本发明属微生物药物领域,涉及一种产除莠霉素的菌株及其应用,该菌株经形态特征、培养特征、生理生化特征及16S?rDNA基因分析,鉴定其为一株吸水链霉菌(<i>Streptomyces?hygroscopicus</i>)。已于2013年12月6日在中国普通微生物菌种保藏管理中心进行登记,保藏编号:CGMCC?No.8544。该菌发酵上清液中活性成分用有机溶剂萃取后,经紫外吸收光谱、红外吸收光谱、核磁共振波谱及质谱检测分析,证实其活性成分为Herbimycin?A(除莠霉素A),属于苯醌安莎类抗生素,发酵水平可达1500μg/mL以上,可实现产业化,提取方便,组分单一,产品质量好。Herbimycin?A具有重要的生物学活性,可作为器官移植抗排斥药物,具有广谱除草活性,有微弱的抗细菌和抗真菌活性。此外,Herbimycin?A对多种肿瘤细胞具有抑制作用。

本发明涉及从自然界分离药用微生物,具体涉及一种产除莠霉素的菌株及其应用,属于微生物药物领域。
背景技术
众所周知,陆地放线菌是抗菌、抗肿瘤以及免疫抑制剂等多种药物的重要来源。土壤放线菌产生的抗生素也已在农业上广泛应用。安莎类抗生素为一个脂肪链(称为安莎桥)连接于芳香环的两个不相邻原子间的一类化合物,其分子结构中的芳香环有两种:苯环和萘环。因此,安莎类抗生素可以分为苯安莎类抗生素和萘安莎类抗生素两种。
由吸水链霉菌产生的HerbimycinA(除莠霉素A)属于苯安莎类抗生素。它具有重要的生物学活性,不但具有广谱的除草作用,还有抗细菌(革兰阳性菌和革兰阴性菌)、抗霉、抗酵母和抗原虫等活性,是一种新型的强效免疫抑制剂。免疫抑制剂的主要功能是异源器官移植抗排斥反应和人自身免疫性疾病治疗,因此,HerbimycinA在器官移植的抗排异反应中,将可能得到广泛应用。此外,HerbimycinA具有很好的抗肿瘤增殖的作用,具有极其重要的开发和研究价值,应用前景广阔。
发明内容
本发明的目的在于提供一种产除莠霉素的菌株及其应用。是从自然界寻找药用微生物,其中一株来源于土壤的放线菌SH035,其发酵代谢产物,含生理活性物质,经提取纯化,确定其化学结构。
一种产除莠霉素的菌株,所述菌株为吸水链霉菌(Streptomyceshygroscopicus)SH035,已于2013年12月6日在中国普通微生物菌种保藏管理中心登记保蒇,保藏编号为CGMCCNo.8544。
所述菌株的代谢产物经HPLC、质谱和核磁共振技术确定,该代谢产物为除莠霉素,与除莠霉素A同质。
所述菌株在抗肿瘤及抗生素药物方面的应用。
具体方法为:
1、该放线菌SH035筛选自福建莆田市坪洋村鸡角髻山上采集的土壤中,经分离获得具有抗细菌、真菌和肿瘤细胞活性的放线菌菌株,命名为吸水链霉菌(Streptomyceshygroscopicus)SH035,于2013年12月6日在中国普通微生物菌种保藏管理中心登记保蒇,保藏编号为CGMCCNo.8544。
2、根据菌株的菌落形态特征(图1)、培养特征、生理生化特征和16SrDNA序列比对分析及其进化树构建(图2),确定该菌株为吸水链霉菌,命名为吸水链霉菌SH035。
3、吸水链霉菌SH035代谢产物结构确定。其发酵上清液中的活性成分,经有机溶剂萃取,浓缩,精制,高效液相色谱(HPLC),质谱(MS)和核磁共振波谱检测分析,确定该活性成分为HerbimycinA,与除莠霉素同质。
4、该出发菌株的除莠霉素发酵单位可达1500μg/mL以上,发酵条件经简单优化后,高达5000μg/mL以上,满足产业化要求。
5、发酵主要培养基组成:葡萄糖1.5%,淀粉3.0%,黄豆饼粉3.5%,玉米粉2.0%,MgSO40.5%,CaCO30.2%,淀粉酶0.01%,pH7.2。
6、代谢产物提取精制。发酵液经酸化,过滤,去除固体物,得到滤液,经溶媒萃取,浓缩,蒸干,洗涤,得到干品,即为HerbimycinA。提取方便,组分单一,产品质量好,收率可达70%以上,含量可达98%以上。
本发明的优点在于:本申请的吸水链霉菌SH035,其生物合成HenbimycinA的发酵单位可达1500μg/mL以上,发酵条件经简单优化后,高达5000μg/mL以上,满足产业化要求。本发明提供的代谢产物HerbimycinA,提取方便,组分单一,产品质量好,收率可达70%以上,含量可达98%以上,这些优势未见报道。

具体实施方式
实施例1:菌株SH035的筛选分离
从福建莆田市坪洋村鸡角髻山上采集的土壤分离获得多个菌株,经过初筛和复筛后得到具有抑菌活性的放线菌,其中一株编号为放线菌SH035。
实施例2:放线菌SH035菌株的鉴定
2.1菌落特征观察:用高氏Ⅰ号培养基、察氏(Czapck)培养基、葡萄糖门冬酰胺培养基、淀粉培养基和马铃薯葡萄糖培养基(PDA),涂布于以上5种培养基平板上,28℃恒温培养3至5天,待其长出孢子后,观察单菌落形态以及孢子颜色。
高氏Ⅰ号培养基:菌落较大,圆形,边缘整齐,表面干燥且有皱状隆起,初期为白色,形成孢子后呈浅灰色,气生菌丝为灰色,基内菌丝为淡黄色。
葡萄糖门冬酰胺培养基:菌落较小,圆形,形成的孢子为白色,菌落周边不产孢子,气生菌丝为灰白色,基内菌丝为淡黄色,菌落表面有水珠产生。
察氏培养基:菌落圆形,边缘整齐,菌落表面干燥,形成的孢子为灰色,气生菌丝灰色,基内菌丝灰色。
马铃薯葡萄糖培养基:菌落圆形,边缘整齐,中间凸起,菌落表面长满了丰富的灰白色孢子,气生灰白色,基内菌丝降黄色到褐色。
淀粉培养基:菌落较小,圆形,不透明,边缘整齐,菌落表面不形成孢子,孢子丝为灰白色,基内菌丝为浅黄色。
附培养基配方法:
高氏Ⅰ号培养基:可溶性淀粉2%,KNO30.1%,MgSO4·7H2O0.05%,K2HPO4·3H2O0.05%,NaCl0.05%,FeSO4·7H2O0.001%,琼脂2%,pH7.4~7.6。
察氏(Czapck)培养基:蔗糖3%,NaNO30.2%,MgSO4·7H2O0.05%,K2HPO4·3H2O0.1%,KCl0.05%,FeSO4·7H2O0.001%,琼脂2%,pH自然。
葡萄糖门冬酰胺培养基:葡萄糖1%,天冬酰胺0.05%,K2HPO4·3H2O0.05%,琼脂2%,pH7.2~7.4。
淀粉培养基:可溶性淀粉0.2%,牛肉膏0.5%,蛋白胨1%,NaCl0.5%,琼脂2%,pH7.0~7.2。
马铃薯葡萄糖培养基(PDA):马铃薯20%,葡萄糖2%,琼脂2%,pH自然。
2.2菌丝体特征观察:采用插片法以及深层培养法观察待测菌菌丝的形态以及孢子特征。插片法:取待测菌菌悬液涂布于高氏Ⅰ号固体培养基上,用镊子夹取已灭过菌的盖玻片,以大约45°角斜插入培养基内,每平板插3-4片。将插片后的平板倒置于28℃恒温培养箱培养,3-5天后用镊子小心取出盖玻片,然后将有菌的一面盖在载玻片上,直接用显微镜观察待测菌的气生菌丝、基内菌丝及孢子的着生情况。深层培养法:由此从形态上初步确定待测菌的分类地位。
2.3生理生化特征观察:明胶液化缓慢,牛奶胨化不凝固,可以水解淀粉,在纤维素上生长缓慢,硝酸盐不还原,不产生黑色素和H2S。该菌能广泛利用多种糖类,其中以葡萄糖、木糖、阿拉伯糖较为合适,蔗糖、棉子糖、甘露醇、果糖、鼠李糖、肌醇和半乳糖次之。
2.4分子生物学鉴定:通过提取SH035基因组DNA和16SrDNAPCR扩增,SH035扩增出的16SrDNA片段长度为1584bp,其DNA序列如SEQIDNO.1所示。与链霉菌属(Streptomycessp.)在遗传上很相近,鉴定该菌株为一株新的吸水链霉菌,其进化树见图2。
实施例3:吸水链霉菌SH035发酵代谢产物的制备
3.1菌株复壮培养:将分离保存的吸水链霉菌SH035,接种至高氏Ⅰ号固体培养基上,于28℃条件下,恒温培养至试管斜面长满孢子,供后续使用。
3.2发酵培养:培养液组成:葡萄糖1.5%,淀粉3.0%,黄豆饼粉3.5%,玉米粉2.0%,MgSO40.5%,CaCO30.2%,淀粉酶适量,pH7.2。每1000mL三角瓶装发酵培养液180mL;配制后121℃灭菌20min,待冷却至室温在无菌条件下用接种环挑取一环吸水链霉菌SH035孢子接种,在28℃条件下,摇床转速180rpm,发酵培养144h,发酵单位可达2000~5000μg/mL。
3.3代谢产物提取:发酵完毕,得到发酵液,加盐酸酸化到pH2.0,静置30min,氢氧化钠溶液回调到pH7.0后,离心收集发酵液,去除固体物。上清滤液加入四分之一体积的乙酸乙酯萃取,萃取液经真空浓缩,得到黄色固体,即为高纯度代谢产物,收率70%以上,含量达98以上,供后续结构分析确定。
实施例4:吸水链霉菌SH035发酵代谢产物的鉴定
发酵提取精制的代谢产物,经HPLC(图4),MS(5)和氢核磁共振氢谱及碳核磁共振碳谱(表1)分析,综合分析结果,分子式为:C30H42N2O9,分子量为:574.3。推测其化学结构为herbimycinA(图3)。

1.一种产除莠霉素的菌株,其特征在于:所述菌株为吸水链霉菌(Streptomyceshygroscopicus)SH035,已于2013年12月6日在中国普通微生物菌种保藏管理中心登记保蒇,保藏编号为CGMCCNo.8544。
2.根据权利要求1所述的一种产除莠霉素的菌株,其特征在于:所述菌株的代谢产物经HPLC、质谱和核磁共振技术确定,该代谢产物为除莠霉素,与除莠霉素A同质。
3.如权利要求1所述的一种产除莠霉素的菌株在制备抗肿瘤及抗生素药物方面中的应用。
