Abstract
本发明公开了半透膜分层一体式生物反应器,包括腔体,设置在腔体一侧的血浆分离器及另一侧的生物反应器;血浆分离器和生物反应器通过分隔半透膜间隔开,生物反应器内设有多层过滤半透膜,相邻的两个过滤半透膜与生物反应器的侧壁一起形成一个独立的滤层空间。本发明的有益效果是:通过将血浆分离器与生物反应器拼接或直接将血浆分离器置于生物反应器中,设计更简单实用;分离的血浆通过分离泵连接管进入生物反应器,然后逐层透过间距设置的过滤半透膜,与滤层空间内的细胞进行物质交换,这样可使血浆始终保持稳定的均匀流速,有利于提高两类物质的交换效率。

技术领域
本发明属于生物人工肝技术领域,尤其涉及一种半透膜分层一体式生物反应器。
背景技术
我国乙型肝炎病毒携带者约占总人口的十分之一,而肝功能衰竭即可由单型病毒或两种甚至两种以上的病毒混合感染引起,另外食物药物中毒和缺氧性肝损伤等原因亦可造成肝衰竭。中国肝衰竭病人众多,寻求新的肝衰竭的有效治疗手段成为亟待解决的问题。肝功能衰竭是一类严重的临床疾病,有效治疗手段少,多预后不良,原位肝移植是目前治疗该病最为有效的手段之一。然而,由于供体器官的严重短缺、手术费用昂贵、长期使用免疫抑制剂引发严重感染、恶性肿瘤及慢性移植物失功等原因,目前只有少部分患者能够获得肝移植的机会,大部分患者因为等待时间过长或在等待肝移植手术过程中死亡。在这样的背景下,旨在取代部分肝功能,为患者等待肝源或自身肝脏再生架起桥梁的人工肝支持系统得以发展。
人工肝支持系统包括物理人工肝和生物人工肝,物理人工肝系统虽可以去除体内代谢产生的一些毒素如胆红素和胆汁酸等,但同时也可吸附一些有益蛋白和凝血因子等,治疗效果不理想。近年来,生物人工肝系统已成为临床研发热点,生物人工肝系统即是将活性肝细胞(或具有肝功能的其他细胞)加入到系统内,由于肝细胞具有解毒、合成、生物转化等肝功能,因此当患者血液流经反应器时,通过半透膜或直接接触的方式与培养的细胞进行物质交换,可有效替代肝脏的解毒功能和合成功能,从而达到支持和治疗目的。
生物反应器是生物人工肝系统的核心部分,一个理想的生物反应器应能达到如下要求:(1)为肝细胞提供良好的长期生长代谢环境;(2)能发挥必需的肝脏功能;(3)体积要尽可能地小;(4)要尽可能的减少死腔。就结构而言,生物反应器可分成以下四种:中空纤维管式、单层/多层平板式、灌流床/支架式和微囊悬液式。目前研究及应用最为广泛的一类为中空纤维型,该反应器虽然设计经典,但也存在一些问题,如容积有限,细胞装载量小,另外,细胞生长时易堵塞纤维丝,影响膜内外的物质交换。因此,如何设计研发生物反应器使其满足日益扩大的需求越发重要。
发明内容
本发明的目的在于克服现有的生物反应器容积有限,细胞装载量小,血浆与肝细胞交换面积有限的问题;提供一种半透膜分层一体式生物反应器,它具有装载细胞方便、装载量大、提高物质的交换效率及均匀程度的特点。
为实现上述目的,本发明所设计的半透膜分层一体式生物反应器,包括腔体,设置在腔体一侧的血浆分离器及设置在腔体另一侧的生物反应器;血浆分离器和生物反应器通过分隔半透膜间隔开;
位于血浆分离器所在一侧的腔体上开设有血液入口和血细胞出口,血浆分离器内部填充有多根中空纤维丝;所述中空纤维丝一端与血液入口连通,另一端与血细胞出口连通;
位于生物反应器所在另一侧的腔体的顶部开设有血浆出口,生物反应器内水平设置有多层过滤半透膜,所述过滤半透膜用于阻隔游离细胞穿过且相邻的两个过滤半透膜与生物反应器的侧壁一起形成一个独立的滤层空间,每个独立的滤层空间对应的腔体上设有肝细胞灌注口;最上层的过滤半透膜与生物反应器顶部保持分离;最下层的过滤半透膜与生物反应器底部形成夹层,所述夹层内填充有透气纤维丝且所述夹层对应的腔体上设有进气口,所述进气口仅与透气纤维丝连通;
所述血浆分离器和生物反应器通过设有分离泵的分离泵连接管将两者连通;所述分离泵连接管的入口端位于所述血浆分离器所在一侧的腔体的侧壁下端,出口端位于所述生物反应器所在一侧的腔体的底部。
进一步地,所述过滤半透膜及分隔半透膜均为平片膜元件,所述平片膜元件的周边与腔体贴合固定。
优选地,所述过滤半透膜的孔径按照远离所述分离泵连接管出口端的方向逐个递减。
上述优选方案的有益效果是:通过将过滤半透膜的表面细孔直径采用逐个递减的方式设置,不仅可以防止过滤半透膜发生堵塞,也能保证血浆流速均匀稳定。
进一步地,所述血浆出口内设有阻止细胞透过的阻隔半透膜。
上述进一步方案的有益效果是:通过阻隔半透膜的作用,能避免细胞或细胞产物进入血液,阻隔大分子物质透过,防止过敏。
再进一步地,所述夹层在所述进气口与透气纤维丝之间还设有用于阻隔血浆/预充液的密封支架板;所述密封支架板与所述进气口间距设置;所述密封支架板上开设有多个小孔;所述透气纤维丝一端卡设在小孔内,且通过密封支架板与进气口之间的空间与所述进气口相连通。
优选地,所述滤层空间对应的腔体上还设有备用出口及压力传感器。
上述优选方案的有益效果是:通过压力传感器检测是否有过滤半透膜发生堵塞,当发生堵塞时,通过备用的连接管接通两个备用出口,防止过滤半透膜发生堵塞时造成循环中断,使血浆通过备用的连接管进入下一个没有过滤半透膜堵塞的滤层空间,使治疗继续进行。
进一步地,所述血液入口处设有与其连通的高氧水入口;所述分离泵连接管上还设有压力检测装置。
上述进一步方案的有益效果是:通过高氧水入口向生物反应器内通入高氧水以氧合血浆同时保证细胞供氧,细胞培养效果更佳。
可选地,所述夹层对应的腔体上设有出气口/预留口。
可选地,所述一体式生物反应器还包括设于腔体外的37℃保温加热套及摇床。
上述可选方案的有益效果是:通过加热套的作用,使得本发明的半透膜分层一体式生物反应器控温为37℃,并通过摇床的作用,使其可持续摇动,避免细胞堆积,使治疗效果发挥到最佳。
进一步地,所述分隔半透膜的孔径为0.3~5微米,最下层的过滤半透膜的孔径为5微米。
本发明的有益效果是:通过改进现有人工肝系统中生物人工肝反应器的结构,将血浆分离器与生物反应器拼接或直接将血浆分离器置于生物反应器中,设计更简单实用;通入高氧水以氧合血浆同时保证细胞供氧,通过透气纤维丝的作用进一步提供足量的氧气,能为细胞提供适宜代谢环境,保障细胞活性,使细胞培养效果更佳,有利于细胞功能的维持,提高治疗效果;分离的血浆通过设有分离泵的分离泵连接管进入生物反应器,通过夹层后,再逐层透过间距设置的过滤半透膜,与滤层空间内的细胞进行物质交换,这样可使血浆始终保持稳定的均匀流速,细胞与血浆之间充分接触,有利于提高两类物质的交换效率,降解血浆效果更好;增大了细胞存放空间,加大了装载量;通过过滤半透膜、阻隔半透膜的作用降低了细胞随血浆进入人体的风险;过滤半透膜采用多层设置,相对于并联生物反应器而言提高了细胞的使用效率;另外通过控温和持续摇动,有利于细胞功能的发挥,使治疗效果发挥到最佳。

具体实施方式
以下结合附图和具体实施例对本发明作进一步的详细描述。
如图1所示的半透膜分层一体式生物反应器,包括腔体21,设置在腔体21一侧的血浆分离器1及设置在腔体21另一侧的生物反应器2;血浆分离器1和生物反应器2通过分隔半透膜3间隔开;
位于血浆分离器1所在一侧的腔体21上开设有血液入口4和血细胞出口5,血浆分离器1内部填充有多根中空纤维丝6;中空纤维丝6一端与血液入口4连通,另一端与血细胞出口5连通;
位于生物反应器2所在另一侧的腔体21的顶部开设有血浆出口7,生物反应器2内水平设置有多层过滤半透膜8,过滤半透膜8用于阻隔游离细胞穿过且相邻的两个过滤半透膜8与生物反应器2的侧壁一起形成一个独立的滤层空间10,每个独立的滤层空间10对应的腔体21上设有肝细胞灌注口9;最上层的过滤半透膜8与生物反应器2顶部保持分离;最下层的过滤半透膜8与生物反应器2底部形成夹层25,夹层25内填充有透气纤维丝13且夹层25对应的腔体21上设有进气口24,进气口24仅与透气纤维丝13连通;
血浆分离器1和生物反应器2通过设有分离泵22的分离泵连接管23将两者连通;分离泵连接管23的入口端位于血浆分离器1所在一侧的腔体21的侧壁下端,出口端位于生物反应器2所在一侧的腔体21的底部。
过滤半透膜8及分隔半透膜3均为平片膜元件,平片膜元件的周边与腔体21贴合固定。
过滤半透膜8的孔径按照远离分离泵连接管23出口端的方向逐个递减。通过将过滤半透膜8的表面细孔直径采用逐个递减的方式设置,不仅可以防止过滤半透膜8发生堵塞,也能保证血浆流速均匀稳定。
血浆出口7内设有阻止细胞透过的阻隔半透膜16。通过阻隔半透膜16的作用,能避免细胞或细胞产物进入血液,阻隔大分子物质透过,防止过敏。
夹层25在进气口24与透气纤维丝13之间还设有用于阻隔血浆/预充液的密封支架板26;密封支架板26与进气口24间距设置;密封支架板26上开设有多个小孔;透气纤维丝13一端卡设在小孔内,且通过密封支架板26与进气口24之间的空间与进气口24相连通。
滤层空间10对应的腔体21上还设有备用出口11及压力传感器12。通过压力传感器12检测是否有过滤半透膜8发生堵塞,当发生堵塞时,通过备用的连接管(图未示)接通两个备用出口,防止过滤半透膜8发生堵塞时造成循环中断,使血浆通过备用的连接管进入下一个没有过滤半透膜8堵塞的滤层空间10,使治疗继续进行。
血液入口4处设有与其连通的高氧水入口14;分离泵连接管23上还设有压力检测装置20。通过高氧水入口向生物反应器内通入高氧水以氧合血浆同时保证细胞供氧,细胞培养效果更佳。
夹层25对应的腔体21上设有出气口/预留口18。
一体式生物反应器还包括设于腔体21外的37℃保温加热套(图未示)及摇床(图未示)。通过加热套的作用,使得本发明的半透膜分层一体式生物反应器控温为37℃,并通过摇床的作用,使其可持续摇动,避免细胞堆积,使治疗效果发挥到最佳。
分隔半透膜3的孔径为0.3~5微米,最下层的过滤半透膜8的孔径为5微米。
血细胞出口5与血浆出口7通过管路17连接汇合,在汇合后的管路17上设有正向蠕动泵15;通过正向蠕动泵15拉动系统循环的进行。血浆分离器1上还开设有血浆分离器备用加样口19。
本发明的原理如下
本发明的肝细胞灌注为微载体灌注法,即将贴附在微载体上的细胞连同微载体通过每个肝细胞灌注口9灌注入,灌注体积约为滤层空间10体积的2/3。
当血液从血液入口4进入血浆分离器1中,在中空纤维丝6的作用下,血液被分离,血细胞和部分血浆从血细胞出口5流出,剩余部分血浆通过分离泵连接管23进入生物反应器2内;血浆首先处于设有透气纤维丝13的夹层25内,进气口24向夹层25内充入氧气,之后血浆透过最下层的过滤半透膜8进入滤层空间10,并与该滤层空间10内的细胞进行充分反应,反应后进入下一个滤层空间10,依次进行,逐层穿过过滤半透膜8,直至血浆透过最上层的过滤半透膜8,最后通过血浆出口7排出,并与血细胞出口5排出的血细胞和部分血浆混合后输入人体。由于靠近分离泵连接管23出口端的过滤半透膜8更易发生堵塞,因此采用将过滤半透膜8的孔径按照远离分离泵连接管23出口端的方向逐个递减的设计,使得过滤半透膜8既可以稳定血浆流速,又能防止堵塞;同时压力传感器12能够检测是否有过滤半透膜8发生堵塞,当发生堵塞时,通过备用的连接管接通两个备用出口11,防止过滤半透膜8发生堵塞时造成循环中断,使血浆通过备用的连接管进入下一个没有堵塞的过滤半透膜8之前,使治疗继续进行。
Claims (10)
1.一种半透膜分层一体式生物反应器,其特征在于:包括腔体(21),设置在腔体(21)一侧的血浆分离器(1)及设置在腔体(21)另一侧的生物反应器(2);血浆分离器(1)和生物反应器(2)通过分隔半透膜(3)间隔开;
位于血浆分离器(1)所在一侧的腔体(21)上开设有血液入口(4)和血细胞出口(5),血浆分离器(1)内部填充有多根中空纤维丝(6);所述中空纤维丝(6)一端与血液入口(4)连通,另一端与血细胞出口(5)连通;
位于生物反应器(2)所在另一侧的腔体(21)的顶部开设有血浆出口(7),生物反应器(2)内水平设置有多层过滤半透膜(8),所述过滤半透膜(8)用于阻隔游离细胞穿过且相邻的两个过滤半透膜(8)与生物反应器(2)的侧壁一起形成一个独立的滤层空间(10),每个独立的滤层空间(10)对应的腔体(21)上设有肝细胞灌注口(9);最上层的过滤半透膜(8)与生物反应器(2)顶部保持分离;最下层的过滤半透膜(8)与生物反应器(2)底部形成夹层(25),所述夹层(25)内填充有透气纤维丝(13)且所述夹层(25)对应的腔体(21)上设有进气口(24),所述进气口(24)仅与透气纤维丝(13)连通;
所述血浆分离器(1)和生物反应器(2)通过设有分离泵(22)的分离泵连接管(23)将两者连通;所述分离泵连接管(23)的入口端位于所述血浆分离器(1)所在一侧的腔体(21)的侧壁下端,出口端位于所述生物反应器(2)所在一侧的腔体(21)的底部。
2.根据权利要求1所述的半透膜分层一体式生物反应器,其特征在于:所述过滤半透膜(8)及分隔半透膜(3)均为平片膜元件,所述平片膜元件的周边与腔体(21)贴合固定。
3.根据权利要求1或2所述的半透膜分层一体式生物反应器,其特征在于:所述过滤半透膜(8)的孔径按照远离所述分离泵连接管(23)出口端的方向逐个递减。
4.根据权利要求1或2所述的半透膜分层一体式生物反应器,其特征在于:所述血浆出口(7)内设有阻止细胞透过的阻隔半透膜(16)。
5.根据权利要求2所述的半透膜分层一体式生物反应器,其特征在于:所述夹层(25)在所述进气口(24)与透气纤维丝(13)之间还设有用于阻隔血浆/预充液的密封支架板(26);所述密封支架板(26)与所述进气口(24)间距设置;所述密封支架板(26)上开设有多个小孔;所述透气纤维丝(13)一端卡设在小孔内,且通过密封支架板(26)与进气口(24)之间的空间与所述进气口(24)相连通。
6.根据权利要求1所述的半透膜分层一体式生物反应器,其特征在于:所述滤层空间(10)对应的腔体(21)上还设有备用出口(11)及压力传感器(12)。
7.根据权利要求1所述的半透膜分层一体式生物反应器,其特征在于:所述血液入口(4)处设有与其连通的高氧水入口(14);所述分离泵连接管(23)上还设有压力检测装置(20)。
8.根据权利要求1所述的半透膜分层一体式生物反应器,其特征在于:所述夹层(25)对应的腔体(21)上设有出气口/预留口(18)。
9.根据权利要求1所述的半透膜分层一体式生物反应器,其特征在于:所述一体式生物反应器还包括设于腔体(21)外的37℃保温加热套及摇床。
10.根据权利要求1所述的半透膜分层一体式生物反应器,其特征在于:所述分隔半透膜(3)的孔径为0.3~5微米,最下层的过滤半透膜(8)的孔径为5微米。
