本发明公开了利用玉米芯残渣同步糖化发酵生产2,3-丁二醇的方法,属于属于生物化工技术领域,提供了以使用玉米芯提取木糖后的残渣为原料进行同步糖化发酵生产2,3-丁二醇的方法:以玉米芯残渣为碳源,尿素为氮源,利用肺炎克雷伯氏菌CICC 10011、产酸克雷伯氏菌CICC 21518、多粘类芽孢杆菌CICC 10010、阴沟肠杆菌CICC 10014及其混合菌液进行同步糖化发酵,2,3-丁二醇与乙偶姻达到玉米芯残渣干重的19.59%-36.13%。本发明以玉米芯残渣为碳源,有效降低了生产成本,实现资源的高值利用,具有巨大的经济效益和社会效益。

技术领域
本发明属于生物化工技术领域,涉及一种发酵生产2,3-丁二醇的方法。
背景技术
2,3-丁二醇(2,3-butanediol,简称2,3-BD)广泛应用于化工、食品以及航空航天等众多领域。其热值为27200kJ/kg,与乙醇(29100kJ/kg)相近,可用作燃料,同时,还可用于制备聚合物、油墨、香水、防冻剂、香薰剂、增湿剂、炸药以及药物的手性载体等。
随着石化资源的日益枯竭以及石化工业对环境的影响,可再生资源引来了越来越多的关注。由于石油价格的不断上涨及其不稳定性,利用可再生资源生产生物基大宗化学品如生物燃料等引起了人们极大的兴趣。利用可再生的生物质资源作为底物,将生物发酵和绿色化学工业有机结合,可以建立起环境友好的、可持续发展的化学工业。
目前2,3-丁二醇的生产方法有化学法和生物转化法。工业上生产2,3-丁二醇的化学法,主要是以石油裂解时产生的四碳类碳氢化合物在高温高压下水解得到,然而化学法生产2,3-丁二醇不仅成本高而且过程繁琐,因此工业化生产较困难。生物发酵法合成2,3-丁二醇使用的生物质原料可再生,较化学法生产2,3-丁二醇工艺更简单、更加环境友好,不仅可以避免化学法合成的困难,而且可以促进人类社会生产,由传统的石油炼制向以可再生资源为原料的生物炼制转型,逐渐减少对日益枯竭的石油资源的依赖。目前利用葡萄糖为底物占生物转化法生产2,3-丁二醇的大多数,但由于粮食危机的存在,利用丰富而且廉价的可再生资源木质纤维类物质为原料发酵生产2,3-丁二醇成为必然趋势。
木质纤维素原料,来源广泛,且再生迅速。玉米芯作为一种木质纤维素原料,在我国有着极其广泛的资源。现阶段,我国大部分工业生产的木糖是由玉米芯水解液提取得到,在木糖过程中产生了一种纤维素含量丰富的废弃物——玉米芯残渣。玉米芯残渣具有纤维素含量高,结构疏松的特点,是一种理想的木质纤维素原料。因此利用玉米芯残渣生产高值平台产物2,3-丁二醇,以草代粮,不仅解决了与人争粮的问题,同时实现了一种工业副产物的高值利用,顺应了环境友好和可持续发展的潮流。
发明内容
本发明的目的是以木糖工业废弃物玉米芯残渣为主要原料,利用生物工程技术生产高值平台化合物2,3-丁二醇,实现资源的综合利用。
鉴于纤维素酶解过程当中存在的葡萄糖阻遏,工业上对纤维素酶解液的浓缩存在能耗过大的问题,因此本发明选择同步糖化发酵方式,避免了纤维素酶解后的浓缩过程。本发明对利用玉米芯残渣进行同步糖化发酵的发酵条件进行了探索,并对各种添加因子进行了响应面优化,最终得到了一种利用玉米芯提取木糖后残渣同步糖化发酵生产2,3-丁二醇的方法。
具体步骤如下:
第一步、将玉米芯残渣原材料烘干粉碎后,按照玉米芯残渣与NaOH溶液重量体积比1∶8-1∶12混合,在60-80℃条件下预处理1-3h,后将残渣洗涤至中性,烘干粉碎后过40目筛备用。所述NaOH溶液浓度为1%-2%。
第二步、将处理后的残渣加入发酵液中,残渣与发酵液的重量体积比为1∶8-1∶15,然后在115℃下灭菌20min。
发酵液组成如下:
尿素1.4-2.2g/L,乳酸1-3g/L,柠檬酸钠0.1-1g/L,EDTA0.002-0.006g/L,磷酸氢二钾1-5g/L,磷酸二氢钾4-20g/L,硫酸镁0.05-0.5g/L,硫酸钙0.03-0.06g/L,硫酸亚铁0.1-0.15g/L,氯化钴0.002-0.006g/L,硫酸铜0.0025-0.0075g/L,硫酸锌0.05-0.15g/L,硫酸锰0.025-0.05g/L。
第四步、接入2%-10%的肺炎克雷伯氏菌(Klebsiella pneumoniae)CICC10011、产酸克雷伯氏菌(Klebsiella oxytoca)CICC21518、多粘类芽孢杆菌(Bacillus polymyxa)CICC10010、阴沟肠杆菌(Bacillus cloacae)CICC10014菌液中的一种或上述菌种的混合菌液,同时向发酵液中加入纤维素酶,所述发酵液中纤维素酶的添加量为:每克玉米芯残渣加入300U-500U纤维素酶。于35-42℃同步糖化发酵36-84h后2,3-丁二醇的产量为玉米芯残渣干重的19.59%-36.13%(2,3-丁二醇/玉米芯残渣干重,W/W)。
所述第四步中优选接入2%-10%的肺炎克雷伯氏菌(Klebsiella pneumoniae)CICC10011、产酸克雷伯氏菌(Klebsiella oxytoca)CICC21518、多粘类芽孢杆菌(Bacilluspolymyxa)CICC10010和阴沟肠杆菌(Bacillus cloacae)CICC10014的混合菌液。
所述混合菌液中,肺炎克雷伯氏菌(Klebsiella pneumoniae)CICC10011优选体积百分比为25-30%,产酸克雷伯氏菌(Klebsiella oxytoca)CICC21518优选体积百分比为20-30%,多粘类芽孢杆菌(Bacillus polymyxa)CICC10010优选体积百分比为10-20%,阴沟肠杆菌(Bacillus cloacae)CICC10014体积百分比为25-35%。
本发明所产生的技术效果:本发明利用玉米芯生产木糖的废料——玉米芯残渣生产2,3-丁二醇,实现了一种工业废料的高值利用,即使原料利用不完全,废液中固液分离容易,剩余残渣可循环利用。本发明以草代粮、变废为宝,减小环境污染的同时降低了2,3-丁二醇的生产成本;而且利用优选的菌种采用科学筛选的添加比例进行发酵,从而使发酵工艺效率更好,并实现更加良好的糖化发酵,使发酵产率有了较大提高。
具体实施方式
实施例1
将玉米芯生产木糖工业的废弃物玉米芯残渣粉碎后按固液比1∶8(纤维素原料干重∶NaOH溶液,W∶V)加入至1.5%NaOH溶液当中,80℃条件下预处理3h,处理结束后将残渣洗涤至中性,烘干后粉碎,测得预处理后的残渣含纤维素88.92%、木质素3.35%。影响纤维素水解的木质素含量明显降低。
将处理后及未处理的玉米芯残渣按照纤维素比发酵液1∶8(W/V)的比例加入至含有1.61g/L尿素的发酵液当中,装液量50mL/250mL,初始pH 5.0,灭菌后按每克玉米芯残渣400U的比例加入纤维素酶,接入10%肺炎克雷伯氏菌(Klebsiellapneumoniae)CICC10011菌液,35℃,150r/min摇瓶发酵36h,发酵液中2,3-丁二醇与乙偶姻浓度和分别为8.525g/L(未处理)和28.482g/L(处理),经碱法预处理的玉米芯残渣较未处理残渣单位时间内产量提高3.34倍。
发酵液组成为:尿素1.6g/L,乳酸2g/L,柠檬酸钠0.3g/L,EDTA0.003g/L,磷酸氢二钾3g/L,磷酸二氢钾8g/L,硫酸镁0.2g/L,硫酸钙0.04g/L,硫酸亚铁0.1g/L,氯化钴0.004g/L,硫酸铜0.0045g/L,硫酸锌0.1g/L,硫酸锰0.04g/L。
实施例2
将玉米芯生产木糖工业的废弃物玉米芯残渣粉碎后按固液比1∶10(纤维素原料干重∶NaOH溶液,W∶V)加入至1.5%NaOH溶液当中,80℃条件下预处理3h,处理结束后将残渣洗涤至中性,烘干后粉碎。
将处理后的玉米芯残渣按固液比1∶10(纤维素原料干重∶发酵液,W∶V)加入至含有尿素1.61g/L,乳酸2g/L,柠檬酸钠1g/L,EDTA0.006g/L,磷酸氢二钾5g/L,磷酸二氢钾20g/L,硫酸镁0.25g/L,硫酸钙0.05g/L,硫酸亚铁0.15g/L,氯化钴0.005g/L,硫酸铜0.0075g/L,硫酸锌0.15g/L,硫酸锰0.05g/L的发酵液中,装液量50mL/250mL,初始pH 5.0,灭菌后按每克玉米芯残渣400U的比例加入纤维素酶,接入5%肺炎克雷伯氏菌(Klebsiella pneumoniae)CICC10011菌液,35℃,150r/min摇瓶发酵36h,发酵液中2,3-丁二醇浓度为23.484g/L,2,3-丁二醇与乙偶姻浓度和为25.041g/L,占玉米芯残渣干重的25.04%。

将玉米芯生产木糖工业的废弃物玉米芯残渣粉碎后按固液比1∶12(纤维素原料干重∶NaOH溶液,W∶V)加入至1.5%NaOH溶液当中,80℃条件下预处理3h,处理结束后将残渣洗涤至中性,烘干后粉碎。
将处理后的玉米芯残渣按固液比1∶12(纤维素原料干重∶发酵液,W∶V)加入至含有尿素1.61g/L,乳酸2g/L,柠檬酸钠1g/L,EDTA0.006g/L,磷酸氢二钾5g/L,磷酸二氢钾20g/L,硫酸镁0.25g/L,硫酸钙0.05g/L,硫酸亚铁0.15g/L,氯化钴0.005g/L,硫酸铜0.0075g/L,硫酸锌0.15g/L,硫酸锰0.05g/L的发酵液中,装液量50mL/250mL,初始pH 5.0,灭菌后按每克玉米芯残渣400U的比例加入纤维素酶,接入5%产酸克雷伯氏菌(Klebsiella oxytoca)CICC21913菌液,35℃,150r/min摇瓶发酵36h,发酵液中2,3-丁二醇浓度为17.618g/L,2,3-丁二醇与乙偶姻浓度和为18.548g/L,占玉米芯残渣干重的22.26%。

实施例5
将玉米芯生产木糖工业的废弃物玉米芯残渣粉碎后按固液比1∶15(纤维素原料干重∶NaOH溶液,W∶V)加入至1.5%NaOH溶液当中,80℃条件下预处理3h,处理结束后将残渣洗涤至中性,烘干后粉碎。
将处理后的玉米芯残渣按固液比1∶15(纤维素原料干重∶发酵液,W∶V)加入至含有尿素1.61g/L,乳酸2g/L,柠檬酸钠1g/L,EDTA0.006g/L,磷酸氢二钾5g/L,磷酸二氢钾20g/L,硫酸镁0.25g/L,硫酸钙0.05g/L,硫酸亚铁0.15g/L,氯化钴0.005g/L,硫酸铜0.0075g/L,硫酸锌0.15g/L,硫酸锰0.05g/L的发酵液中,装液量50mL/250mL,初始pH 5.0,灭菌后按每克玉米芯残渣400U的比例加入纤维素酶,接入5%阴沟肠杆菌(Bacillus cloacae)CICC10014菌液,40℃,150r/min摇瓶发酵48h,发酵液中2,3-丁二醇浓度为15.295g/L,2,3-丁二醇与乙偶姻浓度和为16.259g/L,占玉米芯残渣干重的24.39%。
实施例6
将玉米芯生产木糖工业的废弃物玉米芯残渣粉碎后按固液比1∶10(纤维素原料干重∶NaOH溶液,W∶V)加入至1.5%NaOH溶液当中,80℃条件下预处理3h,处理结束后将残渣洗涤至中性,烘干后粉碎。
将处理后的玉米芯残渣按固液比1∶10(纤维素原料干重∶发酵液,W∶V)加入至含有尿素1.61g/L,乳酸2g/L,柠檬酸钠0.5g/L,EDTA0.004g/L,磷酸氢二钾1g/L,磷酸二氢钾4g/L,硫酸镁0.25g/L,硫酸钙0.034g/L,硫酸亚铁0.136g/L,氯化钴0.0044g/L,硫酸铜0.005g/L,硫酸锌0.096g/L,硫酸锰0.0275g/L的发酵液中,于5L发酵罐中发酵,装液量3L,按每克玉米芯残渣400U的比例加入纤维素酶,接入2%肺炎克雷伯氏菌(Klebsiella pneumoniae)CICC10011菌液,发酵过程中控制pH5.0,搅拌桨转速400r/min,通气量1.4vvm。38℃发酵84小时后,发酵液中2,3-丁二醇浓度为31.857g/L,2,3-丁二醇与乙偶姻浓度和为33.754g/L,占玉米芯残渣干重的33.75%。
实施例7
将玉米芯生产木糖工业的废弃物玉米芯残渣粉碎后按固液比1∶10(纤维素原料干重∶NaOH溶液,W∶V)加入至1.5%NaOH溶液当中,80℃条件下预处理3h,处理结束后将残渣洗涤至中性,烘干后粉碎。
将处理后的玉米芯残渣按固液比1∶10(纤维素原料干重∶发酵液,W∶V)加入至含有尿素1.61g/L,乳酸2g/L,柠檬酸钠0.5g/L,EDTA0.004g/L,磷酸氢二钾1g/L,磷酸二氢钾4g/L,硫酸镁0.25g/L,硫酸钙0.034g/L,硫酸亚铁0.136g/L,氯化钴0.0044g/L,硫酸铜0.005g/L,硫酸锌0.096g/L,硫酸锰0.0275g/L的发酵液中,于5L发酵罐中发酵,装液量3L,按每克玉米芯残渣400U的比例加入纤维素酶,接入6%的混合菌液,所述混合菌液中含有体积百分比为26%的肺炎克雷伯氏菌(Klebsiellapneumoniae)CICC10011、体积百分比为24%的产酸克雷伯氏菌(Klebsiella oxytoca)CICC21518、体积百分比为23%的多粘类芽孢杆菌(Bacillus polymyxa)CICC10010、体积百分比为27%的阴沟肠杆菌(Bacillus cloacae)CICC10014。发酵过程中控制pH5.0,搅拌桨转速400r/min,通气量1.4vvm。42℃发酵72小时后,发酵液中2,3-丁二醇浓度为34.879g/L,2,3-丁二醇与乙偶姻浓度和为36.128g/L,占玉米芯残渣干重的36.13%。
Claims (2)
1.一种利用玉米芯残渣同步糖化发酵生产2,3-丁二醇的方法,包括如下步骤:
第一步、将含有纤维素的玉米芯残渣原材料粉碎,烘干备用;
第二步、粉碎后玉米芯残渣与NaOH溶液按照重量体积比1:8-1:12混合,在60-80℃条件下预处理1-3h,后将残渣洗涤至中性,烘干粉碎后过40目筛备用,所述NaOH溶液浓度为1%-2%;
第三步、将烘干粉碎后残渣加入发酵液中,烘干粉碎后残渣与发酵液的重量体积比为1:8-1:15,然后在115℃下灭菌20min;
第四步、接入2%-10%的肺炎克雷伯氏菌(Klebsiella pneumoniae)CICC10011、产酸克雷伯氏菌(Klebsiella oxytoca)CICC21518、多粘类芽孢杆菌(Bacillus polymyxa)CICC10010、阴沟肠杆菌(Bacillus cloacae)CICCl0014上述菌种的混合液,同时向发酵液中加入纤维素酶,所述发酵液中纤维素酶的添加量为:每克玉米芯残渣加入300U-500U纤维素酶,于35-42℃同步糖化发酵36-84h;
所述第四步中接入2%-10%的混合菌液,所述混合菌液中肺炎克雷伯氏菌(Klebsiellapneumoniae)CICC10011体积百分比为25-30%,产酸克雷伯氏菌(Klebsiella oxytoca)CICC21518体积百分比为20-30%,多粘类芽孢杆菌(Bacillus polymyxa)CICC10010体积百分比为10-20%,阴沟肠杆菌(Bacillus cloacae)CICC10014体积百分比为25-35%;
所述发酵液组成为:尿素1.4-2.2g/L,乳酸1-3g/L,柠檬酸钠0.1-1g/L,EDTA0.002-0.006g/L,磷酸氢二钾1-5g/L,磷酸二氢钾4-20g/L,硫酸镁0.05-0.5g/L,硫酸钙0.03-0.06g/L,硫酸亚铁0.1-0.15g/L,氯化钴0.002-0.006g/L,硫酸铜0.0025-0.0075g/L,硫酸锌0.05-0.15g/L,硫酸锰0.025-0.05g/L。
2.根据权利要求1所述一种利用玉米芯残渣同步糖化发酵生产2,3-丁二醇的方法,其特征在于,所述发酵液中纤维素酶的添加量为:每克玉米芯残渣加入400U纤维素酶。
