本发明涉及一种白及粉快速鉴别方法,其具体步骤包括:粉碎干燥白及至50~100目,所获得白及粉作为标准样品备用;取市场白及粉,直接作为检测样品,分别取一定量的标准样品和检测样品,加入沸水,搅拌冷却50~60℃,再加入2~5mL淀粉酶溶液,使淀粉酶的终浓度为500U以上,50~60℃下处理30~60min,4000~6000rpm离心3~10min得酶解液;用葡萄糖氧化酶法或试纸显色法进行测定酶解前后溶液中葡萄糖的含量,市场检测样品中葡萄糖含量超出标准样品的20%或显色时色泽度差异明显,判定为该白及粉中人为掺杂淀粉。

技术领域
本发明属于中药材检测的技术领域,涉及一种白及粉快速鉴别方法,更具体地涉及到对白及粉中掺淀粉的快速鉴别方法。
背景技术
白及(Bletilla striata),又名白给、连及草、紫兰根,为兰科白及属多年生草本植物,白及假鳞茎是常用中药,含淀粉、葡萄糖.尚含挥发油、黏液质、白及甘露聚糖。入药内服主治肺结核咯血、支气管扩张咯血、胃溃疡吐血、尿血、便血等症;外用治外伤出血、烧烫伤、手足皲裂等。近年来研究发现,白及假鳞茎中的白及胶是主要药效部位,其主要成分是白及水溶性多糖(其分子构成为葡萄糖与甘聚糖相互连接的聚合物)。白及水溶性多糖的理化性质与阿拉伯胶、西黄蓍胶类似,但黏性特殊。
白及胶目前广泛用于胃、食道、十二指肠等消化道溃疡,鼻腔、口腔等呼吸道粘膜溃疡,子宫、阴道等妇科疾病的粘膜溃疡,以及皮肤损伤、疮疡、烧烫伤等临床治疗,总有效率在90%以上,有些病例类型的治愈有效率高达100%,而且治愈效果受到年龄大小、病程长短、个体差异及不同职业等因素的影响较小。
白及胶属于分子量较小的天然聚合物,但无抗原性、无致热原性,与动物机体组织较好地相容,并可被缓慢地降解吸收。医药工作者利用白及胶的优良性能来修补破损组织,使某些外科疾病治疗无须手术也能解决。例如,朱志安等将白及胶与经过特殊处理的脱钙骨制成复合性生物颅骨材料,用于修复颅骨缺损,在相容性、可塑性、自由定型性,以及消毒硬化性方面均取得非常满意效果。对某些溃疡穿孔、吻合术后的痰管病例,运用口服白及胶或白及粉治疗,均有较好的填充修复作用。
从2016年以来,白及胶在皮肤美容的广泛应用,市场上开始白及粉直接用于护肤,再加白及价格波动很大,淀粉本身便宜、易得、分散好、吸潮性一般等优点,一些白及粉厂家向其中加入大量淀粉,虽然白及本身含有淀粉,但还会影响其使用效果。因此本发明提供一种用于白及粉掺杂淀粉的快速鉴别方法,便于消费者能快速识别其白及含量。
发明内容
为了防止白及粉人为掺淀粉,影响其使用效果,本发明提供一种用于白及粉掺杂淀粉的快速鉴别方法。
本发明第一个目的是提供一种白及粉比色快速鉴别方法,其具体步骤包括:
(1)粉碎干燥白及至50~100目,所获得白及粉作为标准样品备用;
(2)取市场白及粉,直接作为检测样品,分别取一定量的标准样品和检测样品,加入沸水,搅拌冷却50~60℃,再加入2~5mL淀粉酶溶液,使淀粉酶的终浓度为500U以上,50~60℃下处理30~60min,4000~6000rpm离心3~10min得酶解液,其中样品量g与沸水体积mL比为0.5~10:100;
(3)用葡萄糖氧化酶法进行测定酶解前后溶液中葡萄糖的含量,市场检测样品中葡萄糖含量超出标准样品的20%,判定为该白及粉中人为掺杂淀粉。
在一个实施方案中,所述步骤(2)中淀粉酶由津市新型发酵有限责任公司提供,也可以从市场上购买,淀粉酶使用终浓度为10000U以上(其中U是指国际酶活力单位)。所用淀粉酶只能水解葡萄糖与葡萄糖连接的糖苷键。
在一个实施方案中,所述步骤(3)中葡萄糖氧化酶法进行测定葡萄糖含量:取200μL酶解液,用Tris-HCl缓冲液(10mM,pH 9)稀释20~50倍,作为测试样品。将20μL葡萄糖氧化酶溶液(10mg/mL)与180μL测试样品混合,60℃下反应30min,然后加入0.05mL的二硫化钼溶液(18mg/L)、0.05mL邻苯二胺(0.3M)和0.2mL Tris-HCl缓冲溶液(10mM,pH 9),混匀,放置30min后,利用紫外-可见分光光度法在450nm波长处测定吸光度,进行计算其葡萄糖含量。
本发明第二个目的是提供一种白及粉试纸快速鉴别方法,其具体步骤包括:
(1)粉碎干燥白及至50~100目,所获得白及粉作为标准样品备用;
(2)取市场白及粉,直接作为检测样品,分别取一定量标准样品和检测样品,加入沸水,搅拌冷却50~60℃,再加入2~5mL淀粉酶溶液,使淀粉酶的终浓度为300U以上,50~60℃下处理50~60min,得酶解液,取1mL,稀释10~30倍作为待测液,其中样品量g与沸水体积mL比为0.5~10:100;
(3)直接用所购买的葡萄糖试纸进行测定酶解前后待测液中葡萄糖的含量,若市场检测样品中其试纸显色比标准样品的色泽度深,直接判定为该白及粉中人为掺杂淀粉。
所述葡萄糖试纸可以直接采用尿糖检测试纸,如上海艾测电子科技有限公司等生产厂家。
技术效果
1、本发明简单、方便,比较快速分辨白及粉中掺杂淀粉的鉴别方法。
2、本发明只需要在尿糖试纸基础上增加淀粉酶试剂,降低了其检测成本,方便使用者进行快速鉴别。
具体实施方式
下面,本发明将用实施例进行进一步的说明,但是它并不限于这些实施例的任一个或类似实例。
实施例1:比色法进行鉴别白及粉
干燥白及1kg,粉碎至50~100目,所获得白及粉作为标准样品备用取100克白及粉再加入100克淀粉作为检测样品,另外白及粉作为标准样品。
分别取1g标准样品和检测样品,加入沸水100mL,搅拌冷却50℃,再加入5mL淀粉酶溶液,使淀粉酶的终浓度为500U以上,50℃下处理30min,6000rpm离心5min得酶解液。
取200μL酶解液,用Tris-HCl缓冲液(10mM,pH 9)稀释20~50倍,作为测试样品。将20μL葡萄糖氧化酶溶液(10mg/mL)与180μL测试样品混合,60℃下反应30min,然后加入0.05mL的二硫化钼溶液(18mg/L)、0.05mL邻苯二胺(0.3M)和0.2mL Tris-HCl缓冲溶液(10mM,pH 9),混匀,放置30min后,利用紫外-可见分光光度法在450nm波长处测定吸光度,进行计算其葡萄糖含量,实验结果为检测样品中葡萄糖含量达到60.046g/100样品,标准样品中葡萄糖含量达到37.709g/100样品。
鉴别结果为:检测样品中葡萄糖含量超出标准样品的59.23%,判定为该白及粉中人为掺杂淀粉。
实施例2:试纸法进行鉴别白及粉
干燥白及1kg,粉碎至50~100目,所获得白及粉作为标准样品备用取100克白及粉再加入100克淀粉作为检测样品,另外白及粉作为标准样品。
分别取1g标准样品和检测样品,加入沸水100mL,搅拌冷却50℃,再加入5mL淀粉酶溶液,使淀粉酶的终浓度为500U以上,50℃下处理30min,6000rpm离心5min得酶解液,取1mL,稀释20倍作为待测液。
直接用所购买的葡萄糖试纸进行测定,将其试纸直接浸入待测液,室温放置30S,取出,标准样品所制得的待测液显黄绿色,检测样品所制得的待测液显砖红色,说明检测样品中其试纸显色比标准样品的色泽度要深,直接判定为该白及粉中人为掺杂淀粉。

1.一种白及粉比色快速鉴别方法,其具体步骤包括:
(1)粉碎干燥白及至50~100目,所获得白及粉作为标准样品备用;
(2)取市场白及粉,直接作为检测样品,分别取一定量的标准样品和检测样品,加入沸水,搅拌冷却50~60℃,再加入2~5mL淀粉酶溶液,使淀粉酶的终浓度为500U以上,50~60℃下处理30~60min,4000~6000rpm离心3~10min得酶解液,其中样品量g与沸水体积mL比为0.5~10:100;
(3)用葡萄糖氧化酶法进行测定酶解前后溶液中葡萄糖的含量,市场检测样品中葡萄糖含量超出标准样品的20%,判定为该白及粉中人为掺杂淀粉。
2.根据权利要求1的方法,所述步骤(2)中淀粉酶由津市新型发酵有限责任公司提供,也可以从市场上购买,淀粉酶使用终浓度为10000U以上。
