    Background: Dental materials, in particular resins used in prosthetic, orthodontic, conservative and gnathological dentistry, are frequently in long-term contact with the tissues of the mouth, and the chronic exposure to these products of a significant proportion of the population means that the release of genotoxic substances by dental resins must be evaluated. Methods: For this purpose, ten methyl metacrylate-based resins, both cold- and hot-polymerising and all of frequent use in clinical practice, were prepared following the indications in Standard UNI 9582-2 now included in Standard ISO 10993. Genotoxicity was evaluated through the Tradescantia/micronuclei test. Results: By using the Tradescantia/micronuclei test, which detects aberrant chromosomes by detecting micronuclei in flower tetrads, nine resins were found not to the positive; one resin caused a significant increase in the frequency of micronuclei compared to negative controls. Conclusions: The results of the Tradescantia/micronuclei test, together with the results of tests cytotoxicity and the Ames and Allium cepa tests performed in previous research, indicate a need for further analyses of these resins for dental use, through other in vivo and in vitro tests, in order to achieve certainty over the risk connected with their use in human populations.

    The aim of this study was to evaluate the mutagenicity of several traditional detergents and that of newer more biodegradable detergents, by using a bacterial test (Ames test), a plant cell test (Allium cepa micronuclei test) and a human leucocyte test (Comet test). All tests were conducted using a wide range of doses (1-2000 mg/l). None of the examined detergents induced mutations in S.typhimurium. One traditional detergent showed a genotoxic effect with the A. cepa test, while all newer detergents and one traditional detergent were shown by the Comet test to be capable of inducing DNA damage.

    Air pollution has well-known harmful effects on human beings, causing both acute and chronic diseases. Some data suggest that genetic damage occurring early in life may influence the risk of having cancer and other chronic diseases in adulthood. Therefore, there is a growing interest in studying the genotoxic activity of air pollution, and especially particulate matter. The aim of this study was to analyze airborne particulate matter (PM10) collected in an industrialized town of Northern Italy. PM10 was sampled in six areas of the town, divided in three distinct dimensional classes (<0.5 μm; 0.5-3 μm and 3-10 μm), and analyzed for the quantification of polynuclear aromatic hydrocarbons and metals contents. A model-based approach using diagnostic ratios and toxic equivalency factor was also followed. Concurrently, biological assays were performed for the assessment of mutagenicity and genotoxicity in bacteria, human and plant cells. Mutagenicity was observed in bacteria and human cells, with a clear dose-response relationship, induced above all by the finest PM samples (PM0.5 and PM0.5-3), which contained the largest number of polycyclic aromatic hydrocarbons. DNA damage, such as chromosomal aberrations and micronuclei, was also found in Allium cepa root cells, but without a clear relationship with the tested doses. The in-vitro models utilized showed to be good indicators of air quality for mutagenicity. Chemical analyses evidenced high content of polycyclic aromatic hydrocarbons, metals and semi-metals in PM extracts. Polycyclic aromatic hydrocarbons, metals and mutagenicity can be ascribed mainly to vehicular traffic (in terms of both exhausted gases emission and mechanical losses), which represents a constant and ubiquitous source of human exposure, and to steel working, carried out within the urban area.

    The widespread use of detergents has raised concern with regard to the environmental pollution caused by their active ingredients, which are biorefractory, toxic and persistent. Since detergents are complex mixtures of different substances, in which synergistic effects may occur, we aimed to assess the mutagenicity of different detergent formulations, taking into account aquatic toxicity and ready biodegradability. We performed a ready biodegradability test (OECD 301 F), Daphnia magna and Vibrio fischeri toxicity tests, and mutagenicity tests (Salmonella/microsome test, Allium cepa test and comet assay). Six detergent formulations were examined, 3 pre-manufacture and 3 commercially available. All detergents presented ready biodegradability. EC50 values varied for all products, according to the marker organism used, but were always higher than the more stringent value considered for aquatic toxicity assessment (V. fischeri 10-60 mg/L; D. magna 25-300 mg/L; A. cepa 250-2000 mg/L). None of the detergents caused mutations in bacteria. However, one commercial ecolabelled product induced an increase in micronucleus frequency in A. cepa root cells. All pre-manufacture detergents and one commercial one, which gave negative results in the Ames and A. cepa tests, induced DNA damage in human leukocytes. A more accurate evaluation of the environmental impact of complex mixtures such as detergents requires a battery of tests to describe degradation, as well as toxicological and mutagenic features.

    The control of the fungal contamination on crops is considered a priority by the sanitary authorities of an increasing number of countries, and this is also due to the fact that the geographic areas interested in mycotoxin outbreaks are widening. Among the different pre- and post-harvest strategies that may be applied to prevent fungal and/or aflatoxin contamination, fungicides still play a prominent role; however, despite of countless efforts, to date the problem of food and feed contamination remains unsolved, since the essential factors that affect aflatoxins production are various and hardly to handle as a whole.     The assessment of the actual impact of discharged wastewater on the whole ecosystem and, in turn, on human health requires the execution of bioassays. In effect, based on the chemical characterization alone, the synergistic/antagonistic effect of mixtures of pollutants is hardly estimable. The aim of this work was to evaluate the applicability of a battery of bioassays and to suggest a smart procedure for results representation. Two real wastewater treatment plants were submitted to analytical campaigns. Several baseline toxicity assays were conducted, together with tests for the determination of endocrine activity, genetic toxicity and carcinogenicity of wastewater.     The aim of this paper was to investigate the relationship between micronuclei and DNA damage in children’s buccal mucosa cells and the genotoxicity and mutagenicity of the different sized fractions of particulate matter as well as the concentration of PAHs (polycyclic aromatic hydrocarbons) and metals in particulate matter. Air particulate matter was collected by high volume samplers located near the schools attended by the children on the same days of biological samplings. The mutagenic activity was assessed in different cells in in vitro tests (Ames test on bacteria and comet test on leukocytes). Our study showed weak positive correlations between (a) the mutagenicity of the PM0.5 fraction and PAHs and (b) the micronuclei test of children’s buccal cells and PAHs detected in PM0.5 and PM0.5–3 fractions. DNA damage was investigated with comet test in saliva leukocytes taken from sputum of 3- to 6-year-old children living in Brescia, Northern Italy, collected during two consecutive winter seasons (2012–2013). The results obtained using PAA were compared to those for the most commonly used disinfectant, sodium hypochlorite (NaClO). Ames test and three genotoxicity plant tests, Allium tests and Tradescantia/micronuclei (TRAD/MCN) test, were used to evaluate the mutagenic activity of disinfected water samples. Chemical analysis, using gas chromatography/mass spectrometry (GC/MS), was also performed to identify disinfection by-products (DBPs).     ABSTRACT A study was carried out on lake Como water, collected before and after potabilization treatment. The water samples were concentrated and the extracts were analyzed by gas‐chromatographic techniques and tested for potential mutagenic and toxic effects. Two different solid phase extraction methods (tC18 and Lichrolut EN) were compared for the evaluation of the more efficient recovery system of toxic compounds. The results showed the presence of toxic compounds and a mutagenic activity of the water samples lower than the one found in previous studies. The source of mutagenic compounds could be contamination of Cosia river influent.

    A chemical and bio-analytical protocol is proposed as a holistic monitoring framework for the assessment of WWTPs (Wastewater Treatment Plants) performance. This combination of tests consists of: i) an analysis of emerging contaminants, to be added to the established physico-chemical parameters in order to understand the causes of (new) pollution phenomena and ii) some of the bio-analytical tools most widely applied in the field of wastewater research, which provide information on groups of chemicals with a common mode of toxic action (baseline toxicity, estrogenicity and mutagenicity/genotoxicity, selected as the most representative for human health). The negative effects of the discharge can thus be highlighted directly and used to assess the global environmental impact of WWTPs. As a validation, this multi-tiered approach was applied to a full-scale WWTP (150,000 p.e.), where different measurements were carried out: EDCs (Endocrine Disrupting Compounds) detection; algal growth inhibition, bioluminescence inhibition and acute toxicity test (for baseline toxicity); an E-Screen-like assay (for estrogenic activity); Ames, Allium cepa and Comet tests (for mutagenic/genotoxic activity). As a result, the WWTP showed good performance for all these issues, displaying its ability to enhance effluent quality, except for residual mutagenic behaviour, probably due to the by-products generated by the tertiary ozonation.

    Genotoxic and ecotoxicologic effects of urban wastewater disinfected with sodium hypochlorite or peracetic acid were analyzed. The formation of genotoxic species was studied by determining clastogenic and mutagenic activity of aqueous samples and their extracts with in vivo and in vitro tests, respectively. In particular, we have applied citogenetic tests to Allium cepa roots and Tradescantia inflorescence (Allium cepa test and Tradescantia/micronuclei test) and reversion test to Salmonella typhimurium according to the microsuspension procedure (Kado test). The latter is the method of choice for the analysis of complex matrices due to its high sensitivity and specificity. The mutagenic activity of disinfected effluents was similar to the corresponding untreated wastewater both sampled in four different periods.     The biomonitoring of genotoxic effects in environmental complex mixtures using higher plants is very useful for hazard evaluation. In this study we evaluated the potential application of plant genotoxicity tests in monitoring mutagens in landfill environment. The clastogenic effects of gaseous emissions (biogas)from three municipal landfills were evaluated by in situ monitoring using the Tradescantia micronucleus assay. The cytotoxicity and genotoxicity of leachates were studied using the Allium cepa test. We found no significant differences in micronuclei frequency in pollen cells of Tradescantia. Leachate samples showed elevated toxicity that inhibited root tip development in Allium cepa. Genotoxicity of the leachates was evaluated in diluted samples only. We found a significant increase in chromosomal metaphase aberrations only in one of the samples analyzed.     The presence of chemical residues in vegetables and fruit is a source of human exposure to toxic and genotoxic chemicals. The mutagenic and carcinogenic action of herbicides, insecticides and fungicides on experimental animals is already known. Several studies have shown that chronic exposure to low levels of pesticides can cause adverse health effects and that many pesticides are mutagenic/carcinogenic. In the present research we monitored concurrently the presence of pesticides and genotoxic compounds extracted from 21 treated vegetables and 8 types of grapes sampled from the markets of a region in Southern Italy. The extracts were analysed for pesticides by gas-chromatography and HPLC, and for genotoxicity with two plant tests in Allium cepa roots: the micronucleus test and the chromosomal aberration test.
