Abstract

Description
本发明涉及包含维生素和角黄素的饲料补充剂,以及这种补充剂用于改善家禽(具体地家禽小母鸡)性能和健康的用途。
已经观察到,当只喂养标准饲料时,受到严重应力(例如接种时的应力)的动物或者当要求高的性能/表现(performance)时的动物会疲劳、腹泻、拒绝摄入饲料、贫血等等。在这些情况下,显然需要向饲料中加入添加剂或补充剂。然而,通常很难界定所观察到的问题是由什么原因引起的,因而很难界定使用哪种添加剂。尽管已知的添加剂和饲料补充剂种类繁多,但是没有一种能够解决所有上述问题。一些添加剂主要用于提高动物的生长,而其他的则声称改善其健康。维生素缺乏可能是问题的一部分,但人们应当意识到即使当预期饲料包含足够量的维生素时为什么这仍然发生。
在比赛家禽(racingpoultry)、具体地小母鸡(pullet)上已经观察到一个特殊的问题。超过一岁的雌性动物被称为母鸡,更年轻的雌性动物被称为小母鸡。当小母鸡开始在15到20周的年龄段开始下蛋时,小母鸡开始成为母鸡。
小母鸡饲养在鸟禽的生活中是一个极其重要的阶段,因为它的体重发育和疾病状态将极大地影响鸟禽群体的产蛋性能。通常在其生命的第一个星期中通过各种途径给小母鸡施用大约15种疫苗。在整个这期间改善鸟禽的免疫应答将不仅增强鸟禽个体的抗体水平,而且有助于抵抗接种感染以及抵抗其他自然感染的能力。这可能导致在产蛋的时间点时更好的鸡群均匀度和更具免疫能力的鸡群。
众所周知,接种引起鸡的性能降低。具体来说,接种引起饲料摄入和饲料效率降低。因此,接种对鸡生产商来说具有巨大的经济影响。
因此,本发明的目标是得到新的饲料补充剂,其会改善小母鸡(尤其是在应力条件下和当要求高的性能时)的健康和性能。
现已惊奇地发现,下文中所限定的饲料添加剂组合物改善了小母鸡的
-饲料转化和生长
-免疫状态
-骨和骨骼发育

本发明的范围和特征如所附权利要求所限定。
根据本发明的饲料添加剂组合物包含角黄素、选自由维生素C和维生素E组成的组的至少一种维生素、微量矿物(具体地硒)和任选地至少两种化合物的混合物,所述化合物选自由麝香草酚、丁香油酚、香兰素和γ-萜品烯组成的组。
本发明的发明人已经能够证明,协同并组合使用这些活性成分的混合物以完全预料不到的方式展现出了本发明所寻求的改善可消化性、生长和骨发育以及提升小母鸡的免疫系统的效果。
在一个具体的实例中已经发现,通过给动物施用有效量的饲料补充剂组合物,能够改善家禽小母鸡中饲料摄入、重量增加和饲料转化率,该饲料补充剂组合物由角黄素、维生素C和维生素E、硒和精油混合物组成,该精油混合物包含麝香草酚和丁香油酚作为主要成分。
在整个说明书和权利要求书中适用如下定义。
术语“饲料转化率”是基于生长试验确定的,该生长试验包括第一处理和第二处理(对照),在第一处理中以每kg饲料中合适的浓度向动物饲料添加根据本发明的组合物,在第二处理中不向动物饲料添加该组合物。
众所周知,改善的FCR比对照FCR更低。在具体的实施方式中,与对照相比,FCR被改善(即减少)至少1.0%或5%。
角黄素和维生素E和C是可商业购买的,或者能够容易地由本领域技术人员使用现有技术中公知的工艺和方法制备得到。例如,维生素E可在商标E50下购得,维生素C可在商标C下购得,角黄素可在商标Red下购得(所有化合物由DSMNutritionalProducts,Kaiseraugst,Switzerland提供)。
硒可以从任何来源获得,其组合物可以使用常规的技术硒(例如,以无机形式Na2SeO3或有机络合物)来制备得到。可商购的产品是Se或2000。
根据本发明的精油混合物是可商业购买的(例如在商标下,由DSMNutritionalProducts,Kaiseraugst,Switzerland供应),或者可以容易地由本领域技术人员使用现有技术中公知的工艺和方法制备得到。精油混合物可以以高度纯化的形式使用或者以天然获得的植物提取物或提取混合物的形式使用。

这个方面也包括用饲料补充剂组合物饲养动物的方法,该饲料补充剂组合物包含如下物质作为主要成分:角黄素,选自由维生素C和维生素E组成的组的至少一种维生素,微量矿物、具体地硒,以及任选地精油混合物,所述精油混合物包含选自由麝香草酚、丁香油酚、香兰素和γ-萜品烯组成的组的至少两种化合物。
角黄素、选自由维生素C和维生素E组成的组的至少一种维生素、微量矿物硒和精油混合物适于与饲料一起施用。术语“饲料”或“饲料组合物”指的是适合用于或意图用于动物摄入的任何化合物、配制物、混合物或组合物。本文中使用时,术语“饲料”包括固体和液体饲料两种以及饮用流体例如水。

本发明的第三个方面涉及包含根据本发明的饲料补充剂组合物的预混物或常规动物饲料。
在制造按照本发明的家禽饲料时,向常规家禽饲料添加
-约2至100ppm、优选地2至10ppm的角黄素,
-约20至200ppm、优选地30至150ppm的维生素E,
-约20至200ppm、优选地50至150ppm的维生素C,
-约0.01至0.5ppm、优选地0.05至0.2ppm的硒,
-约20至250ppm、优选地30至150ppm的精油混合物。
在家禽喂养概念的一个优选实施方式中,根据本发明的活性成分被以如下的量用于最终的常规家禽饲料中:
-2、4或6ppm角黄素,
-35、70或105ppm维生素E,
-50、100或150ppm维生素C,
-0.05、0.1或0.15ppm硒,
-34、67或101ppm的精油混合物。
尤其优选的精油混合物的实例包含:
-2%至10%、优选地5%的麝香草酚,
-10%至20%、优选地17%的丁香油酚,
-0.5%至5%、优选地1%的γ-萜品烯,
-5%至10%、优选地7%的香兰素,
-55%至82.5%的填料、载剂和乳化表面活性剂。
精油混合物任选地包含其他化合物,例如选自如下的组的至少一种额外化合物(以每kg饲料中用量计算),所述组为:
至多约1mg的亚丙基苯酞、亚丁基苯酞、苯酞、姜辣素、薰衣草油;
至多约2mg的癸内酯、十一内酯、十二内酯、紫罗兰酮、鸢尾酮、间苯二酚、桉树脑、薄荷醇、薄荷油、α-蒎烯;
至多约3mg的苎烯、愈创木酚、茴香脑、里哪醇、二氢茉莉酮酸甲酯;
至多约4mg的香芹酚、丙酸、乙酸或丁酸、迷迭香油、丁香油、香叶醇、萜品醇、香茅醇;
至多约5mg的水杨酸戊酯和/或苄酯、肉桂醛、植物多酚(鞣质);
和至多约5mg的姜黄根粉末或姜黄提取物
乳化剂可以有利地选自具有相当亲水性质的那些,例如脂肪酸的聚丙三醇酯之一,诸如经酯化的蓖麻油酸,或脂肪酸的丙二醇酯、糖-酯(saccharo-esters)或糖-甘油酯、聚乙二醇、卵磷脂等。
在实践中使用浓缩物或预混物,将本文中所举例的饲料补充剂组合物掺入家禽饲料中。预混物是指一种或多种微量成分与稀释剂和/或载剂的优选均匀的混合物。在较大的混合中,预混物便于微量成分均匀地分散。预混物可以以固体形式(例如可水溶粉末形式)或液体形式被添加到饲料成分中或被添加到饮用水中。
而且,可选的饲料添加剂成分为芳香化合物;稳定剂;抗菌多肽;多不饱和脂肪酸(PUFAs);产反应性氧试剂;和/或选自如下的至少一种酶:植酸酶(EC3.1.3.8或3.1.3.26)、木聚糖酶(EC3.2.1.8)、半乳聚糖酶(EC3.2.1.89)、α-半乳糖苷酶(EC3.2.1.22)、蛋白酶(EC3.4.)、磷脂酶Al(EC3.1.1.32)、磷脂酶A2(EC3.1.1.4)、溶血磷脂酶(EC3.1.1.5)、磷脂酶C(EC3.1.4.3)、磷脂酶D(EC3.1.4.4)、淀粉酶(例如α-淀粉酶(EC3.2.1.1))、和/或β-葡聚糖酶(EC3.2.1.4或EC3.2.1.6)。
抗菌多肽(AMP)的实例为CAP18、明串珠菌素A(LeucocinA)、Protegrin-1、Thanatin、防御素(Defensin)、乳铁蛋白(Lactoferrin)、乳铁蛋白肽(Lactoferricin)和Ovispirin,诸如Novispirin(RobertLehrer2000)、Plectasins和他汀(Statins)。
多不饱和脂肪酸的实例是C18、C20和C22多不饱和脂肪酸,诸如花生四烯酸、二十二碳六烯酸、二十碳五烯酸和γ-亚油酸。
产反应性氧试剂的实例是诸如过硼酸盐、过硫酸盐或过碳酸盐的化学品;和诸如氧化酶、加氧酶或合成酶(syntethase)的酶。
通常,脂溶性维生素和水溶性维生素以及微量矿物形成所谓的预混物,从而添加到饲料中,而常量矿物通常单独添加到饲料中。
例如每吨家禽饲料中,预混物可以包含50至200g的活性化合物混合物的丙二醇溶液,20至1000g乳化剂,50至900g谷类和副产物,20-100g蛋白质支持物(奶粉、干酪素等)和50至300g矿物成分(膨胀硅土、饲料级石灰、磷酸氢钙(bi-calciumphosphate)等)。
本文所述发明和所要求保护的发明的范围不应受限于本文所公开的具体实施方式,因为这些实施方式意欲举例说明本发明的各个方面。任何等同的实施方式应包括在本发明的范围内。事实上,除了本文所示和所述的那些实施方式以外,由上述说明书对本发明进行各种修正对于本领域技术人员来说是明显的。上述修正也落在所附权利要求的范围内。

实施例
1.实验设计
整体设计
实验涉及4组260只小母鸡。每组被分成10份26只鸟禽的重复组。在饲料期间将动物放置在笼中。
计划在日龄(day-old)和3、8、12、17周龄时进行体重测量。在相同的年龄还计算饲料摄取。因此,我们能够确定每个时期的饲料转化率。
在6、12、15和17周龄时评价免疫状态。
通过在3个不同的年龄(4、8和17周龄)时协调的胫骨强度来评估骨和骨骼发育。
实验组
对比4种处理方式:
-A批次:对照,无补充剂,
-B批次:饲料中具有饲料补充剂组合物(FSC)(剂量为2kg/t),
-C批次:具有饲料补充剂组合物(FSC)(剂量为4kg/t),
-D批次:具有饲料补充剂组合物(FSC)(剂量为6kg/t)。
2.试验期间鸟禽的管理

疫苗接种程序附于实验报告(附录1)。
饲养
饲养程序被分为3个时间段:
-从日龄到3周的年龄:育雏料(PLD),
-从4周到10周的年龄:生长料(PLC),
-从11周到17周的年龄:小母鸡料(PLE)。
在饲养期间所使用的不同饲料的完整描述附于实验报告(附录2)。下表中给出了饲料组合物的详细内容。
生产的FSC以小麦麸(wheatmiddling)为载体。
3.实验程序和特征测量
测试日历
1天和123天之间进行数据收集。下面列出主要的事件:
骨和骨骼发育
通过评价胫骨强度来表征骨和骨骼发育。
Synergie200MTS压缩机允许测量胫骨的硬度、最大应力和断裂力。每次处理在4、8和17周的3个不同的年龄分析20根左胫骨。
4.实验产品的效果评估准则
-体重:在4个不同年龄(3、8、12和17周龄)时对体重(平均值和均匀性)进行比较。
-饲料消耗:在4个不同年龄(3、8、12和17周龄)时对饲料摄入进行比较。
-饲料转化率是饲料摄入和体重之间的比例。也可以在4个不同年龄(3、8、12和17周龄)时在该特征方面对各处理进行比较。
-免疫状态:在4个不同年龄(6、12、15和17周龄)时对免疫状态进行比较。
-骨和骨骼发育:在3个不同年龄(4、8和17周龄)时对胫骨强度(硬度和断裂力)进行比较。
5.统计分析
实验单位/统计单位
实验单位为每个重复组26个小母鸡。
统计单位为鸡禽或笼。
初始组的可比性
日龄(Day-old)的鸡随机分布在不同的重复组中。
此外,日龄重量测量给我们提供在体重的平均值和均匀性方面初始组的可比性信息。

α风险(Alpharisk)将为5%。

6.结果
用根据本发明的饲料补充剂组合物饲养动物在如下方面表现出显著的效果:
-平均体重:实验处理>对照处理,
-饲料转化率:对照处理>实验处理,
-胫骨强度:实验处理>对照处理。
提高剂量没有改善这些效果。因此,剂量B看起来在改善体重、饲料转化和胫骨强度方面是令人满意的。
补充剂也在新城疫(NewcastleDisease)方面对免疫状态具有积极的影响,对C和D处理的鸟禽提供更好且更均匀的保护。
死亡率和体重测量
在1、22、56、85和123天的年龄时根据处理的体重

一般来说,死亡率在每个处理中相当低。在不同的组中,其在1.2%和2.3%之间变化。在17周龄时各处理之间没有明显的统计学上的显著差异(χ2:p=0.677)。
在第1天,不同处理的平均体重是相类似的(大约34g)。在第22天,B组表现出比其他3组更低的体重。随后,在其他3个年龄(56、85和123天)时,3个实验批次(B、C和D)的平均体重比对照组(A)高。在17周龄(半生长周(halfaweekofgrowth))时,差异大约为40g。
在4组中,鸡群均匀度是相类似的。因此,用来比较方差均匀度的Bartlett测试是不显著的。在123天年龄时,在不同的组中,体重CV在7.1和7.7%之间变化。

在40天年龄(第一次ND接种疫苗之前)时,未检测到抗体。
在滴度的对数上进行统计。在任何年龄时特征的正态性都不被接受,因而未呈现疫苗分析的结果。
在123天年龄时,Bartlett测试接近于显著。因此,A和B处理的滴度看上去表现出在该年龄时更高的CV。
为了比较在123天年龄的处理,我们选择建立2个滴度类别:
第0类:滴度值<4096,
第1类:滴度值≥4096。
我们使用χ2测试来比较每种类别中鸟禽的比例。此处,各组之间的差异接近于显著(p=0.069)。采用较大的样品大小,我们将很可能得到显著差异(p<0.05)。因此,A和B处理将表现出更高比例的具有较低抗体滴度值的鸟禽。
传染性法氏囊病

在123天的年龄时胫骨最大应力和硬度差异接近于显著(分别为p=0.080和p=0.117),而在更小的年龄时(27和56天的年龄)时不接近于显著。因此,与实验批次相比,对照组的特征在于更弱的胫骨。采用较大的样品大小时,我们可能会得到显著的差异(p<0.05)。3个实验处理具有较高的值的部分原因是:这些组的体重和胫骨重量更高。因此,在数学模型中引入这些特征,我们显著地影响p值:

Claims (9)
1.在家禽小母鸡饲料中角黄素与选自由维生素C和维生素E组成的组的至少一种维生素、硒和任选地至少两种如下化合物的混合物的组合用于改善家禽中饲料摄入、重量增加、饲料转化率和/或骨发育和/或用于提升该动物的免疫系统的用途,所述化合物选自由麝香草酚、丁香油酚、香兰素和γ-萜品烯组成的组。
2.根据权利要求1所述的用途,其中角黄素与维生素C、维生素E、硒以及精油混合物组合,所述精油混合物包含麝香草酚和丁香油酚作为主要成分。
3.用于家禽的饲料补充剂组合物,其包含:角黄素,选自由维生素C和维生素E组成的组的至少一种维生素,微量矿物、具体地硒,以及任选地精油混合物,所述精油混合物包含至少两种选自由麝香草酚、丁香油酚、香兰素和γ-萜品烯组成的组的化合物。
4.根据权利要求3的饲料补充剂组合物,其包含角黄素、维生素C、维生素E、硒以及精油混合物组合,所述精油混合物包含麝香草酚和丁香油酚作为主要成分。
5.预混物或常规动物饲料,其包含根据权利要求3或4所述的饲料补充剂组合物。
6.动物饲料,其包含
-约2至100ppm、优选地2至10ppm的角黄素,
-约20至200ppm、优选地30至150ppm的维生素E,
-约20至200ppm、优选地50至150ppm的维生素C,
-约0.01至0.5ppm、优选地0.05至0.2ppm的硒,
-约20至250ppm、优选地30至150ppm的精油混合物。
7.根据权利要求6所述的动物饲料,其包含
-2、4或6ppm角黄素,
-35、70或105ppm维生素E,
-50、100或150ppm维生素C,
-0.05、0.1或0.15ppm硒,
-34、67或101ppm的精油混合物。
8.根据权利要求6或7所述的动物饲料,其中精油混合物包含
-2%至10%、优选地5%的麝香草酚,
-10%至20%、优选地17%的丁香油酚,
-0.5%至5%、优选地1%的γ-萜品烯,
-5%至10%、优选地7%的香兰素,
-55%至82.5%的填料、载剂和乳化表面活性剂。
9.用饲料补充剂组合物饲养动物以改善家禽中饲料摄入、重量增加、饲料转化率和/或骨发育和/或以提升该动物的免疫系统的方法,其中所述饲料补充剂组合物包含如下物质作为主要成分:角黄素,选自由维生素C和维生素E组成的组的至少一种维生素,微量矿物、具体地硒,以及任选地精油混合物,所述精油混合物包含选自由麝香草酚、丁香油酚、香兰素和γ-萜品烯组成的组的至少两种化合物。
