本发明公开了一种环糊精葡萄糖基转移酶突变体及其应用,属于酶工程技术领域。本发明对来源于Bacillus circulans的环糊精葡萄糖基转移酶的234位的甲硫氨酸进行定点突变,M234I、M234A、M234L和M234V突变体对麦芽糖的受体亲和力均比野生型提高了45.6%、34.7%、30.3%和36.9%,本发明的环糊精葡萄糖基转移酶突变体能够应用于海藻糖制备中,使海藻糖的转化率得到进一步的提高。

本发明涉及一种环糊精葡萄糖基转移酶突变体及其应用,尤其是一种在歧化反应中对麦芽糖的受体亲和力提高的环糊精葡萄糖基转移酶突变体及其应用,属于基因工程和酶工程技术领域。
背景技术
海藻糖(Trehalose)由两个吡喃葡萄糖分子通过α,α-1,1-糖苷键连接而成,是一种稳定的非还原性二糖,同时具有独特的高保湿性,高度安全性和有良好的稳定性,被广泛应用于医药、食品、化妆和农业等领域。
自20世纪80年代后,各国相继开展了海藻糖对生理功能作用的研究,现已成为国际上开发研究的主要低聚糖之一。
海藻糖的合成主要有磷酸化酶法、海藻糖合成酶法和双酶法这三种方法。其中,以玉米淀粉为底物,使用双酶法生产海藻糖转化率高达80%以上,其作用机理为以淀粉为底物经普鲁兰酶脱支为麦芽糊精、麦芽寡糖基海藻糖合成酶作用于底物还原性末端的α-1,4-糖苷建,通过分子内转糖苷作用将α,α-1,1-糖苷键转为α,α-1,1-糖苷键,形成中间产物麦芽寡糖基海藻糖,麦芽寡糖基海藻糖水解酶则专一的内切该中间产物中麦芽寡糖基与海藻糖相连的α,α-1,1- 糖苷键,使之分解产生海藻糖和减少两个葡萄糖单位的新麦芽寡糖,减少两个葡萄糖单位的新麦芽寡糖作为新底物进行下一轮反应,如此反复交替进行两种酶反应就可以将麦芽寡糖转化成主要为海藻糖,及少量葡萄糖、麦芽糖、麦芽三糖的产物。双酶法生产海藻糖以淀粉为底物,具有低成本的优点。但是麦芽寡糖基海藻糖合成酶对麦芽四糖和麦芽三糖的亲和性较低,导致反应液中的小分子麦芽寡糖难以利用,使得在工业生产中淀粉底物的利用率降低,尤其是当使用大米淀粉为底物时,体系中的无法被利用的小分子糖较玉米淀粉更多,这无形之中提高了生产成本。
环糊精葡萄糖基转移酶(Cyclodextrin glycosyltransfer,CGT酶,EC2.4.1.19)是一种胞外酶,同时也是一种多功能酶,能催化三种转糖基反应(歧化、环化和耦合反应)和一种水解反应。有研究表明,在多酶体系复配生产海藻糖的酶反应中,CGTase的存在可以将小分子糖的糖链接长,进而使之更容易被利用于合成海藻糖,该复配方法提高了底物利用率。而分析酶转化结果发现,反应结束后,体系中仍存在较多的麦芽糖,因此使用一种在歧化反应中对麦芽糖受体的亲和性高的CGTase能够有助于利用这一部分麦芽糖。目前国内外文献中关于 CGTase对麦芽糖受体的亲和性的报道较少,仅Bacillus circulans和Paenibacillus sp.有相关报道,其中Bacillus circulans来源的CGTase对麦芽糖的受体亲和性远高于Paenibacillus sp.。若能进一步提升Bacillus circulans CGTase对麦芽糖为受体的亲和性,进一步利用体系中的麦芽糖,就可以进一步提高底物利用率,从而继续降低成本,这对海藻糖的双酶法生产具有重要意义。
发明内容
基于上述现状,本发明利用基因工程和酶工程的手段,提高环糊精葡萄糖基转移酶在歧化反应中对麦芽糖的受体亲和力,进一步提高底物利用率,从而提高海藻糖的产量。
本发明的第一个目的是提供了具有酶活提高的环糊精葡萄糖基转移酶的突变体,包含了将来源于环状芽孢杆菌(Bacillus circulans)的环糊精葡萄糖基转移酶的第234位的氨基酸进行取代得到的突变体。这些突变体与其亲代的环糊精葡萄糖基转移酶相比,歧化反应中对麦芽糖的受体亲和力提高。

在本发明的一种实施方式中,所述来源于环状芽孢杆菌(B.circulans)的环糊精葡萄糖基转移酶的氨基酸序列如SEQ ID NO.2所示。编码所述来源于环状芽孢杆菌(B.circulans) 的环糊精葡萄糖基转移酶基因的核苷酸序列如SEQ ID NO.1所示。
在本发明的一种实施方式中,所述突变体是将第234位置的甲硫氨酸(Met)变成异亮氨酸(Ile),突变体命名为M234I。
在本发明的一种实施方式中,所述突变体是将第234位置的甲硫氨酸(Met)变成丙氨酸 (Ala),突变体命名为M234A。
在本发明的一种实施方式中,所述突变体是将第234位置的甲硫氨酸(Met)变成亮氨酸 (Leu),突变体命名为M234L。
在本发明的一种实施方式中,所述突变体是将第234位置的甲硫氨酸(Met)变成缬氨酸 (Val),突变体命名为M234V。
本发明的第二个目的是提供一种环糊精葡萄糖基转移酶突变体的制备方法,包括如下步骤:
(1)根据确定的突变位点,设计定点突变的突变引物,以携带环糊精葡萄糖基转移酶基因的载体为模板进行定点突变,以构建含有编码突变体的基因的质粒载体。
(2)将突变体质粒转化进宿主细胞。
(3)挑选阳性克隆进行发酵培养,离心收集发酵上清,发酵上清液即为环糊精葡萄糖基转移酶突变体的粗酶液。
所述质粒载体,在本发明的一种实施方式中,是pET系列、pUC系列或pGEX中的任意一种。
所述宿主细胞为细菌或真菌细胞。
所述的细菌为革兰氏阴性菌或革兰氏阳性菌。
本发明的第三个目的是提供一种将环糊精葡萄糖基转移酶突变体在多酶偶联生产海藻糖中的应用。
本发明提供了上述一种歧化反应中对麦芽糖的受体亲和力提高的环糊精葡萄糖基转移酶突变体或上述基因或上述重组质粒或上述宿主细胞在生产海藻糖方面的应用。
有益效果:
本发明构建了一种在歧化反应中对麦芽糖的受体亲和力提高的环糊精葡萄糖基转移酶突变体。M234I、M234A、M234L和M234V突变体对麦芽糖的受体亲和力均比野生型提高了 45.6%、34.7%、30.3%和36.9%,本发明的环糊精葡萄糖基转移酶突变体能够应用于海藻糖制备中,使海藻糖的转化率得到进一步的提高。
具体实施方式
本发明的实施例仅作为本发明内容的进一步说明,不能作为本发明的限定内容或范围。
下述实施例中涉及的培养基和检测方法如下:
LB培养基(g·L-1):胰蛋白胨10,酵母粉5,氯化钠10。
TB培养基(g·L-1):胰蛋白胨12,酵母粉24,甘油5,KH2PO4 2.31,K2HPO4·3H2O16.43,甘氨酸7.5。
歧化活力测定方法:以50mmol/L pH 5.5的磷酸盐缓冲液为溶剂,分别配置12mM的EPS 和20mM麦芽糖溶液,各取300μL的12mM EPS和20mM麦芽糖溶液置于50℃水浴锅中预热,加入100μL稀释后的酶液,精确反应10min后,立刻煮沸10min终止反应,加入30 μL稀释后的粗酶液,精确反应75min,于沸水中煮沸10min,冷却,加入100μLα-葡萄糖苷酶和100μL去离子水,混匀,于60℃水浴锅反应60min以上,加入100μL 1M Na2CO3溶液,混匀,最后于400nm测定吸光值。歧化活力定义为每分钟转化一微摩尔EPS的酶量。
对麦芽糖受体亲和力测定方法:用50mmol/L pH 5.5磷酸缓冲液溶解底物,分别配成0.25 mM,0.5mM,0.75mM,1mM,1.5mM,2mM,3mM,5mM,10mM,20mM,50mM, 100mM的麦芽糖溶液,将底物置于50℃水浴锅中预热,并以此为底物,根据上述歧化活力测定方法,测定酶在此麦芽糖浓度下的歧化活力。使用GraphPad Prism软件进行拟合分析,结合拟合分析结果计算得到Km值,即表示歧化活力中对以麦芽糖为受体的亲和力。
实施例1:野生型环糊精葡萄糖基转移酶的表达
从实验室前期保藏的甘油管中,接种Cgt/pET20b(+)/BL21(DE3)(杨玉路,王蕾,陈晟, 等.重组β-环糊精葡萄糖基转移酶生产β-环糊精的工艺条件优化[J].生物技术通报,2014, 8:175-181.)于LB液体培养基(含100mg/L氨苄青霉素)生长8h,按5%接种量将种子接入TB液体发酵培养基(含100mg/L氨苄青霉素)。大肠杆菌在25℃摇床培养发酵48h后,将一定体积发酵液于4℃、12000rpm离心15min,取发酵上清,即为发酵粗酶液。
实施例2:环糊精葡萄糖基转移酶单突变体的制备及表达
(1)突变体的制备
根据Bacillus circulans环糊精葡萄糖基转移酶的基因序列,分别设计并合成引入M234I、 M234A、M234L、M234V突变的引物,对环糊精葡萄糖基转移酶基因Cgt进行定点突变,分别测序确认环糊精葡萄糖基转移酶突变体的编码基因是否正确;将携带突变体基因的载体导入大肠杆菌中进行表达,得到单突变环糊精葡萄糖基转移酶。
定点突变体编码基因的PCR扩增:利用快速PCR技术,以携带编码野生型环糊精葡萄糖基转移酶的基因的表达载体Cgt/pET-20b(+)为模板。
引入M234I突变的定点突变引物为:
核苷酸序列为SEQ ID NO.3的正向引物:

核苷酸序列为SEQ ID NO.4的反向引物:

PCR反应体系均为:正向引物(20μM)0.5μL,反向引物(20μM)0.5μL,dNTPMix 4 μL,5×PS Buffer 10μL,2.5U/μL的PrimeStar聚合酶0.5μL,模板0.5μL,加双蒸水补齐50 μL。
PCR扩增条件为:94℃预变性4min;随后进行25个循环(94℃10s,55℃5s,72℃7min 50s);最后72℃延伸10min,4℃保温。PCR产物用1%琼脂糖凝胶电泳进行检测。
将上述验证正确的PCR产物进行Dpn I消化,转入E.coli JM 109感受态细胞,转化产物涂布于含100mg/L氨苄青霉素的LB平板上,经37℃过夜培养,平板上挑取2个单菌落,接入LB液体培养基,8h后提取质粒并测序,结果正确。
(2)突变体的表达
突变体表达过程如实施例1所述。
实施例3:环糊精葡萄糖基转移酶歧化活力及其对麦芽糖受体亲和力分析
将实施例1和实施例2所得的发酵上清粗酶液进行酶活测定。野生型环糊精葡萄糖基转移酶(WT)和突变体酶活力和酶动力学参数Km列于表1,结果表明,所有突变体的对麦芽糖的受体亲和力均高于野生型。

Claims (8)
1.一种环糊精葡萄糖基转移酶的突变体,其特征在于,将来源于环状芽孢杆菌(Bacillus circulans)的环糊精葡萄糖基转移酶的第234位的氨基酸进行取代得到的突变体;所述来源于环状芽孢杆菌(Bacillus circulans)的环糊精葡萄糖基转移酶的氨基酸序列如SEQ ID NO.2所示;
所述突变体是将第234位置的甲硫氨酸(Met)变成异亮氨酸(Ile),突变体命名为M234I;
或,所述突变体是将第234位置的甲硫氨酸(Met)变成丙氨酸(Ala),突变体命名为M234A;
或,所述突变体是将第234位置的甲硫氨酸(Met)变成亮氨酸(Leu),突变体命名为M234L;
或,所述突变体是将第234位置的甲硫氨酸(Met)变成缬氨酸(Val),突变体命名为M234V。
2.编码权利要求1所述环糊精葡萄糖基转移酶的突变体的基因。
3.携带权利要求2所述环糊精葡萄糖基转移酶的突变体的基因的载体或重组细胞。
4.制备权利要求1所述环糊精葡萄糖基转移酶的突变体的方法,其特征在于,包括如下步骤:
(1)根据确定的突变位点,设计定点突变的突变引物,以携带环糊精葡萄糖基转移酶基因的载体为模板进行定点突变;构建含有编码突变体的基因的质粒载体;
(2)将突变体质粒转化进宿主细胞;
(3)挑选阳性克隆进行发酵培养,离心,收集发酵上清液即为突变体环糊精葡萄糖基转移酶的粗酶液。
5.权利要求1所述环糊精葡萄糖基转移酶的突变体在生产海藻糖中的应用。
6.权利要求2所述环糊精葡萄糖基转移酶的突变体的基因在生产海藻糖中的应用。
7.权利要求3所述环糊精葡萄糖基转移酶的突变体的基因的载体或重组细胞在生产海藻糖中的应用。
8.生产海藻糖的酶制剂,其特征在于,包括权利要求1所述环糊精葡萄糖基转移酶的突变体。
