背景技术
近年来人们发现肽类具有独特的生理功能,如促进钙的吸收、降血压、增强免疫力、抗肿瘤等;且其在人体内的消化吸收明显优于单个氨基酸,营养学实验也证明:肽对人体内蛋白质的合成无任何不良影响。现代科学的许多实验也证明,蛋白质水解成多肽后,具有更强的生物活性。肽还具有良好的理化性质:如水合性提高(蛋白质水解物离子基团增加)、溶解度增加(蛋白质水解物体积变小,离子性增强)、粘度降低(蛋白多聚体的解聚和离子基团的增加,使蛋白质的有序性增加,蛋白分子的表现体积减少)、胶凝性降低(蛋白质水解物疏水性下降)、发泡性降低(粘度降低)等。基于以上优点,相信肽在功能性食品领域将会有广阔的应用前景。可用于食品的功能性肽获得的途径有三种,一是从天然生物体中直接提取天然活性肽;二是在消化过程中产生或体外水解蛋白质产生;三是通过化学方法、酶法,重组DNA等技术合成。从天然生物体中提取是可取的,但成本较高;而合成法虽可按人们的意愿合成任意活性肽,但目前因为成本高,副反应多及残留化合物等问题制约着发展。目前常用的方法是体外水解法,而酶水解能在温和的条件下进行,且可通过选择酶的种类,控制反应时间来得到特定的活性肽。
现有技术中对以大豆、乳制品为原料制备低分子肽的研究较多,市场上的产品也较丰富,例如:三九系列的减肥、降糖肽,核桃肽,特别是核桃肽。桃渣中蛋白质含量高达43.8%。核桃蛋白是一种很好的植物蛋白,它由清蛋白、球蛋白、醇溶谷蛋白和谷蛋白组成,分别占核桃蛋白总量6.8%、17.6%、5.3%和70.1%。所述核桃蛋白肽是以核桃蛋白粉为原料制成,其制法包括:酶解、分子蒸馏工序。所述的酶解工序采用风味蛋白酶作为酶制剂,工艺条件为:底物浓度为10%,水解时间为2h,温度为55°C,pH值为6.5,酶用量为1.5%。
雄蜂蛹的蛋白质含量较高,我国是世界养蜂大国,现拥有700万群(箱)蜜蜂,雄蜂蛹资源丰富,随着养蜂业的发展,雄蜂蛹的来源也将更加广泛。因此,通过酶法将雄蜂蛹水解成多肽,开发成相应的保健食品,不仅能促进我国蜂蛹深加工技术的发展,而且能促进养蜂业的发展和农民的增收。雄蜂蛹蛋白质是一种完全蛋白,富含18种氨基酸,其中8种人体所必需氨基酸占总量的17%以上。雄蜂蛹中谷氨酸和天门冬氨酸的含量达到总氨基酸的20%以上,这不但使蜂蛹美味可口,而且提示具有良好的健脑功能,因为这两种氨基酸能通过大脑屏障,起到健脑的作用。所以,雄蜂蛹有望作为营养丰富的高蛋白食品进行深入的开发。将雄蜂蛹蛋白进一步水解成多肽和氨基酸后,将更容易为人体所吸收,在食品、保健品行业具有广阔的应用前景。蛋白质水解时可以采用酸水解、碱水解和酶水解的方式,但是酸水解导致色氨酸全部破坏,丝氨酸、酪氨酸和苏氨酸部分破坏;碱水解引起消旋作用而降低营养价值;酶水解时反应条件温和,营养价值得以保障;因此,现有技术中多采用酶解法提取雄蜂蛹。
中国专利申请CN89101221.4公开了一种利用酶解反应提取雄蜂营养成分的工艺,并指出所提取的雄蜂营养成分的主要用途。所述的雄蜂营养成分的提取工艺是将雄蜂、雄蜂幼虫、雄蜂蛹等采用蛋白酶经酶解反应提取其各种营养成分。由于所提取的雄蜂营养成分中氨基酸、微量元素、维生素等主要指标均超过蜂王浆、胜过花粉,可用所提取的雄蜂营养成分做保健药品和营养食品的原料,做成各种滋补、保健药品和高级营养食品。
发明内容
本发明提供了一种雄蜂蛹多肽提取物,该雄蜂蛹多肽提取物具有改善记忆和抗氧化功能,可作为具有改善记忆和/或抗氧化功能的产品或者用于制备具有改善记忆和/或抗氧化功能的产品。
本发明另外提供了一种雄蜂蛹多肽的提取方法,提供包括步骤:
将雄蜂蛹用水洗净匀浆后溶于丙酮中脱脂,离心所得残渣用水配成悬浊液,将悬浊液在90°C加热10分钟,冷却后调pH值至8,加木瓜蛋白酶和胰蛋白酶水解,水解时间为2h,温度为55°C,pH值为6.5,酶用量为1%,离心或过滤所得上清液为多肽溶液,将所述的多肽溶液用葡聚糖G-50(SephadexG-50)(1.5cm×90cm)分离,20mmol/LHCl溶液洗脱,流速1.3mL/分钟,收集洗脱产物,调节溶液至pH6.0~7.0,10000转/分钟离心15分钟,上清液冷冻干燥备用;经十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE),回收小分子量的条带,经过验证,获得了24个具有生物学活性的小肽。通过测序,所述小肽的序列为:

根据现有技术的提示,利用本发明的组合物可以制成用于脑记忆增强的保健品,其组分为核桃油35克~55克、白茯苓6克~8克、蜂蜜6克~9克、远志15-20克、大枣40-90克、提取的多肽100mg。
本发明具有如下有益效果:
本发明的产品具有高营养易吸收、促进新陈代谢、调节脑细胞生长,促进脑记忆等多种生理功能。同时具有很好的滋补作用,特别是用于血虚体弱等症状,具有明显改善效果,常服食之,对改善身体健康情况大有裨益。
具体实施方式
实施例1:多肽的制备
将雄蜂蛹用水洗净匀浆后溶于丙酮中脱脂,离心所得残渣用水配成悬浊液,将悬浊液在90°C加热10分钟,冷却后调pH值至8,加木瓜蛋白酶和胰蛋白酶水解,水解时间为2h,温度为55°C,pH值为6.5,酶用量为1%,离心或过滤所得上清液为多肽溶液,将所述的多肽溶液用葡聚糖G-50(SephadexG-50)(1.5cm×90cm)分离,20mmol/LHCl溶液洗脱,流速1.3mL/分钟,收集洗脱产物,调节溶液至pH6.0~7.0,10000转/分钟离心15分钟,上清液冷冻干燥备用;经十二烷基硫酸钠-聚丙烯酰胺凝胶电泳(SDS-PAGE),回收小分子量的条带,共得到89个小肽的序列。根据色谱柱中不同的峰值分离时间,可以批量获得相应的小肽。
实施例2多肽保健食品的制备
取白茯苓7克、远志18克、大枣80克、蜂蜜8克、核桃油40g混合加水煎煮2次,每次煎煮2小时,合并煎液,滤过、去杂质,滤液浓缩至80°C下相对密度为1.15的清膏,加入多肽(89种分离多肽的一种,共89种形式)100mg、搅拌、混匀,冷冻干燥,制成饼状食品。
实施例3多肽保健食品的效果验证
六周龄健康小鼠(体重22g-25g,雌雄各半),将试验小鼠随机分成3组,即空白组,口服生理盐水的对照组、活性肽处理组。其中,活性多肽组小鼠的口服食品以多肽的含量为计量单位(20mg/kg),I次/d,连续6周,对照组为等量的生理盐水。
各组小鼠处理6周后,进行水迷宫试验,迷宫是80cmX50cmX20cm双层不透明有机玻璃箱,内设起始点、终点平台和四个盲端,平台附近为安全区。试验时迷宫水深12cm,水温保持(25±1)°C,迷宫周围一切空间标志在整个试验过程中保持不变。正式试验前一天,帮助每只小鼠在迷宫中熟悉水环境15min,如小鼠在60s内未到达平台,可帮助式地将小鼠赶至平台,并使小鼠在平台上停留20s。第2天开始正式试验。小鼠面向壁在起始点放入水中,记录小鼠放入水中到达终点平台时间(游全程时间)和进入盲端的次数(错误次数),统计结果见表I,没有明显效果的多肽在此不一一列举。

根据试验结果可以看出,多肽组与空白组或对照组相比,水迷宫试验游全程时间明显减少(P<0.01),错误次数显著减少(P<0.05),这说明本发明制备的多肽食品能够提高衰老小鼠的学习和记忆能力。具有较好的应用前景。
另外本发明的多肽食品具有较好的刺激小鼠红细胞生长的功效,与未使用本发明的多肽保健品相比,小鼠红细胞数目提高大约25%。
通过12个月的连续服用,本发明的多肽具有较好的安全性,小鼠使用无任何异常情况发生。
