Abstract
本发明涉及农药复配领域,主要涉及一种含吩嗪‑1‑甲酰胺和乙蒜素的复配杀菌剂及其应用。本发明采用生长速率法测试了吩嗪‑1‑甲酰胺和乙蒜素单剂及其复配剂的毒力,同时依据共毒系数法评价了复配剂的增效作用。结果显示:吩嗪‑1‑甲酰胺和乙蒜素均对龙牙百合灰霉病菌有抑菌活性,其EC50分别为21.0859μg/mL和138.9683μg/mL。将两者进行复配,当吩嗪‑1‑甲酰胺和乙蒜素的重量比为1:1时,其共毒系数为172,大于120,具有明显的增效作用。离体叶片防治效果试验结果显示:吩嗪‑1‑甲酰胺和乙蒜素以1:1复配,对龙牙百合灰霉病的防治效果可达83.85%,具有进一步研制新农药的潜力。

技术领域
本发明属于农药复配领域,特别涉及一种含吩嗪-1-甲酰胺和乙蒜素的复配杀菌剂,同时涉及该杀菌剂在防治龙牙百合灰霉病中的应用。
背景技术
龙牙百合(Lilium brownii var.viridulum)属百合科百合属,是多年生草本植物。龙牙百合鳞茎片似龙牙,故称之为“龙牙百合”。龙牙百合中含有丰富的对人体有益的微量元素,且可溶性有害重金属等含量较少,是集食品、药品于一身的绿色保健食品。龙牙百合在生长的过程中常受多种因素的干扰,如:温度、湿度、肥料、光照、病虫草害等,其中龙牙百合灰霉病(简称:百合灰霉病)是影响龙牙百合产量和品质的重要因素。该病主要是由灰葡萄孢菌引起的真菌性病害,由于其寄主范围广、易变异、适生性强、繁殖快等特点,防治十分困难。生产上为有效控制该病的发生和危害常采用农业防治、生物防治和化学防治,其中化学防治是主要手段。目前,常用的化学农药主要有腐霉利、嘧霉胺、乙霉威等,但长期大量使用存在病菌产生抗药性的风险。前人的研究结果显示:灰霉病菌已对腐霉利等多种化学药剂产生抗药性,因此,探索防治灰霉病的新方法迫在眉睫。
将两种作用机理不同的药剂复配不仅可延长药剂的使用寿命,而且还可延缓病菌的抗药性,是解决病菌产生抗药性的重要方法。吩嗪-1-甲酰胺是一种天然的吩嗪类化合物,具有广谱的抗真菌活性,本课题组研究结果显示其作用机理主要通过抑制细胞壁合成酶和线粒体复合酶活性达到控制菌丝的目的。乙蒜素是一种植物仿生农药,一般制剂的含量是80%的乙蒜素,是高效无公害广谱仿生杀菌剂,兼具植物生长调节作用,能促进萌芽、提高发芽率、增加产量和改善品质,其作用机理是与菌体内含硫基的物质作用,从而抑制菌体的正常代谢。由于两者作用机理不同,且尚未见二者复配的报道,因此,本发明通过测定该二种单剂及其复配物对龙牙百合灰霉病菌的室内毒力,确定增效作用和最佳配方,并进一步通过离体防效试验明确增效潜力,旨在为有效防治龙牙百合灰霉病选用合适的杀菌剂做铺垫,同时有助于更好地指导农业生产。
发明内容
本发明所要解决的技术问题是,针对上述现有技术的不足,提供一种含吩嗪-1-甲酰胺和乙蒜素的复配杀菌剂,同时提供该复配杀菌剂在防治龙牙百合灰霉病中的应用。
为解决上述技术问题,本发明所采用的技术方案是:一种含吩嗪-1-甲酰胺和乙蒜素的复配杀菌剂,该复配杀菌剂含有吩嗪-1-甲酰胺和乙蒜素,该吩嗪-1-甲酰胺与乙蒜素的重量比为1:1。
本杀菌剂可与现有常规助剂(制备农药中可接受的其他辅助成分)配制成常规的农药剂型。
本发明同时提供上述含吩嗪-1-甲酰胺和乙蒜素的复配杀菌剂在防治龙牙百合灰霉病中的应用。
吩嗪-1-甲酰胺和乙蒜素均对龙牙百合灰霉菌均有抑菌活性,其EC50分别为21.0859μg/mL和138.9683μg/mL。将两者进行复配,当吩嗪-1-甲酰胺和乙蒜素的配比为1:1时,其共毒系数为172,大于120,具有明显的增效作用。离体叶防效试验结果显示:吩嗪-1-甲酰胺和乙蒜素按上述比例复配,对龙牙百合灰霉病菌的防治效果达83.85%,分别是吩嗪-1-甲酰胺和乙蒜素单一使用时的1.51倍和1.48倍。
与现有技术相比,本发明具有的优点为:
1、吩嗪-1-甲酰胺和乙蒜素以1:1复配对龙牙百合灰霉病菌具有较强的毒力,其共毒系数为172,大于120,具有明显的增效作用。
2、将吩嗪-1-甲酰胺和乙蒜素两种作用机理不用的药剂复配,可以提高药剂的使用寿命,且不易使病菌产生抗药性。
3、本发明提供了一种用于防治百合灰霉病的复配杀菌剂,丰富了防治龙牙百合灰霉病的新农药。
具体实施方式
下面结合具体试验和实施例对本发明的技术方案作进一步说明,本发明实施例中涉及的百分比均为质量百分比。
实施例1吩嗪-1-甲酰胺和乙蒜素复配防治龙牙百合灰霉病菌的增效作用
(1)供试材料
本试验所用的供试病菌龙牙百合灰霉病菌来自湖南农业大学植物保护学院生物防治实验室。
本试验所采用的药剂是乙蒜素(80%,北京中农佳瑞有限公司),吩嗪-1-甲酰胺(95%,上海秦巴化工有限公司)。
供试培养基:PDA培养基(200g马铃薯、20g葡萄糖、20g琼脂,pH值自然,加水定容至1L,于1个大气压,121℃灭菌20min)。
(2)吩嗪-1-甲酰胺和乙蒜素单剂毒力测定
分别称取吩嗪-1-甲酰胺和乙蒜素于50mL的烧杯中,然后在两个烧杯中分别加入2mL丙酮和适量吐温80充分震荡搅拌至原药完全溶解,最后分别加入去离子水至100mL,配成1000μg/mL的母液。
采用生长速率法测试单剂的毒力。将吩嗪-1-甲酰胺和乙蒜素配置成6.25,12.5,25,50,100μg/mL系列浓度的含药PDA培养基,用打孔器从培养3d的龙牙百合灰霉菌菌落边缘取灰霉病菌菌饼,菌丝面朝下接种在含药PDA培养基上,每个处理3个重复,以加无菌水的无药培养基作为空白对照(CK),倒置培养在20℃的培养箱中。待对照菌落直径长至整个培养皿2/3时,用十字交叉法测量菌落直径,计算抑制率,抑制率=(对照菌落直径-处理菌落直径)/(对照菌落直径-菌饼直径)×100%。
试验结果利用DPS数据处理平台统计处理,计算个单剂对龙牙百合灰霉病菌毒力的抑制率—剂量(浓度)对数方程及其相关系数(R)、抑制中浓度(EC50)。
(3)吩嗪-1-甲酰胺和乙蒜素复配剂的毒力测定及增效作用
取适量吩嗪-1-甲酰胺和乙蒜素母液分别按1:9,2:8,3:7,4:6,1:1,6:4,7:3,2:8,1:9的比例混合成不同混剂,按(2)的方法测定各混剂对龙牙百合灰霉病菌的毒力,并计算共毒系数。
共毒系数的计算:以药剂的剂量对数值与死亡率的机率值计算毒力回归方程和EC50(μg/mL),按照孙云沛等报道的杀菌剂联合毒力计算方法计算共毒系数(CotoxldityCoefficient)。
共毒系数(CTC)=((A+B)的实测毒力指数/(A+B)的理论毒力指数)×100;
混剂中(A+B)的实测毒力指数(ATI)=(A单剂EC50/(A+B)EC50)×100;
(A+B)的理论毒力指数(TTI)=A剂的毒力指数×A剂在混剂中的质量百分含量+B剂的毒力指数×B剂在混剂中的质量百分含量;
单剂毒力指数(TI)=(标准杀菌剂的EC50/供试单剂的EC50)×100。
杀虫剂复配后的联合毒力表现为3种情况,即增效、相加和拮抗作用。若共毒系数在100左右为相加作用,明显大于120为增效作用,明显小于80为拮抗作用。
(4)结果与分析
4.1吩嗪-1-甲酰胺和乙蒜素单剂对龙牙百合灰霉病菌的毒力测定
由表1可知,吩嗪-1-甲酰胺和乙蒜素两种药剂的浓度越高,抑菌效果越好。吩嗪-1-甲酰胺对龙牙百合灰霉病菌的抑制效果优于乙蒜素。其中吩嗪-1-甲酰胺对龙牙百合灰霉病菌的毒力回归方程为y=3.5439+1.0998x,相关系数为0.9946,EC50为21.0859μg/mL;乙蒜素对龙牙百合灰霉病菌的毒力回归方程为y=1.2124+2.4020x,相关系数为0.9961,EC50为138.9683μg/mL;由于抑制中浓度越低杀菌剂的防治效果越佳,因此,吩嗪-1-甲酰胺对龙牙百合灰霉菌的毒力最强,防治效果最佳。

4.2复配剂的室内毒力测定
由表2可知,吩嗪-1-甲酰胺和乙蒜素以不同比例复配对龙牙百合灰霉病菌的抑制效果与浓度呈正比,浓度越高,抑制效果越好。吩嗪-1-甲酰胺和乙蒜素以不同比例复配均对龙牙百合灰霉病菌具有抑制作用,其中以1:1时的抑制效果最佳。由表3可知,吩嗪-1-甲酰胺和乙蒜素以1:1复配对龙牙百合灰霉病菌的毒力最强,其次为4:6,其他复配剂的毒力效果按照从强到弱顺序依次为3:7、1:9、1:1、2:8、6:4、7:3、8:2、9:1等。

4.3复配剂的共毒系数
由表4可以看出吩嗪-1-甲酰胺与乙蒜素以1:1,4:6,3:7,1:9制成的药液对龙牙百合灰霉菌的共毒系数依次为172,153,133,130,表现出明显的增效作用,而其他比例复配表现为拮抗作用。

实施例2吩嗪-1-甲酰胺和乙蒜素复配对龙牙百合叶灰霉病的离体防效
(1)供试材料
本试验所用的供试病菌龙牙百合灰霉病菌来自湖南农业大学植物保护学院生物防治实验室。
本试验所采用的药剂是吩嗪-1-甲酰胺(95%,上海秦巴化工有限公司)和80%乙蒜素乳油(80%,北京中农佳瑞有限公司),腐霉剂(95%,上海秦巴化工有限公司)。
供试培养基:PDA培养基(,200g马铃薯、20g葡萄糖、20g琼脂,pH值自然,加水定容至1L,于1个大气压下,121℃灭菌20min)。
(2)离体防治效果
分别取吩嗪-1-甲酰胺和乙蒜素以及两者复配物制备成浓度为0.210859mg/mL、1.389683mg/mL、0.213349mg/mL,备用。本试验共设3个处理,1个药剂对照(0.80mg/mL),1个空白对照,3个处理为吩嗪-1-甲酰胺处理,乙蒜素处理,吩嗪-1-甲酰胺和乙蒜素复合物处理,对照药剂为腐霉利,并以无菌水为空白对照。将均匀一致的幼龙牙百合叶分别用药剂浸泡30s,晾干,于叶正中央接种龙牙百合灰霉病菌菌饼,每处理重复3张叶,不同处理随机分布,药后72h,测量病斑总长度,计算防治效果,防治效果(%)=((对照病斑长度-处理病斑长度)/(对照病斑长度-菌饼直径))×100%。
结果与分析:由表5可知,吩嗪-1-甲酰胺和乙蒜素以1:1复配对龙牙百合灰霉病的防治效果可达83.85%,是单一使用吩嗪-1-甲酰胺和乙蒜素的1.51和1.48倍,且比对照药剂腐霉利的效果好,具有较好的应用潜力。
