本发明属于农药技术领域,特别涉及瑞香狼毒中3,7,11-三甲基-2,6,10-十二碳三烯-1-醇在农药中的用途。
背景技术
蚕豆蚜(Aphis craccivaro)是蚜虫的一个种类,分布于除西藏外的全国各地,属于半翅目,蚜科,寄主为蚕豆、菜豆、豌豆等豆科作物。其危害特点是:成虫和若虫刺吸嫩叶、嫩茎、花和豆荚的汁液,使叶片卷缩发黄,嫩荚变黄,严重时影响生长,造成作物减产。粘虫(Leucania separata)属于鳞翅目,夜蛾科,分布于全国各地,寄主为麦、稻、粟、玉米等禾谷类粮食作物及棉花、豆类、蔬菜等16科104种植物上。其危害特点是:幼虫食叶,可将作物叶片全部食光,造成严重损失。因其具有群聚性、迁飞性、杂食性、暴食性,已成为全国性重要农业害虫。
瑞香狼毒(Stellera chamaejasme L.)为瑞香科狼毒属植物,又名狼毒、断肠草,广泛分布于我国西北、西南等高原草地,是一种重要的多年生有毒草本植物,对畜牧业的发展和草原保护有着严重的危害,是需要防除的对象。但它又有可利用的一面,其根在中药中称为狼毒,对某些疾病如结肠癌和胃癌等有一定的治疗效果。最新研究还发现它具有抗病毒特别是HIV病毒的作用,对某些害虫也有一定的毒杀活性。因此,国内外研究人员对瑞香狼毒的化学成分和生物活性进行了广泛而深入的研究并从中分离和鉴定了多种成分。到目前为止,分离得到的物质主要包括下列四类:(1)黄酮类;(2)香豆素类;(3)二萜类;(4)木脂素类,但未见有分离出法呢醇的报道。
近年来,对瑞香狼毒的杀虫活性进行了广泛的研究并申请了不少的专利。张杰克等(专利申请号02133769.1)发明的狼毒素水乳剂对蚜虫和粘虫等有较好的毒杀效果,但该制剂的杀虫活性成分为新狼毒素A。刘世贵等(专利申请号02133971.6)发明的狼毒酮杀虫剂对多种害虫有较好的毒杀效果,但其有效成分为狼毒素B。侯太平等(专利申请号200510021333.3)公开了1,5-二苯基-3-羟基-1-戊酮和4-羟基倍半萜烯丙酯的杀虫活性,但其作用靶标为菜青虫。韩明理(专利申请号02100771.3)发明的狼毒素杀虫剂虽对禾谷缢管蚜、小菜蛾、菜青虫、桃蚜等害虫均有较好的毒杀活性,但其活性成分不清楚,因而不利于活性成分的人工合成和杀虫机理的研究,也不利于制剂的质量控制。
法呢醇广泛存在于天然植物的精油中,可以用分离提取的方法得到,也可以化学合成获得。其用途主要为:昆虫保幼激素和雄性性激素引诱剂;精神抑制药和皮肤保护的外用药;高级香精、香料和化妆品的配制;抗菌、杀菌、防霉、除臭等。例如,胜山雅子(专利号ZL00106930.6)发明了含有法呢醇和木糖醇的抗菌组合物;罗纳德·H·巴特利特等(专利号ZL87101769.5)公开了含有法呢醇和橙花叔醇的杀螨组合物,但到目前为止未见有法呢醇对蚕豆蚜和粘虫具有毒杀活性的报道。
发明内容
本发明所要解决的第一个技术问题是提供瑞香狼毒中3,7,11-三甲基-2,6,10-十二碳三烯-1-醇(结构如式I所示)在农药中的应用。

本发明通过生物活性测定结果表明法呢醇对两种不同的害虫(蚕豆蚜和粘虫)均有较好的毒杀效果,其24小时的LC50分别为20.2mg·L-1和15.2mg·L-1。
本发明还提供了法呢醇在制备杀灭蚕豆蚜的药剂中的用途,药剂中法呢醇的浓度范围为100~200mg·L-1(害虫的死亡率在80%以上)。以及法呢醇在制备杀灭粘虫的药剂中的用途,药剂中法呢醇的浓度范围为50~100mg·L-1(害虫的死亡率在80%以上)。
本发明所要解决的第二个技术问题是提供一种杀虫剂,它是以法呢醇为活性成分制备而成的,杀虫剂中法呢醇的浓度范围为50~200mg·L-1(害虫的死亡率在80%以上)。
本发明的法呢醇结构简单,化学合成容易,可以通过市场购买获得,是一种新型、高效、低毒、对非靶标生物安全的杀虫剂活性成分。

具体实施方式
本发明以蚕豆蚜为靶标,采用生物活性跟踪法分离得到了一个对蚕豆蚜和粘虫均有较好毒杀活性的天然产物-法呢醇,这是一种从天然植物中发现具有杀虫活性物质的行之有效的方法(如图1所示)。具体为将狼毒根粉置于索氏提取器中,加8倍样品量的石油醚(60~90℃),在70℃下提取8小时,减压过滤并浓缩滤液得粗提物,然后对粗提物进行大柱初分,得到7个活性馏分,再对杀虫活性第二强的馏分(F6)进行中柱细分又得到5个活性馏分,然后对毒杀活性最高的馏分(F63)进行反相硅胶柱分离得到了一个纯化合物,再对其进行结构鉴定和杀虫谱测定。在整个分离纯化过程中,以蚕豆蚜为靶标对粗提物和柱层析得到的各馏分进行毒力测定。
以下结合实施例对本发明作进一步的阐述。实施例仅用于说明本发明,而不是以任何方式来限制本发明。
实施例1法呢醇的提取
(1)将瑞香狼毒根洗净,切成碎片,放入鼓风式烘箱内于60℃下烘2天,用粉碎机粉碎成粉末,保存于冰箱内备用。
(2)将步骤(1)制得的瑞香狼毒根粉置于索氏提取器中,加入8倍重量的石油醚(60~90℃),于70℃提取8小时,减压过滤并浓缩滤液至半固体状,得粗提物。
(3)大柱粗分
柱径:6cm;柱高:40cm;固定相:硅胶H(200~300目);常规湿法装柱。以粗提物为分离样品,每次上样量10g,流速10ml·min-1,梯度洗脱(体积比为5∶1的石油醚-乙酸乙酯,用量约为1500ml和体积比为1∶1的石油醚-乙酸乙酯,用量约为1500ml),每段收集100ml,经薄层检测后合并相同的馏分,以蚕豆蚜为靶标进行生物活性测定得到7个具有杀虫活性的馏分(F1~F7)。再对杀虫活性第二强的馏分(F6)作进一步的分离提纯。
(4)中柱细分
柱径:3cm;柱高:40cm;固定相:硅胶H(200~300目);常规湿法装柱。以步骤(3)得到的馏分(F6)为分离样品,上样量6g,流速10ml·min-1,梯度洗脱(体积比为8∶1的乙酸乙酯-甲醇,用量约为1500ml和体积比为1∶1的乙酸乙酯-甲醇,用量约为1000ml),每段收集100ml,经薄层检测后合并相同的馏分,以蚕豆蚜为靶标进行生物活性测定得到5个具有杀虫活性的馏分。杀虫活性最强的馏分(F63)作进一步的分离提纯。
(5)反相硅胶柱分离
柱径:2cm;柱高:30cm;固定相为反相硅胶Lichroprep?RP-18;流动相为甲醇-水(体积比为8∶1);用水常规湿法装柱。以步骤(4)得到的F63馏分为分离样品,上样量2g,流速5ml·min-1,每段收集100ml,经薄层检测后合并相同的馏分,浓缩后得到的产品为无色油状液体,溶于二甲亚砜、甲醇、乙醇等溶剂。产品检测数据如下:

分析上述实验数据可知,EI-MS给出了分子离子峰M+为222,经高分辨质谱推断其化学式为C15H26O。
红外光谱中,3322cm-1出现强而宽的吸收峰表明有羟基存在;2964cm-1、2920cm-1和2856cm-1表示有甲基或亚甲基存在;1666cm-1和831cm-1表示有三取代烯烃存在;1445cm-1表示有饱和亚甲基存在;1375cm-1表示有甲基存在;1000cm-1表示有伯醇存在。
1H NMR谱中,δ3.91为1位上的2个H,它和2位上的H、羟基上的H相互偶合裂分为多重峰;δ5.26为2位上的H,它和1位上的H相互偶合裂分为多重峰;δ1.91-2.06为4,5,8,9位上的8个H,它们和邻位上的H相互偶合裂分为多重峰;δ5.08为6位上的H,它和邻位上的H相互偶合裂分为多重峰;δ5.04为10位上的H,它和邻位上的H相互偶合裂分为多重峰;δ1.65为12位上的3个H;δ1.62为13位上的3个H;δ1.56为14位上的3个H;δ1.55为15位上的3个H;δ4.37为羟基上的H。
13C NMR谱中有15个不同的吸收蜂,表明有15种不同化学环境的碳存在,这与高分辨质谱测得的化学式中的碳数是相同的。
上述实验数据与法呢醇的文献值进行了对比,结果基本一致,因此本实施例分离得到的杀虫活性最强的馏分为法呢醇,结构如式2所示:

本实施例步骤(3)、(4)分离纯化过程中各馏分对蚕豆蚜的毒力测定
分别将各馏分浓缩后溶解于二甲亚砜,然后每个馏分用重量百分比浓度为O.1%的吐温-80水溶液稀释成5种不同浓度的待测溶液。并以空白溶剂为阴性对照。
采集虫口密度大的蚕豆叶,仔细挑选使健康的无翅成蚜(30~50头)留在叶面上,分别将带虫的蚕豆叶浸入待测溶液5s后拿开,沥干,将叶上的蚕豆蚜转入装有新鲜蚕豆叶的培养皿中,放置在温度为22~24℃,相对湿度为60%,光照时间为14:10h的实验室中继续饲养,24h后记录死亡情况,每一浓度的待测溶液重复实验3次并按Abbott公式对死亡率进行校正。测定结果如表1所示:

实施例2法呢醇对蚕豆蚜的毒力测定
以超高效杀虫剂吡虫啉为比较标准,按实施例1所述的毒力测定方法分别对法呢醇和吡虫啉进行蚕豆蚜室内毒力测定,再根据几率值分析法,采用SPSS软件计算LC50值及其置信区间,并求出相关系数和毒力回归方程,结果见表2(本实施例所用的法呢醇可以市场购买所得,也可以通过实施例1记载的方法制得)。
结果显示,法呢醇对蚕豆蚜有较好的毒杀活性,其LC50值为20.2mg·L-1,而且浓度与药效间存在着显著的相关关系,其相关系数为0.9716。卡方X2=2.925,df=3,p>0.05)检验表明测定结果是可靠的。
另外,实验中观察到蚕豆叶浸入药液后并未显示出任何异常,这表明法呢醇对其没有药害。

实施例3法呢醇对粘虫的毒力测定
以高效氯氰菊酯为比较标准,分别将法呢醇、氯氰菊酯溶解于二甲亚砜并用重量百分比为0.1%的吐温-80水溶液稀释成5种不同浓度的待测溶液。将玉米叶片剪成2X5cm的小段,在待测溶液中浸5s后拿开,沥干后放入底部铺有滤纸的培养皿(6cm)中,接入10头3龄幼虫,再将其放置在温度为22~24℃,相对湿度为60%,光照时间为14:10h的实验室中继续饲养,24h后记录死亡情况,每个浓度的待测溶液重复实验3次(本实施例所用的法呢醇可以是市场购买所得,也可以通过实施例1记载的方法制得)。
按Abbott公式对死亡率进行校正,再根据几率值分析法,采用SPSS软件计算LC50值及其置信区间,并求出相关系数和毒力回归方程,结果如表3所示。

另外,实验中观察到玉米叶浸入药液后并未显示出任何异常,这表明法呢醇对其没有药害。

1.瑞香狼毒中3,7,11-三甲基-2,6,10-十二碳三烯-1-醇在制备杀灭蚕豆蚜的药剂中的用途。
2.根据权利要求1所述的用途,其特征在于:药剂中3,7,11-三甲基-2,6,10-十二碳三烯-1-醇的浓度范围为100~200mg·L-1。
