适用于本发明中的动物消化物可以是本领域技术人员已知的任何动物消化物,例如从化学和/或酶水解干净并且未分解的动物组织得到的任何材料。适用于本发明的干啤酒酵母可以是本领域技术人员已知的任何干啤酒酵母,例如作为酿造工业副产品的干燥的、无活性的试剂。已经发现动物消化物和干啤酒酵母可以增强膳食补充剂或含有膳食补充剂的食物或其它组合物的可口性。存在于补充剂中时,动物消化物占膳食补充剂的约10%到约90%,干啤酒酵母占膳食补充剂的约1%到约30%。
维生素C、维生素E、β胡萝卜素、蛋白锌、蛋白锰、硫酸亚铁、蛋白铜、碘酸钙和亚硒酸钠是技术人员已知的化合物。已经发现这些化合物增强免疫并增进益生菌在本发明的膳食补充剂中的作用。已经发现与不在组合物中的益生菌相比,所有这些化合物、动物消化物和干啤酒酵母的组合可以延长益生菌的寿命。存在于补充剂中时,膳食补充剂包含从约0.1%到约10%的维生素C、从约0.1%到约10%的维生素E、从约0.01%到约3%的β胡萝卜素、从约0.1%到约5%的蛋白锌、从约0.01%到约3%的蛋白锰、从约0.01%到约3%的硫酸亚铁、从约0.01%到约1%的蛋白铜、从约0.01%到约1%的碘酸钙,和从约0.001%到约0.1%的亚硒酸钠。在另一实施方案中,膳食补充剂还含有从约0.1%到约2%的牛磺酸。
膳食补充剂可以被配制为多种制剂,如粉末、颗粒、丸(pellet),或任何其它合适的递送形式。在优选的实施方案中,膳食补充剂制剂是含有在生物聚合物基质中被微囊化的益生菌的粉末。粉末膳食补充剂可以被洒在食物或其它组合物上或以其它方式应用于食物或其它组合物并与之混合,所述食物或其它组合物尤其是宠物食物如犬食或猫食。粉末膳食补充剂可以被特别地配制为用于特定动物如陪伴动物的食用。在一个实施方案中,粉末膳食补充剂包含高浓度的益生菌,使得补充剂可以被小量施用给动物,或者可以在施用给动物之前稀释。本领域技术人员可以设计其它施用途径,如单独提供膳食补充剂,或者在宠物零食(pet treat)之中、之上或与宠物零食一起喂食。
膳食补充剂可以经配制以在室温下长期储存,或者可以被冷藏、冻干或冷冻。优选地,膳食补充剂储存于室温,且在室温下储存时膳食补充剂中的益生菌至少22天保持基本上是活的。预测性模拟显示即使在储存温度中有周期性的短期温度尖峰(spike),益生菌仍基本上是活的,所述尖峰可 超过室温高达60℃。
一般而言,以下述用量对动物施用益生菌,用于肠内微生物区系的健康维持:每天从约10亿到约200亿个菌落形成单位(CFU),优选每天从约1000万到约100亿个活的细菌。尽管可以应用更高浓度的益生菌,但是膳食补充剂可以被配制为每克补充剂含有从约105到约1010个菌落形成单位(CFU)。

在多个实施方案中,本发明的膳食补充剂还可包含从约15%到约60%的粗蛋白质。优选地,组合物包含约40%到约55%的粗蛋白质。粗蛋白质材料可包含植物性蛋白质如大豆、玉米、水稻、棉籽和花生,或动物性蛋白质如酪蛋白、白蛋白和肉蛋白质。适用于本文的肉蛋白质的非限制性实例包括猪肉、羔羊肉、马肉、家禽、鱼及其混合物。

为了增加膳食补充剂可以被储存的时间长度,膳食补充剂优选地具有按补充剂重量计在约2%和约10%之间的总水分含量。优选地,总水分含量按补充剂的重量计少于5%。类似地,膳食补充剂优选具有0.20到0.6范围内的水活度。优选地,水活度为少于0.55。

当膳食补充剂在高达50℃的温度下储存11天、高达60℃的温度下储存2天,或高达65℃的温度下储存20小时时,膳食补充剂中的益生菌,尤其是微囊化的益生菌,保持基本上是活的。

容器上或与容器相连的标签可以指出膳食补充剂的可用适应症。试剂盒也可任选地包含另外的容器,所述容器具有本文所述的另一补充成分或试剂。产品或试剂盒可包含使用膳食补充剂在陪伴动物如犬或猫中治疗多种病症或支持总体健康和/或康健的说明书。在一些实施方案中,多个容器被包装在一起成为纸板箱。例如,但不仅限于,纸板箱可包含等同于一周膳食补充剂供应的7个容器,或等同于一个月膳食补充剂供应的30个容器,等等。容器可装有单剂的补充剂(即单次使用),或多剂补充剂。容器还可以与宠物食品容器包装在一起,例如与犬食或猫食罐头、或干燥的犬食或猫食、或其它膳食补充剂如零食包装在一起。
本发明另一方面提供了下述食物组合物,所述食物组合物包含一种或多种适合被动物食用的成分和本发明的膳食补充剂。膳食补充剂可与所述成分混合产生食物组合物,或可以被施加于所述成分上以产生食物组合物。在一个实施方案中,通常通过喷洒或喷雾将膳食补充剂施加于食物组合物上。在一个优选的实施方案中,所述成分形成营养全面的陪伴动物食物,如犬或猫食,并优选地将膳食补充剂施加于食物上。通常将犬或猫食物置于合适的容器中,并将膳食补充剂施加于所述食物上,例如喷洒在食物上。
本发明另一方面提供了用于对动物施用益生菌的方法,所述方法包括对动物施用下述食物组合物,所述组合物包含一种或多种适于被动物食用的成分和本发明的膳食补充剂。
本发明另一方面提供了用于制造含益生菌的食物组合物的方法,所述方法包括将一种或多种适于动物食用的成分与本发明的膳食补充剂混合,或将本发明的膳食补充剂施加于食物组合物上。本发明再一方面提供了使用该方法制造的食物组合物。
本发明另一方面提供了用于促进动物的健康或康健的方法,所述方法包括对动物施用促进健康或康健的量的本发明膳食补充剂。在一个实施方案中,膳食补充剂与抗腹泻剂联合施用。
本发明另一方面提供了用于促进动物胃肠健康的方法,所述方法包括对动物施用促进胃肠健康的量的本发明膳食补充剂。在一个实施方案中,膳食补充剂与抗腹泻剂联合施用。
对这些方法而言,可以使用任何合适的方法施用补充剂,但是优选通过将补充剂与食物组合物混合或将补充剂施加于食物组合物上来施用,所述食物组合物优选全营养食物组合物。
为了促进健康和康健或促进胃肠健康,通过本发明的膳食补充剂以下述用量向动物施用益生菌,所述用量为每天从约10亿到约200亿个菌落形成单位(CFU),优选每天从约1000万到约100亿个活的细菌。
本发明另一方面提供了用于交流关于下述一个或多个内容的信息或说明的工具:(1)使用膳食补充剂对动物施用益生菌,尤其是为了维持或改善 胃肠健康,(2)将膳食补充剂与本发明的其它组分(食物组合物)混合,(3)对动物施用膳食补充剂,单独地或与本发明的其它成分组合地施用,和(4)使用本发明的试剂盒对动物施用益生菌,尤其是为了维持胃肠健康或促进动物的健康或康健,例如预防和治疗腹泻或用于改善粪便质量。该工具包括含有所述信息或说明的文件、数码存储介质、光学存储介质、音频显示(audio presentation)或视频显示。在某些实施方案中,交流工具是含有这类信息或说明的展示网站、可视化展示信息站(visual display kiosk)、宣传册、产品标签、包装说明书(package insert)、广告、传单、公告、录音带、录像带。DVD、CD-ROM、计算机可读芯片、计算机可读卡片、计算机可读盘片、计算机存储器或其组合。有用的信息包括下述之一种或多种:(1)用于组合和施用膳食补充剂和/或其它组分的方法和技术,和(2)供动物或它们的看护者对本发明及其用途有疑问时使用的联系信息。有用的说明包括用于混合的量和施用的量和频率。交流工具可用于说明使用本发明的益处和交流经批准的对动物施用本发明的方法。

实施例1
益生菌粉末混合过程
使用标准的干混装置制造用于犬和猫的益生菌粉末制剂,包括螺带混合机、v-混合机或双搅拌叶片混合机。混合时间是可变的,取决于使用的混合机。最大混合时间为10分钟。向混合机中添加益生菌组合物成分的顺序不是关键性的。混合后的粉末立即包装进用PVC套保护的大袋中或具有PVC衬里的袋子中。针对猫制备的制剂在表1中公开,针对犬的制剂在表2中公开。

实施例2
益生菌粉末包装
使用Cloud Packaging Solutions,Des Plains,II制造的专用包装设备将约1.15±0.1克的经混合的益生菌粉末包装进独立单次使用的容器中。如表3所列构建容器。容器尺寸为3″X 2″。容器由纸、低密度聚乙烯、箔和 Dupont 组成。WVTR和OTR在表3中给出。

粉末组合物中的益生菌稳定性
膳食补充剂包括使用屎肠球菌(SF68)作为代表性益生菌微生物的益生菌粉末组合物,所述微生物被微囊化于生物聚合物基质中。经包囊的组合物在五种不同的温度下储存多达96小时。所述温度为25℃、33℃、45℃、55℃和65℃。在从0时间开始于如表4所示的不同时间点采取来自各储存组合物的样品,并分析屎肠球菌的生活力。生活力测量的结果在表4中概括。
在粉末膳食补充剂中针对SF68观察到的结果和拟合的Arrhenius模型。该数据显示在四种温度(25℃、33℃、45℃、55℃和65℃)下短期储存期间观察和预测的屎肠球菌平均对数(计数)。使用的Arrhenius模型为Y=a+(b*小时)*(e(-k/temp)),其中系数a=9.3348;b=-73214.6和k=-914.7。

还在持续高温的条件下评价包囊化的组合物中益生菌生物的生活力。将使用屎肠球菌SF68的微囊化的益生菌组合物包装在如实施例2所述的容器中,并在45℃、50℃、55℃和60℃储存11天。一式四份进行实验。以对数菌落形成单位测量的结果展示于表5中。经七天的储存期未在50℃或以下的温度观察到显著的生活力丧失 。

表4和表5中所示结果单位是log cfu/克膳食补充剂。
在粉末膳食补充剂中针对SF68观察到的结果和拟合的Arrhenius模型。该数据显示在四种温度(45℃、50℃、55℃和60℃)下长期储存期间观察和预测的屎肠球菌平均对数(计数)。使用的Arrhenius模型为Y=a+(b*小时)*(e(-k/temp)),其中系数a=9.2249;b=-3.0488和k=-1187.4。
实施例4
犬和猫中益生菌粉末组合物的接受度
针对犬和猫特别地配制益生菌粉末组合物。如下施用组合物:在动物的干燥挤压成形的膳食上局部喷洒1.0克组合物或将1.0克组合物与干燥的挤压成形的膳食均匀混合。
连续数天评价三组犬,每组二十只。第一天第1组测量相对于对照而言对局部施加的益生菌组合物的偏好,第2组测量相对于对照而言对均匀混合的益生菌组合物的偏好,第3组测量相对于均匀混合的组合物而言对 局部施加的益生菌组合物的偏好。第二天颠倒膳食的放置。在各组中,在喂养时间给犬提供两种可能的食物选择,各含有700克无补充剂的膳食(对照)。如喂养方案中所述,在喂养犬之前添加补充剂。在食用期前对各碗食物称重。给予动物二十分钟进食。20分钟进食期后通过称重各碗食物(对照和有补充剂的)确定偏好。
连续数天评价三组猫,每组二十只。第一天第1组测量相对于对照而言对局部施加的益生菌组合物的偏好,第2组测量相对于对照而言对均匀混合的益生菌组合物的偏好,第3组测量相对于均匀混合的益生菌组合物而言对局部施加的益生菌组合物的偏好。第二天颠倒膳食的放置。在各组中,在喂养时间给猫提供两种可能的食物选择,各含有约100克无补充剂的膳食(对照)。如喂养方案中所述,在喂养猫之前添加补充剂。在进食期前对各碗食物称重。给予动物十八分钟进食。通过在自动秤上记录每份膳食重量的减少,确定偏好。
总起来说,含有益生菌制剂(喷洒或与膳食混合)的膳食显著地被猫偏好,或同等地或显著地被犬偏好。
在说明书中,公开了本发明一般的优选实施方案,并且虽然采用了特定术语,但其仅以一般性和描述性意义使用,而不用于限制目的,本发明的范围由权利要求说明。明显地,根据上述教导,本发明可以有许多修改和变化。因此,应当理解,在所附权利要的范围内,本发明可以以除具体描述之外的方式实施。
Claims (24)
1.膳食补充剂,其包含至少一种益生菌和动物消化物、维生素C;维生素E、β胡萝卜素、蛋白锌、蛋白锰、硫酸亚铁、蛋白铜、碘酸钙和亚硒酸钠,其中益生菌包括屎肠球菌。
2.权利要求1的补充剂,其中益生菌包括原核生物、真核生物或古细菌益生菌。
3.权利要求1的补充剂,其中益生菌包含至少一种任何适宜的肠球菌属、链球菌属、乳杆菌属、乳球菌属、双歧杆菌属或酵母属的菌株或亚种。
4.权利要求1的补充剂,其中屎肠球菌为保藏为NCIMB 10415的SF68。
5.权利要求1的补充剂,其还包含牛磺酸。
6.权利要求1的补充剂,其还包含益生元。
7.权利要求1的补充剂,其还包含抗腹泻剂。
8.权利要求1的补充剂,其中益生菌被微囊化。
9.权利要求8的膳食补充剂,其中微囊包含用紫胶包衣的生物聚合物基质。
10.权利要求1的膳食补充剂,其中膳食补充剂针对陪伴动物配制。
11.权利要求10的膳食补充剂,其中陪伴动物是猫或犬。
12.适用于对动物施用益生菌的试剂盒,所述试剂盒视试剂盒组分而定在单包装的分开容器中或在虚拟包装的分开容器中包含根据权利要求1-11之任一项的膳食补充剂和至少下述之一:(1)一种或多种适于动物食用的成分,(2)一种或多种益生元,(3)一种或多种抗腹泻剂,(4)如何将该膳食补充剂和其它试剂盒组分组合的说明书,和(5)如何使用该膳食补充剂和其它试剂盒组分的说明书。
13.食物组合物,其包含一种或多种适于动物食用的成分和权利要求1-11之任一项的膳食补充剂。
14.权利要求13的食物组合物,其中所述成分形成全营养陪伴动物食品。
15.权利要求13的食物组合物,其中膳食补充剂与所述成分混合。
16.权利要求13的食物组合物,其中膳食补充剂被施加于所述成分上。
17.权利要求1-11之任一项的膳食补充剂在制备对动物施用益生菌的食物组合物中的用途,其中所述食物组合物包含一种或多种适于动物食用的成分和所述膳食补充剂。
18.用于制造含益生菌的食物组合物的方法,所述方法包括将一种或多种适于动物食用的成分和权利要求1-11之任一项的膳食补充剂混合,或将权利要求1-11之任一项的膳食补充剂施加于食物组合物上。
19.权利要求1-11之任一项的膳食补充剂在制备用于促进动物健康或康健的试剂盒中的用途。
20.权利要求19的用途,其中通过将补充剂与下述食物组合物混合或将补充剂施加于所述食物组合物上施用膳食补充剂,所述食物组合物包含一种或多种适于动物食用的成分。
21.权利要求19的用途,其中膳食补充剂与一种或多种抗腹泻剂联合施用。
22.权利要求1-11之任一项的膳食补充剂在制备用于促进动物胃肠健康的试剂盒中的用途。
23.权利要求22的用途,其中通过将补充剂与下述食物组合物混合或将补充剂施加于所述食物组合物上施用膳食补充剂,所述食物组合物包含一种或多种适于动物食用的成分。
24.权利要求22的用途,其中膳食补充剂与一种或多种抗腹泻剂联合施用。
