    Members of the genus Flavobacterium occur in a variety of ecological niches and represent an interesting diversity of lifestyles. Flavobacterium branchiophilum is the main causative agent of bacterial gill disease, a severe condition affecting various cultured freshwater fish species worldwide, in particular salmonids in Canada and Japan. We report here the complete genome sequence of strain FL-15 isolated from a diseased sheatfish ( Silurus glanis ) in Hungary. The analysis of the F. branchiophilum genome revealed putative mechanisms of pathogenicity strikingly different from those of the other, closely related fish pathogen Flavobacterium psychrophilum , including the first cholera-like toxin in a non- Proteobacteria and a wealth of adhesins. The comparison with available genomes of other Flavobacterium species revealed a small genome size, large differences in chromosome organization, and fewer rRNA and tRNA genes, in line with its more fastidious growth.     In the opinion of many workers, one of the major difficulties in studying furunculosis of salmonids has been the experimental reproduction of the disease (Amend, 1969; Anderson, 1972). Notwithstanding some success obtained with parenteral injection of low doses of the responsible agent, Aeromonas salmonicida (Groberg et al., 1978), this problem has never been well solved. We recently described a standardized method using intramuscular injection of highly virulent bacteria to rainbow trout (Salmo gairdneri), which has given consistent results and has proven fairly reliable in testing the efficacy of therapeutic treatments (submitted for publication). The difficulties that we had to overcome during this work and the proposed solutions are summarized in Table 1. In the present work we have tried to give some additional features of this model, concerning the fate of the causative organism, both when it multiplies in the fish and when it is shed in water by infected fish. Especially in the latter case, serious hazards of superinfections could have interfered with standardization procedure.

    La lutte contre les maladies des poissons d’elevage repose sur des techniques de diagnostic precoce, des mesures zootechniques (hygiene assurant le « bien-etre » animal et evitant le stress) et genetiques (exploitant la variabilite de la sensibilite aux maladies), des mesures de police sanitaire (evitant les transferts de bioagresseurs contagieux) et, enfin, des mesures medicales (therapeutique et vaccination). Les recherches menees a l’Inra portent sur l’etude d’agents pathogenes bien etablis ou nouveaux qui sont des virus (septicemie hemorragique virale ou SHV), des bacteries (groupe des cytophagales, Aeromonas salmonicida), des parasites (agent de la prolifenttive kidney disease). L’effort principal porte sur la pathologie virale avec le virus de la SHV pour modele dans lequel virologie classique, biologie moleculaire et experimentation in vivo se combinent dans des travaux de pathogenie et des methodes d’intervention anti-virales fondees sur la vaccination, la genetique de la resistance du poisson et, en arriere-plan, des visees d’immunisation genetique de la cellule et de l’animal.

    In order to compare the respective roles of specific response and non-specific mechanisms in the enhancement of resistance to furunculosis, 2 groups of rainbow trout (Oncorhynchus mykiss) were immunized with supernatants of Aeromonas salmonicida or Escherichia coli cultures. The fish received 4 weekly intraperitoneal injections (70 micrograms of crude proteins). One control group was injected with phosphate-buffered saline, while another was untreated. Specific agglutinins were produced only in the fish immunized with A salmonicida supernatant. Polyspecific natural antibodies were measured using different antigens in an enzyme immunoassay. A significant rise (up to 3 times the mean titer of untreated controls) was observed in the 3 injected groups (even with PBS), which also displayed significant protection against experimental furunculosis (32 to 42% mortality versus 72% in controls).     Abstract Text: ABSTRACT: Flavobacterium psychrophilum is one of the most significant bacterial pathogen in salmonids worldwide. Selective breeding for resistance seems a promising method to control the disease. We searched for resistance QTL in a doubled haploid F2 progeny from two clonal lines chosen for opposite resistance. About 280 fish (6.3g mean body weight) were infected through intramuscular injection (4.2 105 cfu/fish) and mortality was recorded daily during one month. Using a genome-scan with 217 microsatellite and 101 SNP markers, QTL for survival were detected on several chromosomes, three of them having a strong effect on survival. To get further insight into the defense mechanisms involved, the location of a set of immune candidate genes was determined and the effect of the QTL on bacterial load in the spleen is being investigated.     Lactic acid bacteria have become a major source of concern for aquaculture in recent decades. In addition to true pathogenic species of worldwide significance, such as Streptococcus iniae and Lactococcus garvieae , several species have been reported to produce occasional fish mortalities in limited geographic areas, and many unidentifiable or ill-defined isolates are regularly isolated from fish or fish products. To clarify the nature and prevalence of different fish-associated bacteria belonging to the lactic acid bacterium group, a collection of 57 isolates of different origins was studied and compared with a set of 22 type strains, using amplified rRNA gene restriction analysis (ARDRA). Twelve distinct clusters were delineated on the basis of ARDRA profiles and were confirmed by sequencing of sodA and 16S rRNA genes.
