使混合均匀。在造粒机上过16~20目筛网造粒。取湿粒摊于烘盘上于65℃烘干至含水量<4%。整粒(取烘干的颗粒过20目筛网,取筛下物过80目筛网,得筛上物),填充胶囊,胶囊分装于密闭的食用包装容器中或压膜于铝塑片中,即为1202多糖冲剂成品。
4)液体饮品(饮料或口服液):以1202多糖∶蔗糖∶柠檬酸∶水=0.5∶5∶0.15∶94.35(W/W)称取原料。1202多糖预先用少量热水充分溶解后,分别加入蔗糖、柠檬酸、水。充分混合溶解后,定容。分装于密闭的耐高温的食用包装容器中,121℃灭菌20分钟。即为1202多糖液体饮品。
上述各种制品中,还可根据口感需要,加入适量的食用香料(如水果香精等),制成各种水果风味的冲剂、片剂或液体饮品。
Claims (1)
1、一种海洋细菌胞外多糖的生产工艺,其特征在于所涉及的微生物为土壤杆菌1202菌株(Agrobacterium sp.1202),保藏单位简称为CCTCC,保藏编号为CCTCCNO.M200019;海洋细菌胞外多糖的生产工艺步骤为1)分离获得生产菌株-土壤杆菌1202菌株;2)制作种子培养液:将菌株斜面接种于牛肉膏蛋白胨液体培养基中,pH:6.8~7.4,在温度24~28℃、搅拌速度180rpm、通风比1∶1培养48~72小时,获种子培养液;3)发酵生产:种子培养液以5%~10%(V/V)的接种量接种于多糖发酵培养基中,培养基组成中各成分的重量比为:蔗糖60~120g,酵母膏2.0~4.5g,玉米浆5~25g,CaCO31.0~2.5g,KH2PO40.5~1.0g,MgSO40.5~1.0g,FeSO40.08~0.15g,NaCl1.0~3.0g,pH7.0~7.4,自来水定容至1000mL,在温度24~28℃、搅拌速度240rpm、通风比1∶0.3~0.5发酵5~7天;4)提取:将发酵液经离心去除菌体,上清液用2~3倍(V/V)的乙醇沉淀,沉淀物调节成0.2%~0.4%(V/V)的多糖水溶液,以多糖液∶氯仿∶正丁醇=30∶10∶1(V/V)沉淀去除蛋白,离心获得上清液,用2~3倍(V/V)乙醇沉淀,沉淀物经等量的70%乙醇洗涤2遍,在65~70℃烘干,再粉碎后得多糖产品。
