The present study was carried out to conduct drought tolerance in three wheat cultivars including susceptible (Gemmiza7) and tolerant (Sakha93 and Sahel1). Molecular characterization was done by 26 SSR markers located on chromosome7 which was associated with drought tolerance in many previous studies. 26 SSR markers were polymorphic and thus showed 100% polymorphism. The number of alleles per locus varied from 2 to 3 alleles with an average (2.62). The polymorphism information content (PIC) value ranged from 0.34 to 0.59, with a mean of 0.51. The discrimination power (Dp) value ranged between 0.67 and 0.78 with an average of 0.71 per locus and Heterozygosity (He) value varied from 0.44 to 0.67 with an average of 0.59. The genetic relationships estimated by the polymorphism of SSR markers revealed a greater level of genetic variability in wheat cultivars of wide adaptability and applicability. Whereas an average of combined probability value for the SSR markers was 6.15 x 10-16, sugg...

Two simple and sensitive spectrofluorimetric (method Ι) and spectrophotometric (method ΙΙ) methods have been developed for the determination of some chloride containing toothpastes and panthenol-containing cosmetic preparations respectively. Method Ι is based on quantitative fluorescence quenching of (terbium-salicylate-hexamine ternary complex) by fluoride which could be measured at λem/λex of 547nm/322nm. The ∆Fconcentration plot was rectilinear over the concentration range of 0.5-20 μg/ml. Method ΙΙ depends reaction of panthenol with nitrobenzoxadiazole chloride (NBD-Cl) and measuring the absorbance of the resultant product at 480nm. The absorbanceconcentration plot was rectilinear over the concentration range of 2-20 μg/ml

The knowledge of the genetic diversity of barley (Hordeum vulgare) genotypes based on protein polymorphism is very important for breeding programs. The purpose of the current study was to determine the genetic diversity and relationships among ten barley genotypes by using Sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) for protein profiles. A total number of 30 bands with molecular weights ranging from 12 to 148 KD were detected. Out of these, five bands were observed monomorphic. Rest of the bands had shown polymorphism to the extent of 83.3% among the test genotypes. The genetic similarity of the ten genotypes tested varied from 0.26 to 1.00 with an average of 0.51. Cluster analysis divided the ten genotypes into two major clusters comprising four subclusters, which was consistent with the systematic classification of barley done in previous studies. The results of this study indicated that the genotypes of barley could effectively be differentiated based on...

Two rapid, simple, sensitive, selective and economic derivative spectrophotometric (first [D1] and second [D2]) and synchronous spectrofluorimetric (FDSFS and SDSFS) methods have been developed for the analysis of fexofenadine hydrochloride (FXD) in the presence of its different degradation products. Derivative spectrophotometry (D1) was used to measure FXD at 223 nm in the presence of its alkaline or acidic degradation products, and at 211 nm in the presence of its oxidative degradation product. Derivative spectrophotometry (D2) was used to determine FXD at 217 nm in the presence of its alkaline or acidic degradation products, and at 215 nm in the presence of its oxidative degradation product; the UV degradation product was measured at 211 nm. Synchronous spectrofluorimetry (FDSFS) was used to measure FXD in the presence of its alkaline or acidic degradation products at 406 nm, and at 367 nm in the presence of its oxidative or UV degradation products. Synchronous spectrofluorimetry...

With a view to understanding the traits which can be used as a quick criteria for drought tolerance, field and laboratory experiments were used to evaluate nine wheat (Triticum aestivum L.) genotypes; seven local varieties with two introduced genotypes from International Center for Agricultural Research in the Dry Areas (ICARDA). The field experiment was grown under two water regimes (stress and non stress treatments). The stress treatment induced by withholding irrigation after emergence and giving two supplementary irrigations, one after 60 days post-sowing and the other after 90 days post-sowing and non stress (well-watered). Combined analysis of variance over two seasons showed highly significant differences among wheat genotypes in all the studied traits and water stress decreased them significantly. The superior genotypes 1,2 and 6 which gave higher relative water content (RWC) accumulated more free proline (Pro) and had lower drought susceptibility index (S) values, whereas g...

The lack of an effective evaluation method for salt tolerance in the screening process is one of the reasons for limited success in conventional salt tolerance breeding. This study was designed during winter 2006/2007 and 2007/2008 to identify useful agronomical and biochemical traits for salt tolerance in wheat. Five wheat cultivars were grown in field experiment in three sites at Sahel El Tenna region, south Port Said with various salinity levels; 0.098 ds m -1), 4.2 ds m -1 and 8.4 ds m -1. Salt stress suppressed seedling growth and simultaneously reduced leaf area, content chlorophyll, spike length, 1000 grain weight, grain yield as well as harvest index. Based on salt tolerance index, Gemmeiza 1 cultivar showed the highest and stable yield across normal and saline conditions. Significant correlations observed between grain yield and some agronomic traits, indicating that these traits might be useful criteria to select salt tolerant wheat cultivars.Gel Electrophoresis (SDS-PAGE)...
