发明领域
本发明涉及包含编码IL-15及其片段的密码子优化的核酸序列的核酸分子、改良的疫苗、预防性和/或治疗性免疫接种个体抵御免疫原的改良方法,和改良的免疫治疗组合物及改良的免疫治疗方法。
发明背景
免疫治疗指调节人的免疫应答反应从而赋予所需的疗效。免疫治疗剂指给予个体后能调节个体的免疫系统,足以最终减轻不良免疫应答反应相关症状,或通过增强所需的免疫应答而最终减轻症状的那些组合物。在一些情况中,所述免疫治疗是疫苗接种方案的一部分,给予个体疫苗使个体与免疫原接触,进而使该个体产生针对该免疫原的免疫应答,在这些情况中,免疫治疗剂可增强免疫应答和/或选择性增强免疫应答的某一部分(例如细胞应答或体液应答),这是治疗或预防具体病症、感染或疾病所需的。

本发明一些方面提供的组合物和方法能预防性和/或治疗性免疫接种个体以抵御病原体或异常的疾病相关细胞。所述疫苗可以是任何类型的疫苗,例如减毒活疫苗、细胞疫苗、重组疫苗或核酸或DNA疫苗。
本发明涉及用于递送免疫调节蛋白质的组合物和这些组合物的使用方法。
可采用几种熟知的技术,包括DNA注射(也称为DNA疫苗接种)、重组载体,如重组腺病毒、重组腺相关病毒和重组牛痘病毒来递送所述核酸分子。

给药途径包括但不限于:肌肉内、鼻内、腹膜内、真皮内、皮下、静脉内、动脉内、眼内和口服以及局部、透皮、吸入或栓剂或递送给粘膜组织,例如通过灌洗递送至阴道、直肠、尿道、口腔和舌下组织。优选的给药途径包括给予粘膜组织、肌肉内、腹膜内、真皮内和皮下注射。可通过以下方式,包括但不限于:常规注射器、无针注射装置或“微粒轰击基因枪”给予所述基因构建物。
当所述基因构建物被细胞摄取时,可作为功能性染色体外分子维持存在于细胞中或整合入细胞的染色体DNA中。可将DNA以一种或多种质粒形式引入细胞,作为分离的遗传物质而维持在细胞中。或者,可将能整合入染色体的线性DNA引入细胞。当将DNA引入细胞时,可加入试剂以促进DNA整合入染色体。还可在DNA分子内包含用于促进整合的DNA序列。或者,可将RNA引入细胞。也考虑了提供的基因构建物为含有着丝粒、端粒和复制起点的线性小染色体。基因构建物可保留减毒活微生物或重组微生物载体在细胞内存活(所需)的遗传物质部分。基因构建物可以是重组病毒疫苗的基因组部分,此时所述遗传物质或整合入细胞染色体中,或维持存在于染色体外。基因构建物包含核酸分子基因表达所需的调控元件。这些元件包括:启动子、起始密码子、终止密码子和聚腺苷酸化信号。此外,为了基因表达编码靶蛋白或免疫调节蛋白的序列,常需要增强子。这些元件必须与所需蛋白的编码序列操作性相连,而且这些调控元件必须能在所给予的个体内可操作。

用于实施本发明,尤其是用于产生人用基因疫苗的启动子例子包括但不限于:猿病毒40(SV40)的启动子、小鼠乳腺瘤病毒(MMTV)启动子、人免疫缺陷病毒(HIV)的启动子如HIV长末端重复序列(LTR)启动子、莫洛尼病毒启动子、ALV启动子、巨细胞病毒(CMV)的启动子例如CMV立即早期启动子、EB病毒启动子、罗斯肉瘤病毒(RSV)启动子以及例如人肌动蛋白、人肌球蛋白、人血红蛋白、人肌肉肌酸和人金属硫蛋白等人基因的启动子。
用于实施本发明,尤其是用于产生人用基因疫苗的聚腺苷酸化信号的例子包括但不限于:SV40聚腺苷酸化信号和LTR聚腺苷酸化信号。具体地说,可采用加利福尼亚州圣迭戈市英杰公司(Invitrogen,San Diego,CA)的pCEP4质粒中的SV40聚腺苷酸化信号,称为SV40聚腺苷酸化信号。
除了DNA表达所需的调控元件外,DNA分子中还可包含其它元件。这些其它元件包括增强子。增强子可选自但不限于:人肌动蛋白、人肌球蛋白、人血红蛋白、人肌肉肌酸和诸如CMV、RSV和EBV等病毒的增强子。
为了使所述构建物维持存在于染色体外并在细胞内产生该构建物的多个拷贝,可提供含哺乳动物复制起点的基因构建物。加利福尼亚州圣迭戈市英杰公司的质粒pVAX1、pCEP4和pREP4含有EB病毒复制起点和核抗原EBNA-1编码区,它们能产生高拷贝附加型复制产物而不整合。

为了最大程度产生蛋白质,可选择调控序列使其充分适合于在给予该构建物的细胞内表达基因。此外,可选择能在细胞内最有效转录的密码子。本领域普通技术人员能够制备在细胞内具有功能的DNA构建物。
在一些实施方式中,可提供基因构建物以产生连接有IgE信号肽的本文所述免疫调节蛋白质的编码序列。
本发明的一种方法包括通过以下途径给予核酸分子的步骤:肌肉内、鼻内、腹膜内、皮下、真皮内或局部给予,或通过灌洗给予选自呼吸道、阴道、直肠、尿道、口腔和舌下的粘膜组织。

可根据所采用的给药方式配制本发明的药物组合物。如果药物组合物是可注射的药物组合物,它们是无菌、无热原和不含颗粒的。优选采用等渗制剂。等渗添加剂一般包括氯化钠、葡萄糖、甘露糖、山梨糖和乳糖。在一些情况下,优选等渗溶液,如磷酸缓冲盐水。稳定剂包括明胶和白蛋白。在一些实施方式中,在所述制剂中加入血管收缩剂。

所述基因构建物可含有操作性连接于基因表达所需调节元件的编码靶蛋白质或免疫调节蛋白质的核苷酸序列。根据本发明,提供的基因构建物组合包括含有编码靶蛋白质的可表达形式核苷酸序列的构建物和含有编码免疫调节蛋白的可表达形式核苷酸序列的构建物。将含有基因构建物组合的DNA或RNA分子掺入活细胞中可导致该DNA或RNA表达,而产生靶蛋白质及一种或多种免疫调节蛋白。结果增强了抵御该靶蛋白质的免疫应答反应。
可采用本发明来免疫接种个体以抵御所有的病原体,例如病毒、原核生物和致病性真核生物,如单细胞致病生物或多细胞寄生虫(的感染)。本发明特别可用于免疫接种个体以抵御能感染细胞和无衣壳的那些病原体,例如病毒和原核微生物,例如淋球菌、李斯特菌属和志贺菌属那些原核病原体。此外,本发明还可用于免疫接种个体以抵御原生动物病原体,这类病原体在其生活周期中的某一阶段为胞内病原体。表1提供了可用于制备本发明疫苗的一些病毒家族和种属的名单。疫苗中可采用含有编码各肽的DNA序列的DNA构建物,所述肽含有至少一个与例如表中所列那些病原体抗原所展示表位相同的或基本上相似的表位。此外,本发明还可用于免疫接种个体以抵御其他病原体,包括原核病原体和真核原生动物病原体以及多细胞寄生虫,如表2所列。
为制备基因疫苗以提供保护力免遭病原体感染,必须在基因构建物中包括编码免疫原性蛋白质的基因物质,例如靶蛋白的编码序列,以引发对其的保护性免疫应答反应。不论病原体是胞内感染(本发明尤其适用)或胞外感染病原体,不是所有的病原体抗原能都引发保护性应答反应。因为DNA和RNA均较小,而制备比较容易,本发明还提供了采用多种病原体抗原进行疫苗接种的额外优点。用于基因疫苗的基因构建物可包含编码多种病原体抗原的基因物质。例如,一个构建物中可包含数种病毒基因,从而提供多种靶抗原。
表1和表2包括一些致病因子和生物的名单,可以制备它们的基因疫苗以保护个体免遭其感染。在一些优选的实施方式中,免疫接种个体抵御病原体的方法针对HIV、HTLV或HBV。
本发明另一方面,提供了赋予抵御过度增殖性疾病特征性的过度增殖性细胞的保护性免疫应答反应的方法,以及治疗患过度增殖性疾病个体的方法。过度增殖性疾病的例子包括所有形式的癌症和银屑病。
业已发现,将包含编码免疫原性“过度增殖细胞”相关蛋白的核苷酸序列的基因构建物引入个体细胞中能导致在个体的疫苗接种细胞内产生这些蛋白质。为了免疫接种以抵御过度增殖性疾病,可给予个体包含编码过度增殖性疾病相关蛋白的核苷酸序列的基因构建物。
为使过度增殖相关蛋白成为有效的靶免疫原,其必须是只产生在过度增殖细胞内,或在过度增殖细胞内产生的水平高于正常细胞。靶抗原包括这样的蛋白质、其片段和肽,它们包含在上述蛋白中发现的至少一个表位。在一些情况中,过度增殖相关蛋白质是编码某蛋白的基团发生突变的产物。突变基因编码的蛋白质与正常蛋白几乎相同,除了其氨基酸序列略有不同因而产生了正常蛋白质上没有的不同表位。这类靶蛋白包括由例如myb、myc、fyn等原癌基因和转座基因bcr/abl、ras、src、P53、neu、trk和EGRF编码的那些蛋白质。除了作为靶抗原的原癌基因产物外,用于抗癌治疗和保护性方案的靶蛋白还包括B细胞淋巴瘤所产生抗体的可变区和T细胞淋巴瘤的T细胞受体可变区,在一些实施方案中,它们也可用作自身免疫疾病的靶抗原。还可采用其它肿瘤相关蛋白作为靶蛋白,例如发现的肿瘤细胞内含量较高的蛋白质,包括单克隆抗体17-IA所识别的蛋白质和叶酸结合蛋白质或PSA。
虽然可采用本发明来免疫接种个体以抵御几种癌症形式中的一种或多种,但本发明尤其适用于预防性免疫接种易患某种特定癌症或曾患癌症因而易复发的个体。遗传学和技术以及流行病学的发展已能测定和评估个体发生癌症的几率和风险。可利用基因筛检和/或家族健康史来预测某特定个体患几种癌症中任何一种的可能性。
类似地,那些曾患癌症和已接受治疗切除癌瘤或处于缓解中的个体尤其易复发。作为治疗方案的一部分,可以针对已诊断(明确)的癌症给予这种个体免疫接种以抵御复发。因此,一旦得知某个体曾患有某类癌症并有复发危险时,可免疫接种他们,使其免疫系统作好准备以抵御任何将出现的癌症。
本发明提供治疗患过度增殖性疾病个体的方法。在这些方法中,引入的基因构建物用作免疫治疗剂,以指导和促进个体的免疫系统抵御产生靶蛋白的过度增殖细胞。
本发明提供了治疗自身免疫疾病和病症个体患者的方法,该方法可赋予对抗自身免疫相关靶抗原,包括细胞受体和能产生针对“自身抗原”的抗体细胞的广泛保护性基础免疫应答反应。
T细胞介导的自身免疫疾病包括类风湿关节炎(RA)、多发性硬化症(MS)、斯耶格伦综合征(Sjogren syndrome)、肉样瘤病、胰岛素依赖性糖尿病(IDDM)、自身免疫甲状腺炎、反应性关节炎、强直性脊柱炎、硬皮病、多肌炎、皮肌炎、银屑病、脉管炎、韦格纳氏肉芽肿病、克罗恩病和溃疡性结肠炎。上述各种疾病的特征是T细胞受体结合内源性抗原,进而引发自身免疫疾病相关的炎性级联反应。针对T细胞(受体)可变区的疫苗接种将引发包括CTL在内的免疫应答而消除那些T细胞。

在硬皮病中,已特征鉴定到参与该疾病的TCR的几个特异性可变区。这些TCR包括:Vβ-6、Vβ-8、Vβ-14和Vα-16、Vα-3C、Vα-7、Vα-14、Vα-15、Vα-16、Vα-28和Vα-12。因此,可用编码这些蛋白中至少一种的DNA构建物作为疫苗接种,以引发靶向参与硬皮病的T细胞的免疫应答。
为治疗T细胞介导的自身免疫疾病患者,尤其是TCR可变区尚未特征鉴定的患者,可以进行滑膜活检。可提取含T细胞的样品,用标准技术鉴定那些TCR的可变区。利用这些信息制备基因疫苗。
B细胞介导的自身免疫疾病包括狼疮(SLE)、格雷夫斯病、重症肌无力、自身免疫溶血性贫血、自身免疫血小板减少症、哮喘、冷球蛋白血症、原发性胆管纤维硬化(primary biliary sclerosis)和恶性贫血。这些疾病各自的特征是抗体结合内源性抗原,进而引发自身免疫疾病相关的炎性级联反应。针对抗体的这种可变区的疫苗接种,可引发包括CTL在内的免疫应答以消除产生这种抗体的B细胞。
为治疗B细胞介导的自身免疫疾病患者,必须鉴定参与自身免疫活动的抗体的可变区。可进行活检,提取炎症部位存在的抗体样品。用标准技术鉴定那些抗体的可变区。利用该信息制备基因疫苗。
在SLE病例中,认为一种抗原是DNA。因此,可以筛检待免疫接种以抵御SLE的患者血清中的抗DAN抗体,制备包含编码在其血清中发现的抗DNA抗体可变区的DNA构建物的疫苗。

7.如权利要求1-6所述的核酸分子,其特征在于,该分子还包含免疫原的编码序列。
8.如权利要求7所述的核酸分子,其特征在于,所述免疫原是病原体抗原、癌症相关抗原或与自身免疫疾病相关细胞相连的抗原。
9.如权利要求8所述的分离的核酸分子,其特征在于,所述免疫原是选自HIV、HSV、HCV和WNV构成之组的病原体的病原体抗原。
10.如权利要求1-9所述的核酸分子,其特征在于,所述分离的核酸分子是质粒。
11.如权利要求1-9所述的核酸分子,其特征在于,所述分子掺入在病毒载体中。
12.如权利要求1-9所述的核酸分子,其特征在于,所述分子掺入在减毒活病原体中。
13.一种包含权利要求1-12所述核酸分子和含有免疫原编码核酸序列的核酸分子的组合物。

17.一种可注射的药物组合物,其包含权利要求1-12所述的核酸分子或权利要求13-15所述的组合物。
18.一种调节个体的免疫应答反应的方法,该方法包括给予所述个体权利要求1-6所述的核酸分子。
19.一种重组疫苗,其包含编码免疫原的核酸序列和权利要求1-6所述的核酸序列。
20.如权利要求19所述的重组疫苗,其特征在于,所述免疫原是病原体抗原、癌症相关抗原或与自身免疫疾病相关细胞相连的抗原。
21.如权利要求20所述的重组疫苗,其特征在于,所述免疫原是选自HIV、HSV、HCV和WNV构成之组的病原体的病原体抗原。
22.如权利要求19-21所述的重组疫苗,其特征在于,所述重组疫苗是重组牛痘疫苗。
23.一种诱导个体抵御免疫原的免疫应答反应的方法,该方法包括给予所述个体权利要求7-12所述的核酸分子或权利要求13-16所述的组合物或权利要求19-22所述的重组疫苗。
