(II) penetration test of body surface of harmful mite
1. Tetranychus citriodorus test
For testing harmful mites: tetranychus citriodorus (A) and (B)Panonychuscitri)
Test and control agents: bifenazate and etoxazole; cyflumetofen
Test synergist and control adjuvant: n-380; NF-100 (glycosyl modified organic silicon assistant) and penetrating agent T
The test method comprises the following steps:
ethyl acetate is used for preparing mother liquor of bifenazate and etoxazole compound (bifenazate: etoxazole =1: 15) with the total concentration of 5000mg/L and cyflumetofen mother liquor with the concentration of 5000mg/L for later use.
Respectively weighing a certain amount of synergist N-380, a contrast aid NF-100 and a penetrating agent T, preparing solutions with the concentration of 50 mg/L by using distilled water respectively, and dividing into a plurality of parts. According to the experimental design shown in the following table 1, a certain amount of pre-prepared mother solution of the miticide is added to the prepared solution to a concentration of 400 mg/L. The weight ratio of the acaricide to the synergist in each solution is 8: 1.
Selecting citrus leaf mite nymphs with consistent sizes. Taking a plurality of finger tubes, placing 100 nymphs in each finger tube, then adding 5ml of each prepared treatment solution, fixing the finger tubes on a shaking table, and keeping the finger tubes to shake gently. After 1 hour, the solution in the vial was poured off and the surface of the nymph mites was washed 3 times with ethyl acetate to wash the remaining agent on the surface of the nymph mites.
Adopting an improved QuEChERS extraction method, mashing nymphae of a cleaned epidermal medicament, putting the smashed nymphae into a centrifugal tube, adding 2ml of ethyl acetate, uniformly swirling for 2 minutes, and standing for 10 minutes; then centrifuging for 10 minutes at 3000 revolutions per minute, taking supernatant, adding the supernatant into a centrifuge tube containing anhydrous sodium sulfate and ethylenediamine-N-Propylsilane (PSA), uniformly swirling for 10 seconds, centrifuging for 10 minutes at 3000 revolutions per minute, filtering by a filter membrane, concentrating the supernatant to be nearly dry, fixing the volume by using 1ml of ethyl acetate, and detecting the pesticide content in the sample by using GC. Each treatment was repeated 3 times. The results are shown in Table 1 below.
TABLE 1 permeation amounts of the respective treatment agents
The result shows that the synergist N-380 can obviously increase the amount of the bifenazate and etoxazole compound penetrating through the body surface of harmful mites, and the penetration amount is 4.21 mu g and is far greater than that of the compound added with NF-100 and the penetrating agent T. However, the penetration effect of the synergist N-380 to the control agent cyflumetofen was not significantly increased compared to the case of adding NF-100 and the penetrant T.
2. Tetranychus apple test
The mites to be tested are tetranychus pomonensis (A)Panonychusulmi)
The remaining test methods were the same as for Tetranychus citriodorus. The results are shown in Table 2 below.
TABLE 2 permeation amounts of the respective treatment agents
The result shows that the synergist N-380 can obviously increase the amount of the bifenazate and etoxazole compound penetrating through the body surface of harmful mites, and the penetration amount is 3.95 mug and is far greater than that of the compound added with NF-100 and the penetrating agent T. However, the penetration effect of the synergist N-380 to the control agent cyflumetofen was not significantly increased compared to the case of adding NF-100 and the penetrant T.
The above examples are merely illustrative and not restrictive of the present invention, and the solution obtained by simple modification of the above solution is also within the scope of the present application.
