Abstract
本发明公开了一种纳米流感疫苗及构建方法和应用,所述的纳米流感疫苗为3M2e‑rHF重组蛋白,其序列为SEQ ID NO.2所示。本发明的重组蛋白将3M2e展示在铁蛋白笼形结构表面,显著提高了M2e免疫原性,从而构建了一种新型流感疫苗:3M2e‑rHF纳米颗粒。本发明利用原核表达系统大肠杆菌表达重组蛋白疫苗,疫苗制备过程不涉及到活病毒,相比传统的鸡胚制备流感疫苗方法操作更加安全、简便,适合进行快速大规模生产;利用M2e在不同亚型流感病毒中序列的高度保守性,实验证实,3M2e‑rHF保护小鼠抵抗了同型和异型流感病毒的感染,有望发展成为具有交叉保护效力的通用流感疫苗。The invention discloses a nano-influenza vaccine and a construction method and application. The recombinant protein of the present invention displays 3M2e on the surface of the ferritin cage structure, which significantly improves the immunogenicity of M2e, thereby constructing a new type of influenza vaccine: 3M2e-rHF nanoparticle. The present invention utilizes the prokaryotic expression system Escherichia coli to express the recombinant protein vaccine, and the vaccine preparation process does not involve live virus, which is safer and easier to operate than the traditional method for preparing influenza vaccine from chicken embryos, and is suitable for rapid large-scale production; The sequence of influenza viruses is highly conserved. Experiments have confirmed that 3M2e‑rHF protects mice against the infection of homotype and heterotype influenza viruses, and is expected to develop into a universal influenza vaccine with cross-protective efficacy.
