Abstract
本发明提供的乳化或分散制造方法中,使含利用水溶性有机溶剂从然的动植物中提取的天然成分的水溶性有机溶剂溶液和水性溶液分别通过最窄部分的截面积为1μm2~1mm2的微流路后,相向冲撞,由此使其混合。优选在混合后除去水溶性有机溶剂。另外,本发明还提供含有由此得到的乳化物和分散物的食品、皮肤外用剂和医药品。
Description
技术领域
本发明涉及乳化物或分散物的制造方法,以及含有利用该制造方法得到的乳化物或分散物的食品、皮肤外用剂和医药品。
背景技术
以往,一直利用醇等水溶性有机溶剂从天然的动植物提取有效成分,并将其作为食品添加物在食品、饮料中使用,或作为功能性化妆品在化妆品中添加。近年来,随着健康热潮的兴盛,各种源于天然素材的提取越来越流行。另外,从至今为止作为废弃物的物质,例如,橡胶叶、柑橘类残渣、洋葱的外皮等中利用醇提取有效成分的做法也越来越流行。
像这样,利用醇等水溶性有机溶剂提取的天然有效成分,一般来说之后要经过从提取液中除去有机溶剂、浓缩、干燥的工序,以天然有效成分的粉末、油或蜡的形式被提取出,再作为食品、饮料的原料来使用。但是,这些有效成分,多数情况下为不易溶于水的物质,因此为了在水性食品、水性饮料以及触感优异的水性化妆品中使用这些有效成分,需要使有效成分溶解于其他溶剂、油脂,或使有效成分本身在高温溶融后再提供于乳化、分散的工序,从而必然导致工序上的高成本。特别是为了有效地吸收有效成分时,需要将有效成分的油滴的尺寸细微化至1μm以下,因此需要大量乳化能量。另外,一旦使有效成分干燥,在使其再溶解或溶融的过程中,有时有效成分会变质,因此,常常还会存在实效成分量的减少、变质成分的副作用这些问题。
因此,本发明人尝试将提取液直接进行乳化分散的做法。即,使用乳化剂,利用通常的乳化法,将含有效成分的水性有机溶剂在水相中进行乳化分散的做法。在此所说的通常的乳化法,是指使用机械力的方法,即通过从外部给与强剪切力使油滴分裂的方法。作为机械力最普遍使用的是高速、高剪切搅拌机。作为这样的搅拌机,有称为均质机(Homomixer)、分散机(dispermixer)和高速均质机(Ultramixer)的市售品。另外,作为对施加高剪切力进行细微化有用的其他装置,有高压匀浆仪(Homogenizer),市售有各种装置。另外,作为能量效率比较好的分散装置,超声波匀浆仪也常在实验装置级别被使用。
即使是使用乳化剂,利用高剪切搅拌机、高压匀浆仪、超声波匀浆仪等乳化装置,以通常的乳化法将含有有效成分的水性有机溶剂在水相中乳化分散,进行含有大量水溶性有机溶剂的体系的乳化的情况也是非常困难的,不能使平均粒径达到1μm以下。另外,粒径分布也会变宽,粗大粒子会在短时间分离出来,因此在食品、化妆品的保存性方面存在问题。将水中不溶的天然有效成分配合于水性食品、水性化妆品时,从有效成分的消化吸收,对于由经时造成的沉降、浮游分离的稳定性,外观上的透明性等观点出发,要求细微的乳化分散的情况很多。一般的说,为了大致满足这些要求,需要将平均粒径设为200nm以下。
进而,作为其他的乳化方法,在欧州专利第1180062号公开了一种利用超微混合机(Micromixer)连续地制造天然物质的微米粒子或纳米粒子的方法。将天然有效成分的粒子形成相、以水作为主成分的水相分别导入微流路,彼此形成层状液再周期性地混合,由此尝试天然有效成分的粒子化。然而,已知其形成的粒子的平均粒径不低于1000nm,离目标的200nm很远。
发明内容
本发明的目的在于,以天然成分的品质劣化极少的状态提供利用水溶性有机溶剂从动植物提取的天然成分的细微乳化物或分散物。
本发明是鉴于上述情况而开展的,提供制造乳化物或分散物的方法,以及含有利用该制造方法得到的乳化物或分散物的食品、皮肤外用剂和医药品。
本发明的第一方式提供一种乳化物或分散物的制造方法,其特征在于,包括使水溶性有机溶剂溶液和水性溶液分别通过最窄部分的截面积为1μm2~1mm2的微流路后,相向冲撞,由此使其混合的步骤,所述水溶性有机溶剂溶液含有利用水溶性有机溶剂从天然的动植物中提取的天然成分。
本发明的第二方式提供一种含有利用上述制造方法得到的乳化物或分散物的食品,其中,上述天然成分为功能性食品成分。
本发明的第三方式提供一种含有利用上述制造方法得到的乳化物或分散物的皮肤外用剂,其中,上述天然成分为皮肤外用剂成分。
本发明的第四方式提供一种含有利用上述制造方法得到的乳化物或分散物的医药品,其中,上述天然成分为医药成分。

具体实施方式
在本发明的乳化物或分散物的制造方法中含有以下步骤:使水溶性有机溶剂溶液和水性溶液分别通过最窄部分的截面积为1μm2~1mm2的微流路后,相向冲撞,由此使其混合的步骤,所述水溶性有机溶剂溶液含有利用水溶性有机溶剂从天然的动植物中提取的天然成分。
在本制造方法中,由于将使用水溶性有机溶剂作为提取液而提取的天然成分以提取后的溶液的形式,即水溶性有机溶剂溶液的形式与水性溶液混合,因此不必使用经干燥工序的方式或干燥后再溶解的方式。因此,可以使因干燥工序的热劣化等天然成分的品质劣化变得极少。另外,由于通过使用微流路将这样的含有天然成分的水溶性有机溶剂溶液作为油相与水性溶液相向冲撞来进行混合,因此可以形成细微的乳化物,或者可以得到细微的分散物。
此外,所谓本发明中的“分散物”,主要指通过乳化或分散或之后的处理得到的固体在分散介质中分散的“固体分散物”以及“固形的分散物”。
另外,在本发明“工序”之类的语言,并非仅指独立的工序,即使在不能与其他工序明确地区别时,只要能实现本工序的所预期的作用,也包括在本用语中。
另外,本发明中的表示数值范围的表述,为包含其前后的数值的表述。
本发明中的天然成分是指利用有机溶剂通过提取操作从动植物中分离的成分。提取是化学上的分离操作法的一种,是指使液体或固体原料与溶剂接触,将原料中所含的可溶于溶剂的成分从不溶于溶剂或难溶性成分中选择性分离的操作。本发明中的提取操作包括在上述一般的提取操作中。
本发明中使用的动植物原料没有特别的限定。主要希望最终用于健康食品、健康饮料时,从气味、味道、健康印象的观点出发,相比于动物原料更优选植物原料。作为植物的部位,可以将叶、茎、皮、果实等所有部位作为原料。另外,藻类、菌丝类、酵母类、菌类也优选适合本发明用的原料。

作为藻类,所有水中的产氧型光合成生物的均为其对象。作为藻类的例,可举出蓝藻、原核绿色藻、灰色藻、红藻、绿枝藻(Prasinophytes)、石莼藻、绿藻、Trebouxiophyceae(トレボキシア藻)、轮藻、隐藻、Chlorarachniophytes(クロララクニオン藻)、裸藻、涡鞭毛藻、黄金色藻、绿色鞭毛藻、真眼点藻、黄绿色藻、褐藻、硅藻、Dictyochophyceae(デイクテイオカ藻)、金藻(Pelagophyceae)、Chrysochomulina(ハプト藻)等。其中该中特别是属于蓝藻纲的螺旋藻、属于绿藻纲的雨生红球藻、属于褐藻纲的海蕴、昆布、裙带菜等为重要的原料。
菌丝类、酵母类、菌类中有各种原料。作为它们的例,可举出姬松茸、酵母、香菇、香蕈、冬虫夏草、纳豆、双歧杆菌、红曲菌(Monascus anka)、银耳、舞茸、松伞蘑、桑黄、山伏茸、荔枝等,但不限于这些。
作为从天然原料中提取的天然成分,有各种各样的物质。作为代表的物质,有动植物所含的脂质成分。
作为脂质成分,有脂肪酸类、甘油酯类、复合脂质类、萜类、甾体类、前列腺素类等。
作为脂质成分中的有效成分的例子,可举出亚油酸、γ-亚麻仁油酸、花生四烯酸、α-亚麻仁油酸、二十碳五烯酸、二十二碳六烯酸、γ-氨基丁酸、硫辛酸等。作为甘油酯类,可举出单酰基甘油酯、二酰基甘油酯、三酰基甘油酯。作为复合脂质类、可举出磷脂酸、磷脂酰胆碱、磷脂酸乙醇胺、磷脂酰丝氨酸、磷脂酰肌醇等磷脂质、鞘氨醇磷酸酯、鞘磷脂等鞘脂质、单半乳糖基甘油酯、葡糖苷鞘氨醇等糖脂质。
作为萜类的有效成分,有沉香醇、香茅醛、香叶烯、苧烯、蒎烯、薄荷醇、桉树脑、樟脑、ロンジホリン、雪松醇、石竹烯等单萜、叶绿醇、松香酸、贝壳杉烯、赤霉素等二萜、鲨烯、达玛烯二醇等三萜、类胡萝卜素为代表的四萜、古塔胶、维生素K2、辅酶Q、维生素K1等多萜。其中、类胡萝卜素为因为抗氧化力而备受关注的素材、作为类胡萝卜素的代表成分、有虾青素、番茄红素、玉米黄素、叶黄素、辣椒红素、岩藻黄素、α-胡萝卜素、β-胡萝卜素等。
作为甾体类的有效成分,可举出鲨烯、胆固醇、麦角甾醇、谷甾醇、去氢胆固醇、胆钙化醇(维生素D3)、25-羟基维生素D3等的甾醇类、睾酮、男性酯酮、孕甾酮、雌酮、雌二醇等性激素类、可的松、地塞米松等肾上腺皮质激素类、洋地黄毒苷元、地高辛、羟基洋地黄毒苷元等强心苷类、薯蓣皂苷元、可的松等甾体皂苷类、胆汁酸、去氧胆酸酸等胆汁酸类。
作为脂质成分以外的一种重要天然成分,可举出多酚。所谓多酚,是指在同一分子内具有多个酚性羟基的植物成分的总称,其构成利用光合成得到的植物的色素、苦味成分,是具有特别优良的抗氧化力的成分。多酚分为黄酮类、绿原酸酸类、没食子酸类、鞣花酸系、木质素类、姜黄素类,香豆素类。在黄酮类中,分为包括染料木苷、灰斑鸻(学名:Pluvialissquatarola)等的异黄酮,包括槲皮素、山奈酚、杨梅素、芦丁等的黄酮醇,包括橙皮素、柚皮苷等的二氢黄酮,包括矢车菊素、飞燕草素的花青素、包括表儿茶酸、表儿茶素、表儿茶素没食子酸酯、茶黄素等的黄烷醇,包括白杨素、芹菜素、木犀草素等的黄酮。多酚中,作为健康食品、美容成分备受注目的素材也很多,可举出高良姜素、漆黄素、查耳酮、葛根素、白藜芦醇等。
作为其他的天然成分,特别是作为在医药领域有用的成分,可举出生物碱类。作为生物碱类、乌头碱、阿托品、麻黄碱、咖啡因、辣椒碱、奎宁、箭毒碱、可卡因、秋水仙碱、东莨菪碱、番木鳖碱、茄碱、紫杉碱、茶碱、多巴胺、烟碱、长春碱、小檗碱、吗啡、石蒜碱等可举出。
另外,作为其他天然成分,可举出从生姜提取的姜烯酚、姜酚,作为芥末或芥子的成分的烯丙基异硫氰酸酯,作为大蒜等的成分的大蒜素、蒜原素、胡蒜素等。
本发明中的水溶性有机溶剂,以含有天然成分的油相的形式,用来与后述水性溶液混合。该水性有机溶剂同时为提取天然成分的提取液的主成分。即,本发明中,天然成分以被以水溶性有机溶剂为主成分的提取液提取了的状态,与水性溶液的混合使用。
所谓本发明中使用的水溶性有机溶剂,是指在水中25℃时的溶解度为10质量%以上的有机溶剂。从得到的乳化物或分散物的稳定性的观点出发,在水中的溶解度优选30质量%以上,更优选50质量%以上。
水溶性有机溶剂在提取时可以单独使用,也可以使用多种水溶性有机溶剂的混合溶剂。另外,可以使用与水的混合物的形式。使用与水的混合物时,上述水溶性有机溶剂,优选在混合物中的含量至少为50容量%以上,更优选为70容量%以上。
作为这样的水溶性有机溶剂的例子,可举出甲醇、乙醇、1-丙醇、2-丙醇、2-丁醇、丙酮、四氢呋喃、乙腈、甲基乙基酮、二丙二醇单甲基醚、乙酸甲酯、乙酰乙酸甲酯、N-甲基吡咯烷酮、二甲基硫醚、乙二醇、1,3-丁烷二醇、1,4-丁烷二醇、丙二醇、二甘醇、三甘醇等以及它们的混合物。其中,限于在食品中的用途时,优选乙醇、丙二醇、丙酮,特别优选乙醇以及乙醇与水的混合液。
利用水溶性有机溶剂从上述动植物原料提取天然成分的方法,可以使用现有方法来进行。由于多使用将干燥的植物等的原料进行微粉碎的固体原料,因此可以主要使用固液提取装置。在实验中,可以使用利用烧杯等在一定温度下一边搅拌一边进行提取操作的简便方法,或使用索氏提取装置等。作为工业上可使用的固液提取装置,作为浸透贯流型的提取机,可举出间歇式固液混相提取机、连续式的博尔曼(ボ一ルマン)提取机、罗托瑟提取机,鲁奇提取机,肯尼迪提取机等。另外,作为固体分散型装置,可使用帕丘卡槽提取机、博瑙特(ボノト一)提取机等。
在本发明中,优选先将用于提取的动植物、藻类、酵母、菌类等充分水洗,然后干燥后或原样通过球磨机、冷冻粉碎,压缩粉碎,超声波处理等物理的破碎处理或酶处理等来进行前处理。由于在从湿的材料中使用有机溶剂提取的情况下,杂质的混入更明显,另外,提取率同样也会下降,因此工业上优选利用干燥菌体进行溶剂提取的方法。
本发明中使用的提取,代表性地,在低于对被提取的有效成分的特性产生不好的影响的温度(或比所选择的提取溶剂的合理的回流温度高的温度)且对于产生有意义的提取足够高的温度下实施。一般地说,该温度为0℃至300℃的范围,也可以是比其更高或更低的温度。一般地说,提取温度因提取溶剂或溶液的沸腾或冻结特性而不同。原则上温度变高提取速度也会变快,因此为了经济的提取速度,优选中等程度的温度。通常的提取温度为20~68℃,优选在20~50℃左右进行。
提取溶剂的选择,代表性地,根据受提取的有效成分的化学性质、溶解性、沸点和凝固点等的主要条件的不同而不同,但是优选可以从上述的水溶性有机溶剂之内进行选择,优选可以使用乙醇、正丙醇等之类的醇的纯的溶剂或者利用水稀释的溶剂作为提取溶剂,特别优选乙醇为提取溶剂。
作为提取溶剂,例如在使用乙醇时,使用提取原料的3~100倍量左右(重量比)的提取溶剂,进行30分钟~2小时左右的提取。使有效成分溶出在溶剂中之后,过滤除去提取残渣,由此得到提取液。其后,根据通常方法对提取液进行稀释、浓缩、精制等处理,得到本发明的有效成分的乙醇溶液。
提取天然成分之后的动植物原料的残渣,可以利用能够分离浸渍液的固体成分与液体成分的方法来除去。例如,可以是过滤、离心分离、静置沉淀或者它们的组合。过滤时,对过滤方式没有限制。例如,可以是自然落下式,减压过滤等压力控制式的任一者。另外,滤器部可以是纸滤器、膜滤器、布滤器、木炭滤器、中空枝丝膜滤器、微滤器、硅藻土过滤器(celitefilter)、硅藻土、它们的组合等中的任一种。滤器部可以为单层或多层。为多层滤器时,可以由相同或不同的多个层构成。进而,为多层滤器时,只要是液体成分可以经过各层的构成即可,不必是上下或左右重叠的层。例如,将填充了滤器成分的多个柱借助配管进行连接以使其可以过滤的情况下,各层也可以分离。
离心分离时,固体成分与液体成分的分离方式没有限制。例如可以是将浸渍液导入多孔管内,在离心机内使其离心,从而回收从孔放出的提取液的方式;可以是导入无孔管内,离心后回收上清的提取液的方式。另外,对于离心的重力加速度(G)而言,只要液体成分与固体成分可以某种程度分离,就没有特别限定。
在利用过滤等的方法从本发明的天然成分的提取液中除去不溶固体成分之前,使其与选自活性炭、酸性白土或活性白土的1种以上的物质接触,可容易地除去不溶固体成分。例如,利用过滤除去不溶固体成分时,使过滤工序以最小次数,例如1次就可以完成。作为所使用的活性炭,一般而言,只要是工业水平可以使用的活性碳就没有特别限制,可以使用例如ZN-50(北越碳公司制)、Kuraraycaol GLC、Kuraraycaol PK-D、Kuraraycaol PW-D(Kuraray Chemical公司制)、白鹫AW50、白鹫A、白鹫M、白鹫C(武田药品工业公司制)等市售品。活性炭的细孔容积优选0.01~0.8mL/g,比表面积优选800~1300m2/g。活性炭相对于乙醇溶液100质量份优选为0.5~5质量份。
本发明中的水溶性有机溶剂溶液中的天然成分的含量根据原料和天然成分的种类不同,但是从乳化物和分散物制造的效率的观点、乳化物和分散物的稳定性的观点出发,一般而言,相对于水溶性有机溶剂溶液的总质量可以设为0.01质量%~30质量%,优选设为0.1质量%~10质量%。
另外,在水溶性有机溶剂溶液中,除了含有天然成分以外,根据需要还可以含有其他的成分。作为这样的其他成分,可以举出加氢油脂,硅油等油性成分,非离子性表面活性剂,离子性表面活性剂,合成磷脂质,合成神经酰胺等表面活性化合物等。
本发明中的水性溶液即水相为以水为主成分的水溶液。
在该水性溶液中可以含有下述所示的非离子表面活性剂,离子性表面活性剂,水溶性的盐类,糖类,多糖类,蛋白质,pH调整剂,抗氧化剂,防腐剂,色素,香料等。
作为离子性表面活性剂的例子,可举出烷基磺酸盐,烷基硫酸盐,单烷基磷酸盐,脂肪酸盐,卵磷脂等。作为盐类,可以使用氯化钠、柠檬酸钠、抗坏血酸钠等。作为糖类,可举出葡萄糖、果糖、蔗糖、阿拉伯糖、纤维二糖、乳糖、麦芽糖、海藻糖等。另外,作为多糖类,可以举出麦芽糊精、寡糖、菊糖、阿拉伯胶、壳聚糖等。作为蛋白质,可举出各种氨基酸类、寡肽、明胶、水溶性骨胶原、酪朊、环糊精等。
作为pH调整剂,可以使用氢氧化钠等碱、盐酸等酸、磷酸盐缓冲液、柠檬酸盐缓冲液等缓冲液。作为抗氧化剂,可以使用抗坏血酸、抗坏血酸衍生物、柠檬酸单甘油酯等。
从乳化分散的细微化的观点出发,这些添加剂相对于水相的添加量可以设为水相的总质量的20质量%以下,优选设为10质量%以下。另外,根据需要可以在水相中预先添加少量的水溶性有机溶剂。从乳化物和分散物的稳定性的观点出发,这时的水溶性有机溶剂的添加量为水相的总质量的20质量%以下,优选10质量%以下。
[非离子性表面活性剂]
本发明中的非离子性表面活性剂,为了提高分散性,优选HLB为10以上且HLB为16以下的非离子性表面活性剂,从乳液的稳定性的观点出发,更优选为12以上且16以下。
另外,上述非离子性表面活性剂可以含有有机溶剂相和水相中的任一者,也可含有两者,优选在有机溶剂相中添加至少一种。这时,在本发明的制造方法中,与水性溶液混合之前,可以还包括在水溶性有机溶剂溶液中添加非离子性表面活性剂,使其均匀化的工序。
从分散粒径的细微化的观点出发,本发明中可以使用的非离子性表面活性剂的至少一种,优选在有机溶剂中可以溶解的非离子性表面活性剂。作为可溶于有机溶剂的非离子性表面活性剂,只要是在有机溶剂中溶解的非离子性表面活性剂,就没有特别的限制。
在此,所谓HLB,是指在通常表面活性剂的领域中使用的亲水性-疏水性平衡值,作为通常使用的计算式,例如可以使用川上式等。
本发明中,采用下述川上式。
HLB=7+11.7log(Mw/M0)
在此,Mw为亲水基的分子量,M0为疏水基的分子量。
另外,可以使用厂商公开发表的目录等中记载的HLB的数值。另外,由上述的式可知,利用HLB的加合性,可以得到具有任意的HLB值的非离子性表面活性剂。
作为本发明中可以优选使用的非离子性表面活性剂的例,可举出(单,二,三)甘油脂肪酸酯,单甘油有机酸酯,多甘油脂肪酸酯,丙二醇脂肪酸酯,多甘油缩合蓖麻酸酯,脱水山梨糖醇脂肪酸酯,蔗糖脂肪酸酯等。其中,从分散物的稳定性向上的观点出发,本发明中更优选的非离子性表面活性剂为聚甘油脂肪酸酯,蔗糖脂肪酸酯或它们的组合。
这些非离子性表面活性剂可以分别单独使用或将它们的2种以上以任意比例并用。
另外,上述的非离子性表面活性剂不必为利用蒸馏等高度精制的成分,可以是反应混合物。
作为本发明中使用的聚甘油脂肪酸酯,平均聚合度为4以上,优选6~10的聚甘油、碳数8~18的脂肪酸与例如辛酸、癸酸、月桂酸、肉豆蔻酸、棕榈酸、硬脂酸、油酸和亚油酸所形成的酯。
作为聚甘油脂肪酸酯的优选例,可举出六甘油单棕榈酸酯、六甘油单肉豆蔻酸酯、六甘油单月桂酸酯、十甘油单油酸酯、十甘油单硬脂酸酯、十甘油单棕榈酸酯、十甘油单肉豆蔻酸酯、十甘油单月桂酸酯等。
这些多甘油脂肪酸酯可以单独使用或混合使用。
作为市售品,例如可举出日光化学(株)公司制的NIKKOL Hexaglyn1-L、NIKKOL Hexaglyn 1-M、NIKKOL Decaglyn 1-L、NIKKOLDecaglyn1-M、NIKKOL Decaglyn1-SV、NIKKOL Decaglyn1-50SV、NIKKOL Decaglyn 1-ISV、NIKKOL Decaglyn 1-O、NIKKOL Decaglyn 1-OV、NIKKOL Decaglyn1-LN、三菱化学食品(株)公司制RYOTOPolygly ester L-10D、L-7D、M-10D、M-7D、P-8D、S-28D、S-24D、SWA-20D、SWA-15D、SWA-10D、O-15D、理研维生素(株)公司制POEM J-0381V、POEM J-0021V等。
本发明中使用的蔗糖脂肪酸酯优选脂肪酸的碳数为12以上的物质,更优选12~20的物质。
作为蔗糖脂肪酸酯的优选例,可举出蔗糖单油酸酯、蔗糖单硬脂酸酯、蔗糖单棕榈酸酯、蔗糖单肉豆蔻酸酯、蔗糖单月桂酸酯等。
在本发明中,这些蔗糖脂肪酸酯可以单独使用或混合使用。
作为市售品,例如可举出三菱化学食品(株)公司制RYOTO sugaresterS-1170、S-1170S、S-1570、S-1670、P-1570、P-1670、M-1695、O-1570、OWA-1570、L-1695、LWA-1570、第一工业制药(株)公司制的、DK ester F140、DK ester F160、DK ester SS等,但并不限于这些。
在本发明中可以与上述的水溶性非离子性表面活性剂并用地使用卵磷脂。本发明中可使用的卵磷脂,由于以甘油骨架和脂肪酸残基以及磷酸残基为必须构成成分,在其上结合碱、多元醇等,因此称为磷脂质。
卵磷脂由于在分子内具有亲水基和疏水基,因此以往在食品,医药品,化妆品领域广泛作为乳化剂使用。
产业上卵磷脂纯度60%以上的物质可以作为卵磷脂利用,本发明中也可以利用,但是从细微的油滴粒径的形成和功能性油性成分的稳定性的观点出发,优选一般称为高纯度卵磷脂的物质,这些成分的卵磷脂纯度为80%以上,更优选纯度90%以上的物质。
作为卵磷脂,可列举从植物、动物和微生物的生物体中提取分离的以往公知的各种物质。
作为这样的卵磷脂的具体例,可举出例如来自大豆、玉米、花生、油菜子、麦等植物或卵黄、牛等的动物和大肠菌等的微生物等的各种卵磷脂。
若化合物名例示这样的卵磷脂,则可举出磷脂酸、磷脂酰基甘油、磷脂酰基肌醇、磷脂酰基乙醇胺、磷脂酰基甲基乙醇胺、磷脂酰胆碱、磷脂酰基丝氨酸、羟乙磷酸(ビスホスアチジン酸)、二磷脂酰基甘油(心磷脂)等甘油卵磷脂;鞘磷脂等的鞘酯卵磷脂等,或它们的组合。
另外,在本发明中,除了上述的高纯度卵磷脂以外,可使用加氢卵磷脂、酶分解卵磷脂、酶分解加氢卵磷脂、羟基卵磷脂等或它们的组合。本发明中可使用的这些卵磷脂可以单独使用或以多种成分的混合物的形态使用。
这些非离子性表面活性剂的添加量,相对于乳化物或分散物总质量,优选为0.1~50质量%,更优选为0.5~20质量%,更优选1~15质量%。
另外,相对于天然成分,优选10~1000质量%,更优选50~500质量%。
通过将非离子性表面活性剂的添加量设定在0.1质量%以上,细微的粒径的分散物变得易于获得,从得到的分散物的稳定性提高的观点出发是优选的。另外,通过设为50质量%以下,分散物的起泡变得剧烈等问题变得难以发生,从这一观点出发是优选的。
另外,在水溶性有机溶剂溶液中,除了作为提取液使用的水溶性有机溶剂之外,也可以混合其他水溶性有机溶剂。另外,除了提取液中用的水溶性有机溶剂之外也可以添加水。后添加的水溶性有机溶剂、水的混合比例,从乳化粒子的细微化的观点出发,优选相对于混合后的水溶性有机溶剂溶液总质量为50容量%以下,更优选为30容量%以下。
(超微混合机)
在本发明中,使水溶性有机溶剂溶液(以下,有时简单称为“油相”或“水溶性有机溶剂相”)和水性溶液(以下,有时简单称为“水相”)通过最窄部分的截面积为1μm2~1mm2的微流路之后,通过相向冲撞使其混合的最适当的装置是对抗冲撞型超微混合机。超微混合机为将两种不同的液体在微小空间中进行混合的机器,一方的液体为含有功能性油性成分的有机溶剂相,另一方为制成水性溶液的水相。
本发明人发现,将超微混合机应用于作为微化学过程的一种的粒径较小的乳液制备时,耗能量较低且发热少,与通常的搅拌乳化分散方式、高压匀浆仪乳化分散相比,可以得到粒径集中、保存稳定性也优异的良好的乳化物或分散物。已判明是用于含有易于热劣化的天然成分的乳化的最适方法。
使用超微混合机进行乳化或分散的方法的概要是,将水相和水溶性有机溶剂相分别分配至微小空间,再使各微小空间相互接触、或者相互冲撞。与仅将一方分配至微小空间、另一方仍为大体积之类的方法即膜乳化法、微型通道乳化法明显不同,实际上仅将一方分配至微小空间不能得到本发明的效果。作为公知的超微混合机,有各种结构的机器。若以与微流路中的液流的混合为着眼点,则可举出维持层流进行混合的方法、扰乱液流即以湍流进行混合的方法这2种方法。在维持层流进行混合的方法中,通过使流路深度比流路宽度的尺寸大,从而使2液的边界面积尽可能的大,通过使两层的厚度变薄来实现混合的高效化。另外,也正在设计将2液的入口分割为多个从而将其变成交替地流动的多层流的方法。
另一方面,以湍流进行混合的方法中,将各液体分配至窄流路并以比较高的速度流动的方法是一般的做法。也在研究使用阵列化的微喷嘴将一方的液体导入微小空间并喷向另一方的液体的方法。另外,使用各种的方式强制地将高速流动的液体彼此接触的方法的混合效果特别良好。一般来说,前者的使用层流的方法产生的粒子的尺寸的分布比较集中,后者的使用湍流的方法可以得到非常细微的乳液,从稳定性和透明性的观点出发,优选使用湍流的方法的情况更多。在作为使用湍流的方法中,以梳齿型和冲撞型为代表。作为上述梳齿型超微混合机,像以IMM公司制为代表的那样形成2个梳齿状的流路相向放置并错综复杂地配置的结构。这时因为,如果将梳齿的宽度设置的足够小,虽然液流变为湍流,但是有机溶剂相和水相并不冲撞,相遇后以并行流的方式沿着同一方向流动,因此强制的接触效果与冲撞型相比并不充分。
与此相对,在以KM混合机为代表的冲撞型超微混合机中,利用运动能量实现强制接触。具体地说,可举出由长泽等(“H.Nagasawa et al,Chem.Eng.Technol,28,No.3,324-330(2005)”,日本特开2005-288254号公报)公开的中心冲撞型超微混合机。对使水相和有机溶剂相相向冲撞的方法而言,由于其混合时间极短,可以瞬时形成油相滴,因此可以形成极细微的乳化物或分散物。
在利用本发明的冲撞型超微混合机进行微混合而乳化的情况下,从得到的乳液的粒径均匀性的观点出发,对于乳化时的温度(乳化温度)而言,需要将超微混合机的上述其他微小空间的温度(超微混合机的微混合部的温度)设为80℃以下进行微混合,优选0~80℃,特别优选5~75℃。通过将上述乳化温度设为0℃以上,由于分散介质的主体为水,从而可管理乳化温度因而优选。优选超微混合机的上述微小空间的保温温度为100℃以下。通过将上述保温温度设为100℃以下,保温温度的管理可以容易地控制,另外,可以消除对乳化性能产生不好影响的微小突沸现象。进一步优选将上述保温温度控制在80℃以下的温度。
超微混合机的分配至上述微小空间的油相(水溶性有机溶剂相)、水相和超微混合机的上述微小空间的保温温度根据水相和有机溶剂相中所含成分的不同而不同,分别独立地优选0~80℃,特别优选5~75℃。油相中含有水溶性有机溶剂时,超微混合机的上述分配至微小空间的油相、水相和超微混合机的上述微小空间的保温温度分别独立地优选0~50℃,特别优选5~25℃。超微混合机的上述微小空间的保温温度、超微混合机的分配至上述微小空间的油相和水相的保温温度、超微混合机的即将分配至上述微小空间之前的油相和水相的保温温度(即油相和水相供给槽的保温温度)可以互不相同,但从混合的稳定性的观点出发,优选设为相同温度。
在本发明中,特别优选:在将超微混合机的即将分配至微小空间前后的水相、水溶性有机溶剂相和超微混合机的上述微小空间以及上述其他微小空间的保温温度设置在比室温高的温度下进行微混合而乳化后,采集利用超微混合机得到的水中油滴型乳液,然后冷却至常温。
本发明中的超微混合机的微小空间(流路)的最窄部分的截面积为1μm2~1mm2,从乳液粒径的细微化和粒径分布的尖锐化的观点出发,优选500μm2~50000μm2。
从混合稳定性的观点出发,本发明中的水相中使用的超微混合机的微小空间(流路)的最窄部分的截面积特别优选1000μm2~50000μm2。
从乳液粒径的细微化和粒径分布的尖锐化的观点出发,油相中使用的超微混合机的微小空间(流路)的最窄部分的截面积特别优选500μm2~20000μm2。
另外,在本发明中的超微混合机中乳化分散时,作为乳化分散时的油相和水相的流量,根据使用的超微混合机的不同而不同,但从乳液粒径的细微化和粒径分布的尖锐化的观点出发,作为水相的流量,优选10ml/min~500ml/min,更优选20ml/min~400ml/min,特别优选50ml/min~300ml/min。
作为油相的流量,从乳液粒径的细微化和混合稳定性的观点出发,优选1ml/min~70ml/min,更优选3ml/min~60ml/min,特别优选7ml/min~40ml/min。
两相的流量除以微型通道的截面积的值,即两相的流速比(Vo/Vw),从粒子的细微化和超微混合机的设计观点出发,优选0.05以上5以下的范围。其中,Vo是含有水不溶性天然成分的有机溶剂相的流速,Vw是水相的流速。另外,从将粒子更加细微化的观点出发,流速比(Vo/Vw)在0.1以上3以下为最优选范围。
另外,作为水相和油相的送液压力,水相与油相优选分别为50KPa~5000KPa和10KPa~1000KPa,更优选100KPa~2000KPa和20KPa~500KPa,特别优选200KPa~1000KPa和40KPa~200KPa。通过将上述水相的送液压力设为50Kpa~5000KPa,可以维持稳定的送液流量的趋势,通过将油相的送液压力设为10KPa~1000KPa,可以得到均匀的混合性,因而优选。
在本发明中,上述流量、送液压力和保温温度更优选各优选例的组合。
接着,关于将上述水相、油相导入超微混合机直至以水中油滴型乳液的形式排出为止的路径,例示作为本发明的超微混合机的一例的微型设备(图1)进行说明。
如图1所示,微型设备100包括均为圆柱状形态的、供给元件102、合流元件104和排出元件106。
在供给元件102的与合流元件104对置的面,本发明的油相或水相的流路的截面为矩形的环状通道108和110形成为同心状。在供给元件102中,形成沿其厚度(或高度)方向贯通至各环状通道的钻孔112和114。
在合流元件104中,形成沿其厚度方向贯通的钻孔116。该钻孔116在用于构成微型设备100而紧固元件时,位于与供给元件102对置的合流元件104的面的钻孔116的端部120以向环状通道108开口的方式形成。在图示的方式中,形成4个钻孔116,它们在环状通道108的周向上等间隔配置。
在合流元件104中,与钻孔116同样地贯通形成钻孔118。钻孔118也与钻孔116同样地,以向环状通道110开口的方式形成。钻孔118也在环状通道110的周向上等间隔配置,且钻孔116与钻孔118以交替定位的方式配置。
在合流元件104的与排出元件106对向的面122形成有微型通道124和126。该微型通道124或126的一端为钻孔116或118的开口部,另一端部为面122的中心128,所有的微型通道均朝向该中心128从钻孔延伸设置并在中心合流。微型通道的截面例如可以为矩形。
在排出元件106中,形成有通过其中心且沿厚度方向贯通的钻孔130。因此,该钻孔在一端向合流元件104的中心128开口,另一端向微型设备的外部开口。
在本微型设备100中从微型设备100的外部供给的流体A和B在钻孔112和114的端部分别经由钻孔112和114而流入环状通道108和110。
环状通道108和钻孔116连通,流入环状通道108的流体A经由钻孔116进入微型通道124。另外,环状通道110与钻孔118连通,流入环状通道110的流体B经由钻孔118进入微型通道126。流体A和B分别流入微型通道124和126之后,流向中心128而合流。
上述合流的流体经由钻孔130,以液流C形式排出微型设备的外部。

对于水相与有机溶剂相发生冲撞的微型通道(图1中,124和126)的尺寸,根据与水相和有机溶剂相的流量的关系,规定为适当范围。
在本发明的制造方法中,使用的水溶性有机溶剂优选流过微流路而乳化或分散后再除去。作为除去溶剂的方法,已知使用旋转蒸发仪、闪蒸器、超声波喷雾器等的蒸发法,超滤膜、反渗透膜等的膜分离法,特别优选超滤膜法。
所谓超滤(Ultra Filter:简称UF),是指通过将原液(水、高分子物质、低分子物质、胶体物质等混合水溶液)加压,并在UF装置中注入水,从而能够将原液分离为透过液(低分子物质)和浓缩液(高分子物质,胶体物质)这2个系统的溶液并取出的装置。
超滤膜是利用罗布石栗(ロブ-スリ一ラ一ヤン)法制作的典型的非对称膜。使用的高分子素材为聚丙烯腈、聚氯化烯-聚丙烯腈共聚物、聚砜、聚醚砜,偏氟乙烯,芳香族聚酰胺,乙酸纤维素等。最近,也开始使用陶瓷膜。超滤法与反渗透法等不同,由于不进行前处理,因此在膜面产生高分子等堆积的污垢。因此一般利用药品和温水定期的对膜进行清洗。因此膜素材需要具有对药品的耐性和耐热性。超滤膜的膜模块为平膜型、管状型、中空丝型、螺旋型各种。超滤膜的性能指标为截留分子量,其为1000~300000为止的各种的膜有市售品。作为市售的膜模块,有MicrozaUF(旭化成化学品(株)),毛细管型Element NTU-3306(日东电工(株))等,但不限于这些。
为了从本发明的乳化物中除去溶剂,从膜的材质有溶剂耐性的观点出发,特别优选聚砜、聚醚砜、芳香族聚酰胺。作为膜模块的形态,在实验室规模主要使用平膜,而工业上使用中空丝型、螺旋型,特别优选中空丝型。另外,截留分子量根据有效成分的种类不同而不同,通常可以使用5000~100000的范围的的超滤膜。
操作温度可以为0℃~80℃,若考虑有效成分的劣化,特别优选10~40℃的范围。
作为实验室规模的超滤装置,使用平膜型模块,有ADVANTEC-UHP(Advantec(株)),Flow type Labotest unit RUM-2(日东电工(株))等。工业上可以将各膜的模块根据需要的能力对尺寸和根数进行任一组合构成成套设备。作为小型实验装置的单元,市售有RUW-5A(日东电工(株))等。
在本发明的制造方法中,在溶剂除去之后,优选将得到的乳化物浓缩。作为浓缩方法,可以使用与蒸发法、过滤膜法等溶剂除去相同的方法、装置。浓缩时也优选超滤膜法的方法。只要可以使用与溶剂除去相同的膜就即可,但根据需要也可以使用截留分子量不同的超滤膜。另外,以与溶剂除去不同的温度进行运转,也可以提高浓缩效率。
由本发明的制造方法得到的乳化物为水中油滴型乳液,从得到的乳化物的透明性的观点出发,乳化物中的油滴粒子的体积平均粒径(中位径)优选为200nm以下,更优选为1nm~100nm,更优选1nm~50nm。
另外,从得到的分散物的透明性的观点出发,根据本发明的制造方法得到的水性固体分散物中的疏水性固体微粒子、即分散粒子的体积平均粒径(中位径)优选为200nm以下,更优选为1nm~100nm,更优选1nm~50nm。
本发明的乳化物粒子或分散物粒子的粒径可以利用市售的粒度分布计等进行测量。作为乳液的粒度分布测定法,已知有光学显微镜法,共焦点激光显微镜法,电子显微镜法,原子间力显微镜法,静态光散射法,激光衍射法,动态光散射法,离心沉降法,电气脉冲测量法,色谱法,超声波减衰法等,市售有与各原理对应的装置。
从本发明中的粒径范围和测定的容易度考虑,本发明的乳化物或分散物的粒径测定优选动态光散射法。作为利用动态光散射的市售的测定装置,可举出Nanotrac UPA(日机装(株))、动态光散射式粒径分布测定装置LB-550((株)堀场制作所)、浓厚系粒径分析仪FPAR-1000(大冢电子(株))等。
本发明中的粒径,采用利用上述动态光散射式粒径分布测定装置在25℃时的测定值。
利用本发明得到的乳化物或分散物是天然成分的品质劣化明显较低的细微的乳化物或分散物。因此,可以优选用于根据天然成分的种类的各种的用途。
作为这样的用途,例如可举出食品、皮肤外用剂和医药品。
即,本发明的食品为含有由上述制造方法得到的乳化物或分散物的食品,上述天然成分为功能性食品成分的物质。在此作为功能性食品成分,只要是上述的天然成分中在食品用途中可以使用的即可,特别地可举出萜类、多酚类等,但并不限于这些。
另外,本发明的皮肤外用剂为含有由上述制造方法得到的乳化物或分散物的皮肤外用剂,上述的天然成分为皮肤外用剂成分的物质。在此作为皮肤外用剂成分,只要是上述的天然成分中的在皮肤外用用途中可以使用的即可,特别地可以举出脂肪酸类、复合脂质类、萜类等,但并不限于这些。
本发明的医药品为含有由上述制造方法得到的乳化物或分散物的医药品,上述的天然成分为医药品成分的物质。在此作为医药品成分,只要是上述的天然成分中的在医药用途中可以使用的即可,特别地可举出生物碱类,甾体类等,但并不限于这些。
实施例
以下利用实施例对本发明进行更具体地说明。此外,在以下的记载中“份”和“%”表示的数值,没有特别说明时为质量基准。
<实施例1>
(乳液A的制备)
作为水相A,准备纯水280g。
另外,将下述成分在65℃条件下溶解1小时后冷却至25℃,得油相A。
·雨生红球藻提取物(虾青素类含有率10质量%)0.6g
·混合生育酚0.1g
·蔗糖硬脂酸酯(HLB=15)0.3g
·单油酸十甘油酯(HLB=12)0.3g
·乙醇38.4g
在此,蔗糖硬脂酸酯使用三菱化学食品(株)制RYOTOSugar ester S-1670(HLB=15),单油酸十甘油酯使用日光化学品(株)制NIKKOLDecaglyn 1-O(HLB=12)。雨生红球藻提取物使用武田纸器(株)制ASTOTS-10O。该雨生红球藻提取物是以已经干燥了的油状物形式提取的成分。混合生育酚使用理研维生素(株)制的理研E OIL 800。乙醇使用和光纯药试药特级。
作为在乳化中使用的超微混合机,准备在流路中分别设置了5μm的烧结金属滤器的、梳齿型SSIMM-SS-Ni25(IMM公司制),将超微混合机、水相A、油相A都置于25℃环境下。
将精密定量泵设定为水相的流量为21.0ml/min.油相的流量为3.0ml/min.,将水相和油相分别导入超微混合机,进行微混合。
接着,在上述流量和送液压力稳定的阶段,收集乳液,制成含有虾青素的乳液A。乳液A的油滴粒径利用大冢电子制FPAR1000粒径测定机进行测定时,中位径为135nm。
(乳液B的制备)
水相和油相的组成、制备方法与乳液A完全相同,用于乳化的超微混合机使用冲撞型的KM型超微混合机100/100。以下是冲撞前的微流路即图1中的、微型通道124与微型通道126的尺寸,以矩形计为,
微型通道124:宽/深/长:100μm/100μm/12.5mm
微型通道126:宽/深/长:100μm/100μm/10mm。
使用该超微混合机,在外环中以21.0ml/min.的流量导入水相,在内环中以3.0ml/min.的流量导入油相进行乳化,除此之外,与乳液A使用相同的方法进行乳化,制成乳液B。乳液B的油滴的中位径为45nm。
(乳液C的制备)
水相和油相的组成、制备方法与乳液A完全相同,但是用于乳化的超微混合机使用KM型超微混合机350/50。以下是冲撞前的微流路即图1中的、微型通道124和微型通道126的尺寸,以矩形计为:
微型通道124:宽/深/长:350μm/100μm/12.5mm
微型通道126:宽/深/长:50μm/100μm/10mm。
使用该超微混合机,在外环(微型通道124)以42.0ml/min.的流量导入水相,在内环(微型通道126)以6.0ml/min.的流量导入油相进行乳化,除此以外,与乳液A采用同样的方法进行乳化,制成乳液C。乳液C的油滴的中位径为19nm。
(乳液D的制备)
水相和油相的组成、制备方法与乳液C完全相同,用于乳化的超微混合机也使用KM型超微混合机350/50。其中,与乳液C不同的是:在内环(微型通道126)以42.0ml/min.的流量导入水相,在外环(微型通道124)以6.0ml/min.的流量导入油相进行乳化,除此以外,采用与乳液C同样的方法进行乳化,制成乳液D。乳液D的油滴的中位径为95nm。
(乳液E的制备)
水相和油相的组成、制备方法与乳液A完全相同,用于乳化的超微混合机使用KM型超微混合机200/100。以下为冲撞前的微流路即图1中的、微型通道124和微型通道126的尺寸,以矩形计为,
微型通道124:宽/深/长:200μm/100μm/12.5mm
微型通道126:宽/深/长:100μm/100μm/10mm。
使用该超微混合机,在外环(微型通道124)以31.5ml/min.的流量导入水相,在内环(微型通道126)以4.5ml/min.的流量导入油相进行乳化,除此以外,采用与乳液A同样的方法进行乳化,制成乳液E。乳液E的油滴的中位径为28nm。
(乳液F的制备)
水相和油相的组成、制备方法与乳液E完全相同,用于乳化的超微混合机也使用KM型超微混合机200/100。但与乳液E不同的是,在内环(微型通道126)以31.5ml/min.的流量导入水相,在外环(微型通道124)以4.5ml/min.的流量导入油相进行乳化,除此以外,采用与乳液E同样的方法进行乳化,制成乳液F。乳液F的油滴的中位径为55nm。
(乳液G的制备)
采用与乳液A相同的组成,将水相A和油相A利用搅拌机混合后,直接利用搅拌式匀浆仪(日本精机(株)制)以10000rpm进行5分钟乳化,从而得到含有虾青素的乳液G。乳液G的油滴中位径为347nm。
(乳液H的制备)
将含虾青素的雨生红球藻冷却至-50℃,通过将其冻结干燥除去水分,然后,添加氯化钠进行粉碎操作,得到雨生红球藻的微粉末。冷藏室中将该雨生红球藻微粉末在乙醇中浸渍24小时,从雨生红球藻提取虾青素。
在上述提取液中加入乙醇,以使上述提取液的478nm的吸光度与油相A的吸光度吻合。在由此得到的雨生红球藻乙醇提取液中,以达到与油相A相同组成的方式添加溶解混合生育酚、蔗糖硬脂酸酯和单油酸十甘油酯,将其作为油相H。
利用与乳液A完全同样的方法对油相H和水相A进行超微混合机乳化,将其作为乳液H。乳液H的油滴中位径为120nm。
(乳液I的制备)
在乳液B中,除了代替油相A而使用油相H以外,与乳液B同样地进行制备。乳液I的油滴中位径为37nm。
(乳液J的制备)
在乳液C中,除了代替油相A而使用油相H以外,与乳液C同样地进行制备。乳液J的油滴中位径为12nm。
(乳液K的制备)
在乳液D中,除了代替油相A而使用油相H以外,与乳液D同样地进行制备。乳液K的油滴中位径为89nm。
(乳液L的制备)
在乳液E中,除了代替油相A而使用油相H以外,与乳液E同样地进行制备。乳液L的油滴中位径为26nm。
(乳液M的制备)
在乳液F中,除了代替油相A而使用油相H以外,与乳液F同样地进行制备。乳液M的油滴中位径为54nm。
(乳液N的制备)
在乳液G中,除了代替油相A而使用油相H以外,与乳液G同样地进行制备。乳液N的油滴中位径为330nm。
针对上述乳液A~N,使用实验室规模超滤装置ADVANTEC-UHP-43K型,超滤膜使用聚砜制Q 0500043E(50000Da),去除乙醇使其含量为0.1%,进而使用相同装置进行浓缩操作,浓缩化至虾青素浓度达到1.0%,制得浓缩乳液A~N。
针对浓缩乳液A~N,寻找男女10名进行异臭有无的评价,按以下进行判断:
可容忍的水平;-,
稍有些在意的水平;+,
需要应对的水平;++。
另外,对于这些乳液,在50℃下保存21日,测定色素残存率。对于色素残存率而言,通过以下方法求得:将保存后的乳液和保存前的乳液分别利用纯水稀释至3000倍,利用分光光度计测定该液的吸光度,根据保存后的478nm吸光度和保存前的478nm吸光度之比求算。
将上述油滴粒径、异臭判定、50℃保存21日的色素残存率的结果汇总在表1中。

由表1可知:使用相向冲撞型超微混合机时,与搅拌乳化机、梳齿形超微混合机相比,可以细微化的性能有压倒性优势,乳液的透明性也高(乳液B~F:参考例,乳液I~M:本发明)。
另外,尽管是从相同的雨生红球藻中提取的虾青素,但在以已干燥的油状物形式提取出并将其溶解于乙醇构成油相的乳液B~F中,有异臭,且随着保存时间延长虾青素色素的保存稳定变差。与此相对,可以判明,通过使从雨生红球藻中乙醇提取了虾青素的液体不干固的连续操作来构成油相的乳液I~M中,异臭情况改良,虾青素的保存稳定性也提高。
另外可以判明,将已干燥的油状虾青素作为原料进行乳化时,乳化油滴径变小,则经时稳定性也表现稍微恶化的趋势,但是对不使提取液干固而连续操作情况下制成的原料进行乳化时,可以更细微化,虾青素的经时稳定性也保持良好的水平(乳液I~M)。
由此,根据本发明,可以以天然成分的品质劣化极少的状态提供利用水溶性有机溶剂从动植物提取的天然成分的细微的乳化物或分散物。

Claims (14)
1.一种乳化物或分散物的制造方法,包括:
使水溶性有机溶剂溶液和水性溶液分别通过最窄部分的截面积为1μm2~1mm2的微流路后,相向冲撞,以此使其混合的步骤,所述水溶性有机溶剂溶液含有利用水溶性有机溶剂从天然的动植物中提取的天然成分。
2.根据权利要求1所述的乳化物或分散物的制造方法,其特征在于,
所得到的乳化物或分散物中的油滴粒子或分散粒子的体积平均粒径在1nm~100nm的范围。
3.根据权利要求1或2所述的乳化物或分散物的制造方法,其特征在于,
混合时上述水溶性有机溶剂溶液的流速Vo与上述水性溶液的流速Vw之比Vo/Vw为0.05~5。
4.根据权利要求1或2所述的乳化物或分散物的制造方法,其中,
水溶性有机溶剂溶液的流量为1ml/min~70ml/min。
5.根据权利要求1或2所述的乳化物或分散物的制造方法,其中,
水性溶液的流量为10ml/min~500ml/min。
6.根据权利要求1或2所述的乳化物或分散物的制造方法,其中,
还包括从上述混合后得到的乳化物或分散物中除去水溶性有机溶剂的步骤。
7.根据权利要求1或2所述的乳化物或分散物的制造方法,其中,
上述水溶性有机溶剂溶液含有至少1种HLB为10~16的非离子性表面活性剂。
8.根据权利要求1或2所述的乳化物或分散物的制造方法,其中,
所述水溶性有机溶剂为乙醇或乙醇与水的混合物。
9.根据权利要求1或2所述的乳化物或分散物的制造方法,其中,上述天然成分为功能性食品成分。
10.一种食品,其是含有利用权利要求9所述的制造方法制造的乳化物或分散物的食品,其中,所制造的乳化物或分散物中的油滴粒子或分散粒子的体积平均粒径为1nm以上50nm以下。
11.根据权利要求1或2所述的乳化物或分散物的制造方法,其中,上述天然成分为皮肤外用剂成分。
12.一种皮肤外用剂,其是含有利用权利要求11所述的制造方法制造的乳化物或分散物的皮肤外用剂,其中,所制造的乳化物或分散物中的油滴粒子或分散粒子的体积平均粒径为1nm以上50nm以下。
13.根据权利要求1或2所述的乳化物或分散物的制造方法,其中,上述天然成分为医药成分。
14.一种医药品,其是含有利用权利要求13所述的制造方法制造的乳化物或分散物的医药品,其中,所制造的乳化物或分散物中的油滴粒子或分散粒子的体积平均粒径为1nm以上50nm以下。
