Abstract

本发明公开了一种由NHPI与含苄基化合物在非金属催化下直接偶联制备PINO衍生物的方法，该方法是在氮气保护下，含苄基化合物与N‑羟基邻苯二甲酰亚胺和K2S2O8，在四烷基季铵盐催化下一锅反应，生成PINO衍生物；该方法在温和条件下一步反应高产率合成PINO衍生物，且成本低，操作简单，满足工业生产要求。

Description

一种由NHPI与含苄基化合物在非金属催化下直接偶联制备 PINO衍生物的方法
技术领域
本发明涉及一种PINO衍生物的制备方法，特别涉及一种由NHPI与含苄基化合物在非金属催化下直接偶联制备PINO衍生物的方法，属于有机合成领域。
背景技术
多年来，自由基化学一直都是研究的热点，因为它在有机合成和工业应用中的确起着重要作用。本领域公知，N-羟基邻苯二甲酰亚胺(NHPI)能产生高反应活性的N-氧邻苯二甲酰亚胺(PINO)自由基，可以用作催化剂或参与各种类型的偶联反应。一些PINO化合物可以很容易地转化为相应的醇或羟胺类化合物，两者都是药物合成中非常有用的中间体。在现有技术中，报道了几种合成PINO化合物方法，主要是通过使用金属盐催化反应生成PINO基团，如采用氯化亚铜或铈(IV)、铵(CAN)等作为催化剂，如反应式(1)～(3)。
然而，在这些报道的合成N-氧邻苯二甲酰亚胺(PINO)化合物的反应中用到了昂贵的过渡金属催化剂，且收率较低，造成生产成本高，难以实现工业化生产。
目前，也有报道采用有机过氧化物来以完成反应的相关报道，如反应式(4)。
在这个报道的合成N-氧邻苯二甲酰亚胺(PINO)化合物的反应必须要使用大大过量甲苯来提高NHPI的转化率，且收率较低，而且还用到了具有安全隐患的过氧化物，对环境破坏，也增加了潜在的危险。
发明内容
针对现有的合成PINO衍生物的方法存在使用过渡金属催化剂及高活性过氧化物作为氧化剂，成本高、安全系数低、产率低，难以适用工业化生产等缺陷，本发明的目的是在于提供一种NHPI与含苄基化合物原料在温和条件下一步高产率合成PINO衍生物的方法，该方法成本低，操作简单，满足工业生产要求。
为了实现上述技术目的，本发明提供了一种由NHPI与含苄基化合物在非金属催化下直接偶联制备PINO衍生物的方法，该方法是在氮气保护下，式1含苄基化合物与N-羟基邻苯二甲酰亚胺和K2S2O8，在四烷基季铵盐催化下一锅反应，生成式2PINO衍生物；
其中，
R1和R3独立地选自为氢、卤素、硝基、烷基酰基或烷基；
R2为氢、烷基或芳基烷基；
或者，R1和R2为环状烷氧链；
或者，R2和R3为环状烷氧链。
优选的方案，R1和R3独立地选自为氢、氯、溴、硝基、C1～C4烷基酰基或C1～C4烷基，R2为氢、C1～C4烷基或苄基；或者，R1和R2为C2或C3环状烷氧链；或者，R2和R3为C2或C3环状烷氧链。较优选的方案，R1和R3独立地选自为氢、氯、溴、硝基、乙酰基或甲基；R2为氢、甲基或苄基；或者，R1和R2为C2或C3环状烷氧链；或者，R2和R3为C2或C3环状烷氧链。
最优选的方案，式1含苄基化合物为以下结构式中一种：
优选的方案，反应的条件为：在70℃～90℃温度下反应8～16h；较优选在80℃温度下反应12h。
较优选的方案，反应采用DCE和/或MeCN为溶剂。最优选为采用DCE为溶剂。
优选的方案，四烷基季铵盐为四丁基溴化铵、四丁基碘化铵、四丁基氯化铵、四丁基氟化铵、四丁基硫酸氢铵中至少一种；较优选为四丁基碘化铵。
优选的方案，四烷基季铵盐的用量为含苄基化合物摩尔量的10～30％。
优选的方案，含苄基化合物与NHPI的最佳反应摩尔比为1∶2。K2S2O8的相对含苄基化合物用量为2.0equiv。
本发明的技术方案，在氮气保护下，含苄基化合物与N-羟基邻苯二甲酰亚胺和K2S2O8，在四丁基碘化铵催化下反应，生成PINO衍生物，反应方程式如下：
基于大量的实验总结以及参考先前文献报道，本发明提出了如下合理的反应机制。以甲苯和N-羟基邻苯二甲酰亚胺为原料，K2S2O8为氧化剂，四丁基碘化铵为催化剂，进行具体说明。
首先，四丁基碘化铵与K2S2O8离子交换生成过硫酸季铵盐，在加热作用下分解成季铵硫酸根自由基，季铵硫酸根自由基捕获N-羟基邻苯二甲酰亚胺的氢质子，生成N-氧基邻苯二甲酰亚胺自由基；一分子N-氧基邻苯二甲酰亚胺自由基捕获甲苯的α氢，生成苄基自由基，一分子N-氧基邻苯二甲酰亚胺自由基与苄基自由基偶联反应，生成目标产物；季铵硫酸根自由基捕获氢质子后生成季铵硫酸氢盐，与碘负离子交换后，转化成四丁基碘化铵，继续催化K2S2O8产生自由基；具体反应式如下：
通过在反应中加入BHT(2，6-二叔丁基-4-甲基苯酚)或加入TEMPO，反应几乎得不到目标产物，进一步验证该反应为自由基反应机理；反应式如下。
相对现有技术，本发明的技术方案带来的有益技术效果：
1)本发明的技术方案首次以过硫酸钾为氧化剂，以季铵盐为催化剂，由含苄基化合物与N-羟基邻苯二甲酰亚胺一步合成PINO衍生物，克服了现有技术中必须使用昂贵金属催化剂或采用安全性能低的过氧化物作为氧化剂的缺陷。
2)本发明的技术方案通过一锅法一步反应生成PINO衍生物，步骤简单，流程短，产率高，成本低，有利于工业化生产，克服了现有合成PINO衍生物的方法存在产率低，成本高，难以工业化生产的缺点。
3)本发明的技术方案反应条件温和，反应选择性高，满足工业生产要求。
4)本发明的技术方案可以在含苄基化合物上修饰各种取代基团，很容易地转变成相应的醇或羟胺物产品，在药物中间体的应用具有重要意义。
具体实施方式
以下实施例旨在进一步说明本发明内容，而不是限制本发明权利要求的保护范围。
以下实施例中的目标产物均为本领域常规的产物，都存在已有文献报道，本发明的实施例旨在重点介绍本发明的新型合成方法，目标产物表征可借鉴现有文献报道，这里不再介绍。
实施例1～15按以下反应方程式进行：
将含苄基化合物(0.5mmol)、N-羟基邻苯二甲酰亚胺(1.0mmol)、K2S2O8(2.0equiv)、TBAI(10mol％)、DCE(2.0mL)加入到10mL反应容器中，通入氮气保护，加热至80℃反应12h，产物采用色谱柱分离。

对照试验组1～17：
将甲苯(0.5mmol)、N-羟基邻苯二甲酰亚胺(1.0mmol)、K2S2O8(2.0equiv)、四烷基季铵盐、溶剂(2.0mL)加入到10mL反应容器中，通入氮气保护，加热至反应12h，产物采用色谱柱分离；各对照试验组的具体反应条件如表1所示。
具体反应方程式如下：

TBAB(四丁基溴化铵)、TBAI(四丁基碘化铵)、TBAC(四丁基氯化铵)、TBAF(四丁基氟化铵)、TBAHS(四丁基硫酸氢铵)。
从上述表中可以看出，四烷基季铵盐催化剂的选择对反应的影响比较大，在其它条件相同的情况下，TBAB、TBAI、TBAC、TBAF作为催化剂，催化效率呈现一定规律变化，采用TBAI具有最佳的催化效率，如实验组1～4所示。而其它四烷基季铵盐催化剂的催化效率比TBAI也要低，如试验组5所示。而采用碘化铵无催化作用(如实验组17)。
从上表还可以看出，溶剂的选择也直接影响反应的产率，发现采用DCE和乙腈等中等偏弱极性非质子溶剂中可以进行反应，而在强极性非质子溶剂和质子溶剂中几乎得不到产物，如实验组6～11；如果不采用其它溶剂，直接以甲苯作为反应剂和溶剂，也几乎得不到产物。
从上表还可以看出，反应温度最好是在70℃以上，最好是在80～90℃反应。

Claims (5)

1.一种由NHPI与含苄基化合物在非金属催化下直接偶联制备PINO衍生物的方法，其特征在于：在氮气保护下，式1含苄基化合物与N-羟基邻苯二甲酰亚胺和K2S2O8，在四烷基季铵盐催化下一锅反应，生成式2 PINO衍生物；
其中，
R1和R3独立地选自为氢、卤素、硝基、烷基酰基或烷基；
R2为氢、烷基或芳基烷基；
或者，R1和R2为环状烷氧链；
或者，R2和R3为环状烷氧链；
所述反应的条件为：在70℃～90℃温度下反应8～16h；
所述反应采用DCE和/或MeCN为溶剂；
所述的四烷基季铵盐为四丁基溴化铵、四丁基碘化铵、四丁基氯化铵、四丁基氟化铵、四丁基硫酸氢铵中至少一种。
2.根据权利要求1所述的由NHPI与含苄基化合物在非金属催化下直接偶联制备PINO衍生物的方法，其特征在于：
R1和R3独立地选自为氢、氯、溴、硝基、C1～C4烷基酰基或C1～C4烷基；
R2为氢、C1～C4烷基或苄基；
或者，R1和R2为C2或C3环状烷氧链；
或者，R2和R3为C2或C3环状烷氧链。
3.根据权利要求2所述的由NHPI与含苄基化合物在非金属催化下直接偶联制备PINO衍生物的方法，其特征在于：
R1和R3独立地选自为氢、氯、溴、硝基、乙酰基或甲基；
R2为氢、甲基或苄基；
或者，R1和R2为C2或C3环状烷氧链；
或者，R2和R3为C2或C3环状烷氧链。
4.根据权利要求1所述的由NHPI与含苄基化合物在非金属催化下直接偶联制备PINO衍生物的方法，其特征在于：所述的四烷基季铵盐为四丁基碘化铵。
5.根据权利要求1或4所述的由NHPI与含苄基化合物在非金属催化下直接偶联制备PINO衍生物的方法，其特征在于：所述的四烷基季铵盐用量为含苄基化合物摩尔量的10～30％。
