Abstract

技术领域
本申请大体上涉及适用于吸收和清洁的产品尤其多孔基质复合物的领域,所述吸收和清洁功能同时提供持续不断的保护而免于细菌和其它有害环境病原体。
背景技术

发明内容
本申请的一个方面涉及一种制造吸收性非滤取抗微生物剂多孔结构复合物的方法,包括:将呈可流动形式(如液体或半液体形式)的可固化基质与1)非滤取且非可溶、精细分散的抗微生物剂和2)可移除微孔形成物质混合,其中微孔形成物质和精细分散的非可溶非滤取抗微生物剂均匀地分布且物理地包封在可固化基质内,就像葡萄干在葡萄干布丁中一样;将可固化基质固化成固体基质,其中固化固体基质同时物理地包封微孔形成物质和非滤取抗微生物剂;通过使固化的基质经受移除固体微孔形成物质的物理条件(如通过融化或溶解)从固体基质移除微孔形成物质,其中固体基质中的微孔通过移除微孔形成物质且因此缺乏其来形成,且其中含微孔固体基质中的微孔在其最窄尺寸处宽度的平均大小不足3mm;其中非可溶不可滤取抗微生物剂现在物理地并入在多孔基质复合物内,且其中不可滤取非可溶抗微生物剂相当于≤5%wt/wt的干燥制成品;和从含微孔固体基质移除任何未物理地并入的不可滤取抗微生物剂以用于任选的回收和再使用。在这个实施例中,非滤取多孔抗微生物剂复合物是在一个步骤中形成。

在某些实施例中,通常呈液体或半液体形式的可流动可固化基质包括天然聚合物(如纤维素、胼胝质、昆布糖、金藻昆布多糖、木聚糖、阿拉伯木聚糖、甘露聚糖、褐藻糖胶、阿拉伯木聚糖、半乳甘露聚糖、半乳甘露聚糖、阿拉伯糖基木聚糖、菊糖、胶淀粉或果胶)、非天然聚合物、热固性聚合物、橡胶(天然或人造的)和/或基于丙烯腈和苯乙烯丁二烯的聚合物。在最优选实施例中,多孔基质能够从其水中自身重量的l×吸收到其水中自身重量的50×。在另外的实施例中,固体微孔形成物质包括硫酸钠、糖(如葡萄糖、海藻糖、果糖和许多其它糖)、可溶或可熔融的天然聚合物、可溶或可熔融的人造聚合物或盐,所述盐包括铵、钙、镁、钾、锂、铁、铝和其它金属盐和阳离子盐、吡啶鎓、季、铵、钠、乙酸盐、硅酸盐、碳酸盐、氯化物、碘化物、柠檬酸盐、硼酸盐、苯甲酸盐、氰化物、氟化物和其它卤化物、硝酸盐、亚硝酸盐、磷酸盐和/或硫酸盐和其它阴离子以及选择其特定溶解度和/或熔融点的材料,如二甲亚砜(DMSO)或其它溶剂、或月桂酸或在0℃到100℃的范围内融化的其它材料。还认为,能够在低温(如0℃)下固化的可流动可固化基质使得水冰(water ice)作为微孔形成物质来使用成为可能。在另一个优选实施例中,含微孔固体基质中的微孔在其最窄尺寸处平均最小尺寸不足2mm。在另一优选实施例中,含微孔固体基质中的微孔在其最窄尺寸处平均最小尺寸不足1mm。
在特定实施例中,不可滤取抗微生物剂包括:铜、银、锌、铋、金、铝、硼、硒、镓、硫、锗、钡、锰、钒、铅、铍、镉、铬、砷、碲、汞、铊、铀、锡、锑、铟、铁、钴或镍、锶、钪、钛、锰、钇、锆、铌、钼、锝、钌、铑、钯、镧、铪、钽、钨、铼、锇、铱、铂、金、汞、锕、钅卢、钅杜、钅喜、铍、钅黑、钅麦、钅达、钅仑、钅哥、铈、镨、钕、钷、钐、铕、钆、铽、镝、钬、铒、铥、镱、镥、钍、镤、铀、镎、钚、镅、锔、锫、锎、锿、镄、钔、锘或铹,所述元素呈其纯形式、调配形式或呈化学组合。

本申请的另一方面涉及一种用于最大化抗微生物剂吸收性多孔复合物的抗微生物效果的试剂盒,所述试剂盒包括:说明书(包含(例如)关于恰当使用和存储的信息);一切割片的抗微生物剂多孔复合物,其适用作清洁,任选地并且优选地粘附到擦洗器;和任选地保持器,其中保持器含有作为抗微生物剂多孔复合物的相同或不同抗微生物剂。在某些实施例中,抗微生物剂多孔复合物和保持器采用选自包括铜或银或所述两种金属的某种组合的群组的金属、或其它非滤取抗微生物剂材料或复合或衍生有抗微生物剂的材料。在某些实施例中,保持器是选自包括封闭或打开的空间(如钵或盒)的群组的容器,其中保持器可以是实心或多孔的(如通过筛网或格栅形成)。保持器可以提供一种汲取液体的装置。

制造具有有效孔径的抗微生物剂多孔基质复合物的方法
在本申请的一个方面中,制造程序可以分解为六个步骤:1)建立(例如)呈液体或半液体形式的可流动、可成形的、可固化基质;2)提供具有特定可控制大小的微孔的基质,任选地并优选地通过并入到具有特定可移除衬底(在其周围,基质可以固化)的可成形的基质(如可溶的或可熔融物质,类似于盐)中,其中在其最窄尺寸处微孔的直径为或约0.1mm到3mm,在一些实施例中,在其最窄尺寸处平均大小不大于3mm,在一优选实施例中,在其最窄测量值处平均大小范围在≤1mm到2mm的范围内;3)物理并入不可滤取抗微生物剂,通过在固化基质同时其含有微孔形成物质期间或在其固化之后物理锚定(类似于物理锚定滞留于葡萄干布丁中的葡萄干)或通过物理化学制共价结合来得到的所述物理并入,提供至少6,000:1m-1和最优选地在50,000m-1到1,000,000m-1的范围内的抗微生物剂的较高表面积比体积;4)将基质固化在微孔形成衬底周围;5)移除微孔形成衬底和6)在分布、出售或使用之前,移除任何未物理地结合的不可滤取抗微生物剂,任选地并优选地用于回收和再循环。应注意,可以在固化基质之前或之后发生物理并入抗微生物剂,且各次序提供不同优点。
在某些实施例中,可固化基质由经加工的天然碳水化合物聚合物(如纤维素)组成。在一些经加工天然聚合物实施例中,类似于纤维素的碳水化合物基质的液化可以通过用氢氧化钠(NaOH)处理适当形式(如浆)的纤维素以通过将纤维素链断裂成较小长度来软化纤维素(所谓“老化”)来实现。接着,用二硫化碳(CS2)处理这种碱性纤维素以制造黄原酸纤维素钠,所述黄原酸纤维素钠在通过真空或其它手段移除CS2之后溶解于苛性碱中以形成粘胶纤维。

在本发明的其它实施例中,基质是非天然聚合物,如热固性聚合物,如可以使用酚-甲醛、脲-甲醛、聚酯、聚胺脂、硫化橡胶、聚氧苯甲基亚甲基乙二醇酐(胶木)、热固性塑料(Duroplast)、脲-甲醛、三聚氰胺或三聚氰胺-甲醛、邻苯二甲酸二烯丙酯(DAP)、环氧树脂、与环氧树脂组合的聚酰胺树脂、聚酰亚胺、氰酸酯或聚氰酸酯、聚酯树脂、乙酸-丁酸纤维素、乙基纤维素、乙烯与偏乙烯的聚合物和共聚物、含有聚乙烯醇的热固性树脂、丙烯酸酯和甲基丙烯酸酯树脂、聚苯乙烯等,这些聚合物包含任选地弹性材料(如橡胶(天然或人造的)、GRS(丁苯橡胶,一种丁二烯和苯乙烯的共聚物))和/或各种合成的胶乳(如丙烯腈和苯乙烯丁二烯基聚合物),所述材料可以制为液体或半液体且可以并入一或多种聚合物的混合物中。所属领域的技术人员应认识到,多种基质是可能的,且根据基质而定,从液体或半液体转化成固体应采用温度(如加热)、pH(如酸化)、化学反应和类似的改变。
接着,赋予可成形的、可固化基质(通常呈液体或半液体形式)特定可移除材料,在所述可移除材料周围微孔可以形成。控制孔径对于最终产品的效用是必需的,因为按照定义的物理地结合的抗微生物剂物质是物理地结合的且因此仅在给定距离(例如通过一定距离的水)上有效。尽管扩散和布朗运动(Brownian motion)最终将使微生物与微生物表面靠近或接触,其中所述微生物将被理想地杀灭,在较大微孔的情况下,其进入的速度较慢。同样,在大孔隙的情况下,总滞留时间(接触抗微生物剂表面的时间)更短。因此,控制孔径并且具体地说预防最窄距离过大对于物理地结合抗微生物剂的微生物杀灭效果是必需的。尽管微孔在其最窄尺寸处可以是对称(圆形)或不对称(针状)或其它形状,但所述微孔的平均直径应不大于3mm,且在一优选实施例中平均直径在≤1mm到2mm的范围内。还强调,孔径的均一性,且最具体地说,不存在极大孔隙对于抗微生物剂效用是必需的:一个较大微孔可以允许有害微生物的储槽,这是因为较大尺寸将允许有害微生物逃离与抗微生物剂表面接触。
在一个优选实施例中,通过将特定可移除衬底并入到可成形的、可固化基质(通常呈液体或半液体形式)中来形成这些微孔。
这种微孔形成物质最初并不溶解到显著程度,从而使微孔形成物质和可固化基质(例如)在回转螺杆混合器或滚筒混合器中均匀混合,且均匀地并入到基质中。
在固化基质之后,通过经由改变温度或化学反应的实例,(例如)通过溶解(例如用水洗涤出)和/或融化来移除微孔形成物质,因此在基质中留下与先前存在固体微孔形成物质的大小和形状精确相关的孔洞(微孔形成物质曾经所处位置)。必须注意,因此通过选择微孔形成物质的大小和形状来直接、精确且容易地控制所得多孔基质的孔径:通过如简单筛分固体微孔形成物质的程序,可以选择和确保最小孔径。
应注意,气体容易地被移除,且气体还可以用作微孔形成物质。然而,使用固体、可移除微孔形成物质控制孔径是直接的、高许多且更具有可再现性。举例来说,气体可以在基质中形成不均匀的气泡(举例来说,在压力较高的底部处具有较小的气泡,且在压力较小的顶部处具有较大的气泡,并且多个气泡可以合并成更大的气泡),气泡可以合并,从而形成更大微孔,气体随着温度扩散,并且因为气体对于环境条件的灵敏度,通过鼓泡气体制造的孔径的再现性可能是较差的。固体微孔形成物质并不压缩且并不受温度改变的影响直到达到其熔融点。重要地是,甚至在固体微孔形成物质的颗粒开始紧密接触的情况下,可能的是,基质材料将仍掉落在其之间,从而维持最小孔径。

在其它实施例中,微孔形成物质是糖的晶体,如葡萄糖、海藻糖、果糖、蔗糖和许多其它糖。在其它实施例中,微孔形成物质是可熔融材料,如天然聚合物(包含但不限于碳水化合物(如麦芽糊精或琼脂糖)、蛋白质(如明胶))、脂类或人造聚合物,其为可熔融、可溶、可熔融和可溶两者或以其它方式可移除的(如但不限于聚(N-异丙基丙烯酰胺)(PNIPAM)和聚丙烯酰胺(PAM)、聚(2-恶唑啉)和聚乙烯亚胺(PEI)、聚(丙烯酸)、聚甲基丙烯酸酯和其它丙烯酸聚合物、聚(乙二醇)和聚(氧化乙烯)、聚(乙烯醇)(PVA)和共聚物、聚(乙烯吡咯烷酮)(PVP)和共聚物)。
在所有实施例中,选择特性,使得微孔形成物质在并入到可流动可固化基质材料的条件期间维持一定形状(最优选地为固体),所述可流动可固化基质材料通常呈液体或半液体形式,但能够在固化基质材料过程之后或期间熔融、溶解或以其它方式移除。这类材料的许多实施例对于所属领域的技术人员是熟悉的。举例来说,尤其聚合物长度和交联(如果存在)的程度可以调变熔融温度和溶解度。理想地选择廉价、可容易获得的某些物质单独可以呈现所需特性而不需要修改。这些实例是出于说明性目的并且并不意图限制本发明的实施例。
在一个实施例中,在可成形的、通常液体到半液体未固化的基质材料与微孔形成物质混合时,在回转滚筒混合步骤期间,添加不可溶抗微生物剂(如但不限于金属铜、金属银或其它不可溶或大部分不可溶)、不可滤取抗微生物剂或抗微生物剂的混合物,从而允许完成和容易的集成到制造程序中而无需任何改变。具有较高表面积与体积的比(至少6,000:1m-1,且最优选地在50,000m-1到1,000,000m-1的范围内)的粉末(均为常规球形和不规则地形状,这些均为固体和多孔的)、碎箔、和其它几何形状是优选的,且在相同时间混合到液体或半液体基质中作为微孔形成物质(如盐)。这种混合方法使基质到基质围绕不可溶或微可溶抗微生物剂形成物理地包封袋:抗微生物剂如葡萄干在葡萄干布丁中一样被保持,一种通过使用不规则形状的不可溶抗微生物剂的另外辅助方法。表面积与体积的比被计算为表面积除以体积。通过实例,1cm(0.01m)的立方体在每一侧上的表面积与体积的比应是(6(0.01m)2)/(0.01m)3=600m-1。其它几何形状的比根据其形状来计算。这些实例是出于说明性目的并且并不意图限制本发明的实施例。
接着,挤压含抗微生物剂基质或形成为所需几何形状,在此时间后即根据基质的特性来固化基质。在经加工纤维素(如呈粘胶纤维形式)的实施例中,这可能包含加热和/或电流和/或在酸性条件(如在硫酸的稀释溶液中)下洗涤的应用。所属领域的技术人员应认识到,基质的选择对于发明的范围不具有限制性。随后移除(通过例如洗涤掉和/或融化)微孔形成物质且进行回收。接着充分洗涤所得多孔吸收性抗微生物剂基质复合物以移除、捕获和回收任何未结合的不可溶抗微生物剂。接着,按需要和期望对得到的洗涤产物进行切割、干燥、精加工和包装。可以用保湿剂(如氯化镁、聚乙二醇)或表面活性剂或类似物来包装产物以维持产物柔软性和柔性。可以用多种不可溶非滤取抗微生物剂材料(包含抗微生物剂金属、抗微生物剂化合物或其混合物)单独或以组合形式取代铜或银。铜代表优选实施例,这是由于其成本、可获得性、毒性、效果和可接受性,随后是铜和银。所属领域的技术人员应认识到,不可溶非滤取抗微生物剂材料的选择对于发明的范围不具有限制性。在这个实施例中,非可滤取不可溶抗微生物剂相当于≤5%wt/wt的干燥、精加工多孔基质复合物。通过实例,如果当洗涤和完全干燥时200cm3的对照多孔基质(其不含有任何不可溶抗微生物剂)重量为95g,那么200cm3的洗涤的、完全干燥的抗微生物剂多孔基质复合物的重量应不大于100g,额外5g(100g-95g=5g)表示不可溶非滤取抗微生物剂的重量且表示5重量%(5g/100g=5%)。
在本发明的其它实施例中,(例如)在已移除微孔形成物质之后,将不可溶或大部分不可溶抗微生物剂复合到备用基质的微孔和表面不规则体中。这个实施例使抗微生物剂并入到预先存在的基质上,例如并入到来自丝瓜属(Luffa genus)植物的天然丝瓜海绵的纤维素主链的微孔中,且同时产生极其高的>6,000:1m-1且通常是>50,000m-1到1,000,000m-1的表面积与体积的比,同时材料通常沉积成<1mm厚的较薄层,且最终因为沉积是在微孔内且精确地镜像微孔的尺寸,所以所述实施例呈现一种固有锚定机制:到微孔的入口通常小于微孔自身,和/或微孔具有不规则体,因此物理地将所沉积的抗微生物剂滞留在微孔内。在优选实施例中,引入抗微生物剂金属,如铜、银或其混合物。所属领域的技术人员应认识到,不可溶非滤取抗微生物剂材料的选择对于发明的范围不具有限制性,只要可以将其添加在预成型的固体多孔基质的微孔内。
在本发明的一些优选实施例中,多孔基质由来自天然生物源(如来源于植物、天然橡胶、纤维素和/或多孔动物(Porifera)门和/或共质体(Symplasma)门的海洋动物)的材料)的材料形成。在实施例中,采用合适的丝瓜葫芦,也被称为“蔬菜海绵”和/或“滤网藤”,通常在葫芦科内的丝瓜属内。在其它实施例中,包含玻璃海绵(有时归类在共质体门中)的海绵,玻璃海绵也被称为具有由四角和/或六角的硅质骨针组成的构架的六放海绵。替代地,除通过本文中所描述的方法来处理天然聚合物以外,可以将来自纤维植物的纤维编织(例如粗糙地编织)、缝合或以其它方式结合在一起以提供通过多个呈三维形式的流道和微孔实现液体和气体连通的众多多孔基质。这类纤维来源包含但不限于椰子或大麻纤维、海参构架(丝瓜)和藤本植物、植物的外皮和纤维集聚物,所述植物如灯芯草、稻谷植物、丝瓜、荞麦、大豆、竹子、木材或海草等。另外,可以以与植物纤维相同的方式采用动物纤维,如毛发、羽毛、表皮和兽皮、蚕丝和羊毛。这类材料是天然可获得的,通常是废产物,是高度多孔、廉价且完全可生物降解的。所属领域的技术人员应认识到,植物表示碳水化合物基质、海绵和动物纤维(一种蛋白质基质)和玻璃海绵(一种二氧化硅基质),所有均来自天然来源。所属领域的技术人员还应了解,许多天然物种是适用和可使用的基质的潜在来源,且所述这些实例决不具有限制性。
在本发明的其它实施例中,在已固化基质之后,将非滤取抗微生物剂并入到多孔抗微生物剂基质复合物中。在这个实施例中,将抗微生物剂物理地以与由基质和抗微生物剂呈现的可获得的官能性化学基团一致的方式共价结合基质,特定来说包含在其微孔内能够吸收其内所含有的包含微生物的溶液的基质。举例来说,抗微生物剂的NHS酯可以与固化柔性固体基质中存在的胺基反应。胺(例如含胺的主链的聚合物的那些化合物,如聚氨酯)可以同样地与抗微生物剂的卤化形式反应。替代地,卤基-环氧基烷基化合物(如C1-C6,如表溴醇的溴-环氧基烷基)可以在强碱(氢化钠(NaH)或二异丙胺基锂(LDA))存在下与固化基质的胺反应,从而安装反应性环氧基环且随后与其它官能团(如抗微生物剂的那些官能团)反应。所属领域的技术人员应认识到,添加所述安装或转化为更便利的反应性官能性的这个主题同样适用于胺外部的其它官能团,且在化学文献中充分发展。同样,活化的酯可以与亲核基团(如羟基或胺)反应,所述酯如酰基氯、R(CH2)COCl或酸酐(R(CH2)O)2O,其中R是抗微生物剂或其混合物。式RSiXlX2X3(其中R是抗微生物剂)的衍生硅烷可以与含有二氧化硅的基板(如海绵)反应。还应注意,带有反应性部分(如环氧化物)的硅烷衍生作用试剂(如GPTMS,(3-甘油基氧基丙基)三甲氧基硅烷)使得安装反应性环氧化物且与其它官能团进一步反应。形式RCOOCOR(其中R为抗微生物剂)的酸酐可以与羟基反应。在所形成的柔性多孔基质下方的羟基还可以与含有羟基的抗微生物剂通过酯化来反应。形式NH2NH-R(其中R是抗微生物剂)的酰肼可以与羧基反应。用于各种实施例中的制造方法不具有限制性,且可以包括并入到具有沉积溶液或化学表面衍生的复合物内。强调,不可溶抗微生物剂或通过在复合物内物理共价结合使得不可溶的抗微生物剂必然地必须存在于能够吸收的复合物的多孔区域的其表面。还强调且所属领域的技术人员已知的是,大量的化学物质可用于影响共价结合,包含与以下物质偶合:AOP、BOP、Brop、碳化二亚胺、COMU、氰基-羟亚胺基-乙酸乙酯(Oxyma)、DCC、DPPA、EDAC、EDC、EDCI、EEDQ、HATU、HDMC、HOSu、氧氯化磷、PyAOP、PyBOP、PyOxim、TATU、TBTU、TFFH、亚硫酰二氯、TOTT和所属领域的技术人员已知的多种其它试剂和化学物质。
应认识到,在使用清洁时,通过包含研磨表面或擦洗器来最大化效用,所述研磨表面或擦洗器必须是粗糙的且其优选地还含有不可溶抗微生物剂、由不可溶抗微生物剂组成或复合有不可溶抗微生物剂。还可以通过将抗微生物剂并入或沉积到经受微生物污染的其它物件来应用抗微生物活性,所述物件如各种大小的刷子(牙刷或马桶刷);表面,如货架;桌子和复合物、陶瓷或玻璃的工作台面;容器,如冰箱的食物抽屉或午餐袋或塑料袋;和其它通常被污染的区域,如冷冻机或洗碗机门周围的密封件。在这些实施例中,应认识到,多种制造方法可以应用于产生充分不可溶抗微生物剂的必需元件,这种不可溶抗微生物剂首先可进入空气,且这种不可溶抗微生物剂其次广泛分布在物件中并具有至少6,000:1m-1,且最优选地在50,000m-1到1,000,000m-1范围内的较大比表面积;和保持不可溶抗微生物剂的基质材料,这种基质材料物理地或共价地或这两者来保持不可溶抗微生物剂。给定的充分不可溶抗微生物剂和分布这些实施例呈现本文中的多孔柔性吸收性抗微生物剂复合物的抗微生物特性。
多种不可溶抗微生物剂(如铜、银和多种其它金属)以及其它不可溶材料(如硫)呈现微生物杀灭效果,且可以类似地通过物理并入或沉积来复合到多孔基质中。制得抗微生物剂沉积溶液但用银替代铜。可以将多种金属组合在相同或随后沉积中。还可以并入不可溶抗微生物剂(如铜或银——纯形式、呈混合物形式或呈化学组合形式)的粉末。还可以添加可以容纳金属离子或其它抗微生物剂的螯合剂和/或离子交换表面或基质和/或沸石或其它多孔或保持结构,例如,可以使用环糊精以将有机抗微生物剂保持在结构中。

实例1

将纤维素基质放置于Ziploc袋中以预防蒸发,在温热环境(30℃到40℃)下静置8小时以促进细菌生长。挤出这些基质,如上文所描述移除任何较大颗粒。将0.5mL的这种溶液涂铺在LB板上,且使其生长隔夜。来自柔性多孔纤维素抗微生物剂复合物的板具有零个菌落。相反,来自对照的板具有数不清的细菌生长的菌苔,从而指示细菌负载的多个对数下降(参见表2)。

实例2
其它实验在市售摇滚滚筒上旋转的塑料瓶子中进行不可溶的抗微生物剂并入。将天然丝瓜基质、市售纤维素基质、海绵基质和聚氨酯基质放置于塑料瓶中,且将以相同方式制备的一升抗微生物剂并入溶液倒入到其上。通过使用塑料棒挤压和扩展多孔基质材料来移除气泡,且如在实例1中将基质并入不可溶抗微生物剂隔夜但伴随摇滚滚筒旋转基质和抗微生物剂沉积溶液。
次日,铜并入到基质/抗微生物剂复合物中在基质上和其中是明显的,如在实例1中。因为海绵的颜色与铜的颜色极其接近,所以如果铜并入出现在其内,不可能以肉眼确定海绵。将所有基质洗涤一个循环。聚氨酯基质上的沉积呈现已清除,从而指示在固化基质期间并入的必要性。丝瓜和纤维素基质随时间的推移变成蓝色,如在实例1中。
针对细菌生长的测试如在实例1中进行,除了以下修改:第一,以1:1000稀释细菌汁液以减少所涂铺细菌的数量且允许进行菌落形成单位(CFU)计数。第二,因为基质的大小不同,所以将所有复合物均浸没在经稀释细菌汁液中,且使其在格栅上滴沥直至无滴落。应注意,当准确含有相同细菌的汁液用于实验和对照时,每ml的细菌初始数量作为不同厨房活动的函数而变化(参见表3)。

对于在室温下而不是30℃到40℃下培育的纤维素抗微生物剂基质复合物,重复测试(参见表4)。

除了用于铜并入反应物的试剂的浓度加倍到20mM硫酸铜、40mM乙酸和40mM抗坏血酸以外,重复测试。正如预期的,注意到额外铜并入。还观测到,在多孔基质接触塑料瓶的侧部的那些位置处,沉积富集。这类沉积可能在塑料瓶的侧部上发生,且并入到复合物中。用洗衣机在一个循环中并不能将这些沉积移除,且所述沉积在用于清洁和吸收期间保持在复合物上(参见表5)。

使用相同细菌汁液的储备液(存储在4℃下)重复测试,除了以下修改:第一,声称为抗微生物剂的其它品牌的纤维素基质。第二,用无菌水将隔夜培育后的细菌汁液以1:10稀释。第三,涂铺两种体积(0.5ml和0.1ml),且使其生长隔夜(参见表6;图1到图3)。

因此,物理并入和因此复合到纤维素基质不可溶的抗微生物剂复合物中的效用比“抗微生物剂海绵”优十倍。另外,通过复合到不可溶抗微生物剂(在这个实例中,铜)的纤维素中来挽救“抗微生物剂海绵”的较差性能。
实例3
为了进一步测试多孔吸收性不可溶抗微生物剂复合物的性能,将纤维素和铜以及来自相同制造商和批次的未复合的多孔纤维素基质的多孔不可溶抗微生物剂基质复合物放置于厨房水槽中两个月。在从0(无气味或仅食物的气味)到5(极其不好的)的等级上对气味进行评级。将两种样本暴露于相同条件。这些条件包含两种:可以显著减少细菌负载(例如暴露于来自如通心面制备物或类似物的沸水;用极热的水清洗餐盘;或暴露于具有漂白剂的清洁)的条件;以及可以促进细菌负载(例如在制备肉或鱼类之后清理;或清洁其中食物变质的容器)的条件。未复合的基质不断地比抗微生物剂复合物闻起来糟糕许多(参见图5)。
按以下测定与先前测试相同的多孔基质(复合的和对照未复合的两种)随时间推移的性能:复合物和对照基质两种均如正常清洁使用,且使其在水槽的底部中变潮。在结束当天,每周一次,将复合的样本和对照未复合的样本两种洗涤掉主要食物颗粒,且通过放置在Ziploc袋中来从额外用途中分离。在室温下培育隔夜之后(在紧挨着水槽的台面上以确保与水槽相同的温度),通过挤压微孔基质来采集来自各样本的微孔内的细菌汁液并对其进行过滤,如上文所描述。将细菌汁液在无菌水中以1:1000稀释,且将0.5ml涂铺。应注意,在不存在恰当线性流动无菌橱的情况下,通过空气传播环境污染也可以是平板中的细菌的贡献来源(参见表7)。

实例4
通过延伸,测试铜和其它固体抗微生物剂(例如其它金属,如银)到较大体积制造程序的柔性多孔抗微生物剂基质复合物中的并入。通过以下来产生粘胶纤维/铜粉末混合物:将300g的铜粉末与400kg的粘胶纤维合并,在回转滚筒中均匀混合,将这种混合物与微孔形成物质(在这种情况下,硫酸钠晶体)合并以形成微孔,将盐/粘胶纤维/铜混合物倒入到模具中,用电流固化混合物以及洗涤所得微孔形成物质以产生多孔固化的基质。对照批次相同地来制得,但未复合(即缺乏不可溶抗微生物剂,此处为铜)。如上文所描述洗涤所得产品,且如类似于上文实验所描述对其进行测试:将纤维素基质静置在水槽的底部处,挤出来自未复合基质的细菌汁液,粗糙地过滤这种汁液以移除食物颗粒,将汁液倒入复合和未复合(对照)的固化基质中,以及在这种情况下在室温下精确静置1小时。在精确1小时之后,挤出复合和未复合的基质,以1:3000稀释所得汁液,且将0.1ml涂铺在LB平板上。在37℃下生长之后,对以下菌落进行计数(参见表7):

实例5
形成作为不可溶、非滤取抗微生物剂的铜和/或银,使得带有钝尖峰的直径≤10微米的颗粒在其外部粗略仿生蝉翼上隔开大约0.5微米到200微米;其还可以是其微孔内部具有尖峰的多孔的。向纤维素粘胶纤维添加这些颗粒,使用滚筒混合器使其平均分布,且添加最小平均尺寸为1mm的硫酸钠晶体。再次将混合物混合直至所有组分平均分布,模塑混合物,固化基质,移除微孔形成物质,移除任何未物理结合的非滤取抗微生物剂,洗涤产物并进行包装。所得多孔吸收性非滤取抗微生物剂基质复合物比相同尺寸的球形呈现更强的抗微生物作用。
实例6
用亚硫酰二氯或其它偶合剂活化多孔固体纤维素基质(或天然丝瓜或经加工多孔纤维素)以制造中间物,所述中间物暴露于抗菌肽,尤其但不限于G3KL(序列(KL)8(KKL)4(KKL)2KKL)和G3RL(序列(RL)8(KRL)4(KRL)2KRL)和/或其树枝状聚合物,这些肽由呈D-立体异构体的氨基酸合成以防止蛋白酶活性。所得多孔吸收性非滤取抗微生物剂基质复合物呈现强力持久的抗微生物作用且降解极其缓慢。
实例7
用亚硫酰二氯或其它偶合剂活化多孔固体纤维素基质(或天然丝瓜或经加工多孔纤维素)以制造中间物,所述中间物暴露于含胺抗微生物剂(如胺基-季胺),其进行反应,使得每立方毫米的多孔基质共价锚定>50分子,且移除未反应的分子。所得多孔吸收性非滤取抗微生物剂基质复合物呈现强力持久的抗微生物作用且降解极其缓慢。
已出于说明和描述的目的呈现本申请的具体实施例的前述描述。这些描述并不意图是穷尽性的或限制本申请和所公开的精确形式的使用方法。显然,根据上文启示可能进行多种修改和变化。应理解,根据情况可能暗示或明示有利,涵盖等效物的各种省略或取代,但在不脱离本申请的权利要求书的精神或范畴的情况下意欲覆盖本申请或实施方案。
Claims (4)
1.一种制造吸收性非滤取抗微生物剂多孔基质复合物的方法,包括:
制作或获得形成含微孔固体基质的柔性多孔固体基质衬底,其中所述固体衬底的所述微孔在其最窄尺寸处宽度的平均大小不足3mm,
在含微孔固体基质内或上物理地并入不可滤取抗微生物剂,其中所述不可滤取抗微生物剂已暴露于所述基质微孔的内部;
从所述含微孔固体基质移除任何未物理地并入的不可滤取抗微生物剂,其中所述不可滤取抗微生物剂选自以下各项中的一种或多种:铜、银、锌、铋、金、铝、硼、硒、镓、硫、锗、钡、锰、钒、铅、铍、镉、铬、砷、碲、汞、铊、铀、锡、锑、铟、铁、钴或镍、锶、钪、钛、锰、钇、锆、铌、钼、锝、钌、铑、钯、镧、铪、钽、钨、铼、锇、铱、铂、金、汞、锕、钅卢(rutherfordium)、钅杜(dubnium)、钅喜(seaborgium)、铍(bohrium)、钅黑(hassium)、钅麦(meitnerium)、钅达(darmstadtium)、钅仑(roentgenium)、钅哥(copernicium)、铈、镨、钕、钷、钐、铕、钆、铽、镝、钬、铒、铥、镱、镥、钍、镤、铀、镎、钚、镅、锔、锫、锎、锿、镄、钔、锘、和铹。
2.一种吸收性非滤取抗微生物剂多孔结构复合物,包括:
通过物理锚定或物理化学共价结合物理地并入在抗微生物剂海绵上的不可滤取抗微生物剂;
在所述吸收性非滤取抗微生物剂多孔结构复合物内的微孔,其中所述微孔的大小在其最窄尺寸处宽度不足3mm,
且其中所述微孔将所述不可滤取抗微生物剂保持在所述微孔内,
且其中所述不可滤取抗微生物剂暴露于所述微孔的内部,
且另外其中所述不可滤取抗微生物剂的表面积与体积的比大于6000:1m-1,
其中所述不可滤取抗微生物剂选自以下各项中的一种或多种:铜、银、锌、铋、金、铝、硼、硒、镓、硫、锗、钡、锰、钒、铅、铍、镉、铬、砷、碲、汞、铊、铀、锡、锑、铟、铁、钴或镍、锶、钪、钛、锰、钇、锆、铌、钼、锝、钌、铑、钯、镧、铪、钽、钨、铼、锇、铱、铂、金、汞、锕、钅卢、钅杜、钅喜、铍、钅黑、钅麦、钅达、钅仑、钅哥、铈、镨、钕、钷、钐、铕、钆、铽、镝、钬、铒、铥、镱、镥、钍、镤、铀、镎、钚、镅、锔、锫、锎、锿、镄、钔、锘、和铹。
3.一种用于最大化本质上抗微生物剂吸收性柔性多孔复合物的效用的试剂盒,所述试剂盒包括:
关于如何恰当地使用抗微生物剂海绵的说明;
多孔抗微生物剂复合物,其中所述多孔抗微生物剂复合物包括不可滤取抗微生物剂,其中所述不可滤取抗微生物剂选自以下各项中的一种或多种:铜、银、锌、铋、金、铝、硼、硒、镓、硫、锗、钡、锰、钒、铅、铍、镉、铬、砷、碲、汞、铊、铀、锡、锑、铟、铁、钴或镍、锶、钪、钛、锰、钇、锆、铌、钼、锝、钌、铑、钯、镧、铪、钽、钨、铼、锇、铱、铂、金、汞、锕、钅卢、钅杜、钅喜、铍、钅黑、钅麦、钅达、钅仑、钅哥、铈、镨、钕、钷、钐、铕、钆、铽、镝、钬、铒、铥、镱、镥、钍、镤、铀、镎、钚、镅、锔、锫、锎、锿、镄、钔、锘、和铹;
以及保持器,其中所述保持器具有表面抗微生物特性。
4.根据权利要求3所述的试剂盒,其中所述保持器为选自由碗和盒组成的群组的容器。
