Abstract
The present invention is related to a device and kitfor pulsing a biological sample with a pulsing agent, subsequently stabilizing the biolog-ical sample so pulsed, which device or kit provides a control reaction. The invention has application in the fields of medical diagnostics, particularly re-lating to immunity.
Claims (37)
1. An assay device (30) for a liquid biological sample, which facilitates exposure of said sample separately to a first substance (2) and a control substance (35), and subsequently to a nucleic acid stabilizing agent (5), the device comprising:
- a first compartment (32) in which said first substance (2) is present, - a second compartment (34) in which said control substance (35) is present, - a third compartment (36) where said stabilizing agent (5) is present, and - a support (52) for a biological sample tube, wherein one or more of the compartments (32, 34, 36) is sealed and comprises one or more areas suitable for puncture by a hollow needle.
- a first compartment (32) in which said first substance (2) is present, - a second compartment (34) in which said control substance (35) is present, - a third compartment (36) where said stabilizing agent (5) is present, and - a support (52) for a biological sample tube, wherein one or more of the compartments (32, 34, 36) is sealed and comprises one or more areas suitable for puncture by a hollow needle.
2. Device according to claim 1, wherein said area is a re-sealable septum (40, 42, 46).

Device according to claim 1 or 2, further comprising a support (54, 56) for demountable attachment to a transfer-tubing (60), which transfer tubing comprises a hollow flexible or rigid tubing (64) disposed with a hollow needle (62, 66) at either end, adapted for transfer of liquid between any two compartments, or between a compartment and the biological sample tube.
4. Device according to claim 3, further comprising the transfer tubing (60) as defined in claim 3.

Device according to claim 3 or 4, wherein one needle (62) of the transfer tubing is attached in parallel alignment to the hollow needle (80) of a pressurising tubing (88), which pressurising tubing comprises a flexible hollow tubing (82) disposed at one end with said needle (80), and is adapted to apply vacuum or pressure to a compartment or biological sample tube to propel liquid through the transfer tubing.
6. Device according to any of claims 1 to 5, wherein said first substance (2) is immobilised on part or all of the inside surface of said first compartment (32).
7. Device according to any of claims 1 to 6, wherein one or more of the compartments (32, 34, 36) comprises an air vent.
8. Device according to any of claims 1 to 7, wherein the first (32) and/or second (34) compartments are held under negative pressure.
9. Device according to any of claims 1 to 8, wherein the first (32) and/or second (34) compartments comprise an indication for dispensing a known volume of stabilising agent therein.
10. Device according to any of claims 1 to 9, wherein said first substance (5) comprises one or more immune system antigens.
11. Device according to claim 10 wherein said immune system antigens are vaccine components.
12. Device according to claim 10 wherein said immune system antigens are antigens which provoke a hyperallergenic response.
13. Device according to claim 10 wherein said immune system antigens are one or more selected from histocompatibility antigens, bacterial LPS, tetanous toxoid, a cancer immunotherapy antigen, MAGE-3, a cat allergen, Feld1, antigen presenting cells from an organ donor, an autoantigen, GAD65.
14. Device according to any of claims 1 to 13 wherein said stabilizing agent (35) is an inhibitor of cellular RNA degradation and/or gene induction.
15. Device according to claim 14 wherein said inhibitor of cellular RNA
degradation and/or gene induction is that as found in a PAXgene.TM. or Tempus .TM. Blood RNA Tube.

16. A kit for assaying a liquid biological sample comprising:
a vessel (1) suitable for accepting liquid biological sample, exposing said sample to a first substance (2) and subsequently a nucleic acid stabilising agent, said vessel comprising:
a) a first substance (2) present inside said vessel (1), b) a container (4, 12, 16) in which said stabilising agent (5) is present, c) a connection between the inside of said vessel (1) and the inside of said container (4, 12, 16), d) a physical barrier (25, 15, 18) that temporarily blocks said connection;
and a control vessel (37, 37') suitable for accepting liquid biological sample, exposing said sample to a control substance and subsequently a nucleic acid stabilising agent, said control vessel comprising:
a) a control substance (35) present inside said control vessel (37, 37'), b) a control container (4', 12', 16') in which said stabilising agent (5') is present, c) a connection between the inside of said control vessel (37, 37') and the inside of said control container (4', 12', 16'), d) a physical barrier (15', 18', 25) that temporarily blocks said connection, wherein the vessel (1) and/or control vessel (37, 37') comprise one or more areas suitable for puncture by a syringe needle.
a vessel (1) suitable for accepting liquid biological sample, exposing said sample to a first substance (2) and subsequently a nucleic acid stabilising agent, said vessel comprising:
a) a first substance (2) present inside said vessel (1), b) a container (4, 12, 16) in which said stabilising agent (5) is present, c) a connection between the inside of said vessel (1) and the inside of said container (4, 12, 16), d) a physical barrier (25, 15, 18) that temporarily blocks said connection;
and a control vessel (37, 37') suitable for accepting liquid biological sample, exposing said sample to a control substance and subsequently a nucleic acid stabilising agent, said control vessel comprising:
a) a control substance (35) present inside said control vessel (37, 37'), b) a control container (4', 12', 16') in which said stabilising agent (5') is present, c) a connection between the inside of said control vessel (37, 37') and the inside of said control container (4', 12', 16'), d) a physical barrier (15', 18', 25) that temporarily blocks said connection, wherein the vessel (1) and/or control vessel (37, 37') comprise one or more areas suitable for puncture by a syringe needle.
17. Kit according to claim 16 wherein said first substance (2) is immobilised on part or all of the inside surface of said vessel (1).
18. Kit according to claim 16 wherein said first substance (2) is immobilised on a solid support.
19. Kit according to any of claims 16 to 18 wherein said first substance (2) is a liquid.
20. Kit according to any of claims 16 to 18 wherein said first substance (2) is a solid.
21. Kit according to any of claims 16 to 20 wherein said area is a re-sealable septum.
22. Kit according to any of claims 16 to 21, wherein the vessel (1) and/or control vessel (37, 37') comprise a fitting suitable for receiving a syringe and transmitting the contents therein to the interior of said vessel (1) or control vessel (37, 37').
23. Kit according to any of claims 16 to 22, wherein the vessel (1) and/or control vessel (37, 37') comprise a fitting suitable for receiving a syringe needle.
24. Kit according to any of claims 16 to 23, wherein the vessel (1) and/or control vessel (37, 37') comprise a valve which is capable of minimising the flow of gas/liquid from vessel, and allowing the flow of liquid biological sample into the vessel.
25. Kit according to any of claims 16 to 24, wherein the vessel (1) and/or control vessel (37, 37') comprise a means through which displaced gas may be expelled.
26. Kit according to any of claims 16 to 25, wherein the vessel (1) and/or control vessel (37, 37') are held under negative pressure.
27. Kit according to any of claims 16 to 26, wherein the physical barrier (15, 18, 25, 15', 18', 25') of item d) is opened by the application of physical force to said vessel or control vessel.
28. Kit according claim 27, wherein said force transmits an opening means to said physical barrier (15, 18, 25, 15', 18', 25').
29. Kit according to claim 27 wherein said force irreversibly opens said physical barrier (15, 18, 25, 15', 18', 25').
30. Kit according to any of claims 16 to 29, wherein said vessel (1) and/or control vessel (37, 37') comprises an indication for dispensing a known volume of stabilising agent therein.
31. Kit according to any of claims 16 to 30, wherein said first substance (2) comprises one or more immune system antigens.
32. Kit according to claim 30 wherein said immune system antigens are vaccine components.
33. Kit according to claim 30 wherein said immune system antigens are antigens which provoke a hyperallergenic response.
34. Kit according to claim 30 wherein said immune system antigens are one or more selected from histocompatibility antigens, bacterial LPS, tetanous toxoid, a cancer immunotherapy antigen, MAGE-3, a cat allergen, Feld1, antigen presenting cells from an organ donor, an autoantigen, and GAD65.
35. Kit according to any of claims 16 to 34, wherein said stabilising agent is an inhibitor of cellular RNA degradation and/or gene induction.
36. Kit according to any of claims 16 to 35, wherein said inhibitor of cellular RNA
degradation and/or gene induction is that as found in a PAXgene.TM. or Tempus .TM. Blood RNA Tube.

37. Kit according to any of claims 16 to 36, wherein the exterior of the vessel (1) and control vessel (37, 37') are joined to form a single entity.
