Abstract

本发明提供一种复合惰性液,它由碳原子数分别为6~8和20的全氟取代的氟碳化合物组成。该复合惰性液用于自动生化分析仪上,测定总蛋白、葡萄糖、尿素氮、尿酸、谷丙转氨酶、无机磷、镁、氯、α-羟丁酸脱氢酶、胆固醇、白蛋白等生化项目,测定结果精密度、准确度符合临床检验要求。

Description

复合惰性液
本发明涉及一种分析用试剂,特别是涉及一种复合惰性液。
目前国内进口美国TECHNICON公司RA一系列自动生化分析仪100台左右,凡购买分析仪单位必须购进价格昂贵的惰性液(200$/100ml),仪器才能运转,经文献检索未查找到该惰性液的具体组份,通过分析研究,确认它是一种沸点为150℃,粘度为4.9mm2/S的氟碳化合物。目前国内仅有三种氟碳化合物,分子式为CnF2n+2,即FC80、FC100和FC170。其中n分别为6,6~8,20。但作为惰性液单独使用均不能满足分析的要求。
本发明的目的在于提供一种价格低廉,且由两种氟碳化合物组成的双组份复合惰性液。
本发明复合惰性液由两种全氟取代的氟碳化合物组成,其中一种为FC100,分子式CnF2n+2,6≤n≤8,n为正整数;另一种为FC170,分子式C20F42,二者配合的体积比为FC100∶FC170=120~150∶1,FC100和FC170的浓度均为100%。其中FC100的沸点为95~107℃,25℃温度下,粘度为0.8毫米2/秒;FC170的沸点为176℃,25℃温度下,粘度为116毫米2/秒,当配合比例超出此范围,如FC100含量过高时,会出现标本粘附于加样针外缘,影响仪器操作功能;而FC170含量过高时,则会出现氟油现象,似油状物附着于反应盘上,不利于反应盘清洗和反复使用。采用此配比范围内的复合惰性液于生化分析仪上,使之在仪器运转时通过液体促动器及任意增进阀的作用将其吸入管腔,当再吸入试剂或样品时,已吸入管腔的惰性液将作为非反应化学胶束把试剂或标本包裹其中,起到试剂,标本间互不污染的作用,使分析仪在无化学污染的条件下,快速、连续地进行任意检验项目的测定。
本发明复合惰性液的优点在于具有抗化学交叉污染能力,耐强酸、强碱、强氧化剂,且不沾污加样针和反应盘,可反复使用,与进口惰性液相比,应用效果相同,所测结果精密度,准确度均符合临床检验要求,但成本大大低于进口惰性液(进口惰性液单价为200$/100ml,该复合惰性液单价为42元/100ml)。专用于生化分析仪上测定各种生化项目。
下面结合实施例详细说明本发明
实施例1:
按体积比,取120份FC100与1份FC170充分混合,测试质控物,所测结果在靶值范围内,加样针外缘无标本粘附物附着,反应盘上无油状物附着。临床应用情况如表2所示。
实施例2:
按体积比,取150份FC100与1份FC170充分混合,测试质控物,所测结果在靶值范围内,加样针外缘无标本粘附物附着,反应盘上无油状物附着。临床应用情况如表2所示。
对照例:
分别取用实施例1和2的复合惰性液及美国Random AccessFluid于RA-XT自动生化分析仪上测定总蛋白、葡萄糖、尿素氮、尿酸、谷丙转氨酶、无机磷、钙、氯、镁、α-羟丁酸脱氢酶、胆固醇和白蛋白等生化项目,进行抗化学交叉污染能力比较,结果如表1所示。

Claims (2)

1、一种复合惰性液,其特征在于由两种全氟取代的氟碳化合物组成,其中一种为FC100,分子式CnF2n+2,其中6≤n≤8,并且n为正整数;另一种为FC170,分子式C20F42,二者配合的体积比为FC100∶FC170=120~150∶1,FC100和FC170的浓度均为100%。
2、如权利要求1所述的复合惰性液,其特征在于FC100的沸点为95~107℃,25℃温度下粘度为0.8毫米2/秒;FC170的沸点为176℃,25℃温度下的粘度为116毫米2/秒。
