Abstract
本发明涉及一种用于防治鲢鱼出血病的饲料添加剂,主要由以下重量份的原料制备而成:鱼粉50-100份、溶菌酶1-10份、鸡腿菇多糖1-10份。与普通饲料添加剂相比,本发明的水产生物饲料添加剂制备方法简单,配伍合理,疗效显著,能够显著降低鱼类的死亡率,实验证明对鲢鱼出血病有特效;由于添加剂中无西药成分,因此饲料添加剂无毒副作用,无耐药性,无污染。

背景技术
鱼产品具有较高的营养价值,对改善膳食结构,提高人体健康水平非常重要。随着健康意识日益增强,消费者对鱼产品的消费需求量和品质要求越来越高。近几年,随着饲料养鱼的迅猛发展,鲢鱼因其生长速度快,已成为国内主养鱼类,尤其是山塘型水库和小型水库,主养、密养鲢鱼已经相当普遍。然而随着养殖密度不断提高,养殖环境污染等原因,各种鱼类疾病发病率也急剧升高,由于鲢鱼出血病的频频发生以及难以治疗而大大降低了其成活率,严重影响了养殖专业户的经济效益。
鲢鱼出血病主要由鲢鱼感染出血病病毒所致,其因来势凶猛、死亡率高,病情易反复且难以控制,引起了广大渔业养殖户的高度重视。每年春季3-5月份及秋季8-10月份为流行季节,以秋季8-9月份最流行;发病特点为急性、暴发性、大批量地死鱼,如不及时采取治疗措施,发病3~5天内,死亡率80%-90%,甚至可达100%。当水质恶劣变化,pH值低于6.5时极易引起暴发流行,且在发生过该病的水域,第二年养殖鲢鱼也易复发流行。发病初期,鲢鱼常浮于水面下10厘米缓慢游动,对人影反应灵敏,病情严重时则反应迟钝或无反应,表现木讷,昼夜浮头,白天太阳照射也不下沉。刚死或即将死亡的鲢鱼,鳃盖外表粗糙无粘膜,鳃盖内膜有血丝,颌下部有条状或散点状红斑,嘴唇外突呈紫色,臀鳍基部发红,尾鳍有白边无粘液,严重的有蛀鳍。剪去鳃盖骨,可见鳃片红肿,粘液增多,鳃丝乌紫分瓣呈木梳齿状分离,表面有一层蓝白色光膜。网状鳃耙布满红色血点,严重的呈灰白色并腐蚀成缺刻。解剖病鱼腹部,可见胆囊、肝、肾颜色变淡,肿大易碎,肠道内无食物,但充满粘液,肛门膨大红肿。
目前生产上主要用抗生素和消毒剂来防治出血病等鱼类疾病,但长期使用会导致十分严重的后果:(1)滥用抗生素造成病菌抗药性增强,使得鱼病治疗越发困难;(2)消毒剂毒副作用大,局部刺激强,容易致癌,造成鱼类应激严重;(3)破坏养殖水体,污染水环境,降低鱼体免疫力;(4)药物残留增加,水产品安全堪忧,质量下降,危害人类健康。
为解决上述问题,科研人员不断探索水产饲料添加剂新品种以代替抗生素和消毒剂。一些学者开始从免疫学角度对水产动物病害防治进行研究,认为在饲料中添加免疫增强剂是增强鱼类免疫功能,抵抗疾病的一种有效实用的方法。但是,动物的免疫系统是相对稳定的,在正常情况下免疫系统会将机体的免疫功能整合在一个适宜的水平。免疫增强剂只可能较大幅度地提高免疫功能低下的水产动物的免疫水平,而对于免疫功能正常的水产动物会因机体自身免疫系统的调节阻止其免疫水平过高而影响免疫增强剂的真实效果,况且免疫水平也并非越高越好,免疫水平过高会导致免疫亢进,引起各种类型的超敏反应、炎症和自身免疫性疾病。在众多的免疫增强剂中,溶菌酶和多糖类成分因其低药残、生物毒性小的优点而引起了学者们重视。
溶菌酶(lysozyme)又称胞壁质酶(muramidase)或N-乙酰基胞壁质聚糖水解酶(N-acetylmuramideglycanohydrlase),是一种能破坏致病菌细胞壁中黏多糖的水溶性蛋白酶。它是由Fleming在1922年在人唾液和眼泪中发现的存在溶解细菌细胞壁的酶,故命名为溶菌酶。此后,在鸟类、家禽的蛋清和哺乳动物泪液、唾液、血浆、乳汁、胎盘以及体液、组织细胞内等发现了溶菌酶,其中以蛋清中含量最丰富(约0.3%);在一些植物体如卷心菜、萝卜、无花果和微生物体内也存在溶菌酶。研究发现枯草芽孢杆菌(Bacillussubtilis)中的溶解子就是溶菌酶的前身。
溶菌酶本身是一种碱性蛋白质,在酸性环境下,溶菌酶的热稳定性很强:pH值3.0时96℃加热处理15min仍能保持87%的酶活性。在自然条件下,大多数溶菌酶化学性质稳定,即使当pH值在1.2-11.3范围内剧烈变化,溶菌酶的空间结构仍可以保持不变,酶活性的变化也不显著。但碱性环境很容易使溶菌酶的空间结构破坏,此时对热的稳定性较差,当处于碱性pH值范围时,温度会使酶活力损失非常快。
溶菌酶能有效水解细菌细胞壁肽聚糖,其水解位点是N-乙酰氨基葡萄糖胺(NAM)的l位碳原子和N-乙酰胞壁酸(NAG)的4位碳原子间的β-l,4糖苷键,可使细菌细胞壁变得松弛,失去对细胞的保护作用,细菌内容物溢出而使细胞壁溶解,最后细胞溶解死亡。溶菌酶之所以会对革兰阳性细菌有很强杀伤力,就是因为革兰阳性菌的细胞壁主要是由胞质壁和磷酸质组成的,其主要成分胞质壁又是杂多糖与多肽组成的糖蛋白,而杂多糖正是由NAM和NAG以β-1,4糖苷键联结的,溶菌酶催化水解该糖苷键使细菌细胞壁破裂而死亡。而革兰阴性菌内壁层为肽聚糖,因此溶菌酶只能破坏革兰阳性菌细胞壁,而对革兰阴性菌作用不大。
溶菌酶在中性体液环境下带有大量正电荷,可与带有负电荷的病毒蛋白直接作用,与DNA、RNA、脱附基蛋白相结合形成复盐,使病毒失活。在被疱疹病毒感染的Hela细胞培养液中加溶菌酶后有抑制细胞变性作用。溶菌酶也可抑制腺病毒生长,可用于带状疱疹、腮腺炎、鸡水痘、肝炎及流感等病毒性疾患治疗。
溶菌酶可保持机体生理平衡,参与体内多种免疫反应,是机体内重要的非特异性免疫因子之一。溶菌酶本身具有T细胞表位,能诱导2型(Th2)T辅助细胞反应。口服溶菌酶能诱导小鼠产生全身性Th1和Th2免疫反应,增强免疫功能,因此,口服溶菌酶具有控制上呼吸道感染和治疗痢疾的功能。此外,溶菌酶还具有激活血小板的功能,可改善组织局部血液循环障碍,分解脓液,增强局部防卫功能,从而体现其止血、消肿及加快机体损伤组织的修复作用。它还可作为一种宿主抵抗因子,对组织局部起保护作用。
溶菌酶还可与各种诱发炎症的酸性物质结合,减少细菌内毒素释放,从而减轻内毒素血症发生,减缓炎症发生过程,并对抗生素有一定增效作用,改善组织基质的黏多糖代谢,起到抗菌消炎、修复组织的作用。经试验证明,用1种纤维素共聚物将人溶菌酶和蛋白水解酶偶联,可显著提高溶菌酶抗菌能力,加速伤口愈合。
近年来,随着研究的深入,人们不仅对溶菌酶的作用机制有更进一步了解,且根据其溶菌特性,已将其广泛应用于医疗、食品防腐、生物工程及动物生产中。世界卫生组织及许多国家都公认溶菌酶的无毒性,并核准其可作为治疗性物质在一些食品中使用。作为一种无毒、无害的高盐基天然蛋白质酶,溶菌酶对动物细胞无任何影响,是安全的营养性饲料添加剂,有非专用性生长激素作用,可改善消化道微生物群落,促进营养消化吸收。
作为鱼类防护病原体入侵的一种重要非特异性免疫因子,溶菌酶能水解致病菌中黏多糖的碱性酶,在抵抗外界细菌入侵时起到重要作用,它广泛存在于动物的血清、皮肤粘液和组织器官中,在其他生物中也有存在。鱼类溶菌酶的研究虽然起步较晚,但因其特殊的机理及其在免疫防御中的重要作用,近年来引起了广泛的关注。科研人员已经在鱼类溶菌酶基因特征、表达活性、分子结构及其作用机理、转基因鱼类的培育、重组溶菌酶的开发等方面进行了深入研究,并应用于食品保鲜剂、饲料添加剂、消炎镇痛药物等领域的生产。
多糖及其缀合物是普遍存在于植物细胞壁及细胞中的一种成分,由于组成结构不同,其理化特点及生物活性也各不相同。有些多糖(如淀粉、果胶、纤维素等)早已成为人类日常生活中的重要组成部分,是一种重要的经济原料,对动物不具有特异性生物活性。从20世纪50年代末对真菌多糖的抗癌效果的发现使人们开始了多糖系列的化学研究。日本从20世纪60年代开始研究担子菌多糖的药物活性,并开发了一些有名的药物。中国对多糖的研究始于20世纪70年代,近年来对植物多糖的研究发展很快,在免疫机理和临床应用方面的研究已由一般的观察发展到分子、受体水平,由动物实验发展到临床应用和保健食品的制作,由菌类多糖到高等植物多科属、多组织、多器官的研究。研究发现不同种植物所含多糖类化合物是一种免疫调节剂,它具有抑制肿瘤生长、激活免疫细胞、改善肌体免疫功能等作用,对正常细胞无毒副作用。到目前为止,从植物中分离出来的多糖类化合物中水溶性多糖最为重要。
多糖对机体免疫系统有重要的增强作用,具有多靶点和普遍存在于特异性及非特异性免疫激活过程的各个环节的特点。大量的药理和临床实验证实,多糖不仅可以激活T淋巴细胞、B淋巴细胞、巨噬细胞、自然杀伤(NK)细胞等多种免疫细胞,而且可以活化补体系统,有效促进IL-1,IL-2,IL-6,TNF等细胞因子的分泌和表达。对多糖的免疫活性、作用机制及构效关系的研究既可以寻找活性多糖类药物及辅助药物,又可以对已有多糖的结构进行优化,从而进一步提高活性。多糖由于其独特的性质逐渐被人们所认识,一些多糖药物已经在临床上作为治疗癌症的辅助药物使用。
多糖类物质具有提高动物机体的免疫力、阻止病毒复制、杀灭细菌、排出毒素等功效,并且具有用量少、作用快、持效久等特点。研究发现在饲料中添加黄芪多糖可使鲤鱼白细胞的吞噬率显著升高,对白细胞活性的促进作用也较为明显。
鸡腿菇(Coprinuscomatus)学名毛头鬼伞,隶属于真菌门层菌纲伞菌目鬼伞科鬼伞属,是药食两用菌。鸡腿菇的营养价值很高,具高蛋白、低脂肪、氨基酸种类齐全的特点,有益脾、清心安神、助食、治痔等功效,被世界卫生组织及联合国粮农组织确定为集“天然、营养、保健”三种功能于一体的16种珍稀食用菌之一,并被推荐进行大力推广。
现代研究表明,鸡腿菇的营养价值很高,具有高蛋白、低脂肪、氨基酸种类齐全的特点。另外,鸡腿菇还有丰富的糖类,据测定干品鸡腿菇中总糖含量为57.65%,还原糖含量为53.54%,多糖含量为4.1l%。鸡腿菇头具有良好的食用价值和药用价值,愈来愈受到国内外研究者的高度重视。目前,鸡腿菇多糖已被证实具有抑菌,提高免疫力,抗肿瘤,降血糖等生物活性。
氨基酸是蛋白质的基本单位,存在于蛋白质中的二十多种氨基酸为脂肪族、芳香族和杂环三大类。根据所含氨基、羧基的多寡及是否含硫或羟基,脂肪族氨基酸又可分为中性、酸性、碱性、含羟基及含硫氨基酸筹。还可从营养角度分类,把饲料蛋白质中所含的氨基酸分为必需氨基酸和非必需氨基酸。所谓必需氨基酸即在动物体内不能合成或合成速度及数量不能满足正常生长需要,必须从饲料来供给的。非必需氮基酸即在动物体内合成较多或需要较少,无须从饲料中供给亦能保持动物正常生长的需要。
蛋氨酸主要有DL-蛋氨酸和蛋氨酸的羟基类似物(MHB)及其钙盐。MHB的营养效价相当于DL-蛋氨酸的80%左右。蛋氨酸在植物性蛋白质中含量很少,禽类要使羽毛丰满需要摄取足够的蛋氨酸(含硫氨基酸)。日本在鱼粉不足的鸡饲料中添加蛋氨酸的用量为0.1%~0.2%。欧美国家因一般不使用鱼粉,因而在玉米加豆饼或小麦加豆饼的饲粮中添加蛋氨酸的用量为日本用量的3倍。美国常以MHB为蛋氨酸添加剂。在饲粮中添加蛋氨酸能把蛋白质强化到鱼粉的水平,每添加1千克蛋氨酸,可代替50千克鱼粉。如以玉米和饼类饲料为主的日粮中添加蛋氨酸的参考比例为,肉鸡和蛋鸡0.1%~0.2%,育肥猪为0.02%~0.05%,仔猪为0.05%~0.10%。
赖氨酸在植物性蛋白中的含量不高,豆饼里含少量,玉米含量很少。因此在缺少鱼粉和豆饼不足的饲粮中需添加赖氨酸。赖氨酸是动物第1限制性氨基酸。根据试验,在动物饲粮中添加赖氨酸,可提高饲粮中的蛋白质水平2%~3%。肉鸡饲料中每添加1千克赖氨酸,可以节约25千克饲料,在猪的饲粮中使用赖氨酸能提高瘦肉率。在猪禽饲粮中添加赖氨酸的比例,视饲粮中赖氨酸的水平而定,一般为0.1%~0.2%,缺少鱼粉和豆饼不足的饲料,可添加到0.2%,一般仔猪和育肥猪为0.1%。
随着饲料工业迅速发展在饲料中广泛应用维生素添加剂。维生素添加剂有天然的与合成的,作为饲料添加剂的维生素主要采用合成维生素。维生素添加剂种类很多,常用的维生素添加剂有14种,按其溶解性可分为脂溶性维生素和水溶性维生素制剂两大类。常用的脂溶性维生素包括维生素A、维生素D3、维生素E和维生素K四种,水溶性维生素包括维生素B族、维生素C、烟酸、泛酸等。维生素虽然需求量较微量,但每一种维生素都起着其他物质不能替代的特殊营养生理作用。动物若缺乏或过多维生素将对生长发育产生明显的不良后果。因此,必须在日常饲料中添加所需量的维生素,以供动物机体需要。
维生素A又名视黄醛、抗干眼病维生素,是含有β-白芷酮环的不饱和一元醇,由异成二烯构件分子生物合成。纯维生素A为淡黄色晶体,缺氧时对热稳定,有氧时对热不稳定,且易被紫外线破坏。维生素A仅存在于动物体内,植物中只有维生素A原——胡萝卜素和类胡萝卜素。维生素A包括A1与A2两种,也存在于哺乳及咸水鱼的肝脏中,A2存在于淡水鱼的肝脏中,二者可直接被动物机体消化吸收。α、β、γ-胡萝卜素和玉米黄素在动物肠壁细胞内及肝脏、乳腺内经胡萝卜素酶作用可转化为维生素A。维生素A能剌激实验动物的生长,也可剌激许多组织中DNA的合成;维生素A能调节动物机体的免疫功能;能恢复某些类型细胞的生长接触性抑制,并提高细胞之间的黏附;维生素A的衍生物在特异的糖蛋白合成中还可作为糖的携带者。
维生素C又称抗坏血酸,在自然界中以两种形式存在,即还原形式L-抗坏血酸和氧化形式L-脱氧抗坏血酸,大部分维生素C以还原形式存在。在制作饲料过程中,维生素C的实际添加量应大于理论需求量。目前,维生素C在饲料中的实际添加量主要依据有两点:一是相关动物的营养需求量;二是使用的品种的稳定性。维生素C在饲料中的推荐用量一般为需求量的4-5倍。在日益发展的水产饲料、哺乳动物的开食补料、畜禽抗应激日粮及速长型肉禽骨骼矿质化特殊日粮中补充稳定性较高的维生素C或维生素C衍生物,已成为饲用维生素C需求增长的重要因素。
维生素E又名生育酚、抗不育维生素,是一组具有生物活性的化学结构相似的酚类化合物,主要存在于植物油中。天然的维生素E有8种,即:α、β、γ、δ-生育酚和α、β、γ、δ-生育三烯酚,均系苯共二氢吡喃的衍生物。维生素E极易被氧化而保护其他物质不被氧化,是动物体内最有效的抗氧化剂。它能对抗生物膜磷脂中不饱和脂肪酸的过氧化反应,避免脂质中过氧化物的产生,保护生物膜的结构和功能;维生素E还可与硒协同通过谷胱甘肽过氧化酶发挥抗氧化作用,防止肝组织坏死和肌肉受损,维持红细胞的稳定性和毛细血管的完整性;α-生育酚能通过影响膜磷脂的结构而影响生物膜的形成;维生素E与动物的繁殖机能密切相关,具有促进性腺发育,促成受孕和防止流产等作用;维生素E可以促进血红素的合成;高剂量的维生素E还能促进免疫球蛋白的生成,提高动物机体对疾病的抵抗力,增强抗应激作用。
维生素K是一类含有甲萘醌结构的抗出血性化合物的总称。维生素K可分为两大类化合物:K1、K2和K3,可在动植物体内提取;另一类为水溶性化合物K4,为人工化学合成。维生素K是动物体内形成凝血酶原所必需的维生素,能促进肝脏合成凝血酶原所必需的维生素,能促进肝脏合成凝血酶原(凝血因子Ⅱ);调节另外几种凝血因子Ⅶ、Ⅸ及Ⅹ的合成;除凝血作用外,维生素K还可能依赖蛋白质和肽参与钙代谢。
目前,将溶菌酶与鸡腿菇多糖组合用于防治鲢鱼出血病的应用尚未见报道。
发明内容
本发明所要解决的技术问题是提供一种能有效提高鲢鱼抗出血病能力,且无毒副作用,无污染,无耐药性的鱼类饲料添加剂及含有该添加剂的饲料。
此外,本发明还提供一种上述添加剂的应用方法。
本发明解决上述问题所采用的技术方案是,提供一种防治鲢鱼出血病的饲料添加剂,主要由以下重量份的组分制成:鱼粉50-100份、溶菌酶1-10份、鸡腿菇多糖1-10份。
优选的,所述饲料添加剂中,各组分的重量份为:鱼粉50份,溶菌酶1-4份,鸡腿菇多糖1-4份。
更优选的,所述饲料添加剂中,各组分的重量份为:鱼粉50份,溶菌酶1份,鸡腿菇多糖3份。
优选的,所述原料进一步包括其他活性成分。
更优选的,所述其他活性成分选自氨基酸、维生素等中的一种或多种。
最优选的,所述氨基酸为蛋氨酸和/或赖氨酸等,所述维生素为维生素A、维生素C、维生素E和/或维生素K等。
本发明还提供一种上述饲料添加剂在制备防治鲢鱼出血病的饲料中的应用。
优选的,所述100重量份的饲料中,含有1-5重量%的饲料添加剂。
本发明具有积极有益的效果:
令人惊奇的是,经过反复多次试验,本发明意外发现将溶菌酶与多糖类物质鸡腿菇多糖组合使用对于鲢鱼出血病的防治具有协同增效的效果,配合其他活性成分使用效果更佳。
与普通饲料添加剂相比,本发明的水产生物饲料添加剂制备方法简单,配伍合理,疗效显著,能够显著降低鱼类的死亡率,实验证明对鲢鱼出血病有特效;由于添加剂中无西药成分,因此饲料添加剂无毒副作用,无耐药性,无污染。
具体实施方式
下面结合实施例对本发明作更进一步的说明,但本发明的实施方式不限于此。下述实施例中所使用的实验方法如无特殊说明,均为常规方法。
实施例1、鸡腿菇多糖的提取
取鸡腿菇干品加30倍重量的水混匀,在90℃浸提4小时,取上清液真空浓缩,用4倍浓缩液体积的95%乙醇沉淀,沉淀物再溶于适量水中,用Sevag法脱蛋白,适量95%乙醇沉淀,真空干燥得鸡腿菇多糖。
实施例2、饲料添加剂的制备
取鱼粉50重量份,溶菌酶5重量份,混合后制成饲料添加剂A1。
取鱼粉50重量份,实施例1制备得到的鸡腿菇多糖5重量份,混合后制成饲料添加剂A2。
取鱼粉50重量份,溶菌酶4重量份,实施例1制备得到的鸡腿菇多糖1重量份,混合后制成溶菌酶和鸡腿菇多糖重量比为4:1的饲料添加剂A3。
取鱼粉50重量份,溶菌酶2.5重量份,实施例1制备得到的鸡腿菇多糖2.5重量份,混合后制成溶菌酶和鸡腿菇多糖重量比为1:1的饲料添加剂A4。
取鱼粉50重量份,溶菌酶1.25重量份,实施例1制备得到的鸡腿菇多糖3.75重量份,混合后制成溶菌酶和鸡腿菇多糖重量比为1:3的饲料添加剂A5。
取鱼粉50重量份,溶菌酶1重量份,实施例1制备得到的鸡腿菇多糖4重量份,混合后制成溶菌酶和鸡腿菇多糖重量比为1:4的饲料添加剂A6。
上述实施例中使用的溶菌酶是采用凝胶法,经冷冻干燥制成直径0.5mm的颗粒,即为溶菌酶包被品。
实施例3、包含溶菌酶和鸡腿菇多糖的饲料添加剂在抗鲢鱼出血病中的应用
为验证本发明的应用效果,本发明的饲料添加剂在7个渔场的鲢鱼中进行试验。将鲢鱼分为实验组(A1-A6)和对照组,每组各为200尾。其中,实验组的日粮按照饲料总重量的1%添加上述饲料添加剂,对照组的日粮为不添加上述饲料添加剂的相同重量的鱼饲料,各组鱼每日投喂5次,饲喂60天后进行感染实验,实验组和对照组均注射可导致出血病的致病菌,连续观察7天,统计实验组和对照组中鲢鱼的死亡数量,并计算死亡率(见表1)。

由实施例3的对比实验可以看出,本发明的饲料添加剂能有效改善鲢鱼的免疫功能,增强抗病菌感染能力,降低鲢鱼的死亡率,特别是溶菌酶和鸡腿菇多糖的组合使用产生了协同效果,减少了药物的使用量。在活性成分总量保持不变的情况下,本发明的饲料添加剂较单独使用溶菌酶或鸡腿菇多糖的饲料添加剂抗鲢鱼出血病的效果显著提高,鱼类死亡率至少降低了29%,其中溶菌酶与鸡腿菇多糖重量比为1:3的饲料添加剂效果最为突出,将鱼类死亡率至少降低了40%,产生了难以预期的优异效果。
Claims (8)
1.一种用于防治鲢鱼出血病的饲料添加剂,其特征在于,主要由以下重量份的原料制备而成:鱼粉50-100份、溶菌酶1-10份、鸡腿菇多糖1-10份。
2.根据权利要求1所述的饲料添加剂,其特征在于,所述原料重量份为鱼粉50-75份,溶菌酶1-4份,鸡腿菇多糖1-4份。
3.根据权利要求2所述的饲料添加剂,其特征在于,所述原料重量份为鱼粉50份,溶菌酶1份,鸡腿菇多糖3份。
4.根据权利要求1所述的饲料添加剂,其特征在于,所述原料进一步包括其他活性成分。
5.根据权利要求4所述的饲料添加剂,其特征在于,所述其他活性成分选自氨基酸、维生素等中的一种或多种。
6.根据权利要求5所述的饲料添加剂,其特征在于,所述氨基酸为蛋氨酸和/或赖氨酸等,所述维生素为维生素A、维生素C、维生素E和/或维生素K等。
7.权利要求1-6任一项所述的饲料添加剂在制备防治鲢鱼出血病的饲料中的应用。
8.根据权利要求7所述的应用,其特征在于,所述100重量份的饲料中,含有1-5重量%的饲料添加剂。
