本发明涉及饲料添加剂,更具体地,涉及一种预防大口黑鲈脂肪肝饲料添加剂。
背景技术
大口黑鲈,隶属鲈形目,鲈亚目、太阳鱼科、黑鲈属,原产于北美洲,是一种世界性的游钓鱼类,在湖泊和河流中均有分布。因其肉质鲜美、营养丰富、生长快等一系列优点,自1973年被引入广东省并于1985年人工繁殖技术取得成功以来,其养殖范围及面积得以迅猛扩展,目前已成为我国最重要的淡水经济养殖鱼类之一。大口黑鲈在传统的养殖过程中主要依赖冰鲜杂鱼,然而,伴随大口黑鲈养殖集约化程度的进一步提高,冰鲜杂鱼的养殖劣势日渐凸现:其获得量及质量均不稳定,饲料系数高,也容易污染水质,导致水体富营养话,且投喂操作繁杂,人力投入成本高。因此,大口黑鲈人工配合饲料的研发和推广成为近年来行业内从业人员关注的热点问题。
目前已有不少企业已陆续推出商品化的大口黑鲈人工配合饲料并投放市场使用,然而在养殖应用过程中发现用配合饲料饲喂的大口黑鲈在生长中期会出现厌食、生长缓慢等一系列负面现象。
而养殖过程中的负面现象已成为当前制约大口黑鲈人工配合饲料应用推广的瓶颈问题。
发明内容
经过进一步解剖鱼体可发现:此阶段鱼肝脏大多肿大呈不健康褐色或花白色且充满脂肪,即出现“脂肪肝”症状。众所周知,肝脏是动物体及其重要的解毒器官,同时还是动物体的脂肪代谢中心,脂类的消化、吸收、运输、分解机合成代谢都和肝脏有密切联系,因此,肝脏的损伤会直接导致动物体的机能受损,影响其健康生长。
因此为了解决这个问题,本发明提供一种预防大口黑鲈脂肪肝病的饲料添加剂,按重量份计,包括,黄芪15~22份;丹参13~20份;陈皮4~8份;佛手4~8份;香橼4~8份;山楂7~12份;炙甘草3~6份;红花5~8份;金钱草4~6份;大黄4~6份;茵陈4~6份;郁金2~4份;维生素C 4~8份;维生素E 3~5份;蛋氨酸3~6份和胆碱1~3份。
进一步提供上述的饲料添加剂在制备预防大口黑鲈脂肪肝病药物中的应用。

本发明的优点在于:
1、本发明优选具疏肝解郁、强肝散结、可修复受损肝细胞,恢复肝功能功效的中草药作为主要原料,能够有效防治大口黑鲈养殖过程中的脂肪肝现象,可极大地降低甚至避免传统抗生素类药物的使用,消除了药物残留所致的食品安全隐患,提高综合经济效益。
2、本发明不仅可有效防治大口黑鲈脂肪肝,还可显著性降低养殖过程中大口黑鲈死亡率,降低饵料系数,提高饲料利用率,可满足大口黑鲈集约化养殖所需。
3、本发明还补充添加了经前期大量研究证实的对大口黑鲈脂肪肝病有显著防治功效的几种维生素和蛋氨酸,其适量添加对本发明所优选的几种中草药有较好的协同促进功效,使得本发明优选的各组分对大口黑鲈脂肪肝的防治功效得以最大化地发挥。
4、本发明作为添加剂使用,在饲料中按照0.15%~0.25%的比例添加即可达到理想效果,且本发明无任何毒副作用、安全、高效、环保。

效果验证试验(试验在海鸥岛实验基地进行)
1、水族箱试验
1.1、材料和方法
1.1.1、试验设计
以目前市场上的普通大口黑鲈配合饲料作为基础配方,设置4个处理组:基础配方组(对照组)、基础配方+0.20%本发明专利实施例1生产的添加剂、基础配方+0.20%本发明专利实施例2生产的添加剂、基础配方+0.20%本发明专利实施例3生产的添加剂,对应投喂不同处理组别的大口黑鲈,分别记做:对照组、实例1组、实例2组、实例3组。每组3个重复。
1.1.2、试验鱼养殖与管理
本实验所用的大口黑鲈鱼苗购于顺德某家鱼苗场。
在正式实验开展之前,将经驯化(驯化期间采用的配合饲料为对照组配合饲料)可摄食配合饲料的大口黑鲈鱼苗(均重6克左右)放在实验系统中暂养1周,以适应环境。暂养结束后即选取大小均一,健康活泼的大口黑鲈鱼苗作为试验对象并开展本次实验。
实验系统使用室内循环水族箱(300L玻璃纤维缸,共18个),水源为曝气自来水。每组实验用饲料设三个平行,每个水族箱里面放30尾大口黑鲈,平均初始体重为6.67±0.03g。实验期间,采取饱食投喂方式,且分别于每天的9:00am和4:00pm投喂两次。实验周期为8周,实验期间每天记录水温及死亡鱼情况,并定时排污一次以清理水族箱底部的粪便。实验期间水温为27.55±1.32℃,溶氧为7.05±0.13mg L-1,氨氮为0.13±0.02mg L-1,pH为7.02±0.10。

实验到第8周结束,逐个清点各个水族箱里大口黑鲈的尾数。实验鱼饥饿空腹24小时后,称重并记录。从每个实验箱内随机选取6尾鱼,用MS-222(Sigma,St Louis,USA at 10mg/l)进行麻醉,分别量体长,称重并做记录,将鱼体进行解剖,分离出内脏团,分离肝脏并观察记录其表观特征,取1g左右的离体肝脏放入离心管中,并将离心管置于液氮罐内,次日转入-80℃冰箱,为后期肝脏酶活的测定作储备。剩下的肝脏称重记录后转置于105℃烘箱中烘干,至重量恒定后称量并记录数据,备后期的常规营养组成分析用。
1.1.4、样品分析方法
采用国际标准方法(AOAC,1995)测定并分析样品的营养成分。肝脏谷胱甘肽过氧化物酶(GSH-PX)活性、过氧化氢酶(CAT)活性的测定采用南京建成相关试剂盒来测定。
1.1.5、统计分析
实验结果用“平均数±SD”来表示,经单因子方差分析后(ANOVA),用Duncan’s多重比较法来分析各实验组之间差异的显著性(P<0.05)。所采用的分析软件为SPSS11.5。
1.2、试验结果
经解剖发现,对照组实验鱼体的肝脏均出现不同程度的病变,出现脂肪肝症状,呈不健康白色,且肝脏缺乏弹性,易碎。而实例1组、实例2组、实例3组这几个试验组别的鱼体肝脏则基本健康,呈现健康的红润色。
由此可知在配合饲料中补充本发明专利生产的大口黑鲈饲料添加剂对鱼体肝脏有良好的保护功效。
1.2.1、不同处理组对大口黑鲈肝脏的水分及脂肪含量的影响(具体结果见表1)。

由表1可知,在饲料中补充添加本发明生产的饲料添加剂,可显著性降低大口黑鲈肝脏的水分及脂肪水平。
1.2.2、不同处理组对大口黑鲈生长及饲料利用效率的影响(具体结果见表2)。

2增重率(WG)=(末均重-初始均重)×100/初始均重
3饲料系数(FCR)=摄食量(g)/增重(g)
4特定生长率SGR=(ln(末均重)-ln(初始均重))×100/天数
由表2可知:本发明在显著提高大口黑鲈养殖成活率的同时,还可显著性提高大口黑鲈的生长效率及饲料利用率。

1CAT:过氧化氢酶,U/mgprot
2GSH-PX:谷胱甘肽过氧化物酶,U/gprot
CAT(过氧化氢酶),又称触酶,是一类广泛存在于动物、植物和微生物体内的末端氧化酶,存在于所有已知的动物的各个组织中,特别在肝脏中以高浓度存在,其生物学功能是催化细胞内过氧化氢分解,防止过氧化。
GSH-PX(谷胱甘肽过氧化物酶)是广泛存在于机体内的一种重要的过氧化物分解酶。它能催化GSH变为GSSG,使有毒的过氧化物还原成无毒的羟基化合物,同时促进H2O2的分解,GSH-Px是细胞内抗脂质过氧化作用的酶性保护系统的主要成分,可催化LPO分解生成相应的醇,防止LPO均裂和引发脂质过氧化作用的链式支链反应,减少LPO的生成从而保护细胞膜的结构及功能不受过氧化物的干扰从而保护机体免受损害。
由表3可知:本发明可显著提高大口黑鲈肝脏中CAT(过氧化氢酶)及GSH-PX(谷胱甘肽过氧化物酶)的活性,从而对肝脏及机体的健康起显著性保护作用。
综上:本发明可有效防治大口黑鲈在用人工配合饲料饲养过程中的脂肪肝的发生,并可显著改善大口黑鲈的生长及对饲料的利用效率和养殖成功率,提高综合养殖效益。
Claims (3)
1.一种预防大口黑鲈脂肪肝病的饲料添加剂,其特征在于,按重量份计,包括,黄芪15~22份;丹参13~20份;陈皮4~8份;佛手4~8份;香橼4~8份;山楂7~12份;炙甘草3~6份;红花5~8份;金钱草4~6份;大黄4~6份;茵陈4~6份;郁金2~4份;维生素C 4~8份;维生素E 3~5份;蛋氨酸3~6份和胆碱1~3份。
2. 一种权利要求1所述的饲料添加剂在制备预防大口黑鲈脂肪肝病药物中的应用。
3. 一种权利要求1所述的饲料添加剂在制备降低大口黑鲈肝脏水分及脂肪的药物中的应用。
