Abstract
本发明属于医药领域,具体涉及一种含有中药提取物的胃蛋白酶包衣片,由片芯和包衣膜组成,以包衣片的质量计,所述的片芯中包含有白刺提取物5~10%、肉桂提取物5~10%、刺玫果提取物5~10%和胃蛋白酶0.01~0.02%。本发明的优点在于:提供了一种新的胃蛋白酶片配方,加入了白刺提取物、肉桂提取物和刺玫果提取物,能够激活胃腺主细胞和胃腺壁细胞能量,恢复正常的分泌胃蛋白酶原和胃酸能力,从本质上解决了胃蛋白酶分泌过少的问题,并且为胃蛋白酶提供了适宜的酸性环境,充分发挥胃蛋白酶的作用,解决消化不良症状。

技术领域
本发明属于医药领域,具体涉及一种含有中药提取物的胃蛋白酶包衣片。
背景技术
功能性消化不良(Functional dyspepsia,FD)是以上腹部疼痛、不适、饱胀、嗳气、食欲不振和恶心等为主要症状,经检查排除引起这些症状的器质性疾病的一组临床综合征。近年来,患病率高,欧美国家统计达19%-41%,平均32%;国内为18%-45%,占消化道疾病患者的20%-40%。
FD的发病机制尚不明确,可能与胃肠道运动功能障碍和感觉功能异常关系密切,心理和神经因素以及环境、饮食、胃肠激素、胃酸和HP感染参与发病,而浓茶、酒精、高脂食物、咖啡可诱发症状加重。(1)胃肠运动功能障碍胃肠运动功能障碍是FD的主要发病机制。主要包括胃排空缓慢(30-80%的FD病人有此症状)、消化间期和消化期动力异常、胃内食物分布异常、十二指肠胃反流、胃电异常。FD病人的动力异常除胃及胃肠外,还存在胆道运动功能障碍。FD患者同时存在胃排空、胆囊排空及再充盈延缓。三者之间相互关系的紊乱可能是消化不良的重要原因。(2)胃感觉功能异常内脏神经的过度敏感可能使FD患者对多种刺激因素的反应性增高。正常人进食时胃出现容受性舒张,胃的顺应性增高使胃内压力或胃壁张力无明显升高,胃张力调节使胃对扩张有不同敏感性。内脏感觉敏感性增高的机制可能会有许多。如胃壁机械受体阈值的改变、感觉信息的传导异常或中枢阈值的改变。(3)胃肠激素、NO与性激素的影响胃动素、促胃泌素等能引起胃电节律加快,诱发峰电位的产生,促进胃的排空。生长抑素、胰高糖素、抑胃肽、胃泌素等对胃肠运动有抑制作用。与刺激神经有关的迷走神经介导的NO释放参与调节胃部的动力。FD病症多见于女性,孕酮、雌二醇和泌乳素影响平滑肌的收缩并使胃肠道通过时间延长。(4)心理精神因素焦虑或抑郁等精神因素及应激状态在FD的发病中起一定作用。(5)HP感染HP在FD发病中的作用是当前最具争议性的问题之一,50%FD病人HP阳性。HP感染根除后对FD病人症状改善的效果存在争议,目前尚未确定HP感染与胃肠运动功能和感觉功能改变有无关系。(6)其他胃酸;自主神经功能紊乱;胃、十二指肠慢性炎症等。
胃蛋白酶片,是一种适应症为用于胃蛋白酶缺乏或消化功能减退引起的消化不良症的药物。1836年,索多·施旺(Theodor Schwann)在对消化过程进行的研究中,发现了一种能够参与消化作用的物质,并将其命名为胃蛋白酶。胃蛋白酶也是第一个从动物身上获得的酶。胃中惟一的一种蛋白水解酶。其最适pH值为1~2。胃蛋白酶作用的主要部位是芳香族氨基酸或酸性氨基酸的氨基所组成的肽键。此酶由胃腺的主细胞合成,以酶原颗粒形式分泌,经胃液中盐酸激活后,具有消化蛋白质的能力。药用胃蛋白酶,可以从猪胃中提取,用于消化不良。现有的胃蛋白酶片只单纯的提供人体所需的胃蛋白酶,但是无法从根本上解决胃腺主细胞分泌胃蛋白酶过少这一本质问题,另外,胃蛋白酶需要借助胃酸激活胃蛋白酶原,使之成为有活性的胃蛋白酶,并为胃蛋白酶提供适宜的酸性环境,胃酸和胃蛋白酶的分泌量反映了胃对食物的消化功能。同时,不同的酶最适pH值不同,胃蛋白酶的最适pH值为1.8,因此,如果不能首先提供适宜的胃内环境,单纯的服用胃蛋白酶片效果往往不够理想,治标不治本。
发明内容
根据以上现有技术的不足,本发明提供一种含有中药提取物的胃蛋白酶包衣片,能够提供适宜的胃内酸性环境,并激活胃腺主细胞分泌胃蛋白酶,提供胃蛋白酶的浓度和活力。
本发明所述的一种含有中药提取物的胃蛋白酶包衣片,由片芯和包衣膜组成,其特征在于:以包衣片的质量计,所述的片芯中包含有白刺提取物5~10%、肉桂提取物5~10%、刺玫果提取物5~10%和胃蛋白酶0.01~0.02%。
本发明中,白刺提取物:甘、酸,温,健脾胃,助消化,安神,解表,下乳,用于脾胃虚弱,消化不良,神经衰弱,感冒,乳汁不下。肉桂提取物:补火助阳,引火归源,散寒止痛,活血通经,暖脾胃,除积冷,通血脉,对人体免疫功能有明显的增强作用,其作用机理是,其可增强人体T淋巴细胞和B淋巴细胞的增殖与分化,并增强其功能,增强杀伤细胞的杀伤功能以及单核吞噬细胞的吞噬功能。刺玫果提取物:健脾消食,活血调经,敛肺止咳。消化不良,气滞腹泻,食欲不振,胃痛,月经不调。三者合用,行气结,以开胃肠三焦之郁,温中助化以消食,促进机体的新陈代谢,激活胃腺主细胞和胃腺壁细胞能量,恢复正常的分泌胃蛋白酶原和胃酸能力。

所述的白刺提取物采用以下方法制备而成:准备原料白刺干果,清洗,用50~95%的乙醇混合冷浸2~4h,过滤,得滤液,滤液压缩至原体积的1/5~1/4,再用纯化水冲洗回用,得初提取物;初提取物用大孔树脂吸附,再用20~60%的乙醇对大孔树脂柱洗脱,洗脱液合并混合,得粗提取物;再对粗提取物减压浓缩至原体积的1/5~1/4,再静置、抽滤、喷雾干燥制得180目以上极细粉,且含水量≤5%。
所述的白刺提取物更优选采用以下方法制备而成:准备原料白刺干果,清洗,用60%的乙醇混合冷浸4h,过滤,得滤液,滤液压缩至原体积的1/5,再用纯化水冲洗回用,得初提取物;初提取物用大孔树脂吸附,再用50%的乙醇对大孔树脂柱洗脱,洗脱液合并混合,得粗提取物;再对粗提取物减压浓缩至原体积的1/5,再静置、抽滤、喷雾干燥制得180目以上极细粉,且含水量≤5%。
所述的肉桂提取物采用以下方法制备而成:采用超临界流体萃取法,将药材粉碎至40~80目,置于萃取池中,选择萃取温度为30~40℃、萃取压力为10~20MPa、萃取时间为2~6h、二氧化碳流量为10~30L/h,萃取液用40~85%的乙醇萃取三次,再用乙醚萃取3次,用无水硫酸钠干燥,蒸除乙醚,经喷雾干燥制得180目以上极细粉,且含水量≤5%。
所述的肉桂提取物更优选采用以下方法制备而成:采用超临界流体萃取法,将药材粉碎至60目,置于萃取池中,选择萃取温度为40℃、萃取压力为16MPa、萃取时间为3h、二氧化碳流量为18L/h,萃取液用80%的乙醇萃取三次,再用乙醚萃取3次,用无水硫酸钠干燥,蒸除乙醚,经喷雾干燥制得180目以上极细粉,且含水量≤5%。
所述的刺玫果提取物采用以下方法制备而成:准备原料刺玫果,去除内部毛刺和种子,清洗,在60~70℃水中煎煮3~4h,然后静置1~2h,过滤,得滤液,滤液压缩至原体积的1/5~1/4;压缩后的滤液用60~90%的乙醇冲洗回用,再用纯化水冲洗回用,得初提取物;初提取物用大孔树脂吸附,再用50~60%的乙醇对大孔树脂柱洗脱,洗脱液合并混合,得粗提取物;再对粗提取物进行发酵10~25h,发酵后的粗提取物减压浓缩至原体积的1/5~1/4,再静置、抽滤、喷雾干燥制得180目以上极细粉,且含水量≤5%。
所述的刺玫果提取物更优选采用以下方法制备而成:准备原料刺玫果,去除内部毛刺和种子,清洗,在70℃水中煎煮3h,然后静置2h,过滤,得滤液,滤液压缩至原体积的1/5;压缩后的滤液用80%的乙醇冲洗回用,再用纯化水冲洗回用,得初提取物;初提取物用大孔树脂吸附,再用50%的乙醇对大孔树脂柱洗脱,洗脱液合并混合,得粗提取物;再对粗提取物进行发酵20h,发酵后的粗提取物减压浓缩至原体积的1/5,再静置、抽滤、喷雾干燥制得180目以上极细粉,且含水量≤5%。
所述的包衣膜为糖衣膜,包括选用虫胶乙醇溶液作为隔离层材料;选用滑石粉作为粉衣层材料;选用糖浆溶液作为糖衣层材料;选用巴西棕榈蜡中的作为打光剂。
所述的糖衣膜的包衣工艺为取合格的片芯,用虫胶乙醇溶液先包3~5层隔离层,再依次包粉衣层及糖衣层,须层层干燥,干燥温度为40~50℃,最后打光。
本发明包衣片的制备方法,采用现有常规工艺即可,将片芯中的原辅料投入到混合机内混合均匀,采用湿法制粒或喷雾制粒,并干燥及整粒,将上述干颗粒混合均匀,并且压片,最后包衣。
本发明的优点在于:提供了一种新的胃蛋白酶片配方,加入了白刺提取物、肉桂提取物和刺玫果提取物,能够激活胃腺主细胞和胃腺壁细胞能量,恢复正常的分泌胃蛋白酶原和胃酸能力,从本质上解决了胃蛋白酶分泌过少的问题,并且为胃蛋白酶提供了适宜的酸性环境,充分发挥胃蛋白酶的作用,解决消化不良症状。

一种含有中药提取物的胃蛋白酶包衣片,由片芯和包衣膜组成,以包衣片的质量计,所述的片芯中包含有白刺提取物5%、肉桂提取物7%、刺玫果提取物5%、胃蛋白酶0.01%、蔗糖粉70%、羧甲淀粉钠2%、硬脂酸镁0.2%。
所述的白刺提取物采用以下方法制备而成:准备原料白刺干果,清洗,用60%的乙醇混合冷浸4h,过滤,得滤液,滤液压缩至原体积的1/5,再用纯化水冲洗回用,得初提取物;初提取物用大孔树脂吸附,再用50%的乙醇对大孔树脂柱洗脱,洗脱液合并混合,得粗提取物;再对粗提取物减压浓缩至原体积的1/5,再静置、抽滤、喷雾干燥制得180目以上极细粉,且含水量≤5%。
所述的肉桂提取物采用以下方法制备而成:采用超临界流体萃取法,将药材粉碎至60目,置于萃取池中,选择萃取温度为40℃、萃取压力为16MPa、萃取时间为3h、二氧化碳流量为18L/h,萃取液用80%的乙醇萃取三次,再用乙醚萃取3次,用无水硫酸钠干燥,蒸除乙醚,经喷雾干燥制得180目以上极细粉,且含水量≤5%。
所述的刺玫果提取物采用以下方法制备而成:准备原料刺玫果,去除内部毛刺和种子,清洗,在70℃水中煎煮3h,然后静置2h,过滤,得滤液,滤液压缩至原体积的1/5;压缩后的滤液用80%的乙醇冲洗回用,再用纯化水冲洗回用,得初提取物;初提取物用大孔树脂吸附,再用50%的乙醇对大孔树脂柱洗脱,洗脱液合并混合,得粗提取物;再对粗提取物进行发酵20h,发酵后的粗提取物减压浓缩至原体积的1/5,再静置、抽滤、喷雾干燥制得180目以上极细粉,且含水量≤5%。
所述的包衣膜为糖衣膜,包括选用虫胶乙醇溶液作为隔离层材料;选用滑石粉作为粉衣层材料;选用糖浆溶液作为糖衣层材料;选用巴西棕榈蜡中的作为打光剂。

包衣片的制备方法,采用现有常规工艺即可,将片芯中的原辅料投入到混合机内混合均匀,采用湿法制粒或喷雾制粒,并干燥及整粒,将上述干颗粒混合均匀,并且压片,最后包衣。

一种含有中药提取物的胃蛋白酶包衣片,由片芯和包衣膜组成,以包衣片的质量计,所述的片芯中包含有白刺提取物8%、肉桂提取物8%、刺玫果提取物7%、胃蛋白酶0.01%、蔗糖粉65%、羧甲淀粉钠3%、硬脂酸镁0.1%。
所述的白刺提取物采用以下方法制备而成:准备原料白刺干果,清洗,用80%的乙醇混合冷浸3h,过滤,得滤液,滤液压缩至原体积的1/5,再用纯化水冲洗回用,得初提取物;初提取物用大孔树脂吸附,再用40%的乙醇对大孔树脂柱洗脱,洗脱液合并混合,得粗提取物;再对粗提取物减压浓缩至原体积的1/5,再静置、抽滤、喷雾干燥制得180目以上极细粉,且含水量≤5%。
所述的肉桂提取物采用以下方法制备而成:采用超临界流体萃取法,将药材粉碎至40~80目,置于萃取池中,选择萃取温度为30℃、萃取压力为14MPa、萃取时间为3h、二氧化碳流量为20L/h,萃取液用60%的乙醇萃取三次,再用乙醚萃取3次,用无水硫酸钠干燥,蒸除乙醚,经喷雾干燥制得180目以上极细粉,且含水量≤5%。
所述的刺玫果提取物采用以下方法制备而成:准备原料刺玫果,去除内部毛刺和种子,清洗,在65℃水中煎煮3h,然后静置2h,过滤,得滤液,滤液压缩至原体积的1/5;压缩后的滤液用80%的乙醇冲洗回用,再用纯化水冲洗回用,得初提取物;初提取物用大孔树脂吸附,再用50%的乙醇对大孔树脂柱洗脱,洗脱液合并混合,得粗提取物;再对粗提取物进行发酵15h,发酵后的粗提取物减压浓缩至原体积的1/5,再静置、抽滤、喷雾干燥制得180目以上极细粉,且含水量≤5%。

所述的刺玫果提取物采用以下方法制备而成:准备原料刺玫果,去除内部毛刺和种子,清洗,在70℃水中煎煮3h,然后静置2h,过滤,得滤液,滤液压缩至原体积的1/4;压缩后的滤液用90%的乙醇冲洗回用,再用纯化水冲洗回用,得初提取物;初提取物用大孔树脂吸附,再用60%的乙醇对大孔树脂柱洗脱,洗脱液合并混合,得粗提取物;再对粗提取物进行发酵20h,发酵后的粗提取物减压浓缩至原体积的1/4,再静置、抽滤、喷雾干燥制得180目以上极细粉,且含水量≤5%。
所述的包衣膜为糖衣膜,包括选用虫胶乙醇溶液作为隔离层材料;选用滑石粉作为粉衣层材料;选用糖浆溶液作为糖衣层材料;选用巴西棕榈蜡中的作为打光剂。
所述的糖衣膜的包衣工艺为取合格的片芯,用虫胶乙醇溶液先包4层隔离层,再依次包粉衣层及糖衣层,须层层干燥,干燥温度为40℃,最后打光。
包衣片的制备方法,采用现有常规工艺即可,将片芯中的原辅料投入到混合机内混合均匀,采用湿法制粒或喷雾制粒,并干燥及整粒,将上述干颗粒混合均匀,并且压片,最后包衣。
实施例4:
采用实施例1~3制备得到的包衣片,进行胃酸胃蛋白酶影响实验。
1.1实验材料
1.1.1实验动物:SPF级SD大鼠50只,雌雄各半,体重为(180~220)g。
1.2实验方法
1.2.1分组与给药
大鼠随机分为正常组、空白对照组、实施例1组、实施例2组、实施例3组。正常组灌服胃蛋白酶片(丹东华盛生物制药有限公司提供)、空白对照组灌服生理盐水,各组灌服量为20g/kg(体重),各组1次/d灌胃,每次用药量均按1g(药片)/100g(水)计算。连续灌胃14天。
1.2.2胃液标本的收集
于最后1次给药后2h,禁食(不禁水)12h,以25mg﹒kg-1戊巴比妥钠麻醉大鼠,解剖上腹部,从幽门向胃插入一直径约为2mm的塑料管,在胃靠近幽门球部结扎固定,以收集胃液,并由口腔经食管用塑料管插入前胃,与食管一起结扎,以pH为7.0、35℃的生理盐水用蠕动泵按10mL﹒h-1的速度进行胃内灌流,收集1小时流出液,供实验用。
1.2.3指标观察与方法
(1)胃液酸度测定:采集到的胃液用pH测定仪直接测定pH值。
(2)胃蛋白酶含量测定:按照ELISA试剂盒说明书操作,测定胃蛋白酶含量。
(3)胃蛋白酶活性单位测定:应用酪氨酸法测定胃蛋白酶的活性:先制备酪氨酸标准曲线,然后分别取酪氨酸对照液,胃液各1mL,置37℃水浴中,保温5分钟,精密加入预热至37℃的血红蛋白试液5mL,摇匀,立即计时,在37℃水浴中反应10分钟,立即精密加入5%三氯醋酸5mL,摇匀,滤清,在275nm处测定吸收度(同时分别做空白对照)。胃蛋白酶活力按CP90方法的公式计算即得:在规定的条件下每分钟能酶解血红蛋白生成1mg的酪氨酸为1个胃蛋白酶活力单位。
1.2.4统计学方法
各组数据以均数±标准差(x±s)表示,使用SPSS软件行单因素方差分析(one-wayANOVA),设定p值小于0.05有统计学意义。
2结果
2.1实验各组大鼠胃液酸度、胃蛋白酶浓度、胃蛋白酶活力的影响的比较,结果见表1。

实验结果表明:本发明制备的胃蛋白酶包衣片能使实验组大鼠胃液氢离子浓度增加(即胃液酸度增加,pH值降低)、胃蛋白酶浓度增加以及胃蛋白酶活力增加,并且伴随片剂中丁香叶提取物和山楂提取物的浓度有正相关,因此,可以认定,白刺提取物、肉桂提取物和刺玫果提取物的合用能够激活胃腺主细胞和胃腺壁细胞能量,恢复正常的分泌胃蛋白酶原和胃酸能力,从本质上解决了胃蛋白酶分泌过少的问题,并且为胃蛋白酶提供了适宜的酸性环境,充分发挥胃蛋白酶的作用。
Claims (10)
1.一种含有中药提取物的胃蛋白酶包衣片,由片芯和包衣膜组成,其特征在于:以包衣片的质量计,所述的片芯中包含有白刺提取物5~10%、肉桂提取物5~10%、刺玫果提取物5~10%和胃蛋白酶0.01~0.02%。
2.根据权利要求1所述的一种含有中药提取物的胃蛋白酶包衣片,其特征在于:所述的片芯中还包含有蔗糖粉50~80%、羧甲淀粉钠1~6%、硬脂酸镁0.1~1%。
3.根据权利要求1所述的一种含有中药提取物的胃蛋白酶包衣片,其特征在于:所述的白刺提取物采用以下方法制备而成:准备原料白刺干果,清洗,用50~95%的乙醇混合冷浸2~4h,过滤,得滤液,滤液压缩至原体积的1/5~1/4,再用纯化水冲洗回用,得初提取物;初提取物用大孔树脂吸附,再用20~60%的乙醇对大孔树脂柱洗脱,洗脱液合并混合,得粗提取物;再对粗提取物减压浓缩至原体积的1/5~1/4,再静置、抽滤、喷雾干燥制得180目以上极细粉,且含水量≤5%。
4.根据权利要求3所述的一种含有中药提取物的胃蛋白酶包衣片,其特征在于:所述的白刺提取物采用以下方法制备而成:准备原料白刺干果,清洗,用60%的乙醇混合冷浸4h,过滤,得滤液,滤液压缩至原体积的1/5,再用纯化水冲洗回用,得初提取物;初提取物用大孔树脂吸附,再用50%的乙醇对大孔树脂柱洗脱,洗脱液合并混合,得粗提取物;再对粗提取物减压浓缩至原体积的1/5,再静置、抽滤、喷雾干燥制得180目以上极细粉,且含水量≤5%。
5.根据权利要求1所述的一种含有中药提取物的胃蛋白酶包衣片,其特征在于:所述的肉桂提取物采用以下方法制备而成:采用超临界流体萃取法,将药材粉碎至40~80目,置于萃取池中,选择萃取温度为30~40℃、萃取压力为10~20MPa、萃取时间为2~6h、二氧化碳流量为10~30L/h,萃取液用40~85%的乙醇萃取三次,再用乙醚萃取3次,用无水硫酸钠干燥,蒸除乙醚,经喷雾干燥制得180目以上极细粉,且含水量≤5%。
6.根据权利要求5所述的一种含有中药提取物的胃蛋白酶包衣片,其特征在于:所述的肉桂提取物采用以下方法制备而成:采用超临界流体萃取法,将药材粉碎至60目,置于萃取池中,选择萃取温度为40℃、萃取压力为16MPa、萃取时间为3h、二氧化碳流量为18L/h,萃取液用80%的乙醇萃取三次,再用乙醚萃取3次,用无水硫酸钠干燥,蒸除乙醚,经喷雾干燥制得180目以上极细粉,且含水量≤5%。
7.根据权利要求1所述的一种含有中药提取物的胃蛋白酶包衣片,其特征在于:所述的刺玫果提取物采用以下方法制备而成:准备原料刺玫果,去除内部毛刺和种子,清洗,在60~70℃水中煎煮3~4h,然后静置1~2h,过滤,得滤液,滤液压缩至原体积的1/5~1/4;压缩后的滤液用60~90%的乙醇冲洗回用,再用纯化水冲洗回用,得初提取物;初提取物用大孔树脂吸附,再用50~60%的乙醇对大孔树脂柱洗脱,洗脱液合并混合,得粗提取物;再对粗提取物进行发酵10~25h,发酵后的粗提取物减压浓缩至原体积的1/5~1/4,再静置、抽滤、喷雾干燥制得180目以上极细粉,且含水量≤5%。
8.根据权利要求7所述的一种含有中药提取物的胃蛋白酶包衣片,其特征在于:所述的刺玫果提取物采用以下方法制备而成:准备原料刺玫果,去除内部毛刺和种子,清洗,在70℃水中煎煮3h,然后静置2h,过滤,得滤液,滤液压缩至原体积的1/5;压缩后的滤液用80%的乙醇冲洗回用,再用纯化水冲洗回用,得初提取物;初提取物用大孔树脂吸附,再用50%的乙醇对大孔树脂柱洗脱,洗脱液合并混合,得粗提取物;再对粗提取物进行发酵20h,发酵后的粗提取物减压浓缩至原体积的1/5,再静置、抽滤、喷雾干燥制得180目以上极细粉,且含水量≤5%。
9.根据权利要求1所述的一种含有中药提取物的胃蛋白酶包衣片,其特征在于:所述的包衣膜为糖衣膜,包括选用虫胶乙醇溶液作为隔离层材料;选用滑石粉作为粉衣层材料;选用糖浆溶液作为糖衣层材料;选用巴西棕榈蜡中的作为打光剂。
10.根据权利要求7所述的一种含有中药提取物的胃蛋白酶包衣片,其特征在于:所述的糖衣膜的包衣工艺为取合格的片芯,用虫胶乙醇溶液先包3~5层隔离层,再依次包粉衣层及糖衣层,须层层干燥,干燥温度为40~50℃,最后打光。
