Abstract
一种利用环糊精水解酶制备特定聚合度麦芽低聚糖的方法,属于麦芽低聚糖生产技术领域。本发明采用环糊精水解酶催化水解底物环糊精,反应结束后通过酶灭活、脱盐和干燥,制得麦芽低聚糖。本发明采用新的环糊精水解酶对环糊精水解,制备得到特定聚合度的麦芽低聚糖,其制备过程简单环保,所生产的特定聚合度的麦芽低聚糖纯度高,在医学、食品、化学材料等领域应用广泛。
Description
技术领域
本发明涉及一种利用环糊精水解酶制备特定聚合度麦芽低聚糖的方法,属于麦芽低聚糖生产技术领域。
背景技术
麦芽低聚糖主要是由2-10个葡聚糖以α-1,4键连接而成的直链麦芽低聚糖。麦芽低聚糖不仅对人的身体有着多种的有益功能,并且具有十分良好的食品加工适应性,因而在食品行业中,得到广泛的应用。麦芽低聚糖具有低糖度、粘度高及抗结晶性能等,可以替代蔗糖在食品中的应用,使食品获得更良好的感官和质构特性。此外,作为功能性低聚糖它还能促进人体对钙的吸收,抑制人体肠道有害菌的生长,促进有益菌的生长,对人体健康大有裨益。
目前麦芽低聚糖主要的应用是添加在食品中以改善食品的相关性质,如甜度、粘度、吸湿性、渗透性、稳定性、凝胶性以及人体的消化吸收性能等。然而,麦芽低聚糖的生产主要是以淀粉为底物,经酶解、转苷、纯化制备,所得产物也主要是不同聚合度的,从麦芽糖、麦芽三糖到麦芽八糖等的混合物。在改善食品的加工适应性和各类物理和生物性质中,常常需要精确定量,而混合的麦芽低聚糖则很难满足精准改善食品性质的需要。因此,这就需要在了解特定聚合度的麦芽低聚糖理化特性的同时,生产高纯度的麦芽低聚糖,以达到对食品特性的良好控制。此外,特定聚合度的麦芽低聚糖在医疗方面也有很重要的作用,如麦芽五糖可测定血液中的a-淀粉酶活性可以用于诊断疾病。
为了解决上述问题,科研工作者做了大量工作,其主要目的是集中于筛选高特异性的麦芽低聚糖生成酶,如麦芽三糖酶、麦芽四糖酶等。然而此法所得特定聚合度麦芽低聚糖纯度仍不高,且目前六糖、七糖、八糖生成酶还无较多报道。本发明的优势在于,突破以往技术手段,采用现已逐渐产业化的环糊精为底物,利用环糊精水解酶的开环水解作用,来制备高纯度的特定聚合度麦芽低聚糖。在避免了化学制备法的残留污染等问题的同时,为高纯度的特定聚合度麦芽低聚糖制备提供了新思路。
发明内容
本发明的目的是克服上述不足之处,提供一种利用环糊精水解酶制备特定聚合度麦芽低聚糖的方法,所提供的环糊精水解酶对环糊精有着高度的特异性。
本发明的另一个目的是提供一种新环糊精水解酶,该酶能耐受80℃及以上高温,适宜pH范围为5.0~8.0。此外,该酶对环糊精有着高度的特异性,而对可溶性淀粉、直链淀粉、支链淀粉以及普鲁兰多糖等相对活性较弱等特性。
本发明的技术方案,一种利用环糊精水解酶制备特定聚合度麦芽低聚糖的方法,步骤为:采用环糊精水解酶催化水解底物环糊精,反应结束后通过酶灭活、脱盐和干燥,制得麦芽低聚糖。
所述环糊精水解酶的氨基酸序列具体如SEQ ID NO.1所示,核苷酸序列为SEQ IDNO.2所示。
所述底物环糊精主要包括以α-1,4键连接,含不同个数的α-D吡喃葡萄糖的环状糊精;底物环糊精聚合度为6-8。
所述底物环糊精具体为α-环糊精、β-环糊精或γ-环糊精。
所述麦芽低聚糖为麦芽六糖、麦芽七糖或麦芽八糖。
制备的具体步骤为:按照底物环糊精/溶液质量浓度为1%-50%(w/w)、pH为5.0-8.0配置底物环糊精的水溶液,按照环糊精水解酶/底物环糊精0.05 -100U/mg添加环糊精水解酶,在40-90℃、pH为5.0-8.0范围内水解反应0.5-48h;将酶灭活,脱盐,干燥,最终得到麦芽低聚糖。
所述酶灭活方法包括酸灭活,碱灭活或高温灭活;
所述酸灭活方式为添加反应总体积0.1%-10%(v/v),浓度为0.1-5M的酸灭活,再添加等量的浓度为0.1-5M的碱中和至pH为5.0-8.0;
碱灭活方式为添加反应总体积0.1%-10%(v/v),浓度为0.1-5M的碱灭活,再添加等量的浓度为0.1-5M的酸中和至PH为5.0-8.0;
高温灭活方式为将温度升到100-120℃,保温15-120min。
所述碱为氢氧化钠、氢氧化钾、碳酸钠或氢氧化铵;所述酸为盐酸、硫酸、硝酸、磷酸、乙酸、柠檬酸、高氯酸或甲酸。
所述脱盐方式为透析脱盐或脱盐柱脱盐。
所述干燥方法包括冷冻干燥、常压干燥、减压干燥或喷雾干燥。
本发明的有益效果:本发明采用新的环糊精水解酶对环糊精水解,制备得到特定聚合度的麦芽低聚糖,其制备过程简单环保,所生产的特定聚合度的麦芽低聚糖纯度高,在医学、食品、化学材料等领域应用广泛。

具体实施方式
以下实施例中环糊精水解酶经过设计后,由北京六合华大基因科技有限公司合成。
实施例1环糊精水解酶的底物特异性证明实验
分别配置pH=6,质量浓度为1%的α-CD、β- CD、γ- CD、可溶性淀粉、直链淀粉、支链淀粉和普鲁兰多糖为底物,分别各取450μL底物,各加入5U环糊精水解酶,用缓冲液定容至500μL,在最适条件下反应10min,用DNS法分别测定对不同底物的酶反应活性,最终结果如图1所示。
从图1可知环糊精水解酶对环糊精水解有着高度的特异性,且水解作用为α-CD>β-CD>γ-CD。
该酶对可溶性淀粉、直链淀粉、支链淀粉和普鲁兰多糖水解作用微弱,其中对可溶性淀粉、支链淀粉和普鲁兰多糖几乎无作用,而对直链淀粉有着微弱的水解作用。
实施例2
本实施例以麦芽六糖的制备举例说明上述方法制备特定聚合度的麦芽低聚糖。
配置质量浓度为100mg/g的pH=5的α-CD溶液30mL为底物,加入30U的环糊精水解酶,在60℃条件下反应8h,反应结束用反应液体积2%、5M的盐酸将酶灭活,随后采用等量的5M的NaOH进行中和;脱盐柱脱盐;最后将产物-80℃下冻干24h,最终得到高纯度的麦芽六糖产品。
实施例3
本实施例以麦芽七糖的制备举例说明上述方法制备特定聚合度的麦芽低聚糖。
配置质量浓度为30mg/g的pH=7的β-CD溶液30mL为底物,加入50U的环糊精水解酶,在40℃条件下反应24h,反应结束后用反应液体积5%、0.4M的KOH将酶灭活,随后采用等量的0.4M的硫酸进行中和;脱盐柱脱盐;最后将产物置于60℃,真空干燥48h。
将产品粉末复溶后,采用高效阴离子交换色谱-脉冲安培检测法(HPAEC-PAD)测定样品的组分与含量,如图2-a所示,麦芽七糖产品的TLC图谱如图2-b所示,产品中主产物为麦芽七糖。
实施例4
本实施例以麦芽八糖的制备举例说明上述方法制备特定聚合度的麦芽低聚糖。
配置质量浓度为200mg/g的pH=8的γ-CD溶液30mL为底物,加入80U的环糊精水解酶,在90℃条件下反应2h,反应结束后采用高温煮沸60min,将酶灭活;透析脱盐;最后将产物置于40℃下,热风干燥72h,最终得到高纯度的麦芽八糖产品。
虽然本发明以巳实施例公开如上,但并非用以限定本发明,任何熟悉此技术的人,在不脱离本发明的精神和范围内,都可做各种的改动与修饰,因此本发明的保护范围应该以权利要求书界定的为准。

1.一种利用环糊精水解酶制备特定聚合度麦芽低聚糖的方法,其特征在于步骤为:采用环糊精水解酶催化水解底物环糊精,反应结束后通过酶灭活、脱盐和干燥,制得麦芽低聚糖。
2.根据权利要求1所述利用环糊精水解酶制备特定聚合度麦芽低聚糖的方法,其特征在于:所述环糊精水解酶的氨基酸序列具体如SEQ ID NO.1所示,核苷酸序列为SEQ IDNO.2所示。
3.根据权利要求1所述利用环糊精水解酶制备特定聚合度麦芽低聚糖的方法,其特征在于:所述底物环糊精主要包括以α-1,4键连接,含不同个数的α-D吡喃葡萄糖的环状糊精;底物环糊精聚合度为6-8。
4.根据权利要求3所述利用环糊精水解酶制备特定聚合度麦芽低聚糖的方法,其特征在于:所述底物环糊精具体为α-环糊精、β-环糊精或γ-环糊精。
5.根据权利要求1所述利用环糊精水解酶制备特定聚合度麦芽低聚糖的方法,其特征在于:所述麦芽低聚糖为麦芽六糖、麦芽七糖或麦芽八糖。
6.根据权利要求1所述利用环糊精水解酶制备特定聚合度麦芽低聚糖的方法,其特征在于具体步骤为:按照底物环糊精/溶液质量浓度为1%-50%w/w,pH为5.0-8.0配置底物环糊精的水溶液,按照环糊精水解酶/底物环糊精0.05 -100U/mg添加环糊精水解酶,在40-90℃、pH为5.0-8.0范围内水解反应0.5-48h;将酶灭活,脱盐,干燥,最终得到麦芽低聚糖。
7.根据权利要求6所述利用环糊精水解酶制备特定聚合度麦芽低聚糖的方法,其特征在于:所述酶灭活方法包括酸灭活,碱灭活或高温灭活;

9.根据权利要求6所述利用环糊精水解酶制备特定聚合度麦芽低聚糖的方法,其特征在于:所述脱盐方式为透析脱盐或脱盐柱脱盐。
10.根据权利要求6所述利用环糊精水解酶制备特定聚合度麦芽低聚糖的方法,其特征在于:所述干燥方法包括冷冻干燥、常压干燥、减压干燥或喷雾干燥。
