Abstract

一种标本防腐固定液,属于防腐剂技术领域。本发明由有机酸、脂肪醇、胺、砜、有机胺盐和水组成,是一种用于人体、动物、植物组织标本防腐、固定的溶液,不含甲醛、乙醛等任何醛类化合物。本发明的标本防腐固定液特点为:(1)无毒,小鼠急性毒性灌胃剂量大于50,000mg/kg,优于世界卫生组织的产品微毒标准(5,000mg/kg),根据国际权威文献报道的食盐、甲醛安全性数据,本发明标本防腐固定液安全性比食盐、甲醛分别高16.7倍和1190倍;(2)防腐、固定性能优于福尔马林,用本发明标本防腐固定液处理的标本可长期存放,用于病理诊断固定时,组织细胞结构清晰、背景对比度高,免疫组化抗原免修复,简化了免疫组织化学切片制作程序。

技术领域
一种标本防腐固定液,属于防腐剂技术领域。本发明是一种用于人体、动物、植物组织标本防腐、固定的溶液,不含甲醛、乙醛等任何醛类化合物。
背景技术
福尔马林(formalin,甲醛稀释溶液)是最常用的人体、动物、植物组织标本防腐剂,福尔马林中的甲醛能与蛋白质的氨基结合,蛋白质发生不可逆变性,其用于人体、动物、植物组织标本的防腐已有一百多年的历史。在医院病理科、外科手术室,福尔马林用于病理组织标本的固定,以防止组织自溶、结构破坏、细胞变形,组织标本离体后用固定液固定,制成病理切片,进行医学病理学诊断。医学病理诊断需要高质量的病理切片,正确的标本固定是制成高质量切片的第一步。
福尔马林具有高毒性、致癌性,它的毒性为甲醇的30倍,长期接触福尔马林可能会引起慢性呼吸道疾病、癌症发生,如鼻咽癌、结肠癌、脑瘤、白血病,导致记忆力和智力下降,严重时可出现胃肠穿孔、肝脏损害甚至昏迷;长期接触福尔马林的人员,男性得癌症死亡的机率比非接触人员高6.8倍,女性高9倍。
发明内容
本发明的目的是提供一种标本防腐固定液,可全面替代福尔马林,用于人体、动物、植物组织标本防腐、固定,产品无毒,避免福尔马林对标本保存、解剖、病理诊断等场所工作人员的职业损害。
本发明的技术方案:一种标本防腐固定液,由有机酸、脂肪醇、胺、砜、有机胺盐和水组成;各组份的重量%含量为:
有机酸    0.2-10
脂肪醇    45-85
胺        0.5-5
砜        0.3-6
有机胺盐  0.3-3
水        10-35
所述的有机胺盐为:二氨基胍盐酸盐、2,4,5,6-四氨基嘧啶盐酸盐、吡啶鎓盐、氨基乙腈盐酸盐、6-硝基苯并咪唑硝酸盐、4-哌啶酮盐酸盐或丙二酸单乙酯钾盐。
其中有机酸的化学结构为:
化学结构1:CH3(CH2)nCOOH,n=0-8;
化学结构2:HOOC(CH2)nCOOH,n=0-8;
或化学结构3:

其中脂肪醇化学结构为:
化学结构4:CH3(CH2)n OH,n=1-8;
化学结构5:HO(CH2)nOH,n=2-8;
或多元醇:丙三醇、1,2,4-丁三醇、三羟甲基丙烷或季戊四醇。
其中胺为:乙胺、二乙胺、三乙胺、三丁胺、二异辛胺、乙醇胺、三乙醇胺或三羟甲基胺基甲烷。
其中砜为:环丁砜、二甲基砜、二乙烯基砜或二甲基亚砜。
本发明的有益效果:
(1)无毒。小鼠急性毒性灌胃剂量大于50,000mg/kg,优于世界卫生组织的产品微毒标准(5,000mg/kg),根据国际权威文献报道食盐、甲醛安全性数据,本发明标本防腐固定液安全性比食盐、甲醛分别高16.7倍和1190倍。
(2)防腐、固定性能优于福尔马林。用本发明标本防腐固定液处理的标本可长期存放,用于病理诊断固定时,组织细胞结构清晰、背景对比度高,免疫组化抗原免修复,简化了免疫组织化学切片制作程序。
具体实施方式
实施例1
标本防腐固定液的组成为丙酸0.2%、乙醇45%、三羟甲基胺基甲烷0.5%、二甲基砜0.3%、6-硝基苯并咪唑硝酸盐0.3%,其余为水。
实施例2
标本防腐固定液的组成为乙酸5%、丙三醇85%、三乙胺5%、环丁砜0.3%、二氨基胍盐酸盐0.3%,其余为水。
实施例3
标本防腐固定液的组成为苯甲酸10%、丙二醇50%、二异辛胺3%、二甲基亚砜6%、吡啶鎓盐3%,其余为水。
实施例4
标本防腐固定液的组成为乙二酸2%、1,2,4-丁三醇61%、三乙胺2.5%、二乙烯基砜3.5%、2,4,5,6-四氨基嘧啶盐酸盐1.8%,其余为水。
实施例5
标本防腐固定液的组成为丙二酸0.5%、季戊四醇47%、二乙胺3%、二甲基砜2.8%、氨基乙腈盐酸盐1%,其余为水。
实施例6
标本防腐固定液的组成为丙酸0.2%、乙醇70%、三羟甲基胺基甲烷0.5%、二甲基砜0.3%、4-哌啶酮盐酸盐1.5%,其余为水。
实施例7
标本防腐固定液的组成为苯乙酸0.8%、丙二醇60%、乙醇胺2.5%、二乙烯基砜1.5%、丙二酸单乙酯钾盐0.3%,其余为水。
实施例8
标本防腐固定液的组成为对甲氧基苯甲酸1%、乙醇50%、三丁胺3%、二甲基亚砜1.6%、吡啶鎓盐2%,其余为水。

1、一种标本防腐固定液,其特征为由有机酸、脂肪醇、胺、砜、有机胺盐和水组成;各组份的重量%含量为:
有机酸        0.2-10
脂肪醇        45-85
胺            0.5-5
砜            0.3-6
有机胺盐      0.3-3
水            10-35
所述的有机胺盐为:二氨基胍盐酸盐、2,4,5,6-四氨基嘧啶盐酸盐、吡啶鎓盐、氨基乙腈盐酸盐、6-硝基苯并咪唑硝酸盐或4-哌啶酮盐酸盐;
所述的有机酸的化学结构为:
化学结构1:CH3(CH2)nCOOH,n=0-8;
化学结构2:HOOC(CH2)nCOOH,n=0-8;
或化学结构3:

2、根据权利要求1所述的标本防腐固定液,其特征是有机酸选用:乙酸、丙酸、乙二酸、丙二酸、苯甲酸、苯乙酸或对甲氧基苯甲酸。
3、根据权利要求1所述的标本防腐固定液,其特征是脂肪醇选用:乙醇、丙二醇、丙三醇、1,2,4-丁三醇或季戊四醇。
