Description
技术领域
本发明涉及一种褐飞虱生存及生长发育相关的基因、编码蛋白及其应用。
背景技术
褐飞虱(Nilaparvata lugens)是一种远距离迁飞性水稻害虫,主要以刺吸韧皮部汁液和传播水稻病毒的方式为害水稻植株。近年来,褐飞虱已发展成为亚洲稻区的重要害虫,常年危害面积在2,000万hm2以上,造成稻谷产量损失数十亿公斤(林拥军等,2011)。目前,针对褐飞虱的主要应急防治措施为喷施化学农药。化学农药的大量滥用,不仅严重污染环境,而且长期施用化学农药可诱使褐飞虱产生抗药性(王鹏等,2013)。在防治褐飞虱的农药中,曾经发挥过较好效果的有效农药,如氟虫腈、吡虫啉、噻嗪酮等,均已淘汰出局或限制使用,而近年出现的取代农药品种数量不多,很多地方在开展防治时,在农药的选择上感到策手无策,化学防治面临防不胜防的尴尬局面。究其原因,此类化学农药多以大规模高强度杀灭害虫为目标,但是,由于褐飞虱种群遗传多样性非常复杂等现实原因,其中往往有部分个体能够存活下来,高选择压的作用结果,最终会导致适应性更强的种群形成。另一方面,化学农药在杀死害虫的同时,也会危及包括天敌在内的非靶标生物,不可避免的会给农田生态系统造成负面影响,生态防治功能没有等到应有的发挥。因此,调整目前的褐飞虱防治策略势在必行。
发明内容
为了克服现有技术的不足,本发明提供了一种在褐飞虱生存及生长发育中发挥重要作用的基因NlPIK3R1、编码蛋白及其应用。对该基因成功实施了RNA干扰,结果表明害虫的生存及生长发育受到明显抑制,存活率下降,体重减轻,成虫羽化时间延迟。
本发明根据NlPIK3R1基因编码的蛋白质相对保守但核酸序列与其他生物同源性低的特点,对靶标基因进行RNA干扰,实现了在核酸水平对褐飞虱生存及生长发育的抑制,显著地降低了褐飞虱的存活率,并避免了在蛋白质水平采用的喷施农药等可能给非靶标生物造成的杀伤。另一方面,靶标基因的RNA干扰,也抑制了害虫的生长发育,使得褐飞虱体重减轻,成虫羽化时间延迟,减少了害虫取食的直接为害。
本发明的技术方案如下:
一种调控褐飞虱生存及生长发育的磷脂酰肌醇3激酶P85Α亚基基因NlPIK3R1,含有SEQ ID NO:1所示的核苷酸序列及其同源突变序列,该基因在维持褐飞虱正常的生存及生长发育过程中发挥重要功能,其功能受到抑制会导致褐飞虱存活率下降,生长变缓,成虫羽化时间推迟。

本发明的有益效果:(1)褐飞虱存活率下降,生长受到抑制,可以减轻害虫取食对水稻作物的直接危害。(2)褐飞虱为r-对策者,采取最大生殖策略,因此,世代更替速度对种群的放大具有非常重要的作用。本发明可以迟滞褐飞虱的羽化时间,减缓了褐飞虱世代更替的速度,降低了在有限时间窗口虫灾爆发的风险。(3)本发明利用靶标基因的核苷酸序列与天敌核苷酸序列同源性低的特点,在核酸水平上进行RNA干扰,避免了由于蛋白质结构保守对天敌等非靶标生物的伤害,有望在实现抑制害虫的同时,充分发挥生态防治的功能。

具体实施方式
以下结合附图和具体实施方式对本发明做进一步的说明。
实施例1
1材料和方法
1.1供试褐飞虱
供试褐飞虱虫源是在人工气候室内用感虫水稻品种TN1上长期饲养的种群,温度为26±2℃,相对湿度为80%±5%,光周期为16L:8D。
1.2褐飞虱PIK3R1基因核心片段的克隆及验证
根据转录组序列信息,获得褐飞虱NlPIK3R1基因的核心序列,经NCBI网站序列比对鉴定。应用Primer Premier 5.0软件设计引物NlPIK3R1-F和NlPIK3R1-R(表1),对序列进行验证。PCR扩增采用50μL反应体系,其中包括PCR Mix 25μL,10μmol/L正反引物各2μL,模板2μL,ddH2O 19μL。PCR反应程序为:94℃4min;94℃30s,58℃30s,72℃3min,30个循环;72℃10min;4℃保存。PCR扩增得到的产物经1%琼脂糖凝胶电泳检测,用AxyPrepTM DNA Gel Extraction Kit(Axygen,USA)回收目的片段,连接到载体PMD-18T(TaKaRa,Japan)4℃过夜,转化到JM109感受态细胞中加1ml LB液体培养基37℃摇床培养2h,取200μL菌液涂布在含有1%Amp的LB固体培养基37℃倒置培养9小时,随机挑取5个单菌落于含1%Amp的LB液体培养基1ml的1.5ml离心管中37℃摇床培养12小时,取1μL菌液进行菌液PCR鉴定阳性克隆菌株并送到上海桑尼生物科技有限公司测序。测序结果用DNAMAN软件和原序列比对,进行验证。
1.3褐飞虱PIK3R1基因全长的克隆
根据前述研究(1.2)获得的褐飞虱NlPIK3R1基因核心序列信息,采用RACE法克隆基因全长。取不同龄期和不同性别成虫的褐飞虱为样本,用TRIzol法进行总RNA的抽提,使用1%琼脂糖凝胶电泳检测RNA的质量,以及使用Nanodrop2000(Thermo,USA)对RNA纯度和浓度的测定。选取质量检测符合要求的RNA样品,用BD SMARTTM RACE cDNAAmplification Kit(Clontech,USA)合成5'-RACE和3'-RACE的模板。分别设计外围引物5O-PIK3R1和3O-PIK3R1(表1),以及内围引物5I-PIK3R1和3I-PIK3R1(表1)。
采用巢式PCR对目的基因的2端进行扩增。扩增体系同上。PCR反应程序为:外围PCR反应条件:94℃ 4min;94℃ 30s,63℃ 30s,72℃ 3min,30个循环;72℃ 10min;4℃保存。内围PCR所用的模板为第一轮PCR得到的产物。内围PCR反应条件:94℃ 4min;94℃ 30s,56℃ 30s,72℃ 3min,30个循环;72℃ 10min;4℃保存。电泳,胶回收,连接,转化等同上。
根据测序结果用DNAMAN软件拼接得到序列全长,并设计全长验证引物PIK3R1-FL-F和PIK3R1-FL-R(表1)对拼接的全长序列进行验证。

1.4褐飞虱PIK3R1基因表达分析
分别对饲养在TN1上褐飞虱按照1-2龄若虫、3-4龄若虫、5龄若虫、刚羽化雌成虫、怀卵雌成虫、雄成虫等类型进行取样,用荧光定量PCR进行基因表达量分析。总RNA的提取及检测方法同上,用于荧光定量PCR检测的cDNA模板按照PrimeScript RT Reagent Kit withgDNA Eraser(Takara,Japan)反转录试剂盒说明步骤操作。荧光定量PCR特异性引物为QPIK3R1-F和QPIK3R1-R(表1),扩增片段经过测序和原序列比对,以β-actin基因为内参(Chen et al.,2010),检测褐飞虱NlPIK3R1基因的相对表达量。荧光定量PCR参考马艳等(2013)的反应体系及方法,其中退火温度改为58℃。
1.5dsRNA的合成
根据cDNA全长序列设计用于合成dsRNA干扰片段的引物dsPIK3R1-F和dsPIK3R1-R(表1),dsGFP及引物修饰参照马艳等(2013)的方法。按照T7 High YieldTranscription Kit(Ambion,USA)试剂盒说明书合成dsRNA。
1.6RNAi实验
取TN1上2龄褐飞虱若虫,采用Fu等(2001)营养液配方和饲喂装置进行人工饲喂,等到上述若虫发育至5龄时进行RNA干扰处理。RNA干扰设置2个浓度:低浓度组(L)为0.1μg/μL dsNlPIK3R1,高浓度组(H)为0.5μg/μL dsNlPIK3R1,同时设置喂食纯营养液作为空白对照组(CK),喂食含0.5μg/μL dsGFP的营养液作为dsGFP对照组(dsGFP)。处理时,每管放入20只发育一致的5龄雌性褐飞虱若虫,设置3个重复,每天更换取食液,清理死亡的褐飞虱及其羽化蜕下的皮,当空白对照组开始出现有羽化个体时,开始对各组进行存活率、羽化率等的统计,并在喂食最后一天(7d)对各组进行称重。
此外,设置平行的RNAi实验组,当空白对照组出现羽化个体时,在第1、3、5、7天对各组分别进行取样,每组3只,并进行RNA的抽提及后续NlPIK3R1基因表达量的荧光定量PCR检测。
2结果与分析
2.1褐飞虱NlPIK3R1基因的cDNA全长克隆及序列分析
以褐飞虱cDNA为模板,对NlPIK3R1的核心片段进行PCR扩增,得到一条与预期长度相符的特异性条带,测序结果表明该片段大小为1087bp,并通过序列比对发现与转录组所得该序列一致。Blast X比对发现,克隆的片段与人体虱Pediculus humanus corporis的PI3K的调节亚基p85α氨基酸序列相识度为59%,表明该片段为褐飞虱编码p85α的核心片段。应用RACE法扩增该基因的cDNA全长,获得5'端序列2300bp和3'序列464bp。用DNAMAN进行序列拼接得到PIK3R1基因全长为2694bp(SEQ ID NO:1),拼接得到的全长序列经两端PCR和测序得到准确验证。
利用ORF Finder(http://www.ncbi.nlm.nih.gov/projects/gorf/)分析,NlPIK3R1基因cDNA全长包含了94bp的5'非编码区(5'UTR),2526bp开放阅读框(ORF)和75bp的3'非编码区(3'UTR),3'端具有典型的polyA结构。NlPIK3R1基因共编码841个氨基酸(SEQ ID NO:2),利用蛋白分子量及等电点预测工具(http://web.expasy.org/compute_pi/),预测得到该蛋白分子量为95.6kDa,等电点为5.84。通过蛋白结构域预测站网(http://smart.embl-heidelberg.de/)预测发现该蛋白具有山姆结构(Sterile alpha motif,SAM)、蛋白激酶C保守区结构域(Protein kinase Cconserved region 1domains,C1)、RhoGTP酶激活蛋白结构域(GTPase-activator protein for Rho,RhoGAP)和2个Src同源结构域(Src homology 2domains,SH2)(图1)。类比赤拟谷盗Triboliumcastaneum(XP_008193465.1,54%)和顶切叶蚁Acromyrm exechinatior(EGI59407.1,57%)的PI3K p85α发现它们都有共同的结构域(图1)。
用来至膜翅目、鞘翅目、蜘蛛目和虱目的10种昆虫的p85α氨基酸序列与褐飞虱p85α氨基酸序列构建系统进化树。进化树分析表明褐飞虱为单独的一类,与人体虱Pediculushumanus corporis较近(图2)。
2.2褐飞虱NlPIK3R1的表达变化分析
荧光定量PCR检测表明,NlPIK3R1基因在褐飞虱所有龄期和不同性别的褐飞虱中均有不同程度表达。在怀卵雌虫时,NlPI3KR1基因表达量急剧增加并达到最高,而在其他阶段和雄成虫中表达量都很低(图3)。
2.3 RNAi对褐飞虱NlPIK3R1 mRNA表达水平的影响
荧光定量PCR检测结果显示,dsGFP对照组和空白对照组的NlPIK3R1 mRNA表达量没有显著差异,并且这两个对照组的mRNA表达量均随着羽化后的时间的增加而增加。低浓度干扰组中,褐飞虱NlPIK3R1 mRNA也随着羽化后的时间而增加,在5天时达到最高值,第7天时开始下降,并且相较于空白对照组和dsGFP对照组其表达量显著降低(图4)。而高浓度的dsNlPIK3R1干扰,在第1天时即有效的抑制了褐飞虱NlPIK3R1 mRNA的表达,其mRNA的表达量一直处于比较低的状态,而在7天时,由于该组用于取样的褐飞虱全部死亡,因而无法得到相应的表达量数据,这也说明高浓度的NlPIK3R1干扰对褐飞虱影响很大。
2.4 RNAi对褐飞虱存活率的影响
在RNA干扰实验中,连续喂养dsNlPIK3R1,对褐飞虱存活率产生了较为明显的影响,从第2天开始,喂食dsNlPIK3R1的2个实验组与对照组就达到了极显著的差异,高浓度处理组的dsNlPIK3R1干扰对褐飞虱的存活率影响尤其明显,在统计的第7天存活率仅为37.50%,而空白对照组和dsGFP对照组分别为87.00%和76.67%(图5)。由此说明,褐飞虱NlPIK3R1基因对褐飞虱的存活有一定的影响,浓度越高对褐飞虱的存活影响越大。连续喂养dsGFP对褐飞虱的存活率也有一定的影响,但是相较于喂食营养液没有显著差异。
2.5 RNAi对褐飞虱羽化率的影响
通过对每天新蜕的皮统计,对空白对照组,dsGFP对照组,低浓度和高浓度dsNlPIK3R1处理喂养的褐飞虱实验组进行羽化率的观测统计(图6)。由图6可以看出喂养dsGFP和纯营养的实验组褐飞虱的羽化率相当,在第5天时全部羽化,而喂食dsNlPIK3R1的2个处理组在第5天的羽化率分别为73.08%和56.09%,与空白对照组和dsGFP对照组均达到了极显著的差异;低浓度干扰组褐飞虱在第7天才全部羽化,而高浓度组在第7天褐飞虱也没全部羽化,其羽化率为93.75%,相较于对照组和dsGFP对照组而言,推迟了至少3天。这表明褐飞虱NlPIK3R1基因在一定程度上调控褐飞虱的发育。
2.6 RNAi对褐飞虱体重的影响
在第7天分别对各组中存活的褐飞虱进行称重统计和分析(图7)。由图7可以看出空白对照组,dsGFP对照组和喂食低浓度dsNlPIK3R1的实验组中褐飞虱的体重没有明显区别,个体平均体重均在2.0mg以上,而喂食高浓度的dsNlPIK3R1实验组中褐飞虱个体平均体重为1.27mg,与其他几组相比存在着显著的差异,说明高浓度的干扰对褐飞虱的生长有很好的抑制效果。

Claims (6)
1.一种调控褐飞虱生存及生长发育的磷脂酰肌醇3激酶p85α亚基的编码基因NlPIK3R1,其特征在于,含有SEQ ID NO:1 所示的核苷酸序列及其同源突变序列,该基因在维持褐飞虱正常的生存及生长发育过程中发挥重要功能,其功能受到抑制会导致褐飞虱存活率下降,生长变缓,体重减轻,成虫羽化时间推迟。
2.一种如权利要求1所述的褐飞虱NlPIK3R1基因编码的蛋白,其特征在于,含有SEQ ID NO:2 所示的氨基酸序列及其同源突变序列,该基因功能受到抑制会导致褐飞虱存活率下降,生长变缓,体重减轻,成虫羽化时间推迟。
3.一种如权利要求1所述的褐飞虱NlPIK3R1基因的RNA干扰在控制褐飞虱方面的应用。
4.如权利要求3所述的应用,其特征在于,所述NlPIK3R1基因的RNA干扰导致褐飞虱的存活率下降,生长变缓,体重减轻,成虫羽化时间推迟,减轻褐飞虱的取食量和危害程度。
5.如权利要求3或4所述的应用,其特征在于,所述NlPIK3R1基因的RNA干扰迟滞褐飞虱的羽化,延长世代周期,抑制种群增长速度,减轻褐飞虱危害程度。
6.如权利要求3所述的应用,其特征在于,基于该核酸和蛋白序列用于农药研发和生物防治。
