Abstract
本发明公开了一种中药型纳米二氧化钛标本保存液,其原料包括:金银花提取液、丹参提取液、纳米二氧化钛、冰醋酸、辅助成分、水。本发明以冰醋酸、金银花提取液、丹参提取液、纳米二氧化钛为标本液主要成分,使标本中富含纤维的组织膨胀,同时固定蛋白质和脂肪,清除细菌;以乙醇和丙三醇为辅助成分,加快固定的同时吸附溶液中的有机溶剂。本发明保存液可使标本在一年内无腐败现象,且标本皮肤黏膜等无显著变化,保存液内无霉菌产生,标本固定良好,标本保存液无刺激性气味。

技术领域
本发明涉及生物解剖技术领域,尤其涉及一种中药型纳米二氧化钛标本保存液。
背景技术
在临床解剖学试验中,通常需要把标本放入保存液中浸泡,以达到使实验用标本保存较长时间、固定蛋白质和脂肪等目的。从1886年发现甲醛是一种有效的防腐剂开始,甲醛作为标本保存液固定剂广泛应用于保存组织、器官及整个遗体。以甲醛作为防腐固定剂的传统标本保存液的优点在于其价格便宜、杀菌能力强、防腐作用彻底、能抑制霉菌和酵母的生长、对组织的固定硬化作用好、能除去腐烂过程遗体中产生的胺类臭味和操作方法简便用量少,但是甲醛易挥发、毒性大,对实验人员的呼吸道黏膜和皮肤刺激极大;长期接触还可引起多系统、多脏器的损害,甚至具有一定的致癌性;且对标本的色泽、质地和组织弹性的保持较差,随着医学、科技、环保技术领域的逐渐发展,传统标本保存液及传统保存液保存的标本已逐渐不能满足人们科研、教学等方面需求。如何制备出一种具有传统保存液防腐抗菌特点,对人体皮肤黏膜刺激性小,对标本的色泽、质地和组织弹性的保持持久的改良保存液,是目前急需解决的技术问题,对临床解剖学的研究具有十分重要的意义。
发明内容
基于背景技术存在的技术问题,本发明提出了一种中药型纳米二氧化钛标本保存液,防腐抗菌效果好、对标本色泽、质地和组织弹性的保持持久,对人体皮肤黏膜刺激小。
本发明提出的一种中药型纳米二氧化钛标本保存液,其原料包括:金银花提取液、丹参提取液、纳米二氧化钛、冰醋酸、辅助成分、水。
金银花提取液具有清热解毒、通经活络、调节免疫,广谱抗菌及抗病毒等功效,被称为“中药抗生素”。金银花富含有机酸、挥发油、黄酮类等成分,有机酸类被认为是抗菌防腐的主要活性物质,包括绿原酸、异绿原酸、咖啡酸以及木樨草素苷等,对伤寒杆菌、大肠杆菌、变形杆菌、绿脓杆菌、葡萄球菌、痢疾杆菌、链球菌、肺炎双球菌、脑膜炎球菌等致病菌有较强的抑制作用。
丹参提取液具有清除人体的氧自由基,抑制脂质过氧化反应,提高人体免疫机能,抗肝脏纤维化、抗肿瘤、抗衰老。丹参富含丹参酮IIA、丹酚酸B、隐丹参酮、二氢丹参酮等成分,其中,隐丹参酮和二氢丹参酮的抗菌效果优异,对大肠杆菌、金黄色葡萄球菌、变性杆菌及链球菌均具有显著抑菌和杀菌作用。
优选地,其原料按重量百分含量包括:金银花提取液5-15%,丹参提取液5-15%,纳米二氧化钛3-8%,冰醋酸3-6%,辅助成分16-28%,余量为水。
优选地,其原料按重量百分含量包括:金银花提取液8-12%,丹参提取液8-12%,纳米二氧化钛4-6%,冰醋酸4-5%,辅助成分19-24%,余量为水。
优选地,金银花提取液和丹参提取液为金银花和丹参经旋转醇提法制得。
优选地,辅助成分按重量份包括:酒精15-25份,丙三醇1-3份。
优选地,辅助成分中酒精为质量分数为3-8%乙醇水溶液。
有益效果:使用本发明标本保存液的标本,能够在一年内无腐败现象,且标本皮肤黏膜等无显著变化,保存液内无霉菌产生,标本固定良好,标本保存液无刺激性气味。
本发明提出了一种中药型纳米二氧化钛标本保存液,纳米二氧化钛的氧化活性较高,稳定性也较强,对人体无毒,在光线照射下,可短时间内就能完全杀死与其接触的微生物,具有杀菌效率高、持久、无二次污染的优点,中药金银花提取液、丹参提取液的抗炎、抗菌、抗氧化性能优异,作用周期长,对人体有益而无害,冰醋酸可促进富含纤维的组织膨胀吗,减少乙醇对组织的高度收缩和硬化,从而使标本软化。以纳米二氧化钛配合金银花提取液、丹参提取液及冰醋酸为标本保存液主要成分,使标本中富含纤维的组织膨胀,同时固定蛋白质和脂肪,清除细菌;以乙醇和丙三醇为辅助成分,乙醇能杀菌消毒、加快固化,而且是一种很好的有机溶剂,丙三醇不仅能吸附纳米二氧化钛、金银花提取液、丹参提取液、冰醋酸和乙醇,增加其抗菌防腐防霉的能力,还能使标本保持持久的色泽、质地和组织弹性,而且本发明中药型纳米二氧化钛标本保存液中不含甲醛,毒性低,无刺激性气味,是一种较理想的标本保存液。

具体实施方式
实施例1
一种中药型纳米二氧化钛标本保存液,其原料包括:金银花提取液、丹参提取液、纳米二氧化钛、冰醋酸、乙醇、丙三醇、水。
实施例2
一种中药型纳米二氧化钛标本保存液,其原料按重量百分含量包括:金银花提取液5%,丹参提取液5%,纳米二氧化钛3%,3%质量分数的乙醇水溶液15%,丙三醇1%,冰醋酸3%,余量为水。
实施例3
一种中药型纳米二氧化钛标本保存液,其原料按重量百分含量包括:金银花提取液15%,丹参提取液15%,纳米二氧化钛8%,8%质量分数的乙醇水溶液25%,丙三醇3%,冰醋酸6%,余量为水。
实施例4
一种中药型纳米二氧化钛标本保存液,其原料按重量百分含量包括:金银花提取液8%,丹参提取液8%,纳米二氧化钛4%,5.6%质量分数的乙醇水溶液18%,丙三醇1%,冰醋酸4%,余量为水。
实施例5
一种中药型纳米二氧化钛标本保存液,其原料按重量百分含量包括:金银花提取液12%,丹参提取液12%,纳米二氧化钛6%,4%质量分数的乙醇水溶液22%,丙三醇2%,冰醋酸5%,余量为水。
实施例6
一种中药型纳米二氧化钛标本保存液,其原料按重量百分含量包括:金银花提取液10%,丹参提取液10%,纳米二氧化钛5%,7%质量分数的乙醇水溶液20%,丙三醇2%,冰醋酸4.5%,余量为水。
细菌培养实验。为观察本发明中药型纳米二氧化钛标本保存液的防腐抗菌情况,通过细菌培养试验,将实施例5制得标本保存液、水加甲醛及纯水分别取10ml与1ml放凉肉汤混合,于培养皿中培养记录,一旦发现霉菌立即停止记录,观察其防腐情况,对抗菌能力,测试试验数据如下表1。

由上表1可以看出,本发明中药型纳米二氧化钛标本保存液的防腐抗菌效果与水加甲醛一致,具有良好的抗菌能力。
保存液刺激性对比实验。取家兔12只,重约2~3kg,健康无疾,随机分为三组,每组3只,分别将实施例5保存液0、1ml及水加甲醛保存液0、1ml分别滴入兔左右眼结膜囊内,并使眼睑被动闭合5s,记录6、24、48、72h至七天家兔眼的局部反应情况,测试结果如下表2,测试标准如下表3和表4。

通过对上表2-4测试数据分析可知:水加甲醛保存液对家兔眼大多具有重度刺激情况,而本发明中药型纳米二氧化钛标本保存液对家兔眼部只有轻度刺激,大大缓解保存液的刺激性。
组织学检测。将用实施例5制得保存液和水加甲醛保存液保存的家兔的上下肢肌肉,进行HE染色,镜下观察肌肉固定效果,测试结果如图1。
通过试验可知,家兔的上下肢肌肉标本肌肉色泽红润,质地柔软,弹性佳,肌纤维保存完好,无臭嗅,未见污染灶,组织无溶解;通过图1可以看出,镜下观察肌纤维条索完整,细胞结构清晰,高倍镜下横纹存在,两种保存液的组织切片难以区分,肌肉固定效果相仿。

Claims (4)
1.一种中药型纳米二氧化钛标本保存液,其特征在于,其原料包括:金银花提取液、丹参提取液、纳米二氧化钛、冰醋酸、辅助成分、水;其中,其原料按重量百分含量包括:金银花提取液5-15%,丹参提取液5-15%,纳米二氧化钛3-8%,冰醋酸3-6%,辅助成分16-28%,余量为水;
其中,辅助成分按重量份包括:酒精15-25份,丙三醇1-3份。
2.根据权利要求1所述中药型纳米二氧化钛标本保存液,其特征在于,其原料按重量百分含量包括:金银花提取液8-12%,丹参提取液8-12%,纳米二氧化钛4-6%,冰醋酸4-5%,辅助成分19-24%,余量为水。
3.根据权利要求1或2所述中药型纳米二氧化钛标本保存液,其特征在于,金银花提取液和丹参提取液为金银花和丹参经旋转醇提法制得。
4.根据权利要求1所述中药型纳米二氧化钛标本保存液,其特征在于,辅助成分中酒精为质量分数为3-8%乙醇水溶液。
