Four new nitroasterric acid derivatives, pseudogymnoascins A-C (1-3) and 3-nitroasterric acid (4)... more Four new nitroasterric acid derivatives, pseudogymnoascins A-C (1-3) and 3-nitroasterric acid (4), along with the two known compounds questin and pyriculamide, were obtained from the cultures of a Pseudogymnoascus sp. fungus isolated from an Antarctic marine sponge belonging to the genus Hymeniacidon. The structures of the new compounds were determined by extensive NMR and MS analyses. These compounds are the first nitro derivatives of the known fungal metabolite asterric acid. Several asterric acid derivatives isolated from other fungal strains have shown antibacterial and antifungal activities. However, the new compounds described in this work were inactive against a panel of bacteria and fungi (MIC > 64 μg/mL).

Antisense-based screening strategies can be used to sensitize a microorganism and selectively det... more Antisense-based screening strategies can be used to sensitize a microorganism and selectively detect inhibitors against a particular cellular target of interest. A strain of Staphylococcus aureus that generates an antisense RNA against SecA,a central member of the protein secretion machinery, has been used to screen for novel antibacterials. Possible inhibitors of the SecA ATP-ase were selected with a high-throughput, two-plate agar-based whole cell differential sensitivity screen. After screening a library of over 115 000 natural products extracts with the SecA antisense strain, an extract of Geomyces pannorum was identified as providing increased activity against the sensitized strain as compared with the wild-type control. Bioassay-guided isolation of the active component from this fungal extract provided a new cis-decalin secondary metabolite, which we have named pannomycin.

Five streptothricin acids (compounds 1-5) were isolated by ion-exchange resin chromatography and ... more Five streptothricin acids (compounds 1-5) were isolated by ion-exchange resin chromatography and preparative RP-HPLC from the fermentation broth of Streptomyces qinlingensis. Their structures were elucidated mainly by analyses of the IR, HR-EIS-MS and NMR spectral data. They were deduced as two known compounds, streptothricin F acid (1) and streptothricin D acid (2), and three new compounds, 12-carbamoylstreptothricin E acid (3), 12-carbamoylstreptothricin D acid (4) and N-acetyl-streptothricin D acid (5), respectively. The antibacterial activities of 1-5 against Escherichia coli, Bacillus subtilis, Staphylococcus aureus, Bacillus cereus and Pseudomonas aeruginosa were assayed by micro-broth dilution. Comparison of the MICs with streptothricin F and D showed that the antimicrobial activities of 1-5 were decreased significantly but varied with the structures.
