Abstract
Un método para determinar la actividad de las enzimas lisosomales presentes en muestras secas de fluidos corporales y de tejidos celulares tales como alfa-L-iduronidasa, beta-D-galactosidasa, beta-D-glucosidasa, citotriosidasa, alfa-D-galactosidasa total y alfa-D-galactosidasa A, hexosaminidasa A y B, alfa-D-manosidasa, beta-D-manosidasa, alfa-L-fucosidasa, N-acetil-alfa-galactosaminidasa, arilsulfatasa, esfingomielinasa, beta-galactocerebrosidasa, iduronato-2-sulfatasa y beta-D-glucuronidasa, entre otras. El método incluye: a) combinar una muestra seca de fluidos corporales o tejidos celulares conteniendo por lo menos un tipo de enzima lisosomal con: 1) un eluyente, 2) una solución amortiguadora de incubación y 3) un sustrato o sustratos capaces de reaccionar con las enzimas lisosomales ensayadas y de producir un producto enzimático correspondiente, b) permitir que la muestra seca de fluido corporal o tejido celular reaccione con el eluyente, la solución amortiguadora de incubación y el sustrato o sustratos durante un tiempo y temperatura adecuados, y c) detectar la mencionada enzima lisosomal. La presente también se refiere a kit de diagnóstico para la determinación de la actividad de las enzimas lisosomales.
