唾液酸(SiAc)是一个衍生自神经氨酸(NeuAc)的带九碳单糖家族。神经氨酸是通常在人体内形成的唯一的唾液酸。在其他脊椎动物中,也存在例如N-羟乙酰神经氨酸(NeuGc)。
在脊椎动物中SiAc对于主要细胞功能是必不可少的。作为神经节苷脂的功能和结构组分,它们在哺乳动物的所有组织中合成。然而,承受高速率凋亡和更新的快速生长的细胞和组织可能需要额外的(例如通过食物提供的)NeuAc。

生产唾液酸的食品级细菌的“部分”包括细菌和/或含有唾液酸的细菌培养物的任何部分。术语生产唾液酸的食品级细菌“部分”还包括该食品级细菌生长于其中的培养基或其部分,因为此培养基自动包含细菌SiAc。另外,术语生产唾液酸的食品级细菌“部分”还包括从细菌培养物中纯化SiAc时获得的任何含SiAc的部分。
在本发明优选实施方案中自然产生的生产唾液酸的食品级细菌在食物中是存活的。这具有优势,即当该食物被消耗后,细菌能够在体内继续生产唾液酸。另外,如果细菌在体内保持存活,它们会繁殖并因此细菌提供给身体的唾液酸数量将大量增加。
对于灭菌产品,如果细菌以灭活形式存在也可以是优选的、或如果该产品富含有纯细菌SiAc或富含不包含任何活细菌的SiAc生产细菌培养物的部分也可以是优选的。
特别是,如果食品级细菌在产品中是存活的,该细菌或至少其一部分将以任意量有效。如果该细菌活着到达肠道,单个细菌足以通过定殖和繁殖实现有用的效果。
然而,对于本发明的食物,如果使用足以在食物产品中获得唾液酸含量增加0.05-2干重%,优选0.4-1.5干重%,更加优选0.6-1干重%的量的细菌或至少其一部分,则这通常是优选的。
本发明的食物可以是营养组合物、营养药、饮料、食物添加剂或药物。食物添加剂或药物可以是例如片剂、胶囊、锭剂或液体形式。
该食物优先选自基于奶的食物(特别是奶、乳清、酸奶、乳酪、发酵产品)、婴儿制剂、固态幼儿食品、后续制剂、幼儿饮料、果汁(至少部分可溶饮料以粉末形式混合,如麦芽饮品、巧克力饮品、卡普西诺、咖啡饮品)、巧克力、谷物产品、糖果、饼干、胶质。
奶可以是取自动物或植物来源的任意奶,并优选牛奶、人奶、绵羊奶、山羊奶、马奶、骆驼奶或豆浆。
也可以使用源自奶的蛋白质部分或初乳代替奶。
该食物还可以包含保护性亲水胶体(如胶、蛋白质、改性淀粉)、粘合剂、成膜剂、包封剂/材料、壁/壳材料、基质化合物、包被物、乳化剂、表面活性剂、助溶剂(油类、脂肪类、蜡类、卵磷脂类等)、吸附剂、载体、装填物、共化合物(co-compound)、分散剂、润湿剂、加工助剂(溶剂)、助流剂、掩味剂、增重剂、成凝胶剂、凝胶形成剂、抗氧化剂和抗菌剂。它们还可能包含传统药物添加剂和佐剂、赋形剂和稀释剂,包括但不限于水、任意来源的胶质、植物胶、木质素磺酸盐、滑石、糖类、淀粉、阿拉伯树胶、植物油、聚亚烷基二醇、增香剂、防腐剂、稳定剂、乳化剂、缓冲剂、滑润剂、着色剂、润湿剂、装填物等等。另外,它们可能包含适于口服或肠内施用的有机或无机载体材料, 还有依照政府机构(如USRDA)建议的维生素、矿物微量元素和其他微量营养素。
本发明的食物可能包含蛋白质来源、碳水化合物来源和/或脂质来源。
可以使用任何合适的食用蛋白质例如动物蛋白质(如奶类蛋白质、肉类蛋白质和蛋类蛋白质)、植物蛋白质(如大豆蛋白、小麦蛋白、稻蛋白和豌豆蛋白)、游离氨基酸的混合物或其组合。特别优选奶类蛋白质如酪蛋白和乳清和大豆蛋白质。
如果该食物包括脂肪来源,该脂肪来源更优选提供该配方5%到40%的能量,例如20%到30%的能量。可以加入DHA。可以使用菜籽油、玉米油和高油酸葵花籽油的混和物获得适当的脂肪谱。
碳水化合物来源可以更优选提供该组合物的40%到80%间的能量。可以使用任何适当的碳水化合物,例如蔗糖、乳糖、葡萄糖、果糖、淀粉糖浆干粉、麦芽糊精和其混合物。
食品级细菌优先选自乳杆菌。
发明者发现如果乳杆菌生产N-乙酰神经氨糖酸裂合酶和/或N-乙酰神经氨糖酸醛缩酶,则该乳杆菌产生特别大量的SiAc。
特别优选的能用于本发明目的的乳杆菌种类是清酒乳杆菌(Lactobacillus sakei)、植物乳杆菌(Lactobacillus plantarum)和/或唾液乳杆菌(Lactobacillus salivarius)。用选自清酒乳杆菌NCC 121、清酒乳杆菌NCC 2935、清酒乳杆菌NCC 2934、清酒乳杆菌NCC 166、清酒乳杆菌NCC 170、清酒乳杆菌NCC 1393、清酒乳杆菌NCC 1428、清酒乳杆菌NCC 1511、清酒乳杆菌NCC 2937、植物乳杆菌NCC 2936、植物乳杆菌NCC 252的细菌或其混合物能获得特别好的结果。
任何细菌部分可以用于本发明目的。特别优选的部分是通过在生长培养基中培养细胞、收集细胞、在酸性条件下水解细胞和收集含有唾液酸的上清液而获得的部分。
例如,可以使用下面方法之一:
方法1:在API培养基(蛋白胨1%、酵母提取物0.5%、聚山梨醇酯800.1%、柠檬酸铵0.2%、乙酸钠0.5%、硫酸镁0.01%、硫酸锰0.005%和磷酸二钾0.2%、葡萄糖1%)中于37℃生长16小时后,收集细胞并在水中洗涤一次。通过在2M乙酸中80℃水解3小时释放唾液酸。冻干离心后得到的上清液。
方法2:细胞在API培养基(见上面)中37℃下生长16小时。在1升发酵培养基中加入250克三氯酸和室温下搅拌1小时。在离心细胞后,加入1升丙酮于上清液中,并4℃孵育过夜和再次离心。用50%丙酮洗涤沉淀并以水重悬。调节pH到7,再次离心。用水透析抽提物并冻干。
方法3:在API培养基(见上面)中37℃下生长16小时后,以冷的磷酸盐缓冲液(137mM NaCl、2.7mM KCl、8.1mM Na2HPO4、1.5mMKH2PO4、pH7.4)洗涤细菌细胞两次和以0.1M乙酸吡啶(pH5)洗涤一次。在0.1V预热的乙酸嘧啶(0.1M,pH5)中重悬细菌并在37℃孵育1小时。冻干离心后得到的上清液。
为本发明目的可以使用任何唾液酸。然而,如果具有下述结构式的唾液酸则是优选的:
R1=H、乙酰基、乳酰基、甲基、硫酸酯、磷酸酯、脱水唾液酸、岩藻糖、葡萄糖或半乳糖
R2=N-乙酰基、N-乙醇酰基、氨基、羟基、N-乙醇酰基-O-乙酰基或N-乙醇酰基-O-甲基
R3=H、半乳糖、N-乙酰基葡糖胺、N-乙酰基半乳糖胺、唾液酸或N-羟乙酰神经氨酸
R1可以选自氢、乙酰基、乳酰基、甲基、硫酸酯、磷酸酯、脱水唾液酸、岩藻糖、葡萄糖和/或半乳糖。
R2可以选自N-乙酰基、N-乙醇酰基、氨基、羟基、N-乙醇酰基-O-乙酰基和/或N-乙醇酰基-O-甲基。
R3可以选自氢、半乳糖、N-乙酰基葡糖胺、N-乙酰基半乳糖胺、唾液酸和/或N-羟乙酰神经氨酸。
位置R1的基团可以相同或互不相同。相似地,位置R2的基团可以相同或互不相同,位置R3的基团也是可以相同或互不相同。
在本发明的特别优选实施方案中,该唾液酸是N-乙酰基神经氨酸,具有R1=氢、R2=N-乙酰基和R3=氢。
本发明还涉及含有天然产生生产唾液酸的食品级细菌或其部分的唾液酸强化食物。该唾液酸强化食物可具有如上文本发明用途中所述特征一样的特征。
如果加入唾液酸或生产唾液酸的细菌到食物中,食物就被唾液酸强化。例如,通过强化,食物中可能天然存在的唾液酸含量增加至少10重量%,优选至少50重量%,更优选至少100重量%。
本发明的食物可用于提供营养给受试者,例如抗衡生产唾液酸的内源性缺乏。特别是在正在生长的生物中,对唾液酸的需求往往超过身体自身内源性生产的唾液酸。因此添加唾液酸的食物产品将帮助支持受试者发育。然而,缺乏内源性唾液酸生产的也可能发生于不再生长的受试者。
因此本发明还涉及用于提供营养给受试者的本发明的食物。该食物可能用于抗衡内源性唾液酸生产的缺乏。
因此本发明的主旨是针对人类或动物,特别是伴侣动物、宠物和/或家畜。本发明的主旨一般不局限于任何特别年龄群体。该食物可以施用于孕期和哺乳期的母亲来治疗婴儿。它也可以施用于婴儿、儿童、青少年、成年人或老年受试者。然而优选提供食物给婴儿。
Wang,B和Brand-Miller,J.,(2003)Eur.J.Clin.Nutr.Nov;57(11):1351-69总结了唾液酸在人类营养中的作用和潜力。
本发明者最近发现施用本发明的食物给受试者导致了(特别是年长受试者)脑内上升的唾液酸化。例如通过富含增加的唾液酸化神经节苷脂(唾液酸乳糖苷神经酰胺)的脑部制品可看到这一点。唾液酸化和特别是神经节苷脂在稳定神经元完整性和允许中枢和周围神经系统的神经元可塑性中是重要因子。
人们还发现施用本发明的食物(优选施用给老年人)导致胃肠道(GIT)特别是近端和远端结肠粘膜中唾液酸化升高。这里粘蛋白唾液酸化的修饰影响了粘膜屏障的物理化学特性。此外,人们发现正如通过结肠中神经节苷脂水平上升所见的,糖脂结合的唾液酸化作用得以增强。
人们还观察到施用本发明食物给受试者导致细胞介导的免疫反应有所改善。伴随地,与对照受试者相比,在消耗本发明食物的受试者体中,以凝集素ConA体外刺激脾细胞时白介素2的水平增强。这个效果在婴儿和儿童中比在老年人中更显著。
因此本发明食物和/或从食品级细菌获取的唾液酸可被用于治疗或防治神经变性,特别是成人的神经变性。它还可以被用在(特别是在儿童体内)改善认知表现和/或支持脑部发育。
本发明食物和/或取自食品级细菌的唾液酸的应用还有支持免疫系统,特别是加强免疫和/或改善消化道功能。
能够用本发明食物和/或取自食品级细菌的唾液酸来治疗或防治的具体的临床病理包括例如炎症性肠病、肠易激综合症、神经系统退行性病理如痴呆症或阿尔茨海默氏病、感染后自身破坏性免疫系统疾病和/或GIT神经元退化。
因此,本发明的食物和/或取自食品级细菌的唾液酸能例如促进健康生长和健康变老、支持婴儿和儿童的脑部发育、改善认知功能、预防或抵抗如衰老、疾病或压力引起的认知功能衰退和/或神经退行、支持免疫成熟和稳态、如在老年人中增加唾液酸化用于免疫保护,例如通过提供饮食NeuAc、用于降低低度炎症、用于改善肠道屏障和/或用于抑制系统炎症。
本发明的添加唾液酸的食物能够贡献于向新生儿提供最适SiAc(例如在母乳或婴儿制剂中)、最佳中枢神经系统发育、恢复根据情况的SiAc缺乏,如孕期、哺乳期间和/或营养不良情况中的SiAc缺乏。
本领域的那些技术人员应当理解,他们可以自由组合在此所述的本发明的所有特征,而不背离公开的本发明的范围。特别是,所述的用于本发明的使用的特征可以被应用到本发明的食物,反之亦然。
本发明的其他优势和特征在下列实施例和附图中得以展现。
图1显示了1,2-二氨基-4,5-亚甲基二氧苯二氢氯化物(DMB)层析图:植物乳杆菌NCC2936(A);加入NeuAC和NeuGc标准品的NCC2936(B)和NeuAC及NeuGc标准品(C)。
实施例
方法
细菌菌株和其生产
细菌菌株自雀巢菌种保藏中心(Nestle Culture Collection,NCC)获得并生长于API培养基(蛋白胨1%、酵母提取物0.5%、聚山梨醇酯800.1%、柠檬酸铵0.2%、乙酸钠0.5%、硫酸镁0.01%、硫酸锰0.005%和磷酸二钾0.2%、葡萄糖1%)。在30℃生长16小时后,收集细菌并冻干。
检测SiAc
以改良的Jourdian等((1971)J Biol Chem 246:430-435)的方法检测SiAc。简单的讲,10微升0.04M高碘酸与50微升样品或50微升NeuAc标准品(0、20、40、60和100微克/毫升)混合并在冰浴中孵育35分钟。加入125微升新鲜混合物(由在6毫升28%盐酸+1毫升6%间苯二酚+3毫升水中的0.04毫克硫酸铜组成)并在4℃孵育5分钟。将样品煮沸15分钟并冷却。加入125微升95%的叔丁醇并在37℃孵育3分钟来稳定颜色。通过和不同浓度的标准品比较样品的蓝色强度来做视觉评分。
SiAc定量
1,2-二氨基-4,5-亚甲基二氧苯二氢氯化物(DMB)法:溶解细菌样品到水中来得到预期总唾液酸浓度为大约2微克/毫升。通过加入200微升蚁酸(1.0M)并在80℃加热2小时来水解200微升的此溶液等份样品以释放所有结合的唾液酸。然后以1,2-二氨基-4,5-亚甲基二氧苯二氢氯化物(DMB)(特异于α-酮酸的荧光标记)衍生唾液酸。通过向200微升等份样品的水解样品中加入200微升DMB溶液(7.0mM,在1.4M乙酸中,含0.75M 2-巯基乙醇和18mM亚硫酸氢钠)进行衍生化,然后在80℃加热混合物50分钟。注射衍生化样品(5微升)到反相HPLC柱(Zorbax SB-Aq,3.5微米,4.6x50毫米)上并使用以2.0毫升/分钟流动的水/甲醇/乙酸(75/25/0.05(v/v/v))流动相洗脱。使用荧光检测((λ ex=373nm,λem=448nm)监控柱子洗脱液。通过绘制来自已知浓度唾液酸的校准曲线和比较样品的峰面积与标准品的峰面积来进行定量。
鉴定NeuAc(GC-MS分析)
通过用甲醇中的1M HCl(25滴)在80℃处理15小时,然后以吡啶(5滴)和乙酸酐(5滴)在甲醇中(20滴)室温下再N-乙酰化1小时,从称重的样品制备甲基糖苷。然后在80℃通过用Tri-Sil(10滴,Pierce)处理15分钟以对样品进行完全O-三甲基唾液酸化。如前所述(Merkle,R.K.和I.Poppe(1994)Methods Enzymol.230:1-15;York,W.A.等(1986)Methods Enzymol 118:3-40)进行这些步骤。使用DB-1柱(30米×0.25毫米ID),在接有5970MSD的HP 5890GC上进行TMS甲基糖苷的GC/MS分析。
结果
检测SiAc生产细菌
用周期法筛选来自雀巢菌种保藏中心的用于SiAc生产的细菌。以下菌株经鉴定为SiAc生产特别有效,并按照布达佩斯条约进行了保藏。
于2008年6月25日向法国巴黎法国巴斯德研究所国家微生物菌种保藏中心(COLLECTION NATIONALE DE CULTURES DE MICROORGANISMES,CNCM)进行了下述菌种的保藏:
清酒乳杆菌NCC 121 (保藏号CNCM I-4020)
植物乳杆菌NCC 252 (保藏号CNCM I-4021)
清酒乳杆菌NCC 1393 (保藏号CNCM I-4022)
清酒乳杆菌NCC 1428 (保藏号CNCM I-4023)
清酒乳杆菌NCC 1511 (保藏号CNCM I-4024)
清酒乳杆菌NCC 2934 (保藏号CNCM I-4025)
植物乳杆菌NCC 2936 (保藏号CNCM I-4026)。
于2008年8月7日向法国巴黎法国巴斯德研究所国家微生物菌种保藏中心进行了下述菌种的保藏:
清酒乳杆菌NCC 2935 (保藏号CNCM I-4064)
清酒乳杆菌NCC 2937 (保藏号CNCM I-4065)
清酒乳杆菌NCC 166 (保藏号CNCM I-4066)
清酒乳杆菌NCC 170 (保藏号CNCM I-4067)。
鉴定SiAc类型
如材料与方法中所述制备SiAc生产菌株的干粉。使用两种不同方法(DMB和GC-MS)分析同一粉末。
a)图1显示了用DMB法产生的分析菌株(NCC2936)的典型层析图。用此方法我们鉴定到几乎与N-乙酰神经氨酸(NeuAc)运行在同一保留时间的峰,但显示轻微的保留时间位移。未检测到与N-羟乙酰神经氨酸(NeuGc)接近的可见峰。如果样品含有可结合于柱子固定相的某些成分,可能发生这样的位移。这样的结合降低了分析物和固定相之间的相互作用,并因此位移了保留时间。可选的是,SiAc可以用另外的化学基团修饰,并因此显示出改变的保留时间。
为了测试是否所述峰因为样本中的干扰化合物而位移,我们进行了“尖峰形成”实验。向一样品制备物中加入确定量的NeuAc标准品并再次于HPLC上运行样品。可能由于稀释效应,在添加(spiked)标准品的样品中观察到的峰保留时间更加接近于标准品NeuAc峰的保留时间。在添NeuAc的样品中只检测到一个峰,说明加入样品中的标准品和原样品峰运行在同样的位置。因此,我们认为这个峰最可能代表了NeuAc, 并且最初观察的保留时间位移可能由于样品中的干扰化合物而并非由于NeuAc的修饰。
b)在GC-MS分析中得到了细菌生产NeuAc的其他证据。用这个方法,分析了细菌样品的糖基组合物(表1)。结果是,细菌样品中存在的SiAc被证实是NeuAc。

定量SiAc
以DMB法和周期分析法进行细菌样品中SiAc的定量。DMB是定量方法,而周期法用于样品的半定量分析。不可能使用周期分析做精确定量,因为高背景色干扰结果的分光光度计读取。表2显示了对于同一批细菌进行定量的结果。

Claims (15)
1.天然产生的生产唾液酸之食品级细菌或至少其包含唾液酸的一部分的用途,所述用途用唾液酸强化食物。
2.根据权利要求1的用途,其中天然产生的生产唾液酸的食品级细菌是存活的,以使得在食物被消耗后其在体内生产唾液酸。
3.根据权利要求1或2的用途,其中所述细菌或所述至少其一部分以足以在食物产品中获得唾液酸含量增加0.05-2干重%,优选0.4-1.5干重%,更优选0.6-1干重%的量使用。
4.根据权利要求1-3的用途,其中所述细菌选自乳杆菌,特别是生产N-乙酰神经氨糖酸裂合酶和/或N-乙酰神经氨糖酸醛缩酶的乳杆菌,更优选清酒乳杆菌(Lactobacillus sakei)、植物乳杆菌(Lactobacillus plantarum)和/或唾液乳杆菌(Lactobacillussalivarius)。
5.根据权利要求1-4的用途,其中所述细菌选自清酒乳杆菌NCC 121、清酒乳杆菌NCC 2935、清酒乳杆菌NCC 2934、清酒乳杆菌NCC 166、清酒乳杆菌NCC 170、清酒乳杆菌NCC 1393、清酒乳杆菌NCC 1428、清酒乳杆菌NCC 1511、清酒乳杆菌NCC 2937、植物乳杆菌NCC 2936、植物乳杆菌NCC 252或其混合物。
6.根据权利要求1-5的用途,其中所述细菌部分通过包括下列步骤的方法获得:在生长培养基中培养细胞、收集细胞、在酸性条件下水解细胞和收集含有唾液酸的上清液。
8.唾液酸强化的食物,包含如权利要求1-7所述的天然产生的生产唾液酸的食品级细菌或其部分。
9.根据权利要求8的唾液酸强化的食品用于提供营养给受试者。
10.根据权利要求8的唾液酸强化的食品用于抗衡内源性唾液酸生产的缺乏。
11.根据权利要求8的唾液酸强化的食品用于治疗或预防神经变性,特别是成年人的神经变性。
12.根据权利要求8的唾液酸强化的食品用于支持免疫系统,特别是加强免疫和/或改善肠道功能。
13.根据权利要求8的唾液酸强化的食品用于改善认知表现和/或支持脑部发育,特别是支持儿童的脑部发育。
14.生产唾液酸的食品级细菌选自清酒乳杆菌NCC 121、清酒乳杆菌NCC 2935、清酒乳杆菌NCC 2934、清酒乳杆菌NCC 166、清酒乳杆菌NCC 170、清酒乳杆菌NCC 1393、清酒乳杆菌NCC1428、清酒乳杆菌NCC 1511、清酒乳杆菌NCC 2937、植物乳杆菌NCC 2936和植物乳杆菌NCC 252。
