Abstract
本发明公开了泥鳅专用复合免疫增强剂及其制备方法,该复合免疫增强剂由以下重量份数的组分组成:大蒜5-10份、大黄提取物5-15份、连翘提取物1-5份、山核桃外果皮粗提物1-5份、月桂叶4-6份、苦楝皮2-3份、杨树叶1-5份、浮萍提取物5-10份、山楂10-15份、食盐10-20份。本发明作为添加剂与常规饲料配合使用,可增强泥鳅机体免疫力,提高抵抗力;无毒副作用和不良反应,对于饲料的适口性无不良影响,大蒜、大黄提取物、连翘提取物、山核桃外果皮粗体物及食盐配合使用,可全年预防水霉病,降低发病率;月桂叶、苦楝皮、杨树叶可对车轮虫及三代虫起到灭杀的效果;浮萍提取物对预防赤皮病有较好的效果。

技术领域
本发明属于饲料添加剂技术领域,具体涉及一种泥鳅专用复合免疫增强剂及其制备方法。
背景技术
泥鳅,别称鱼鳅,体细长,前段略呈圆筒形。后部侧扁,腹部圆,头小。口小、下位,马蹄形。眼小,无眼下刺。须5条。鳞极其细小,圆形,埋于皮下。体背部及两侧灰黑色,全体有许多小的黑斑点,头部和各鳍上亦有许多黑色斑点,背鳍和尾鳍膜上的斑点排列成行,尾柄基部有一明显的黑斑。泥鳅广泛分布于亚洲沿岸的中国、日本、朝鲜、俄罗斯及印度等地。
泥鳅富含蛋白质、脂肪、碳水化合物和钙、磷、铁等矿物元素以及大量的维生素,其中维生素B1的含量比鱼、黄鱼、虾高出3~4倍,而维生素A、维生素C和铁的含量也比其他鱼类要高。泥鳅肉质清淡细嫩,肉味鲜美,营养丰富,并且具有良好的食疗作用。近年来泥鳅的市场需求量大增,供不应求。泥鳅的出口形势很好,日本、韩国需求量很大,订单逐年增加。因此,天然捕捞泥鳅远不能满足市场需求,泥鳅人工养殖开始兴起,泥鳅在养殖过程中经常容易出现各种病虫害,尤其是每年的三、四月份很容易出现水霉病,泥鳅在苗种培育阶段常见有车轮虫、舌杯虫、三代虫寄生而引起死亡,气泡病及赤皮病也是泥鳅在养殖过程中,很容易感染病害,现有技术中,针对泥鳅常见病虫害,通常是在发现病虫害后再采用化学药剂进行灭杀,而不是采用提前预防的措施,该种化学试剂灭杀,对轻微的病虫害可以起到一定的效果,但对于来势比较凶猛的病虫害,通常情况下,效果不是很好,会导致大批泥鳅的死亡,给养殖户带来一定的经济损失,该种方式进行病虫害的灭杀,只是起到临时的灭杀,并不能从根本上进行治愈, 病虫害的发生,导致泥鳅的免疫力下降,死亡率大增,严重影响了泥鳅的生长速度和养殖的经济效益,如何在病虫害发生前进行预防,提高鱼体抵抗病虫害的入侵,降低发病率, 成为关键。在泥鳅的养殖过程中,饲料的投喂是最关键的一步,而泥鳅本身抵抗力的提高,也跟饲料的投喂密切相关,因此,在饲料中添加复合免疫增强剂,以通过投喂饲料来调节机体健康、增强机体免疫力、抑菌杀菌抗虫,对泥鳅的人工养殖非常重要。
发明内容
针对现有技术中存在的问题,本发明目的在于在提供一种泥鳅专用复合免疫增强剂及其制备方法。
本发明通过以下技术方案加以实现:
所述的泥鳅专用复合免疫增强剂,其特征在于该复合免疫增强剂由以下重量份数的组分组成:大蒜5-10份、大黄提取物5-15份、连翘提取物1-5份、山核桃外果皮粗提物1-5份、月桂叶4-6份、苦楝皮2-3份、杨树叶1-5份、浮萍提取物5-10份、山楂10-15份、食盐10-20份。
所述的泥鳅专用复合免疫增强剂,其特征在于由以下重量份数的组分组成:大蒜7份、大黄提取物10份、连翘提取物3份、山核桃外果皮粗提物2份、月桂叶5份、苦楝皮3份、杨树叶3份、浮萍提取物8份、山楂15份、食盐20份。
所述的泥鳅专用复合免疫增强剂,其特征在于该复合免疫增强剂添加到泥鳅复合饲料中,复合免疫增强剂与泥鳅复合饲料的使用比例为1:50-80。
所述的泥鳅专用复合免疫增强剂,其特征在于所述复合免疫增强剂添加到泥鳅复合饲料中,复合免疫增强剂与泥鳅复合饲料的使用比例为1:65。
所述的泥鳅专用复合免疫增强剂的制备方法,其特征在于按下列步骤进行:
1)分别称取配方量的大月桂叶、苦楝皮、杨树叶、山楂,清洗干净后于烘干箱中烘干,烘干后各成分分别粉碎,磨成细粉,过40目筛,得各成分干粉,备用;
2)取配方量的去皮后的新鲜大蒜头,用粉碎机将大蒜头加工成蒜泥,将蒜泥放入烘箱,以文火烘,温度控制在65℃,直至蒜泥成块状,烘干过程中,每隔1个小时翻动蒜泥一次,使蒜泥受热均匀,将烘干后的蒜泥于粉碎机中粉碎,过60目筛,得蒜粉;
3)将步骤2)制得的蒜粉、步骤1)制得的各成分干粉混合后,添加配方量的大黄提取物、连翘提取物、山核桃外果皮粗提物、浮萍提取物、食盐混合,搅拌均匀即可。
所述的泥鳅专用复合免疫增强剂的制备方法,其特征在于所述大黄提取物的制备方法为:大黄粉碎,过40目筛,用90%乙醇溶液回流提取,每次提取1-2小时,重复提取2-3次,大黄粉与乙醇的重量体积比为1:15-20g/ml,提取液浓缩至浸膏,浸膏在165℃的温度条件下喷雾干燥,得大黄提取物。
所述的泥鳅专用复合免疫增强剂的制备方法,其特征在于所述连翘提取物的制备方法为:连翘叶粉碎,过40目筛,加15倍重量的水回流提取2-3小时,提取液过滤,滤液浓缩至浸膏,浸膏于165℃的温度条件下喷雾干燥,得连翘提取物。
所述的泥鳅专用复合免疫增强剂的制备方法,其特征在于所述山核桃外果皮粗提物的制备方法为:山核桃外果皮烘干、粉碎,过40目筛,加75%的乙醇浸提48小时,浸提液浓缩至浸膏,浸膏在170℃的温度条件下喷雾干燥,得山核桃外果皮粗提物。
所述的泥鳅专用复合免疫增强剂的制备方法,其特征在于所述浮萍提取物的制备方法为:浮萍全草烘干粉碎,按重量加入20倍浓度为90%的乙醇溶液,加热回流提取1-3次,每次1-1.5小时,提取液合并,浓缩至浸膏,浸膏在170℃的温度条件下喷雾干燥,得浮萍提取物。
大蒜营养丰富:每100克含水分69.8克,蛋白质4.4克,脂肪0.2克,碳水化合物23.6克,钙5毫克,磷44毫克,铁0.4毫克,维生素C3毫克。此外,还含有硫胺素、核黄素、尼克酸、蒜素、柠檬醛以及硒和锗等微量元素。含挥发油约0.2%,油中主要成分为大蒜辣素,是大蒜中所含的蒜氨酸受大蒜酶的作用水解产生。尚含多种烯丙基、丙基和甲基组成的硫醚化合物等。大蒜具有强大的杀菌能力,能消灭侵入体内的病菌。它还有助维生素B1的吸收,促进糖类的新陈代谢以产生能量,并缓解疲劳。大蒜集100多种药用和保健成分于一身,其中含硫挥发物43种,硫化亚磺酸(如大蒜素)酯类13种、氨基酸9种、肽类8种、甙类12种、酶类11种。另外,蒜氨酸是大蒜独具的成分,当它进入血液时便成为大蒜素,这种大蒜素即使稀释10万倍仍能在瞬间杀死伤寒杆菌、痢疾杆菌、流感病毒等。
大黄、连翘均为药用植物,具有抗菌消炎的功效,两者复配,其抗菌杀菌能力增强,大黄、连翘、山核桃外果皮粗提物与大蒜及食盐混合使用,可有效的防止水霉病的发生,还可提高肌体的免疫能力。月桂叶与苦楝皮及杨树叶复配在一起可有效避免泥鳅车轮虫及三代虫的发生。
浮萍性味辛、寒、归肺(鳃)、膀胱经。具清热解毒、透疹、利水消肿等功效。 浮萍全草主含荭草素、牡荆素、芹菜糖、木犀草黄素-7-葡萄糖甙。尚含醋酸钾、氯化钾、碘、溴、叶绿素、鞣质及类脂化合物(亚麻酸 、棕榈酸、亚油酸)等。其中,鞣质是一类复杂的酚类化合物,有涩味及收敛性,能与蛋白质、黏液、生物碱盐、重金属盐结合生成沉淀。酚类具有抗菌作用,它对细菌的有害作用可能主要是使蛋白质变性,同时又有表面活性剂的作用,破坏细胞膜的通透性,使细胞内含物外溢。当浓度高时是致死因子,反之则起抑菌作用。故常用作为收敛止血、止泻 、抗菌消炎。此外还可作为生物碱和重金属的解毒剂(与重金属结合生成配位化合物,而排出体外)。叶绿素的可溶性盐类有一定的抗菌消炎作用和促进肉芽生长的作用。碘是强杀菌剂,能杀死细菌、真菌,并使病毒灭活。醋酸钾是强碱弱酸盐,呈碱性,可不断中和细菌产生的酸,强碱对革兰氏阴性细菌与病毒比革兰氏阳性细菌作用强。赤皮病的病原体是萤光极毛杆菌,均为革兰氏阴性茵。因此,浮萍不仅可抗鱼害黏球菌和赤皮病的萤光极毛杆菌,而且可抗病毒。另外,浮萍还含有亚麻酸、棕榈酸、亚油酸等不饱和脂肪酸,其中亚油酸是鱼类最重要的不饱和脂肪酸,可合成前列腺素。
与现有制剂相比,本发明具有以下优点:
1)作为添加剂与泥鳅常规饲料配合使用,大蒜、大黄提取物、连翘提取物、山核桃外果皮粗体物及食盐配合使用,可全年预防水霉病,降低发病率;
2)月桂叶、苦楝皮、杨树叶与饲料配合使用,可对车轮虫及三代虫起到灭杀的效果;
3)浮萍提取物的添加,对预防赤皮病有较好的效果;
4)各成分的配合,可增强泥鳅机体免疫力,提高对病虫害的抵抗力;
5)无毒副作用和不良反应,对于饲料的适口性无不良影响,山楂的添加,便于提高饲料的适口性。
具体实施方式
通过以下具体实施例对本发明作进一步详述。
实施例1
泥鳅专用复合免疫增强剂,组成为:大蒜7份、大黄提取物10份、连翘提取物3份、山核桃外果皮粗提物2份、月桂叶5份、苦楝皮3份、杨树叶3份、浮萍提取物8份、山楂15份、食盐20份。其制备方法为:分别称取配方量的大月桂叶、苦楝皮、杨树叶、山楂,清洗干净后于烘干箱中烘干,烘干后各成分分别粉碎,磨成细粉,过40目筛,得各成分干粉;取配方量的去皮后的新鲜大蒜头,用粉碎机将大蒜头加工成蒜泥,将蒜泥放入烘箱,以文火烘,温度控制在65℃,直至蒜泥成块状,烘干过程中,每隔1个小时翻动蒜泥一次,使蒜泥受热均匀,将烘干后的蒜泥于粉碎机中粉碎,过60目筛,得蒜粉;将上述制备的各成分干粉及蒜粉与配方量的大黄提取物、连翘提取物、山核桃外果皮粗提物、浮萍提取物、食盐混合在一起,搅拌均匀即制得泥鳅专用复合免疫增强剂。取配制好的泥鳅专用复合免疫增强剂1份,与65份的泥鳅常规饲料混合均匀,对泥鳅进行投喂。
其中:大黄提取物的制备方法为:大黄粉碎,过40目筛,用90%乙醇溶液回流提取,每次提取1-2小时,重复提取2-3次,大黄粉与乙醇的重量体积比为1:15-20g/ml,提取液浓缩至浸膏,浸膏在165℃的温度条件下喷雾干燥,得大黄提取物。
连翘提取物的制备方法为:连翘叶粉碎,过40目筛,加15倍重量的水回流提取2-3小时,提取液过滤,滤液浓缩至浸膏,浸膏于165℃的温度条件下喷雾干燥,得连翘提取物。
山核桃外果皮粗提物的制备方法为:山核桃外果皮烘干、粉碎,过40目筛,加75%的乙醇浸提48小时,浸提液浓缩至浸膏,浸膏在170℃的温度条件下喷雾干燥,得山核桃外果皮粗提物。
浮萍提取物的制备方法为:浮萍全草烘干粉碎,按重量加入20倍浓度为90%的乙醇溶液,加热回流提取1-3次,每次1-1.5小时,提取液合并,浓缩至浸膏,浸膏在170℃的温度条件下喷雾干燥,得浮萍提取物。
实施例2
泥鳅专用复合免疫增强剂,组成为:大蒜5份、大黄提取物15份、连翘提取物1份、山核桃外果皮粗提物5份、月桂叶4份、苦楝皮3份、杨树叶1份、浮萍提取物10份、山楂10份、食盐20份。其制备方法为:分别称取配方量的大月桂叶、苦楝皮、杨树叶、山楂,清洗干净后于烘干箱中烘干,烘干后各成分分别粉碎,磨成细粉,过40目筛,得各成分干粉;取配方量的去皮后的新鲜大蒜头,用粉碎机将大蒜头加工成蒜泥,将蒜泥放入烘箱,以文火烘,温度控制在65℃,直至蒜泥成块状,烘干过程中,每隔1个小时翻动蒜泥一次,使蒜泥受热均匀,将烘干后的蒜泥于粉碎机中粉碎,过60目筛,得蒜粉;将上述制备的各成分干粉及蒜粉与配方量的大黄提取物、连翘提取物、山核桃外果皮粗提物、浮萍提取物、食盐混合在一起,搅拌均匀即制得泥鳅专用复合免疫增强剂。取配制好的泥鳅专用复合免疫增强剂1份,与50份的泥鳅常规饲料混合均匀,对泥鳅进行投喂。

实施例3
泥鳅专用复合免疫增强剂,组成为:大蒜10份、大黄提取物5份、连翘提取物5份、山核桃外果皮粗提物1份、月桂叶6份、苦楝皮2份、杨树叶5份、浮萍提取物5份、山楂5份、食盐10份。其制备方法为:分别称取配方量的大月桂叶、苦楝皮、杨树叶、山楂,清洗干净后于烘干箱中烘干,烘干后各成分分别粉碎,磨成细粉,过40目筛,得各成分干粉;取配方量的去皮后的新鲜大蒜头,用粉碎机将大蒜头加工成蒜泥,将蒜泥放入烘箱,以文火烘,温度控制在65℃,直至蒜泥成块状,烘干过程中,每隔1个小时翻动蒜泥一次,使蒜泥受热均匀,将烘干后的蒜泥于粉碎机中粉碎,过60目筛,得蒜粉;将上述制备的各成分干粉及蒜粉与配方量的大黄提取物、连翘提取物、山核桃外果皮粗提物、浮萍提取物、食盐混合在一起,搅拌均匀即制得泥鳅专用复合免疫增强剂。取配制好的泥鳅专用复合免疫增强剂1份,与80份的泥鳅常规饲料混合均匀,对泥鳅进行投喂。

对照组1
泥鳅专用复合免疫增强剂,组成为:大蒜4份、大黄提取物17份、连翘提取物6份、山核桃外果皮粗提物0.5份、月桂叶2份、苦楝皮5份、杨树叶0.5份、浮萍提取物15份、山楂8份、食盐5份。制备方法同实施例1。取泥鳅专用复合免疫增强剂1份,与65份的泥鳅常规饲料混合均匀,对泥鳅进行投喂。
对照组2
采用同实施例1相同的常规饲料进行投喂,不添加复合免疫增强剂。
试验例
将池塘分隔成五个相同面积的区域,编号为1、2、3、4、5,于六月初,向每个分隔区分别投放体长为3cm左右的泥鳅种苗200尾,其中,1按照实施例1的配方含量进行投喂, 2按照实施例2的配方含量进行投喂, 3按照实施例3的配方含量进行投喂, 4按照对照组的配方含量进行投喂, 5按照对照组2配方含量进行投喂,饲喂6个月,饲喂效果见表1。

实验结果表明:在外界条件相同的情况下,本发明泥鳅专用复合免疫增强剂能明显提高泥鳅的增长速度及体重,降低泥鳅的发病率,提高泥鳅的存活率,降低饲料系数。由表1可看出,采用实施例1含量配方投喂的1号隔离区的泥鳅,各项指标均优于其他各组,采用对照组1含量配方投喂的4号隔离区的泥鳅,由于对照组1各成分的含量均不在本专利申请的保护范围内,其饲喂的各项指标均低于实施例1-3的饲喂效果,从表中可看出,其成活率偏低,是因为浮萍提取物的添加量偏多,导致泥鳅鱼苗的死亡数量增加。

1.泥鳅专用复合免疫增强剂,其特征在于该复合免疫增强剂由以下重量份数的组分组成:大蒜5-10份、大黄提取物5-15份、连翘提取物1-5份、山核桃外果皮粗提物1-5份、月桂叶4-6份、苦楝皮2-3份、杨树叶1-5份、浮萍提取物5-10份、山楂10-15份、食盐10-20份。
2.根据权利要求1所述的泥鳅专用复合免疫增强剂,其特征在于由以下重量份数的组分组成:大蒜7份、大黄提取物10份、连翘提取物3份、山核桃外果皮粗提物2份、月桂叶5份、苦楝皮3份、杨树叶3份、浮萍提取物8份、山楂15份、食盐20份。
3.根据权利要求1或2所述的泥鳅专用复合免疫增强剂,其特征在于该复合免疫增强剂添加到泥鳅复合饲料中,复合免疫增强剂与泥鳅复合饲料的使用比例为1:50-80。
4.根据权利要求4所述的泥鳅专用复合免疫增强剂,其特征在于所述复合免疫增强剂添加到泥鳅复合饲料中,复合免疫增强剂与泥鳅复合饲料的使用比例为1:65。
5.泥鳅专用复合免疫增强剂的制备方法,其特征在于按下列步骤进行:
1)分别称取配方量的大月桂叶、苦楝皮、杨树叶、山楂,清洗干净后于烘干箱中烘干,烘干后各成分分别粉碎,磨成细粉,过40目筛,得各成分干粉,备用;
2)取配方量的去皮后的新鲜大蒜头,用粉碎机将大蒜头加工成蒜泥,将蒜泥放入烘箱,以文火烘,温度控制在65℃,直至蒜泥成块状,烘干过程中,每隔1个小时翻动蒜泥一次,使蒜泥受热均匀,将烘干后的蒜泥于粉碎机中粉碎,过60目筛,得蒜粉;
3)将步骤2)制得的蒜粉、步骤1)制得的各成分干粉混合后,添加配方量的大黄提取物、连翘提取物、山核桃外果皮粗提物、浮萍提取物、食盐混合,搅拌均匀即可。
6.根据权利要求5所述的泥鳅专用复合免疫增强剂的制备方法,其特征在于所述大黄提取物的制备方法为:大黄粉碎,过40目筛,用90%乙醇溶液回流提取,每次提取1-2小时,重复提取2-3次,大黄粉与乙醇的重量体积比为1:15-20g/ml,提取液浓缩至浸膏,浸膏在165℃的温度条件下喷雾干燥,得大黄提取物。
7.根据权利要求5所述的泥鳅专用复合免疫增强剂的制备方法,其特征在于所述连翘提取物的制备方法为:连翘叶粉碎,过40目筛,加15倍重量的水回流提取2-3小时,提取液过滤,滤液浓缩至浸膏,浸膏于165℃的温度条件下喷雾干燥,得连翘提取物。
8.根据权利要求5所述的泥鳅专用复合免疫增强剂的制备方法,其特征在于所述山核桃外果皮粗提物的制备方法为:山核桃外果皮烘干、粉碎,过40目筛,加75%的乙醇浸提48小时,浸提液浓缩至浸膏,浸膏在170℃的温度条件下喷雾干燥,得山核桃外果皮粗提物。
9.根据权利要求5所述的泥鳅专用复合免疫增强剂的制备方法,其特征在于所述浮萍提取物的制备方法为:浮萍全草烘干粉碎,按重量加入20倍浓度为90%的乙醇溶液,加热回流提取1-3次,每次1-1.5小时,提取液合并,浓缩至浸膏,浸膏在170℃的温度条件下喷雾干燥,得浮萍提取物。
