Abstract
本发明涉及一种用于延缓衰老的枸杞发酵液及其护肤品,属于生物技术领域。本发明选用枸杞为主要原料,同时添加蜂蜜、番茄、葡萄、柚子、火龙果、花椰菜、大蒜、百合、甘草、白芷、黄芪等辅料进行发酵,得到枸杞发酵液(枸杞酵素),该发酵液具有一定的美白及抗氧化活性,给人体补充多种酶、维生素、氨基酸和矿物质,具有延缓衰老的功效。将发酵液与化妆品其他原料进行配方,得到具有延缓衰老功效的酵素护肤品。

技术领域
本发明涉及生物工程技术领域,特别是指一种用于延缓衰老的枸杞发酵液及其护肤品。
背景技术
从生物学上讲,衰老是生物随着时间的推移,自发的必然过程,它是复杂的自然现象,表现为结构的退行性变和机能的衰退、适应性和抵抗力减退。
衰老的自由基学说是DenhamHarman在1956年提出的,认为衰老过程中的退行性变化是由于细胞正常代谢过程中产生的自由基的有害作用造成的。生物体的衰老过程是机体的组织细胞不断产生自由基积累的结果,自由基可以引起DNA损伤从而导致突变,诱发肿瘤形成。自由基是正常代谢的中间产物,其反应能力很强,可使细胞中的多种物质发生氧化,损害生物膜,还能够使蛋白质、核酸等大分子交联,影响其正常功能。
随着年龄的增长、外界环境的影响(如辐射、风吹日晒)以及自身压力、生活习惯等作用,人类肌肤逐渐老化,具体表现在:皮肤松弛、弹性减弱、皱纹、色泽暗淡、毛孔粗大、毛细血管孔张、各种色素斑点等。
化妆品对抗衰老的原理:一是根据自由基衰老学说清除过量的自由基,拥有清除自由基功能的活性原料以维生素C、维生素E、辅酶Q为代表。二是预防紫外线,日光中的紫外线可引起皮肤红斑和延迟性黑色素沉着,破坏皮肤的保湿能力,使皮肤变得粗糙,皱纹增多。三是抑制体内酪氨酸酶的活性,减少黑色素产生,同时,化妆品还有促进皮肤细胞新陈代谢,补充胶原蛋白和弹性蛋白,保湿和修复皮肤屏障功能等作用。
酵素一类发酵液的总称,是以新鲜水果、蔬菜等为原料,经过天然微生物发酵(或接种食品可用益生菌进行发酵)得到的功能性微生物发酵产品,其中包括原料本身的营养物质和微生物次级代谢产物以及微生物菌体本身等多种成分,含有丰富的酶、氨基酸、维生素、有机酸、矿物质和微量元素等,具有抑菌消炎、排毒养颜、净化血液、促进细胞新生等功效。
酵素对抗皮肤老化的原理是:酵素可清除体内废物,平衡肠道细菌;为机体补充脂肪酶、蛋白酶、超氧化物歧化酶,具有一定的抗氧化活性,有些酵素具有一定的酪氨酸酶抑制活性,可减少体内黑色素的产生;为机体补充维生素、矿物质、氨基酸,强化细胞活性,促进新陈代谢。
发明内容
本发明的主要目的即在于开发一种用于延缓衰老的枸杞发酵液及其护肤品。
本发明采用如下的技术方案:
一种用于延缓衰老的枸杞发酵液,主要由如下重量份数的原料制成:枸杞50~55份、蜂蜜20~25份、番茄4~6份、葡萄3~5份、柚子2~4份、火龙果4~6份、花椰菜2~4份、大蒜3~5份、百合1~2份、甘草2~3份、白芷1~2份、黄芪1~2份;
制备方法如下:
步骤一,将枸杞、番茄、葡萄、柚子、火龙果、花椰菜和大蒜分别洗净、打浆,其中火龙果、大蒜去皮,葡萄籽完好,各果蔬浆混匀得到混合果蔬浆;百合、甘草、白芷和黄芪粉碎后过40目筛,备用;
步骤二,在混合果蔬浆中加入0.05~0.1wt%的纤维素酶和0.05~0.1wt%的果胶酶进行酶解,接着再加入0.02~0.05wt%的蛋白酶和0.1wt%的α-淀粉酶进行酶解;
步骤三,将酶解液置于密闭发酵罐中,加入蜂蜜,再加入酵母菌和乳酸菌,混合均匀,一定温度下进行发酵,每天用无菌木棍搅拌三次以减少酒精产生,将上浮的原料搅拌到发酵液中,发酵时间为50d;
步骤四,在发酵液中加入百合、甘草、白芷和黄芪,一定温度条件下发酵30d,每天用无菌木棍搅拌三次以减少酒精产生,将上浮的原料搅拌到发酵液中;
步骤五,往发酵液中接种醋酸菌,密闭条件下发酵30d,再沉降过滤除渣;
步骤六,将发酵液进行膜过滤除菌、75℃杀菌20分钟,再进行无菌灌装或者喷雾干燥成酵素粉。
所述步骤二纤维素酶和果胶酶的水解pH范围为3.0~4.5,温度范围为40℃~50℃,酶解5~10小时;蛋白酶和α-淀粉酶的水解pH范围为6.0~7.5,温度范围为35℃~45℃,进行酶解3~7小时。
所述步骤三所用的酵母菌为高糖型面包酵母和产朊假丝酵母,乳酸菌为嗜酸乳杆菌和保加利亚乳杆菌,接种方式为按100g/t的接种量接种活性菌数为1×109cfu/g的活性菌粉。
所述步骤五所用的醋酸菌为巴氏醋酸杆菌,接种方式为按100g/t的接种量接种活性菌数为1×109cfu/g的活性菌粉。
所述步骤三、四和五的发酵温度为28~32℃;所述步骤六膜过滤除菌的过滤膜的孔径为0.1~0.2μm。
所述步骤六中,先加入占发酵液0.8wt%的低聚木糖,混匀后进行膜过滤除菌、75℃维持20分钟杀菌及无菌灌装。
上述枸杞发酵液在制备具有延缓衰老作用的产品中的应用。

一种枸杞酵素润肤霜,其重量配方如下:去离子水28.5~31.5%、上述枸杞发酵液3~6%、液体石蜡25%、石蜡1%、橄榄油30%、凡士林2%、羊毛脂2%、倍半油酸失水山梨酯3%、甘油2.5%。
一种枸杞酵素面膜,其重量配方如下:聚乙烯醇10%、Carbopol9410.5%、丙二醇5%、乙醇20%、三异丙醇胺0.6%、水解蛋白5%、上述枸杞发酵液3~6%、对羟基苯甲酸甲酯0.1%、玫瑰精油适量、去离子水52.8~55.8%。
本发明中:
枸杞为被子植物茄科枸杞属,人们日常食用和药用的枸杞子多为宁夏枸杞的果实“枸杞子”,宁夏枸杞也是唯一载入《2010年版中国药典》的品种,与琼珍灵芝、长白山人参、东阿阿胶并称为中药四宝。枸杞多糖是一种水溶性多糖,是枸杞中最主要的活性成分,成为国内外研究热点。其中又以枸杞多糖的免疫调节和抗肿瘤作用的研究最多,现已有很多研究表明枸杞多糖具有促进免疫、抗衰老、抗肿瘤、清除自由基、抗疲劳、抗辐射、保肝、生殖功能保护和改善等作用。枸杞色素是存在于枸杞浆果中的各类呈色物质,是枸杞籽的重要生理活性成分,主要包括胡萝卜素、叶黄素和其他有色物质。枸杞所含有的类胡萝卜素则具有非常重要的药用价值。很多研究已经证明枸杞籽色素具有提高人体免疫功能、预防和抑制肿瘤及预防动脉粥样硬化等作用。胡萝卜素是枸杞色素的主要活性成分,具有抗氧化和作为维生素A的合成前体等重要的生理功能。
番茄营养丰富,有“维生素仓库”之称。它含有蛋白质、脂肪、糖类、多种矿物质、微量元素。番茄中含有丰富的抗氧化剂,而抗氧化剂可以防止自由基对皮肤的破坏,具有明显的美容抗皱的效果,其中维生素C和维生素P的含量非常高。番茄红素不仅仅是当今工业上重要的天然食品着色剂,更为重要的是它是很强的抗氧化剂。补充番茄红素,可以抵抗衰老、增强免疫系统、减少疾病的发生。番茄红素还能降低眼睛黄斑的退化、减少色斑沉着。故番茄拥有“长寿果”之美誉。
葡萄籽95%的成份为原花青素,火龙果内也含有丰富的花青素,因此,葡萄和火龙果能抗衰老,并可清除体内自由基,让人永葆青春,另外葡萄中的类黄酮也是强力抗氧化剂;甘草、白芷、百合内含有美白成分,具有一定的美白功效。
本发明将枸杞与其他果蔬和草本药材进行混合发酵,旨在提供一种具有延缓衰老的枸杞发酵液,并将发酵液作为一种天然健康无刺激护肤品原料。
本发明中,之所以选用枸杞、蜂蜜、番茄、葡萄、柚子、火龙果、花椰菜、大蒜、百合、甘草、白芷、黄芪配伍来制备枸杞发酵液,是因为本申请的发明人经过无数次的配伍实验,选用了数十种原料、各种不同的配方、配比与枸杞配伍,都无法达成本发明的发明目的,抗氧化和美白效果均不佳,直到采用了本发明的配方及配比,才发现其制得的枸杞发酵液的抗氧化和美白效果出乎意料,十分优良,说明本发明选用的枸杞、蜂蜜、番茄、葡萄、柚子、火龙果、花椰菜、大蒜、百合、甘草、白芷、黄芪,在发酵后产生了协同增效作用,从而带来了意向不到的效果,从而达成了本发明的发明目的。
本发明的一种具有延缓衰老作用的枸杞发酵液,具有一定的美白及抗氧化活性,给人体补充多种酶、维生素、氨基酸和矿物质,加入低聚木糖,在人体内可以促进双歧杆菌增殖同时产生多种有机酸,降低肠道pH值,抑制有害菌生长,使益生菌在肠道内大量增殖,由此发挥治疗便秘和慢性腹泻,保护肝脏,防治高血压和动脉硬化以及抗衰老、降低血清胆固醇、预防癌症和抑制肿瘤生长的作用。本发明的枸杞发酵液可以直接饮用或者加工成其他产品,如护肤品等。

具体实施方式
实施例一
本发明的一种具有延缓衰老功效的枸杞发酵液,其详细的制备方法包括如下步骤:
(1)选用新鲜且成熟的水果和蔬菜,剔除病虫害果和霉烂蔬果,洗净并控干表面水分,其中火龙果、大蒜去皮,称重。各蔬菜水果重量如下:枸杞5000g、番茄500g、葡萄400g、柚子300g、火龙果500g、花椰菜300g、大蒜400g,各种果蔬分别用打浆机打碎,葡萄籽完好,各果蔬浆混匀得到混合果蔬浆;百合100g、甘草300g、白芷100g和黄芪100g粉碎后过40目筛,备用;
步骤二,在混合果蔬浆中加入8g的纤维素酶和8g的果胶酶进行酶解,pH范围为3.0~4.5,温度范围为40~50℃,酶解5~10小时;接着再加入4g的蛋白酶和8g的α-淀粉酶进行酶解,pH范围为6.0~7.5,温度范围为35~45℃,进行酶解3~7小时;
步骤三,将酶解液置于密闭发酵罐中,加入2000g的蜂蜜,再接种活性菌数为1×109cfu/g的高糖型面包酵母、产朊假丝酵母、嗜酸乳杆菌和保加利亚乳杆菌活性菌粉,每种菌种接种1g,混合均匀,30℃下进行发酵,每天用无菌木棍搅拌三次以减少酒精产生,将上浮的原料搅拌到发酵液中,发酵时间为50d;
步骤四,在发酵液中加入百合、甘草、白芷和黄芪,搅拌均匀,30℃条件下发酵30d,每天用无菌木棍搅拌三次以减少酒精产生,将上浮的原料搅拌到发酵液中;
步骤五,往发酵液中接种1g活性菌数为1×109cfu/g的巴氏醋酸杆菌粉,密闭条件下发酵30d,再沉降过滤除渣;
步骤六,将发酵液进行膜过滤除菌、75℃杀菌20分钟,再进行无菌灌装或者喷雾干燥成酵素粉。
采用DPPH法测定实施例一的酵素液的抗氧化活性,DPPH法实验操作如下:
用无水乙醇配制0.2mmol/L的DPPH溶液,稳定半小时备用。加样方法为:A组加100μL酵素液和2mL无水乙醇;B组加100μL酵素液和2mLDPPH溶液;C组加100μL水和2mLDPPH溶液,各组混匀后常温避光放置半小时,于517nm测定吸光度A、B、C。DPPH清除率=[1-(B-A)/C]*100%。
经实验测定,实施例一的酵素液对DPPH自由基清除率为72.6%。
采用斑马鱼胚胎测定酵素液的美白效果,实验方法为:选用健康的斑马鱼受精卵,每10枚为一组放于六孔板中,每孔加2mL受精卵培养液,受精9小时后加药,实验组加1.97mL培养液和0.03mL酵素液,对照组加2mL培养液。37℃恒温培养48小时后,剥开受精卵膜,在显微镜下观察斑马鱼黑色素形成情况,并拍照,用Photoshop和ImageJ软件计算斑马鱼侧面积和黑色素面积,从而计算酵素液对斑马鱼黑色素形成的抑制率。
图1为实施例一的对照组对斑马鱼黑色素形成的抑制效果,图2显示了实施例一的酵素液对斑马鱼黑色素形成的抑制效果,经过软件计算,其抑制率为41.2%。
实施例二
本发明的一种具有延缓衰老功效的枸杞发酵液,其详细的制备方法包括如下步骤:
(1)选用新鲜且成熟的水果和蔬菜,剔除病虫害果和霉烂蔬果,洗净并控干表面水分,其中火龙果、大蒜去皮,称重。各蔬菜水果重量如下:枸杞5000g、番茄500g、葡萄400g、柚子300g、火龙果500g、花椰菜300g、大蒜400g,各种果蔬分别用打浆机打碎,葡萄籽完好,各果蔬浆混匀得到混合果蔬浆;百合100g、甘草300g、白芷100g和黄芪100g粉碎后过40目筛,备用;
步骤二,在混合果蔬浆中加入8g的纤维素酶和8g的果胶酶进行酶解,pH范围为3.0~4.5,温度范围为40~50℃,酶解5~10小时;接着再加入4g的蛋白酶和8g的α-淀粉酶进行酶解,pH范围为6.0~7.5,温度范围为35~45℃,进行酶解3~7小时;
步骤三,将酶解液置于密闭发酵罐中,加入2000g的蜂蜜,再接种活性菌数为1×109cfu/g的高糖型面包酵母、产朊假丝酵母、嗜酸乳杆菌和保加利亚乳杆菌活性菌粉,每种菌种接种1g,混合均匀,30℃下进行发酵,每天用无菌木棍搅拌三次以减少酒精产生,将上浮的原料搅拌到发酵液中,发酵时间为50d;
步骤四,在发酵液中加入百合、甘草、白芷和黄芪,搅拌均匀,30℃条件下发酵30d,每天用无菌木棍搅拌三次以减少酒精产生,将上浮的原料搅拌到发酵液中;
步骤五,往发酵液中接种1g活性菌数为1×109cfu/g的巴氏醋酸杆菌粉,密闭条件下发酵30d,再沉降过滤除渣得到7900g枸杞发酵液;
步骤六,在发酵液中加入63.2g低聚木糖,搅拌溶解后再将发酵液进行膜过滤除菌、75℃杀菌20分钟,最后进行无菌灌装或者喷雾干燥成酵素粉。
应用例一
将实施例一的枸杞发酵液用于制备具有延缓衰老功效的枸杞酵素爽肤水,其配方如下:

应用例三
将实施例一的枸杞发酵液用于制备具有延缓衰老功效的枸杞酵素面膜,其配方如下:
利用本发明所制备的枸杞发酵液,含有丰富的胡萝卜素及多种氨基酸、维生素、矿物质等营养成分,能及时清除体内多余的自由基,延缓衰老。添加低聚木糖,可以在人体内促进双歧杆菌增殖同时产生多种有机酸,抑制有害菌生长,使益生菌在肠道大量增殖,促进人体及时排除毒素,达到延缓衰老的效果。
上述仅为本发明的具体实施方式,但本发明的设计构思并不局限于此,凡利用此构思对本发明进行非实质性的改动,均应属于侵犯本发明保护范围的行为。
Claims (10)
1.一种用于延缓衰老的枸杞发酵液,其特征在于主要由如下重量份数的原料制成:枸杞50~55份、蜂蜜20~25份、番茄4~6份、葡萄3~5份、柚子2~4份、火龙果4~6份、花椰菜2~4份、大蒜3~5份、百合1~2份、甘草2~3份、白芷1~2份、黄芪1~2份;
制备方法如下:
步骤一,将枸杞、番茄、葡萄、柚子、火龙果、花椰菜和大蒜分别洗净、打浆,其中火龙果、大蒜去皮,葡萄籽完好,各果蔬浆混匀得到混合果蔬浆;百合、甘草、白芷和黄芪粉碎后过40目筛,备用;
步骤二,在混合果蔬浆中加入0.05~0.1wt%的纤维素酶和0.05~0.1wt%的果胶酶进行酶解,接着再加入0.02~0.05wt%的蛋白酶和0.1wt%的α-淀粉酶进行酶解;
步骤三,将酶解液置于密闭发酵罐中,加入蜂蜜,再加入酵母菌和乳酸菌,混合均匀,一定温度下进行发酵,每天用无菌木棍搅拌三次以减少酒精产生,将上浮的原料搅拌到发酵液中,发酵时间为50d;
步骤四,在发酵液中加入百合、甘草、白芷和黄芪,一定温度条件下发酵30d,每天用无菌木棍搅拌三次以减少酒精产生,将上浮的原料搅拌到发酵液中;
步骤五,往发酵液中接种醋酸菌,密闭条件下发酵30d,再沉降过滤除渣;
步骤六,将发酵液进行膜过滤除菌、75℃杀菌20分钟,再进行无菌灌装或者喷雾干燥成酵素粉。
2.如权利要求1所述的一种用于延缓衰老的枸杞发酵液,其特征在于:所述步骤二纤维素酶和果胶酶的水解pH范围为3.0~4.5,温度范围为40℃~50℃,酶解5~10小时;蛋白酶和α-淀粉酶的水解pH范围为6.0~7.5,温度范围为35℃~45℃,进行酶解3~7小时。
3.如权利要求1所述的一种用于延缓衰老的枸杞发酵液,其特征在于:所述步骤三所用的酵母菌为高糖型面包酵母和产朊假丝酵母,乳酸菌为嗜酸乳杆菌和保加利亚乳杆菌,接种方式为按100g/t的接种量接种活性菌数为1×109cfu/g的活性菌粉。
4.如权利要求1所述的一种用于延缓衰老的枸杞发酵液,其特征在于:所述步骤五所用的醋酸菌为巴氏醋酸杆菌,接种方式为按100g/t的接种量接种活性菌数为1×109cfu/g的活性菌粉。
5.如权利要求1所述的一种用于延缓衰老的枸杞发酵液,其特征在于:所述步骤三、四和五的发酵温度为28~32℃;所述步骤六膜过滤除菌的过滤膜的孔径为0.1~0.2μm。
6.如权利要求1所述的一种用于延缓衰老的枸杞发酵液,其特征在于:所述步骤六中,先加入占发酵液0.8wt%的低聚木糖,混匀后进行膜过滤除菌、75℃维持20分钟杀菌及无菌灌装。
7.权利要求1-6中任一枸杞发酵液在制备具有延缓衰老作用的产品中的应用。
8.一种枸杞酵素爽肤水,其特征在于其重量配方如下:去离子水70~73%、EDTA-Na20.1%、尿囊素0.2%、丙二醇8%、甘油12%、氨基酸保湿剂3.4%、甲基异噻唑啉酮0.3%、权利要求1-6中任一枸杞发酵液3~6%。
9.一种枸杞酵素润肤霜,其特征在于其重量配方如下:去离子水28.5~31.5%、权利要求1-6中任一枸杞发酵液3~6%、液体石蜡25%、石蜡1%、橄榄油30%、凡士林2%、羊毛脂2%、倍半油酸失水山梨酯3%、甘油2.5%。
10.一种枸杞酵素面膜,其特征在于重量配方如下:聚乙烯醇10%、Carbopol9410.5%、丙二醇5%、乙醇20%、三异丙醇胺0.6%、水解蛋白5%、权利要求1-6中任一枸杞发酵液3~6%、对羟基苯甲酸甲酯0.1%、玫瑰精油适量、去离子水52.8~55.8%。
